The widespread occurrence of 2ngametes (i.e. gametes with the somatic chromosome number) in the Medicago sativa-coerulea-falcata complex supports the concept that gene flow from diploid to tetraploid species occurs continuously in nature and plays a key role in alfalfa evolution. Breeders realized early that gene transfer between ploidy levels via 2n pollen and 2n eggs would have had potential use in cultivated alfalfa improvement. Cytological investigations provided insights into the types of meiotic abnormalities responsible for the production of 2n gametes. Alterations were defined as genetically equivalent to first (FDR) or second division restitution(SDR) mechanisms. For breeding purposes,data have proven that 2n gametes of the FDR type are more advantageous than those obtained by SDR for transferring parental heterozygosity and retaining epistatic interactions. The use of diploid meiotic mutants that produce 2ngametes is now recognized as one of the most effective methods available for exploiting heterosis and introgressing wild germplasm traits into cultivated tetraploid alfalfa via unilateral (USP) and bilateral sexual polyploidization (BSP) schemes. Both2n egg and 2n pollen producers could be used for direct gene transfer from wild diploid relatives into cultivated alfalfa by means of 2x-4x and4x-2x crosses. Although data have shown that forage yield improvement can be achieved when plants are sexually tetraploidized, problems related to reduced plant fertility and seed production remain largely unexplained. Apomixis has the potential of cloning plants through seed and thus provides a unique opportunity for developing superior tetraploid cultivars with permanently fixed heterosis and epistatic effects. A main goal in alfalfa breeding could be the introduction of functional apomixis (i.e. Apomeiosis and parthenogenesis) in cultivated alfalfa stocks. In the future, the efficiency of alfalfa breeding programs based on the use of reproductive mutants could be improved by direct selection at the genotype level using RFLPs and PCR-based markers. Suitable DNA markers and detailed linkage maps of alfalfa mutants should help to discover apomictic mutants and address basic genetic issues such as the extent of genomicre combination in polyploid hybrids and the effect of sexual polyploidization on heterosis. Molecular markers have recently been used in alfalfa for studying the inheritance of 2n gamete formation and identifying polymorphisms associated to genes involved in meiotic abnormalities. Molecular tagging of 2n egg and 2n pollen formation not only should explain the genetic control and regulation of these traits, but may also be an essential step towards marker-assisted selection of 2n gamete producers and implementation of USP and BSP breeding schemes. Future perspectives include strategies for the map-based cloning of genomic DNA markers,and screening of EST mini-libraries related to flowers at different developmental stages from meiotic mutants and wild-type scan lead to the identification of mRNAs and thus of candidate genes that control 2n gamete formation in alfalfa. With the aim of utilizing allotriploid (2n = 3x = 36) lily hybrids (Lilium) in introgression breeding, different types of crosses were made. First, using diploid Asiatic lilies (2n = 2x = 24), reciprocal crosses (3x − 2x and 2x − 3x) were made with allotriploid hybrids (AOA) obtained by backcrosses of F1 Oriental × Asiatic hybrids (OA) to Asiatic cultivars (A). Secondly, the AOA allotriploids were crossed with allotetraploid (OAOA, 2n = 4x = 48), in 3x − 4x combination. Finally, the AOA allotriploids where crossed to 2n gamete producer F1 OA hybrids (3x − 2x (2n)). Two types of triploids were used as parents in the different types of crosses, derived from: (a) mitotic polyploidization and (b) sexual polyploidization. Ploidy level of the progeny was determined by estimating the DNA values through flowcytometry as well as chromosome counting. The aneuploid progeny plants from 3x − 2x and reciprocal crosses had approximate diploid levels and in 3x − 4x crosses and 3x − 2x (2n) the progeny had approximate tetraploid levels. Balanced euploid gametes (x, 2x and 3x) were formed in the AOA genotypes. Recombinant chromosomes were found in the progenies of all crosses, except in the case of 2x − 3x crosses through genomic in situ hybridization (GISH) analyses. Recombinant chromosomes occurred in the F1 OA hybrid when the triploid AOA hybrid was derived through sexual polyploidization, but not through mitotic polyploidization with two exceptions. Those recombinant chromosomes were transmitted to the progenies in variable frequencies.  相似文献

Sexual polyploids were induced in diploid (2n = 2x = 24) interspecific F1 hybrids of Longiflorum × Asiatic (LA) and Oriental × Asiatic (OA) Lilium hybrids by backcrossing to Asiatic (AA) parents as well as by sib-mating of the F1 LA hybrids. A majority of the BC1 progenies were triploid and the progenies from sib-mating were tetraploid or near tetraploids. Genomic in situ hybridization (GISH) technique was applied to assess the intergenomic recombination in the BC1 populations of LA and OA hybrids obtained after unilateral sexual polyploidization. A total of 63 LA (LA × AA and AA × LA) and 53 OA hybrids were analysed. LA hybrids were originated through the functioning of 2n gametes either as 2n eggs or 2n pollen while those of OA hybrids originated through functional 2n pollen of diploid OA genotype. In both type of crosses, a majority of the progenies had originated through First Division Restitution (FDR) mechanism of functional 2n gamete either with or without a cross over. However, there were nine LA- and four OA-genotypes where Indeterminate Meiotic Restitution (IMR) was the mechanism of 2n gamete formation. Based on GISH, total amount of introgression of Longiflorum and Oriental genome into Asiatic genome was determined. Most of the BC progenies exhibited recombination and the amount of recombination was higher in LA hybrids as compared to OA hybrids. Intergenomic recombination was also determined cytologically in the 16 plants of sib-mated LA hybrids where both parents had contributed 2n gametes. Based on these results the nature of interspecific lily hybrids obtained from uni- and bilateral sexual polyploidization leading to allotriploid and allotetraploid formation is discussed in the context of introgression and intergenomic recombination.  相似文献

Biomass yield heterosis has been shown to exist between Medicago sativasubsp. sativa and Medica gosativa subsp. falcata. The objective of this study was to gain a better understanding of what morphological and genetic factors were most highly correlated with total biomass yield heterosis. We calculated genetic distances among nine sativa and five falcate genotypes based on amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) DNA markers. Genetic distance did not correlate with specific combining ability (SCA) or mid-parent heterosis. In contrast, a morphological distance matrix based on seventeen agronomic and forage quality traits was significantly correlated with heterosis; the agronomic traits of maturity, midseason regrowth, and autumn regrowth showed strong association with heterosis. Heterosis was also correlated with subspecies. We suggest that in many cases progeny heterosis can be accounted for by the interaction of genes controlling morphologically divergent traits between the parents. In other cases, progeny heterosis could also be due to divergence between the parents at particular genetic loci that do not control field-level phenotypic differences. Genetic distanceper se between parental genotypes, based on neutral molecular markers, however, does not reflect the potential of individual genotypes to produce heterosis in their progeny. Summary InDactylis glomerata L. subsp.lusitanica, triploid and tetraploid plants were obtained by bilateral sexual polyploidization in crosses between diploid parents known to produce 2n gametes. The polyploid and diploid progeny were compared for allozyme diversity (allele number and heterozygosity), phenological (pollen fertility, inflorescence emergence date), cellular (stomatic cell size) and morphological characters (vegetative biomass, seed weight, total seed number per plant, panicle number and seed number per panicle length) in four contrasting growing conditions. Stomatal cell size and the total number of alleles were significantly lower in diploids than in polyploids which sometimes recovered more than two distinct alleles at a locus via 2n gametes possessing heterozygous loci. Pollen fertility was similar in diploids and tetraploids, but significantly lower in the triploids. Seed weight was significantly higher in polyploids, although the average number of seeds per panicle length and the total number of seeds was higher in the diploid sibs. No phenological differences occurred among ploidy levels whereas such differences were observed inDactylis in all the natural areas of sympatry between diploids and their related polyploids. Our results provide evidence for an instantaneous effect of polyploidization on certain traits and suggest that natural selection would act subsequently on several other traits to allow the establishment of new polyploids.  相似文献

Intersubspecific hybrids between Medicago sativa subsp. sativa and subsp. falcata show biomass yield heterosis in space planted plots. Relative biomass yield of alfalfa is known to differ in space planted versus sward plots, although the magnitude of the difference is variable. The objective of this study was to determine if and how much of the biomass yield heterosis observed in space planted alfalfa hybrids was retained in sward planted plots. Two falcata genotypes (WISFAL-6 and PI502453-1) and three elite sativa genotypes were crossed in a diallel mating design. Progeny were transplanted into space-planted plots and harvested three times per year or drilled into sward plots harvested four times per year. Hybrids of WISFAL-6 with the sativa genotypes produced as much first harvest biomass yield as intra-sativa hybrids in space-planted plots and more in sward plots. Yield in subsequent harvests was lower than intra-sativa hybrids in sward plots only. Hybrids of PI502453-1 with sativa were generally lower yielding than intra-sativa crosses. A moderate correlation was observed between biomass yield in space planted and sward plots. Heterosis expression in swards was lower than that in spaced planted nurseries for progeny of both falcata genotypes. The increased plant-to-plant competition in swards is the likely cause of the loss of heterosis.  相似文献

Semi-hybrid alfalfa cultivars offer the possibility of capturing non-additive genetic variation. Medicago sativa subsp. falcata and subsp.sativa have been shown to form a heterotic pattern for biomass yield. Objectives of this study were to examine morphological variation in a broad range of falcate germplasm and to determine how falcate morphological variation per se is related to the performance of falcate germplasm in hybrid crosses with subsp. sativa. Falcata genotypes from 40 populations spanning the subspecies native range were selected and biomass yield, plant width, plant height, growth angle, biomass density, plant maturity, and regrowth after cutting were measured on the genotypes and their hybrid progeny three times throughout the growing season. In addition weekly plant heights were measured and growth rates were determined with a Gompertz function. Falcata parental genotypes exhibited a full range of phenotypes for plant width, plant height, growth angle, density, and maturity. Heterosis was not only observed for biomass yield but also for plant width, plant height, and more erect growth habit. The top yielding sativa-falcata hybrids had increased plant width, plant height, and plant density. European germplasm was taller and had faster regrowth than Asian material. Sativa-falcata hybrids produced biomass yield superior to the mid-subspecies mean only after two to three weeks of growth prior to first and third harvests. Prior to second harvest, biomass production was inferior to the mid-subspecies mean for 30 days. Hybrids using falcata as one parent are not currently adapted to intensive harvest management due to their slower regrowth. Summary Verticillium wilt is a serious disease of potato and is caused by the soil-borne fungi Verticillium dahliae and V. albo-atrum. No major cultivar is resistant to this disease. Two diploid interspecific potato clones, C287 and C545, were previously identified with consistently high levels of Verticillium wilt resistance and are thought to have the same genotype for the loci that confer resistance to V. dahliae stem colonization. The purpose of this study was to determine whether resistance to V. dahliae stem colonization could be transferred to the tetraploid level in potato via unilateral sexual polyploidization (USP). Progenies in eight families obtained by crossing C287 and C545 to two tetraploid breeding clones, S438 and S440, and the cultivar ‘Atlantic’ were planted in a V. dahliae-infested field and a field without a significant V. dahliae infestation. Resistance was evaluated relative to C545 and C287. There were differences among families for stem colonization and yield. No differences between the mean stem colonization of C545 and C287 progeny were detected. Family differences due to the tetraploid parents indicate that they contributed resistance to the progeny. Stem colonization data from this experiment were consistent with the proposed complementary two-gene model for Verticillium wilt resistance in the diploid parents. Unilateral sexual polyploidization is an effective method for transferring V. dahliae stem colonization resistance to the tetraploid level.  相似文献

Feral populations of cultivated crops may act as reservoirs for novel traits and aid in trait movement across the landscape. Knowledge on the genetic diversity of feral populations may provide new insights into their origin and evolution and may help in the design of efficient novel trait confinement protocols. In this study, the genetic diversity of 12 feral alfalfa (Medicago sativa) populations originating from southern Manitoba (Canada) and 10 alfalfa cultivars and a M. falcata germplasm were investigated using eight SSR markers (i.e., microsatellites) and 14 phenotypic traits. We found that the genetic diversity observed in feral populations was similar to the diversity detected among the 10 cultivars. Analysis of molecular variance revealed that there was great genetic variation within (99.8%) rather than between different feral populations. Cluster analysis (unweighted pair-group method using arithmetic average) revealed no differentiation between feral populations and cultivars for neutral loci. High levels of population differentiation for phenotypic traits (and not for neutral markers) suggest the occurrence of heterogeneous selection for adaptive traits. The phenotypic traits we studied did not distinctly separate feral populations from cultivars but there was evidence of natural selection in feral populations for traits including winter survivability, rhizome production, and prostrate growth habit. Our results suggest that feral alfalfa populations need to be considered in the risk assessment of alfalfa containing novel genetically modified (GM) traits. Further, feral alfalfa populations may be regarded as a source of new germplasm for plant improvement.  相似文献

A triploid hybrid, which was obtained from interspecific crosses between tetraploid Primula denticulata (2n = 4x = 44) and P. rosea (2n = 2x = 22), successfully produced 11 plants by backcrossing with pollen of tetraploid P. denticulata. Analysis of ploidy level using flow cytometry and chromosome counting in the 11 BC1 plants revealed that all progeny had much larger DNA contents and chromosome number than both parents. In this triploid-tetraploid (3x–4x) crossing, progeny was predominantly true or near pentaploid presumably produced by the fertilization between true or near triploid female gamete produced from triploid hybrid and diploid pollen of tetraploid P. denticulata. These results suggest that unreduced (3x) or near triploid female gametes were partially produced by single step meiosis, either first-division restitution or second-division restitution process. The zygotes formed by the fertilization between true or near triploid egg produced by single step meiosis in triploid hybrid and diploid pollen produced by normal meiosis of tetraploid P. denticulata might be the only survivors in embryogenesis.  相似文献

To transfer new traits into the gene pool of garden dahlias (Dahlia variabilis), crosses between garden dahlias (2n = 64) and the epiphytic species Dahlia macdougallii (2n = 32) from the section Epiphytum were conducted. Six hybrid plants were obtained. The hybrid status was verified using three SSR markers. In addition, flow cytometry was performed to determine the genome size of the hybrids and to show that the hybrids were hexaploid as expected from crosses between tetraploids and octoploids. The open pollinated progeny of the hybrids produced four progeny with octoploid genomes. The hybrids exhibited indeterminate vegetative growth and the formation of flowers from axillary buds, similar to the father D. macdougallii. This result is of interest for breeding new varieties of dahlia with traits that are not present in the current gene pool.  相似文献

Fourteen interspecific hybrids in sexual diploid Allium senescens var. minor× apomictic tetraploid Allium nutans L. crosses, and eight interspecific hybrids in sexual diploid A. senescens var. minor× apomictic hexaploid A. senescens L. crosses were produced. The number of chromosomes was 2n= 24 in interspecific hybrids of diploid × tetraploid, and 2n= 32 in diploid × hexaploid crosses. Triploid and tetraploid interspecific hybrids showed intermediate parental morphological characteristics. Tetraploid interspecific hybrids of A. senescens var. minor×A. senescens crosses formed two groups based on leaf colour and leaf width. Seeds were formed in 11 out of 14 triploid interspecific hybrids under natural conditions. In cytological observations of parthenogenesis, three out of 12 triploid interspecific hybrids and five out of eight interspecific tetraploid hybrids were observed. Parthenogenesis ranged from 26.0% to 86.0% in five tetraploid interspecific hybrids. Non-parthenogenesis to parthenogenesis segregated in a 3:5 ratio in A. senescens var. minor×A. senescens crosses.  相似文献

The potential for gene flow between Camelina sativa, a promising edible and industrial oil crop in Canada, and its wild North American relatives C. alyssum, C. microcarpa and C. rumelica subsp. rumelica, was investigated. Sequence analysis of the nuclear ribosomal internal transcribed spacer region (ITS) was used to differentiate Camelina species and/or accession‐specific nucleotide markers and identify interspecific F1 hybrids. ITS analysis identified hybrids in progeny of C. alyssum × C. sativa, C. microcarpa accession 36010 × C. sativa and C. sativa × C. rumelica subsp. rumelica. Seed set on C. alyssum × C. sativa F1 and F2 progeny was similar to the parents; few seeds were produced on hybrid progeny of C. microcarpa accession 36010 × C. sativa and C. sativa × C. rumelica subsp. rumelica. The study provided evidence that should the species have sympatric distributions and overlapping flowering periods, gene flow between C. sativa and its wild North American relatives is possible and that it would most likely occur with C. alyssum given the high fertility of the F1 hybrids recovered.  相似文献

Summary Sexual polyploidization via the action of 2n gametes (gametes with the sporophytic chromosome number) has been identified as the most important evolutionary mode of polyploidization among plant genera. This study was conducted to determine whether 2n gametes are present in the tetraploid level of the genus Avena (2n=4×=28) Twenty tetraploid Avena lines, representing four species and one interspecific hybrid, were screened for pollen grain size in order to differentiate between n and 2n pollen. Avena vaviloviana (Malz.) Mordv. line PI 412767 was observed to contain large pollen grains at a 1.0% frequency. Cytogenetic analyses of pollen mother cells of PI 412767 revealed cells with double the normal chromosome number (i.e., 56 chromosomes at metaphase I and anaphase I). The mode of chromosome doubling was found to be failure of cell wall formation during the last mitotic division that preceded meiosis. The resulting binucleate cells underwent normal meiotic divisions and formed pollen grains with 28 chromosomes. Based on the formation and function of 2n gametes, three models involving diploid and tetraploid oat lines are proposed to describe possible evolutionary pathways for hexaploid oats. If stable synthetic hexaploid oat lines could be developed by utilizing 2n gametes from diploid and tetraploid oat species through bilateral sexual polyploidization, the resulting hexaploids could be used in breeding programs for transferring genes from diploids and tetraploids to cultivated hexaploids.  相似文献

Semi‐hybrids between genetically distant alfalfa (Medicago sativa subsp. sativa) populations may display heterosis whose extent is affected by the structure of genetic diversity across populations. This study aimed to assess the genetic diversity across three putative heterotic populations, one Italian, one Egyptian and one of semi‐erect germplasm from Eastern Europe, Canada and Spanish Mielga (EECM population). Each population was bred from ten parents after various selection cycles. Fifteen genotypes per population were characterized by 20 polymorphic SSR markers. The among‐population variance was over eightfold smaller than the average within‐population variance (2.05 vs. 17.24) and accounted for 10.6% of the total variation. GST = .090 across markers indicated modest population differentiation. Various diversity measures, multidimensional scaling, and cluster analysis of the genetic structure indicated that the Italian population was more distant from the EECM population than the Egyptian one. The EECM and Egyptian populations were the most distant geographically and genetically. EECM displayed widest intrapopulation variation, accordingly to its constitutive geographical diversity. In conclusion, this study indicates modest genetic differentiation between alfalfa populations even for geographically distant germplasm.  相似文献

The development of transgenic oilseed Camelina sativa (2n = 40) and the potential for hybridization with its weedy relative Capsella bursa‐pastoris (2n = 36) necessitates a careful evaluation of the reproductive compatibility between the species. Here, we conducted over 1800 crosses (emasculation and manual pollination) to examine the ability of 10 Canadian C. bursa‐pastoris (♀) accessions to hybridize with five accessions of C. sativa (♂). Seven hybrids were confirmed among 586 putative hybrids screened with species‐specific markers, indicating a hybridization rate of 1.5 hybrids per 10 000 ovules pollinated. All seven hybrids had intermediate DNA content compared to their parents, were morphologically distinct, had low (1.9%) pollen fertility and failed to produce selfed or backcrossed seed. Given the abundance of C. bursa‐pastoris along field margins, hybrids will likely be generated in the wild, but they will be unable to establish lineages unless fertility is restored. The large number of crosses and the diversity captured by the use of multiple accessions resulted in strong statistical power and a high degree of confidence in the estimated hybridization rate.  相似文献

The objective of the present study was to evaluate the fertility of tetraploid plants obtained from diploid mutants that produced 2n gametes via bilateral (BSP) and unilateral sexual polyploidization (USP) schemes. Controlled matings between selected plants from BSP and USP were carried out according to a complete diallel. The level of male and female fertility of each plant was estimated within full-sib, half-sib and non-inbred crosses. Crosses with unrelated self-fertile and male-sterile testers were also performed. Pollen size and stainability were determined by acetocarmine staining. The number of ovules per ovary was counted and the callose deposition within ovules examined to evaluate embryo sac fecundity. Cross-fertility was generally much higher for USP than BSP plants (on average, 0.3 vs. 0.03 seeds per flower pollinated). Both male and female fertility were inversely related to the inbreeding level of cross combinations. The in vitro pollen germinability was positively correlated to male fertility of BSP plants. The level of in vivo embryo sac sterility was not correlated to female fertility in either BSP or USP plants. Female fertility was restored in both BSP and USP groups when plants were crossed with unrelated tetraploid testers. Male fertility also increased in USP plants but remained rather low in the BSP plants. Scaling up the ploidy level by means of 2n gamete union resulted in tetraploid plants with low fertility. In particular, the BSP process yielded virtually self-sterile and highly cross-sterile plants. The well documented positive effect of sexual tetraploidization on forage yield was accompanied by worsening of fertility traits. Summary A collection of 90 accessions of different germplasm groups from the Medicago sativa complex was evaluated at Lodi, Italy, for dry-matter yield, time to anthesis, height of regrowth, plant diameter, stem density, vegetative recovery after winter, and presence of deep-set crown and ability to spread with creeping roots or rhizomes, these latter characters possibly contributing to grazing tolerance. The germplasm groups proved rather distinct for these traits, but all included types potentially suitable for grazing. Interesting types combining deep-set crown and high yield occurred mainly in subsp. x varia and materials originated from artificial crosses involving at various extent the subspecies falcata, sativa and x varia. Though belonging to subsp. sativa, mielga wild populations from Spain had aerial traits similar to those of the long-dormant, low-yielding subsp. falcata. Deep-crowned plants had greater plant diameter than shallow-crowned plants, and could be categorised into four distinct top-growth models according to their habit and vigour. One of these models combined spreading ability with high yield. The about 150 plants selected for their combination of deep crown, wide diameter, and good yield were characterised by a branch-rooted system. There was general correspondence between top-growth and root morphology of each model. Of the four deep-crowned models, three had rhizomes, with wide range of underground development and morphology, while the fourth had creeping roots. The latter model was the most rare among the examined accessions, never appearing in either cultivated or wild subsp. sativa. The implications of the results on the development of grazing-tolerant lucerne varieties are discussed.  相似文献

Crosses were carried out between tetraploid Dianthus caryophyllus cv ‘Butterfly’ (2n = 4×= 60) and seven diploid cultivars (2n = 2×= 30). Fewer seeds were obtained and the low seed germination was found which suggested the post-fertilization barrier in 4×–2× crosses. 12 progeny were obtained from 5 crossing combinations. Chromosome analysis revealed that they consisted of 5 triploid hybrid plants and 7 tetraploid hybrid plants, suggesting that unreduced male gamete maybe be involved in polyploid formation. Various flower shapes and colours were observed in the polyploid progenies, showing that sexual polyploidization results in greater variability and fitness. The hybrids obtained by 4×–2× crosses showed the flower-size intermediate between the parents or larger than the parents. Some favourable characters of parents such as flower shape, flower colour and resistance to Fusarium oxysporum, were successfully transmitted to the hybrids. Since polyploid hybrids have some of the profitable characters of the parents, they are expected to be used for future breeding in carnation.  相似文献

Summary Bilateral sexual tetraploidization was achieved by intercrossing diploid Phureja × haploid Tuberosum hybrids. Both parents contribute gametes with the somatic number of chromosomes (diplogynoids and diplandroids). More than 100 tetraploid progeny appeared in nine 2x–2x families. The tetraploids are generally more vigorous and significantly outyield their diploid full-sibs. The superiority of the tetraploids is interpreted on the basis of the mode of diplandroid (2n pollen) formation by first meiotic division restitution (FDR). Heterozygosis, epistasis and genetic diversity, if present in the parental diploid genotype, are largely maintained in the 2n gametes formed by FDR. and syngamy brings about more opportunities for heterotic responses in conjunction with polyploidization. Differences in tuber yield among tetraploid clones within families were found to be statistically significant (P <0.05). Thus, unlike somatic chromosome doubling, which can merely increase chromosome numbers, or somatic hybridization, which could in addition bring about heterosis, sexual polyploidization is also associated with genetic variability. The degree of inbreeding and the genotypic structure in the population of newly arisen tetraploids is discussed. The relationship among asexual reproduction, preservation of favorable nonadditive genetic effects by FDR. restoration of sexuality, and polyploidization is explored. The possible evolutionary significance of these factors is emphasized.  相似文献
