Path analysis was used to determine the interrelationships between postpartum administration of gonadotrophin releasing hormone and cloprostenol and the occurrence of reproductive disease and reproductive performance in dairy cows. The data analysed were those collected on 226 Holstein-Friesian cows calving in a commercial dairy herd during a 17 month period (May 1, 1981 to October 1, 1982). Cows administered gonadotrophin releasing hormone at day 15 postpartum experienced an improved rate of uterine involution as determined by rectal palpation nine days later. Although this improved rate of uterine involution reduced the risk of pyometritis, it actually directly delayed conception. Also, gonadotrophin releasing hormone therapy directly resulted in an increased incidence of pyometritis which in turn resulted in an increase incidence of cystic ovarian disease and anestrus. The occurrence of these abnormalities resulted in increased intervals from calving to first observed estrus, first service and conception. In addition to this effect, the administration of gonadotrophin releasing hormone was also associated with increased plasma progesterone concentrations at days 24 and 28 postpartum which delayed conception. Cloprostenol therapy at day 24 postpartum resulted in a decreased plasma progesterone concentration at day 28 postpartum which was directly and indirectly associated with a decrease in the calving to conception interval. The indirect effects were mediated by a reduction in days to first estrus. Cloprostenol therapy also directly resulted in a decreased calving to first observed estrus interval for reasons not attributable to the level of progesterone at day 28.  相似文献

Path analysis was used to determine the interrelationships between ambient temperature, age at calving, postpartum reproductive events and reproductive performance in dairy cows. The data used in the analysis were collected on 226 Holstein-Friesian cows calving in a commercial dairy herd during a 17 month period (May 1, 1981 to October 1, 1982). The data were obtained from a double blind study evaluating the effects of gonadotrophin releasing hormone and cloprostenol in postpartum cows. Rectal palpation to assess uterine involution and ovarian activity was performed on each cow on days 15, 24 and 28 postpartum. At the same time, blood samples were collected for subsequent progesterone assay. Data were recorded on the occurrence of reproductive diseases and events from the time of parturition until the diagnosis of pregnancy or until the cow left the herd in the case of culled cows. There was an increase in the incidence of retained placenta, in the percentage of cows with abnormal vaginal discharge in the early postpartum period as well as a delay in uterine involution during the winter months. In addition, cows calving during the winter had prolonged intervals to first estrus, first service and conception compared to cows calving during the summer. Thirty-six postpartum Holstein cows consisting of eighteen cows that shed the placenta soon after calving (NRP) and eighteen cows that retained placenta greater than 24 h (RP) were used. There were four treatment groups. Group I consisted of 9 NRP cows which received intramuscular injection of sterile saline on day 15 postpartum. The second group consisted of 9 NRPN cows which received 100 micrograms of gonadotrophin releasing hormone on day 15 postpartum (NRPT). The third group consisted of 9 RP cows which received saline on day 15 postpartum (RPN) and the fourth group consisted of 9 RP cows which received 100 micrograms of gonadotrophin releasing hormone on day 15 postpartum (RPT). Blood samples were collected once daily during the first month and once every other day during the second month postpartum. In addition fourteen cows (RPT, n = 7; NRPT, n = 7), were used to study short-term changes in serum luteinizing hormone concentrations following gonadotrophin releasing hormone treatment on day 15 postpartum. Blood samples were obtained from these cows every 15 min during 1 h before and 6 h after gonadotrophin releasing hormone administration. Sera from all samples were assayed for progesterone and luteinizing hormone concentrations. Starting from four days after calving rectal palpations and ultrasound examinations of the ovaries were carried out once every four days until day 28 postpartum in order to monitor ovarian changes. The effects of gonadotrophin releasing hormone (GnRH) on the reproductive performance of dairy cows with retained placenta were studied. Three hundred and seventy-eight cows diagnosed as having retained placenta received intramuscular injections of either 2 mL sterile water or 200 micrograms of GnRH in 2 mL sterile water between day 8 and day 14 postpartum. Rectal palpation was performed at the time of treatment and ten to 20 days after treatment in order to determine the rate of uterine involution. Thereafter, monthly rectal examinations were carried out until insemination. Pregnancy diagnosis was made by rectal palpation at 40 days or more after breeding. Using the entire experimental population, there were no significant differences between GnRH-treated and control cows for the rate of uterine involution, the occurrence of reproductive problems, the interval from parturition to first observed estrus, the interval from parturition to first insemination, the interval from parturition to conception, the number of services per conception, the total number of services per cow regardless of conception and the incidence of culling for infertility. When the data for herds in which breeding began earlier in the postpartum period (herds having a mean less than or equal to 80 days from parturition to first service for retained placenta cows) were considered, the GnRH treatment resulted in a significantly shorter (p less than or equal to 0.01) calving to conception interval as compared to control cows. 本试验选用276头高产中国荷斯坦牛为研究对象,其中120头初产奶牛和156头经产奶牛均表现为分娩期异常或产后期异常,如难产、胎衣不下、双胎难产、早产或子宫感染。试验牛被分为处理组与对照组。处理组在产后20d间隔811连续两次皮下注射氯前列烯醇(0.4mg/次),对照组注射4.0mL灭菌生理盐水。然后观察其首次发情的时间、适合第一次人工授精的时间、首次授精妊娠率、150d妊娠率、产后的空怀天数、重复配种奶牛的比例和每头牛的授精次数。结果表明,产后应用前列腺素对产后首次发情的时间和产后首次人工授精的时间没有影响,但处理组奶牛首次人工授精的妊娠率明显升高,这在头胎牛更为明显;产后应用氯前列烯醇可明显减少平均授精次数、缩短产后空怀时间、增加产后150d泌乳奶牛的妊娠率、降低产犊后重复配种率。  相似文献

The objective of this study was to determine whether an antiestrogen (enclomiphene) would shorten the interval to first estrus and conception in postpartum beef cows. Sixty postpartum Angus beef cows were stratified by age, body condition, and calving date and were randomly assigned to one of two treatment groups. Group 1 cows (n = 24) received three silastic implants, each containing 150 mg of enclomiphene, on d 20 postpartum. Implants were removed on d 30 postpartum. Group 2 cows (n = 28), received empty implants and served as controls. Cows were artificially inseminated at first detected estrus. Estrus detection and ovulation were further verified by increased serum progesterone. Concentrations and pulse frequencies of LH were determined from blood samples collected at 15-min intervals for 6 h on d 20, 25, 30, and 40 postpartum. Hypothalami and pituitaries were collected from four cows in each treatment group on d 30 postpartum and analyzed for concentrations of estradiol receptors. Concentrations of total and unoccupied hypothalamic and pituitary estradiol receptors were reduced by enclomiphene. Neither concentrations nor pulse frequencies of LH differed significantly between treatment groups on any of the 4 d. Days to first estrus did not differ (P greater than .05) between enclomiphene-treated (57 +/- 6; n = 24) and control (56 +/- 4; n = 28) cows. Days to conception did not differ between treated (81 +/- 9) and control (79 +/- 8) cows. The dose of enclomiphene used in this study reduced hypothalamic and pituitary estrogen receptors but did not alter secretion of LH or days to first estrus in the postpartum beef cow.  相似文献

Recently, it has been demonstrated that administration of equine chorionic gonadotropin (eCG) in the postpartum period in dairy cows can enhance follicle growth, reduce the interval from calving to first ovulation and increase plasma estradiol concentrations, and, thus, could enhance reproductive performance in a dairy herd when administered on day 6 postpartum. The aim of this study was to investigate the effects of a single dose of eCG between days 9 and 15 postpartum on parameters of reproductive performance in dairy cows. German Holstein cows (n = 1937; primiparous cows: n = 748; pluriparous cows: n = 1189) in a commercial dairy farm were randomly assigned to three experimental groups. Animals within the group eCG received a single dose of 600 IU eCG intramuscularly (i.m.) between days 9 and 15 postpartum followed by an i.m. administration of 500 μg cloprostenol after 14 days. Those of treatment group PG received cloprostenol only between days 23 and 29 postpartum. Cows of the control group remained untreated. Starting on day 49 postpartum, cows were subjected to a Presynch‐Ovsynch protocol and inseminated artificially. The impact of application time (days postpartum) of eCG on the intervals calving to first service and calving to conception was statistically not significant. Outcomes of reproductive performance (i.e. first service conception rate, proportion of pregnant cows until 100 and 150 days in milk [DIM], number of inseminations until 150 DIM, calving to first service interval and calving to conception interval) did not differ significantly between treatment group eCG and group PG compared to control group. Regarding postpartum eCG administration, significant interactions between treatment and parity, season, milk yield, and early puerperal disorders, respectively, could not be shown. In conclusion, an eCG treatment of dairy cows between days 9 and 15 postpartum to increase reproductive performance cannot be recommended under the given circumstances.  相似文献

The efficacy of gonadotropin releasing hormone (GnRH) on the fertility of dairy heifers and postpartum cows was studied. In one study, 300 cows were assigned randomly to 1 treatment from a 2 X 2 design, and 2 ml of 0.15M saline solution or 100 micrograms of GnRH was given IM on postpartum day (PPD) 14 and at the time of 1st postpartum breeding. In a 2nd study, repeat breeder cows (n = 346) were given saline solution or GnRH at the time of the 3rd breeding. In a 3rd study, heifers (n = 185) were given saline solution or GnRH at 1st breeding. Animals were observed for repeat estrus, and pregnancy diagnoses were made by rectal palpation 45 to 60 days after breeding. Conception rates were 15% to 18% higher for cows given GnRH, whether at PPD 14 or at the first postpartum breeding. When GnRH was given at the first breeding, conception rates were significantly higher (P less than 0.05). Conception rates for repeat breeder cows given GnRH were 25% higher than those in controls (P less than 0.005). Conception rates for heifers did not differ between treatments.  相似文献

During the fourth week postpartum, 443 healthy Holstein cows milked thrice daily were randomly divided among four groups to receive 0, 2, 8 and 32 micrograms of the GnRH analog HOE 766. Intervals from calving to first estrus and to first breeding, from breeding to conception and conception rates at first breeding were calculated to measure treatment response, and progesterone was measured in the fat-free portion of milk samples collected twice weekly during the first 4 wk following treatment. Uterine involution at the time of treatment was estimated by palpation per rectum. Twenty percent of the cows examined were classified as having delayed uterine involution (abnormal). By analyzing milk progesterone patterns it was determined that 38% of the animals were in the luteal phase of an estrous cycle when treated. Cows without luteal tissue (less than 1 ng of progesterone/ml milk) given 8 or 32 micrograms of HOE 766 increased in progesterone to greater than or equal to 1 ng/ml within 7 d in 77 and 72% of the cows compared with 40 and 57% for cows receiving 0 and 2 micrograms (P less than .05). This increase in progesterone was followed by a normal estrous cycle within 4 wk in a higher proportion of cows treated with the two higher doses of GnRH analog (87 and 86%) compared with 67 and 70% of those receiving 0 or 2 micrograms of the analog (P less than .005). There were no treatment differences (P greater than .05) for other traits analyzed, but cows with a normal progesterone cycle were observed in estrus and were bred sooner (P less than .01) than those with irregular progesterone patterns. It was concluded that the GnRH analog hastened the onset of normal ovarian cycles in cows milked thrice daily.  相似文献

In a randomized double-blind clinical trial, 75 cows with ovarian cysts were treated with the synthetic gonadotropin releasing hormone, gonadorelin acetate (GnRH). Forty-two of these cows were simultaneously treated with cloprostenol (CP), and the remaining 33 cows received sterile saline. Milk progesterone (P4) was measured at treatment and two days later. Clinical response 30 days after treatment was determined by palpation per rectum, and estrus and breeding dates were recorded up to 90 days after treatment. Cows were examined for pregnancy by palpation per rectum 40 days or more after breeding. Milk progesterone levels two days after treatment were significantly lower and the 30-day clinical response rate was significantly higher in the GnRH + CP group than in the GnRH group. Intervals to first estrus and to conception, proportion in heat by day 21 after treatment, and pregnancy rate by 90 days did not differ significantly between the groups. The same relationships held in a subset of cows with P4≥1 ng/mL at treatment. Fewer cows in the GnRH + CP group became pregnant by day 90 after treatment, but this difference was not significant. These results suggest that simultaneous GnRH and cloprostenol treatment of all cows with cystic ovaries cannot be recommended at this time.  相似文献

The objective of this experiment was to determine the effect of sequential treatment with buserelin (a GnRH agonist) and cloprostenol (a prostaglandin F2 alpha analog) on estrous response and fertility in beef cattle with different ovarian conditions. On d 0 (1st d of treatment), the control group (n = 52, 10 heifers and 42 cows) and the GnRH group (n = 48, 10 heifers and 38 cows) received 2 mL of saline or 2 mL of Receptal (8 micrograms of buserelin), respectively. On d 6, all cows that had not exhibited spontaneous estrus were given i.m. 500 micrograms of cloprostenol (PGF). Ultrasonography on d 0 and assays of progesterone in blood on d -11, 0, and 6 were used to identify follicular and luteal status of animals. Cattle were observed for estrus from d 0 to 10. Cows showing estrus were bred artificially 12 h after onset of estrus. Over the 10-d period, the number of cows detected in estrus and pregnancy and conception rates were identical for the two groups. However, between d 0 and 6, the proportion of cows exhibiting estrus was lower (P less than .01) in the GnRH group than in the control group. Between d 6 and 10, the synchronization rate and precision of estrus were greater (P less than .01) in the buserelin-treated group than in the control group. Conception rate and interval from PGF injection to onset of estrus were not different between the two treatment groups. This study evaluated the effect of microencapsulated LHRH agonist (D-Trp6-LHRH) on gonadotropin release and occurrence of estrus in early postpartum beef cows. Angus cows (n = 54) were assigned randomly to two treatment groups at d 5 postpartum. Group 1 received a single i.m. injection of D-Trp6-LHRH (LHRH-A) encapsulated in poly-DL-lactide-coglycolide, calculated to release 15 micrograms of LHRH-A per day for 30 d (n = 23). Group 2 received vehicle only (control, n = 31). Blood samples (15-min intervals for 6 h) were obtained on d 5, 10, 20, 30, and 40 postpartum for evaluation of LH and FSH concentrations (n = 12 per group). Days to first postpartum estrus were reduced by treatment with LHRH-A (Group 1, 43.7 +/- 4.2 d vs Group 2, 55.9 +/- 4.7 d; P < .05). However, days to conception were similar between groups (68.9 +/- 7.9 vs 76.7 +/- 6.7 d, respectively). On the day of treatment, cows treated with LHRH-A had higher mean concentrations of LH and FSH than did controls (8.3 +/- 1.4 vs 2.0 +/- .4 ng/mL for LH and 211.0 +/- 8.6 vs 51.2 +/- 2.7 ng/mL for FSH (P < .05). There were no differences in mean concentrations of LH or FSH between treatment groups on d 10, 20, 30, and 40 postpartum. Cows given LHRH-A had more (P < .05) LH pulses on d 10 and 30 postpartum than did controls. This study demonstrated that microencapsulated D-Trp6-LHRH reduced the postpartum anestrous interval in suckled beef cows.  相似文献

Mature beef cows were slaughtered at 5 (n = 6), 10 (n = 6), 20 (n = 6) or 30 (n = 5) d after calving to identify endocrine events that may affect the duration of postpartum anestrus. Additional cows (n = 6) were slaughtered 12 to 14 d after their first postpartum estrus (luteal phase cows). Anterior pituitary concentrations of luteinizing hormone (LH) were low at d 5 (383 +/- 69 micrograms/g), averaged 445 +/- 103 and 682 +/- 207 micrograms/g at d 10 and 20, respectively, and were elevated (P less than .05) by d 30 (1,097 +/- 174 micrograms) to a concentration similar to luteal phase cows (1,208 +/- 148 micrograms/g). Concentrations of follicle-stimulating hormone (FSH) averaged 12.4 +/- 1.1, 9.6 +/- 2, 8.6 +/- 1.8 and 7.4 +/- 3.3 mg/g at d 5, 10, 20 and 30, respectively. Affinity (1.6 +/- .2 X 10(9) M-1) of anterior pituitary receptors for the GnRH (gonadotropin-releasing hormone) analog (DAla6; des-Gly10, [D-Ala6]-LH-RH ethylamide) and weights (2.1 +/- .1 g) of the anterior pituitaries did not differ among groups (P greater than .05). Number of receptors for GnRH averaged 37 +/- 7, 39 +/- 9, 25 +/- 5 and 23 +/- 5 X 10(-14) M/mg protein at d 5, 10, 20 and 30, respectively. Postpartum infertility is caused by four factors: general infertility, lack of uterine involution, short estrous cycles and anestrus. The general infertility component is common to any estrous cycle and reduces potential fertility by 20 to 30%. Incomplete uterine involution prevents fertilization during the first 20 d after calving but is not related to anestrus. Short estrous cycles prevent fertility during the first 40 d after calving by causing the cow to return to estrus before pregnancy recognition occurs. Anestrus is the major component of postpartum infertility and is affected by several minor factors: season, breed, parity, dystocia, presence of a bull, uterine palpation and carryover effects from the previous pregnancy as well as two major factors: suckling and nutrition. These major factors have direct effects on anestrus but also interact with one or more other factors to control postpartum anestrus. Physiological mechanisms associated with anestrus involve blockage of the GnRH "pulse generator" in the hypothalamus, but other pathways also must be involved because bypassing the pulse generator is not an effective treatment for all cows. The primary cause of anestrus probably is different for different stages of anestrus. The mediating mechanisms for anestrus are not involved with prolactin, oxytocin, the adrenal or direct neural input from the mammary gland but are at least partially involved with blood glucose and the endogenous opioid peptide system. Management options to decrease the impact of anestrus and infertility include: 1) restrict breeding season to less than or equal to 45 d; 2) manage nutrition so body condition score is 5 to 7 before calving; 3) minimize effects of dystocia and stimulate estrous activity with a sterile bull and estrous synchronization; and 4) judicious use of complete, partial or short-term weaning.  相似文献

AIMS: To determine the effects of oral Ca bolus administration in the early postpartum period of cows on milk yield and composition, blood metabolites, early-lactation health status, and reproductive performance.

METHODS: Multiparous Holstein dry cows (n=66) with a mean parity of 3.1 (SD 0.35) were fed a diet with a positive dietary cation–anion difference (DCAD) prior to calving. They were randomly assigned to receive no treatment (Control; n=33) or two oral Ca boluses (n=33, 45?g of Ca per bolus); one was administered immediately after calving (Day 0) and the second 24 hours (±30 minutes) later. Blood samples were collected at calving, and on Days 2 and 7 to determine concentrations in serum of Ca, P, Mg, glucose, non-esterified fatty acids (NEFA), and β-hydroxybutyric acid (BHBA). Milk yield was recorded daily and milk composition was determined weekly from calving until 28 day postpartum. Health and outcomes were determined during the first 30 days postpartum and reproductive outcomes to 180 days postpartum.

RESULTS: Mean milk yields and composition over the first month of lactation were similar between cows in the two treatment groups (p>0.1). Mean concentrations of Ca in serum were not different between treatment groups on Day 0, but were higher on Day 2 for cows that received oral Ca boluses (1.77 (SE 0.07)) compared with Control cows (1.54 (SE 0.08)) (p=0.04). Concentrations in serum of P, Mg, glucose, NEFA and ΒHBA did not differ between treatment groups on any day of measurement. Fewer cows that received oral Ca were diagnosed with hypocalcaemia (total concentrations of Ca in serum <1.5?mmol/L) by Day 2 (2/33; 6%) compared with Control cows (12/33; 36%) (p=0.01). There was no difference in the prevalence of other health outcomes between treatment groups. The proportion of cows conceiving to first insemination was greater in cows that received an oral Ca bolus (19/29; 65%) than Control cows (12/29; 41%) (p=0.01).

CONCLUSIONS: Oral Ca bolus administration increased concentrations of Ca in serum on Day 2 postpartum, and increased first service conception rates, in cows fed a diet with a positive DCAD prior to calving compared to cows that received no oral Ca bolus supplementation. Because of the small number of cows used in this study, further studies in large-scale dairy farms should be carried out to confirm these findings.  相似文献

Eighty multiparous Holstein cows were assigned randomly at calving to receive either 100 microg of GnRH or saline 13 or 14 d postpartum (PP). From 4 to 28 d PP the cows' reproductive organs were palpated weekly per rectum, and cows were subclassified within each group as undergoing slow (delayed) cervical and uterine involution (abnormal) or as normal cows. Last milk obtained after removing the milking machine was assayed for progesterone 3 times a week for 120 d PP. Fourteen of the 80 cows were removed from the experiment because of culling or various veterinary treatments of pathologic conditions that could confound analysis of the GnRH treatment effects. As expected, the treatment of normal cows with GnRH had no significant effects on the first estrus or the first estrous cycle PP, on services per conception, days open, or any other reproductive trait measured. However, in the abnormal group of cows receiving saline, first rebreeding after calving was delayed (81 vs. 67 d), fewer were pregnant by 105 d PP (23 vs. 64%), and number of days open was greater (121 vs. 87 d) compared with those receiving GnRH; all were significant (P<.05). Treated abnormal cows were equivalent to the control normal cows. Thus, GnRH given 13 to 14 d PP to cows characterized as undergoing slow involution of the reproductive system, but with no other clinical problems, seems to assist in promoting rapid normal reproductive function. The objective of these studies was to evaluate whether exposing primiparous, suckled beef cows to the biostimulatory effect of bulls alters breeding performance associated with an estrus synchronization protocol that included GnRH followed 7 d later by PGF(2alpha) and fixed-time AI (TAI). This was a composite analysis of 3 experiments that evaluated (1) the effects of bull exposure at different days after calving (yr 1); (2) the biostimulatory effects of bull excretory products (yr 2); and (3) the biostimulatory effects of familiar and unfamiliar bulls (yr 3) on the resumption of ovarian cycling activity. In all studies, cows were exposed (biostimulated; n = 94) or not exposed (nonbiostimulated; n = 67) to bulls or excretory products of bulls for at least 60 d before the beginning of the estrus synchronization protocol. Average calving day did not differ among years and was 52 +/- 5 d. Year did not affect the proportions of biostimulated and nonbiostimulated cows that were cycling at the beginning of the estrus synchronization protocol; however, a greater (P < 0.001) proportion of biostimulated than nonbiostimulated cows were cycling at this time. In each year, cows were given GnRH followed by PGF(2alpha) 7 d later. Cows were observed for estrus twice daily (am and pm) after PGF(2alpha). Cows that exhibited estrus before 54, 60, and 64 h after PGF(2alpha) were inseminated by AI 12 h later in yr 1, 2, and 3, respectively. Cows that failed to show estrus were given GnRH and TAI at 62, 72, and 72 h after PGF(2alpha) in yr 1, 2, and 3, respectively. Conception rates were determined by transrectal ultrasonography 35 d after TAI in each year. The percentages of cows that exhibited estrus after PGF(2alpha) and before TAI, the interval from PGF(2alpha) to estrus, and the percentages of cows inseminated 12 h after estrus or at TAI did not differ between biostimulated and nonbiostimulated cows and were 51%, 54.7 +/- 7.3 h, 35%, and 65%, respectively. Conception rates for cows bred by AI 12 h after estrus did not differ between biostimulated and nonbiostimulated cows; however, the TAI conception rate was greater (P < 0.05) for biostimulated cows (57.6%) than for nonbiostimulated cows (35.6%). We conclude that TAI conception rates in an estrus synchronization protocol that includes GnRH followed 7 d later by PGF(2alpha) may be improved by the biostimulatory effect of bulls in postpartum, primiparous cows.  相似文献

At 11 to 13 d after insemination, 1028 dairy cows on 19 farms were given an intramuscular injection of 10 micrograms of the GnRH analogue buserelin. Their reproductive performance was compared to 1022 control cows receiving a placebo. The GnRH analogue-treated cows had fewer short interoestrous intervals compared to the control cows, but there were no significant differences in pregnancy rates to either the insemination preceding or following treatment, in calving to conception interval, or to percent pregnant by the end of mating. There were no differences between treatments in herds of high, average or low pregnancy rate, in 2-year-old cows or cows greater than 2-years-old and in cows calved 40 d or less, or greater than 40 d. The use of 10 micrograms of the GnRH analogue, buserelin, 11 to 13 d after insemination, cannot be recommended as a method of improving reproductive performance in dairy herds.  相似文献
