The effects of dietary β‐hydroxy‐β‐methylbutyrate (HMB) supplementation during gestation on reproductive performance of sows and the mRNA expression of myogenic markers in skeletal muscle of neonatal pigs were determined. At day 35 of gestation, a total of 20 sows (Landrace × Yorkshire, at third parity) were randomly assigned to two groups, with each group receiving either a basal diet or the same diet supplemented with 4 g/day β‐hydroxy‐β‐methylbutyrate calcium (HMB‐Ca) until parturition. At parturition, the total and live litter size were not markedly different between treatments, however, the sows fed HMB diet had a decreased rate of stillborn piglets compared with the sows fed the control (CON) diets (p < 0.05). In addition, piglets from the sows fed HMB diet tended to have an increased birth weight (p = 0.08), and a reduced rate of low birth weight piglets (p = 0.05) compared with piglets from the CON sows. Nevertheless, lower feed intake during lactation was observed in the sows fed the HMB diet compared with those on the CON diet (p < 0.01). The relative weights of the longissimus dorsi (LD) and semitendinosus (ST) muscle were higher (p < 0.05) in neonatal pigs from the HMB than the CON sows. Furthermore, maternal HMB treatment increased the mRNA levels of the myogenic genes, including muscle regulatory factor‐4 (MRF4, p < 0.05), myogenic differentiation factor (MyoD) and insulin‐like growth factor‐1 (IGF‐1, p < 0.01). In conclusion, dietary HMB supplementation to sows at 4 g/day from day 35 of gestation to term significantly improves pregnancy outcomes and increases the expression of myogenic genes in skeletal muscle of neonatal piglets, but reduces feed intake of sows during lactation.  相似文献

This study was aimed to investigate the effects of rumen‐protected γ‐aminobutyric acid (RP‐GABA) on apparent nutrient digestibility, growth performance and health status in heat stressed beef cattle. Fifty Jinjiang Yellow cattle were randomly assigned to 5 treatments (10 animals/treatment). Treatments 1 to 5 were basal diets affixed with 0 (control), 8, 16, 24 and 32 mg of RP‐GABA/kg of body weight (BW) respectively. The trial lasted 45 days. Apparent digestibility of crude protein (CP), crude fiber (CF) and calcium (Ca) quadratically increased with increasing RP‐GABA (< .01), while apparent digestibility of phosphorus (P) tended to quadratically increase (= .09). Dietary supplementation with increasing RP‐GABA linearly increased DM digestibility and average daily gain (ADG) (< .01), whereas the feed to gain (F:G) ratio linearly decreased with increasing RP‐GABA (< .01). The average daily feed intake (ADFI) value tended to linearly increase with RP‐GABA supplementation (= .08). Total protein (TP), blood urea nitrogen (BUN) and malondialdehyde (MDA) levels quadratically decreased (< .01) with increasing RP‐GABA, however albumin (ALB), glucose (GLU), superoxide dismutase (SOD), triiodothyronine (T3) and thyroxine (T4) levels quadratically increased (≤ .01). In conclusion, the present results indicated that dietary supplementation with RP‐GABA led to improved nutrient digestibility, growth performance and antioxidant status in heat stressed beef cattle.  相似文献

Two hundred and forty 1‐day‐old male Arbor Acres broiler chickens were randomly assigned to five dietary treatments with six replicates of eight chickens per replicate cage for a 42‐day feeding trial. Broiler chickens were fed a basal diet supplemented with 0 (control), 250, 500, 750 or 1000 mg/kg betaine, respectively. Growth performance was not affected by betaine. Incremental levels of betaine decreased the absolute and relative weight of abdominal fat (linear P < 0.05, quadratic P < 0.01), low‐density lipoprotein cholesterol (LDL‐C), triglyceride (TG) and total cholesterol (TC) (linear P < 0.05), and increased concentration of nonesterified fatty acid (NEFA) (linear P = 0.038, quadratic P = 0.003) in serum of broilers. Moreover, incremental levels of betaine increased linearly (P < 0.05) the proliferator‐activated receptor alpha (PPARα), the carnitine palmitoyl transferase‐I (CPT‐I) and 3‐hydroxyacyl‐coenzyme A dehydrogenase (HADH) messenger RNA (mRNA) expression, but decreased linearly (P < 0.05) the fatty acid synthase (FAS) and 3‐hydroxyl‐3‐methylglutaryl‐CoA (HMGR) mRNA expression in liver of broilers. In conclusion, this study indicated that betaine supplementation did not affect growth performance of broilers, but was effective in reducing abdominal fat deposition in a dose‐dependent manner, which was probably caused by combinations of a decrease in fatty acid synthesis and an increase in β‐oxidation.  相似文献

This study was designed using 360 21‐day‐old chicks to determine the influences of diet supplementation with glutamine (5 g/kg), γ‐aminobutyric acid (GABA, 100 mg/kg) or their combinations on performance and serum parameters exposed to cycling high temperatures. From 22 to 35 days, the experimental groups (2 × 2) were subjected to circular heat stress by exposing them to 30–34 °C cycling, while the positive control group was exposed to 23 °C constant. The blood of broilers was collected to detect serum parameters on days 28 and 35. Compared with the positive control group, the cycling high temperature decreased (p < 0.05) the feed consumption, weight gain and serum total protein (TP), glucose, thyroxine (T4), insulin, alkaline phosphatase (ALP), glutamine, GABA and glutamate levels, while increased (p < 0.05) the serum triglyceride (TG), corticosterone (CS), glucagon (GN), creatine kinase (CK), glutamic oxaloacetic transaminase (GOT), nitric oxide synthase (NOS), glutamate pyruvate transaminase (GPT) and lactate dehydrogenase (LDH) levels during 22–35 days. However, dietary glutamine (5 g/kg) increased (p < 0.05) the feed consumption, weight gain and serum levels of glutamine, TP, insulin and ALP, but decreased (p < 0.05) the serum TG, CK, GOT, NOS and GPT levels. Diet supplemented with GABA also increased (p < 0.05) weight gain and the serum levels of TP, T4, ALP, GABA and glutamine. In addition, the significant interactions (p < 0.05) between glutamine and GABA were found in the feed consumption, weight gain and the serum ALP, CK, LDH, GABA, T3 and T4 levels of heat‐stressed chickens. This research indicated that dietary glutamine and GABA improved the antistress ability in performance and serum parameters of broilers under hot environment.  相似文献

The leucine metabolite β‐hydroxy‐β‐methylbutyrate (HMB) has been studied by many researchers over the last two decades. In particular, the utility of HMB supplementation in animals has been shown in numerous studies, which have demonstrated enhanced body weight gain and carcass yield in slaughter animals; positive immunostimulatory effect; decreased mortality; attenuation of sarcopenia in elderly animals; and potential use in pathological conditions such as glucocorticoid‐induced muscle loss. The aim of this study was to summarize the body of research on HMB supplementation in animals and to examine possible mechanisms of HMB action. Furthermore, while the safety of HMB supplementation in animals is well documented, studies demonstrating efficacy are less clear. The possible reasons for differences in these findings will also be examined.  相似文献

β‐carotene is one of the most abundant carotenoids, has potential anti‐inflammatory effect, it has been reported that β‐carotene could suppress LPS‐induced inflammatory responses by inhibiting nuclear factor kappa B (NF‐κB) translocation, but the more detailed molecular mechanisms underlying the anti‐inflammatory action of β‐carotene remain to be fully understood. In this study, we investigated the influence of β‐carotene on the activation of JAK2/STAT3, MAPK, and NF‐κB signaling pathway induced by LPS in RAW264.7 cells and peritoneal macrophages. Cells were treated with different concentrations of β‐carotene for 3 hr after LPS treatment for 24 hr. The mRNA expression and the release of IL‐1β, IL‐6, and TNF‐α were evaluated by RT‐PCR and ELISA, and the level of signaling proteins of JAK2/STAT3, MAPK, and NF‐κB signaling pathway were detected by Western blot. The results showed that β‐carotene significantly suppressed (p < 0.05) LPS‐induced release of IL‐1β, IL‐6, and TNF‐α and their mRNA expression. LPS‐induced JAK2/STAT3, IκB/NF‐κB p65, JNK/p38 MAPK signal activation were significantly attenuated (p < 0.05) by β‐carotene in a dose‐dependent manner. In conclusion, β‐carotene could attenuate LPS‐induced inflammation via inhibition of the NF‐κB, JAK2/STAT3, and JNK/p38 MAPK signaling pathways in macrophages.  相似文献

This study evaluated the effects of rice whole crop silage (RWCS) on growth, plasma levels of vitamin A, β‐carotene, vitamin E and IGF‐1, and expression of genes involved in muscle protein degradation and synthesis in Japanese Black calves. Eleven calves were divided into RWCS (fed RWCS ad libitum and concentrate, n = 5) and control groups (fed hay ad libitum and concentrate, n = 6). Final body weight and dairy gain were significantly larger in the RWCS group compared with the control group. Plasma β‐carotene and vitamin E concentrations were significantly higher in the RWCS group compared with control group. Although plasma vitamin E concentration in the RWCS group significantly increased from 4 to 9 months of age, it did not increase in the control group. At 6 months of age in the RWCS group, ubiquitin B (< 0.05) and calpain 1 (= 0.097) mRNA expression were lower than control group, but they were not different between groups at 9 months of age. These results indicate that RWCS increases plasma β‐carotene level and promotes muscle growth because of a decrease in the rate of protein degradation, but the effect is lost with the increase in plasma vitamin E level.  相似文献

α‐Ketoglutarate (AKG) is an extensively used dietary supplement in human and animal nutrition. The aim of the present study was to investigate effects of dietary AKG supplementation on the energy status and anti‐oxidative capacity in liver and intestinal mucosa of Cherry Valley ducks. A total of 80 1‐day‐old ducks were randomly assigned into four groups, in which ducks were fed basal diets supplemented with 0% (control), 0.5%, 1.0% and 1.5% AKG, respectively. Graded doses of AKG supplementation linearly decreased the ratio of adenosine monophosphate (AMP) to adenosine triphosphate (ATP) in the liver, but increased ATP content and adenylate energy charge (AEC) in a quadratic and linear manner, respectively (< 0.05). Increasing dietary AKG supplemental levels produced linear positive responses in ATP content and AEC, and negative responses in AMP concentration, the ratio of AMP to ATP and total adenine nucleotide in the ileal mucosa (P < 0.05). All levels of dietary AKG reduced the production of jejunal hydrogen peroxide and hepatic malondialdehyde (P < 0.05). Hepatic and ileal messenger RNA expression of AMP kinase α‐1 and hypoxia‐inducible factor‐1α were linearly up‐regulated as dietary AKG supplemental levels increased (P < 0.05). In conclusion, dietary AKG supplementation linearly or quadratically enhanced hepatic and intestinal energy storage and anti‐oxidative capacity of Cherry Valley ducks.  相似文献

The objective of the present study was to evaluate a potential of Schizochytrium microalga oil to alleviate possible negative effects of high‐fat‐high‐energy diets. Forty adult male rats (Wistar Albino) were fed 7 weeks the diet containing beef tallow + evaporated sweetened milk (diet T) intended to cause mild obesity and low‐grade systemic inflammation. Consequently, the animals were divided into four groups by 10 animals each and fed either the T‐diet (control) or the diet containing 6% of safflower oil (S), 6% of fish oil (F) and 6% of Schizochytrium microalga oil (A), respectively, for another 7 weeks. The A‐diet decreased (p < 0.05) live weight to 86% and glycaemia to 85% of control, respectively; an effect of the S‐ and F‐diet on these markers was insignificant (p > 0.05). In comparison with control, higher (p < 0.05) deposition of eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) in the epididymal adipose tissue (EAT) of the A‐rats correlated with increased (p < 0.05) plasma adiponectin concentration, but it was without the effect (p > 0.05) on cellular adiponectin content in the EAT. Higher (p < 0.05) EPA+DHA deposition in the liver of the A‐rats correlated with higher expression (149% of control; p < 0.05) of the gene coding for peroxisome proliferator‐activated receptor gamma, and with lower expression (82% and 66%; p < 0.05) of the genes coding for adiponectin receptors AdipoR1 and AdipoR2; no relationship to the expression of receptor GPR120 was found. The A‐diet did not affect amount of the nuclear fraction of the nuclear factor kappa B in the liver, but increased plasma level of anti‐inflammatory cytokine TGF‐β1 (p < 0.05). The presented data agree with results of other in vivo rodent and human studies, but not with literature data regarding in vitro experiments: it can be concluded that the effects of dietary oils on inflammatory markers need further investigation.  相似文献

Ascites is a cardiovascular metabolic disease characterized by accumulation of fluid around the heart and in the abdominal cavity that eventually leads to death. This syndrome is the end‐point result of a series of metabolic incidents that are generally caused by impaired oxygen availability. Mitochondria are the major sites of oxygen consumption, therefore major contributors to oxidative stress. Genetic, metabolic and dietary factors can influence variations in mitochondrial biogenesis (mitochondrial size, number and mass) that might have an effect on oxygen consumption and reactive oxygen species production. This study evaluated the effect of genotype on PGC‐1α mRNA gene expression and mitochondrial biogenesis. These parameters were examined in male broiler chickens at 22 weeks of age from the SUS and RES lines divergently selected for ascites phenotype. From each line, five birds were sampled for right ventricle and breast muscle. Gene expression and mtDNA copy number were assessed by quantitative PCR. Results showed that birds from SUS had significantly higher PGC‐1α mRNA gene (p = .033) and mitochondrial DNA copy number (p = .038) in breast muscle. There was no difference in right ventricle PGC‐1α expression or mitochondrial DNA copy number between the two lines. These findings indicate that mitochondrial biogenesis and PGC‐1α mRNA gene expression differ between male broiler chickens from RES and SUS lines in a tissue‐specific manner.  相似文献

Amylose plays important role in body health. It is controversial whether changing dietary amylose/amylopectin ratio (DAR) will improve meat quality in growing‐finishing pigs. A total of 48 Duroc × Landrace × Large White castrated male pigs (initial body weight 49.8 ± 2.8 kg) were randomly allotted to two treatments, and fed ad libitum either with a low DAR diet (LR, amylose/amylopectin: 12/88) or a high DAR diet (HR, amylose/amylopectin: 30/70) for 68 days. Feed intake was recorded every day, body weight was weighed at 46th and 68th day to calculate average daily gain (ADG), average daily feed intake (ADFI) and Feed:gain ratio. Blood was collected at ?30 min (fasting 12 hr), 60, 90, 120, 180 min postprandial at 64th day and then serum was obtained by centrifugation of blood at 1,500× g at 4°C. After pigs were slaughtered, samples such as longissimus dorsi, iliopsoas and semitendinosus were collected. Density, diameter and types of muscle fibres were analysed. Results showed that ADG, ADFI, Feed:Gain ratio, cross‐sectional area of longissimus dorsi muscle, backfat thickness, colour scores were not affected by DAR. Ingestion of LR diet increased the fasting glucose (< 0.05) and insulin (< 0.05) concentrations in serum. The drip loss and firmness were decreased significantly in LR vs. HR animals (< 0.05). Densities of muscle fibre in longissimus dorsi, iliopsoas and semitendinosus were greater in LR pigs (< 0.05). Moreover, ingestion of LR diet significantly increased myosin heavy chain (MyHC) IIa mRNA level and decreased MyHC IIb gene expression in longissimus dorsi muscle (LM) (< 0.05). Therefore, intake of diet low in amylose/amylopectin ratio induces a better meat quality (lower drip loss and lower firmness), which could attribute to smaller myofibres, a shift to slower and/or more oxidative fibres.  相似文献

Thirty‐six Malpura lambs (28 day old and 6.7 ± 0.25 kg BW) were distributed equally in three groups having six males and six female. They were ad libitum fed individually three different experimental diets containing calcium soap of fatty acids (CA‐FA) at 0 (T1) and 40 (T2 and T3) g/kg concentrate up to six months of age. Animals in T3 were supplemented additionally with 40 mg DL‐α‐tocopherol acetate/kg of concentrate. The roughage moiety included ad libitum dry Prosopis cineraria and fresh Azadirachata indica leaves. All the lambs were allowed to suckle from their dam up to weaning (90 day of age). Supplementation of Ca‐FA improved weight gain and feed conversion ratio during both pre‐ (28–90 days) and post‐weaning (91–180 days) phases; however, no effect of DL‐α‐tocopherol was observed. Metabolic parameters during post‐weaning phase revealed non‐significant effect on digestibility but improved nitrogen balance in the test groups. The effect on biochemical attributes did not show any significant alteration in ruminal parameters, blood biochemicals and urinary purine derivatives. Carcass traits revealed higher (p < 0.05) dressing yield and loin eye area with Ca‐FA supplementation. The value of thiobarbituric reactive substances for nuggets prepared from frozen carcasses revealed significant (p < 0.05) reduction in T3 compared to the other dietary groups. Fatty acid profile of adipose tissue revealed higher (p < 0.001) 9‐octadecanoic, 9‐12‐octadecadienoic, polyunsaturated fatty acids (PUFA), higher ratio of PUFA/saturated fatty acids (SFA), ω‐6/ω‐3 and lower SFA in Ca‐FA‐supplemented groups. It is concluded that supplementation of 40 g/kg calcium soap prepared from industrial grade rice bran oil in lamb ration provided additional energy intake, improved N utilization, gain and feed conversion ratio besides improving dressing yield and meat quality with CLA enriched fatty acid profile. DL‐α‐tocopherol acetate when supplemented at 40 mg/kg feed reduced lipid oxidation of meat products thus improving its keeping quality.  相似文献

To examine the effects of dietary β‐carotene (βC) or retinyl palmitate (RP) on fatty acid (FA) profile and mRNA expression, samples were collected from 24 Angus‐cross calves that were allotted to four treatments consisting of RP supplemented at 2200 IU/kg, and synthetic β‐carotene (SβC) supplemented at one, five or 10 times RP. Longissimus muscle (LM) cis‐9, trans‐11 conjugated linoleic acid was greater in RP compared to SβC1X (= 0.04). The polyunsaturated:saturated FA increased linearly (= 0.04) in the LM as dietary SβC increased. Expression of βC oxygenase 2 (βCO2), an enzyme that cleaves β‐carotene, was greater in the LM for SβC1X compared to RP and decreased linearly as SβC increased (P  0.02). Peroxisome proliferator activated receptor γ (PPARγ) expression in the LM increased in SβC1X compared to RP (= 0.03); however, PPARγ and retinoic acid X receptor α (RXRα) expression decreased linearly (P = 0.02) in the LM with increasing SβC. Retinoic acid receptor α (RARα) expression tended (= 0.10) to decrease linearly in the LM with increased SβC. In conclusion, SβC supplementation increased mRNA expression of some lipogenic genes in the LM, but increasing dietary SβC inhibited their expression and tended to increase polyunsaturated FA.  相似文献

α‐1,3‐Galactosyltransferase (α‐GalT), an enzyme creating Galα1‐3Gal (α‐Gal) epitope on the cell surface in some mammalian species such as pigs, is known to be a key factor that causes hyperacute rejection upon transplantation from pigs to humans. To establish the RNA interference‐based suppression of endogenous α‐GalT messenger RNA (mRNA) synthesis in porcine preimplantation embryos, we determined the suitable embryonic stage at which stage such approach is possible by using the semi‐quantitative RT‐PCR (qRT‐PCR) and the cytochemical method using a fluorescence‐labeled Bandeiraea simplicifolia Isolectin B4 (BS‐I‐B4). Staining with BS‐I‐B4 demonstrated that α‐Gal epitope expression was first recognized at the 8‐cell stage, and increased up to the hatched blastocyst stage. Single embryo‐based qRT‐PCR also confirmed this pattern. These results indicate that creation of α‐Gal epitope is proceeded by de novo synthesis of α‐GalT mRNA in porcine preimplantation embryos with peaking at the blastocyst stage.  相似文献

This study was conducted to evaluate the effects of dietary supplementation of protease derived from Pseudoalteromonas arctica (PPA) in finishing pigs. A total of 160 pigs were used in this 10‐week trial. Dietary treatment groups were as follows: CON (basal diet); TRT1 (basal diet + 0.1% PPA); TRT2 (basal diet + 0.2% PPA); and TRT3 (basal diet + 0.3% PPA). During weeks 1–5, pigs fed with different levels of PPA‐supplemented diet showed linear increase (p < .05) in the apparent total tract digestibility (ATTD) of nitrogen (N) and linear decrease (p < .05) in the concentrations of serum total protein. During weeks 6–10, pigs fed with different levels of PPA‐supplemented diet showed a linear decrease in feed conversion ratio (p < .05). During the overall period, there was a linear decrease in feed conversion ratio (p < .05) associated with the inclusion of PPA. Pigs fed diets with 0.2% PPA supplementation had lower (p < .05) feed conversion ratio than those fed CON diet during weeks 6–10 and the overall period, and had higher (p < .05) ATTD of N than those fed CON diet during weeks 1–5. Pigs fed diets with PPA supplementation had lower (p < .05) concentrations of serum total protein than those fed CON diet on week 5. In conclusion, dietary supplementation with PPA diet has beneficial effects on growth performance, nutrient digestibility, backfat thickness and the concentrations of serum total protein.  相似文献

Forty‐eight Duroc × Large White × Landrace pigs with an average initial body weight of 77.09 ± 1.37 kg were used to investigate the effects of combination of leucine (Leu) with arginine (Arg) or glutamic acid (Glu) on muscle growth, free amino acid profiles, expression levels of amino acid transporters and growth‐related genes in skeletal muscle. The animals were randomly assigned to one of the four treatment groups (12 pigs/group, castrated male:female = 1:1). The pigs in the control group were fed a basal diet (13% Crude Protein), and those in the experimental groups were fed the basal diet supplemented with 1.00% Leu (L group), 1.00% Leu + 1.00% Arg (LA group) or 1.00% Leu + 1.00% Glu (LG group). The experiment lasted for 60 days. Results showed an increase (p < 0.05) in biceps femoris (BF) muscle weight in the L group and LG group relative to the basal diet group. In longissimus dorsi (LD) muscle, Lys, taurine and total essential amino acid concentration increased in the LG group relative to the basal diet group (p < 0.05). In LG group, Glu and carnosine concentrations increased (p < 0.05) in the BF muscle, when compared to the basal diet group. The Leu and Lys concentrations of BF muscle were lower in the LA group than that in the L group (p < 0.05). A positive association was found between BF muscle weight and Leu concentration (p < 0.05). The LG group presented higher (p < 0.05) mRNA levels of ASCT2, LAT1, PAT2, SANT2 and TAT1 in LD muscle than those in the basal diet group. The mRNA levels of PAT2 and MyoD in BF muscle were upregulated (p < 0.05) in the LG group, compared with those in the basal diet group. In conclusion, Leu alone or in combination with Glu is benefit for biceps femoris muscle growth in fattening pig.  相似文献

Data from 18 β‐carotene‐deficient Japanese Black cows were collected to clarify the effects of feeding β‐carotene‐enriched dry carrots on β‐carotene status and colostral immunoglobulin (Ig) in cows. Cows were assigned to control or carrot groups from 3 weeks before the expected calving date to parturition, and supplemental β‐carotene from dry carrots was 138 mg/day in the carrot group. Plasma β‐carotene concentrations in the control and carrot groups at parturition were 95 and 120 μg/dL, and feeding dry carrots slightly improved plasma β‐carotene at parturition. Feeding dry carrots increased colostral IgA concentrations in cows and tended to increase colostral IgG1, but colostral IgM, IgG2, β‐carotene and vitamin A were not affected by the treatment. Feeding dry carrots had no effects on plasma IgG1, IgA and IgM concentrations in cows, but plasma IgG1 concentrations decreased rapidly from 3 weeks before the expected calving date to parturition. These results indicate that feeding β‐carotene‐enriched dry carrots is effective to enhance colostral IgA and IgG1 concentrations in β‐carotene‐deficient cows.  相似文献
