The free amino acids (FAA) profile was determined for newly fertilized eggs and resultant larvae from wild-caught red snapper Lutjanus campechanus induced to spawn with hCG. Yolk sac and oil globule volumes of eggs and larvae were monitored over time from digital photographs. FAA profiles of the eggs and larvae were measured in picomoles (pmol) of FAA/mg of eggs by HPLC. Newly fertilized eggs had a mean total FAA content of 21.72 ± 3.55 nmoles/egg (92.81 ± 9.71 nmoles/mg eggs). Leucine, valine, lysine, and isoleucine were the most abundant essential FAA comprising 35.9% of the total FAA. Alanine, serine, asparagine, and glycine were the most abundant non-essential FAA comprising 34.2% of the total FAA. At 24 h post-hatch (hph) the mean total FAA had decreased by 81% since egg fertilization. The bulk of the FAA decrease was between the time of hatch and 12 hph. Only 8.5 ± 1.5% of the initial concentration in fertilized eggs of isoleucine, 9.7 ± 2.5% of arginine, and 9.9 ± 2.0% of threonine remained at 12 hph. Among the non-essential FAA, alanine dropped the most by 12 hph with 4.6% of the concentration found in a recently fertilized egg remaining, while cysteine had increased 254.7 ± 26.2%. The yolk sac volume decreased rapidly in the first 12 hph and was further reduced 77.0 ± 2.5% from 12 to 24 hph. The oil globule depletion rate was a more linear decline from fertilized egg to 36 hph.  相似文献

This study was conducted to determine the effects of dietary highly unsaturated fatty acids (HUFA) on flame angelfish (Centropyge loriculus) reproduction, and egg and larval quality. In the experiment, formulated diets containing 1.8, 2.9 or 3.6% n‐3 HUFA were fed to flame angelfish broodstock (n = 4) for 5 months. Mean fecundity (daily egg production), egg fertilization rates and embryo viability were used as indicators of egg quality. In addition, mean egg diameter, oil globule diameter, per cent hatch, larval size at hatch, per cent survival to yolk exhaustion and larval size at yolk exhaustion were recorded for each treatment. Flame angelfish fed the diet containing 3.6% n‐3 HUFA exhibited significantly increased fecundity, fertilization rates and embryo viability than fish that were fed the other two formulated diets. Egg diameter, egg oil globule diameter, larval size at hatch, larval survival to yolk exhaustion and larval size at yolk exhaustion from the 3.6% n‐3 HUFA broodstock treatment group were not significantly different from those derived from the Control broodstock treatment. These data revealed that flame angelfish egg quality could respond rapidly (within weeks) to maternal dietary changes. Results from this study further support that dietary HUFA composition can significantly affect broodstock reproductive performance as well as subsequent performance of eggs and larvae.  相似文献

Sexual maturation and induced spawning treatments were carried out with captive spotted rose snapper, Lutjanus guttatus. A total of 3013 × 106 eggs (64.7% were floating) were produced from eight treated females in 42 spawns induced with GnRHa implants during the course of the present study. GnRHa ethylene‐vinyl acetate copolymer effective doses were 204 ± 11 µg/kg in June 2005, and 224 ± 13 µg/kg in July 2005. General fertilization was 50.9 ± 34.5% and 12–14 h after spawning, viability of floating eggs was 90.4 ± 12.4%. Mean incubation period at 29–31 C was 18–20 h, and mean hatching was 94.4 ± 8.2% (73–100%). Newly hatched larvae were 2.18 ± 0.15 mm in total length (TL). One month after the last hormone experiment, previously GnRHa‐treated and untreated fish began spawning voluntarily. Hormone‐treated breeders had higher fecundity than untreated fish, producing 72.5 million eggs versus 13.9 million eggs for the untreated fish, over the following 11 mo. Combined data of volitional spawning for total egg fertilization, viability, hatching, and larval TL were 77.7 ± 1.8%, 90.3 ± 1.3%, 87.9 ± 2%, and 2.50 ± 0.12 mm, respectively. This is the first report on the successful year-round natural spawning and larval rearing of Epinephelus polyphekadion (Bleeker) in captivity and under hypersaline water conditions of 42-43%0 salinity in the Red Sea coast of Saudi Arabia. Although the fish spawned naturally once or twice a year during 1992-94 culture period, incorporation of cod-liver oil in the broodstock diet during the 1995 culture period enabled the fish to spawn continuously for 2-3 days in each month during March, April, May and August. The egg fertilization and hatching rates also increased during the 1995 spawning period. The egg fertilization rate varied from 90 to 100% with a mean of 96.5 ± 3.38%. The egg hatching rate varied from 70 to 95% with a mean of 83.1 ± 10.12%. The fertilized egg diameter averaged 757.3 ± 37.36 μm. There was a linear relation between the fertilized egg size and the egg hatching rate. The increase in the hatching rate relevant to the egg size was statistically significant (P < 0.01). The egg development time until hatching lasted for 19 h at 29°C. The newly hatched larval size ranged from 1.55-1.71 mm with a mean of 1.65 ± 0.052 mm in total length. The larval growth was slow in the early stages and the growth curve until metamorphosis showed a curvilinear pattern. Wide variations in larval size, range 22-47 mm with a mean of 33.40 ± 7.01 mm, were observed during the metamorphosis stage at day 50. No significant difference (P > 0.05) in growth and survival was observed between the larvae reared using white and grey coloured tanks. The larval survival up to metamorphosis was 1.6-4.7% with a mean of 2.98 ± 1.56% in the grey coloured tanks and 1.6-1.9% with a mean of 1.73 ± 0.16% in the white tanks. The results demonstrated the possibility of breeding E. polyphekadion under captive culture conditions. However, methods to improve the larval survival have to be pursued further for commercial farming of this species.  相似文献

A reliable breeding technique was developed for the mangrove red snapper, Lutjanus argentimaculatus (Forsskal 1775), to help sustain the aquaculture of this immensely popular species in Southeast Asia. Using standardized indices of female maturity (based on mean oocyte diameter of ≥0.40 mm), time of injection (1000–1130) and sex ratio (one female to two males), a single injection of 100 μg kg?1 luteinizing hormone‐releasing hormone analogue (LHRHa) (n=16 fish), but not 50 μg kg?1 (n=five fish), successfully induced egg (62.5% success rate) and larval (43.8%) production. Human chorionic gonadotropin (hCG) at 500 IU kg?1 (n=five fish) also failed to induce spawning, but doses of 1000 (n=22 fish) and 1500 IU kg?1 (n=15 fish) gave spawning (77.3% and 80.0% respectively) and hatching success rates (72.7% and 60.0% respectively) that were not significantly different from those of 100 μg kg?1 LHRHa. No spawning was observed in saline‐injected controls (n=seven fish). While mean spawning latency, egg diameter, egg production per spawn, percent egg viability, hatching rate, percent of normal larvae and cumulative survival of eggs to normal larvae did not differ significantly among the effective hormones and doses, 1000 IU kg?1 hCG had a higher percentage (76.5%) of total spawns with egg production per spawn in excess of one million than those of 1500 IU kg?1 hCG (50.0%) and 100 μg kg?1 LHRHa (40.0%). Mangrove red snapper spontaneously spawned from March–April to November–December with a peak of egg collection and spawning in May–June. Egg collection per spawn ranged from 0.05 to 6.35 million. Spontaneous spawning of mangrove red snapper exhibited lunar periodicity with spawns mostly occurring 3 days before or after the last quarter and new moon phases and occurred consistently between 02:00 and 04:00 hours. High fecundity and good egg quality, coupled with the ability to respond to induce spawning or natural spawning in captivity, provide a sound basis for improving the sustainability of red snapper aquaculture in Southeast Asia.  相似文献

Twenty egg batches spawned naturally from 17 different females over two spawning seasons were used to evaluate the egg quality of cobia Rachycentron canadum. A reduction in egg size was observed towards the end of the spawning season for both years. The proportion of floating eggs demonstrated a positive linear relationship with both yolk‐sac larval survival (r2=0.91, P<0.05) and batch larval production (r2=0.80, P<0.01). Viable egg batches (i.e. fertilization success >50%) were of higher batch fecundity, had larger eggs and a higher proportion of floating eggs than non‐viable batches (i.e. 0% fertilization success). Also, biochemical analyses revealed that these viable eggs had significantly higher protein and amino acid contents. A multiple regression model based on the proportion of floating eggs, batch fecundity and fertilization success provided the most accurate predictions of batch larval production (r2=0.95, P<0.001). Similarly, using the egg content of arginine/glycine and methionine significantly increased the correlation coefficient in the multiple regression model predicting larval deformity (r2=0.92, P=0.002). This study reveals that accurate determination of egg quality in cobia can be improved using a combination of several variables rather than a single variable.  相似文献

This study was conducted to determine the effects of broodstock diet on flame angelfish (Centropyge loriculus) reproduction and generate baseline spawning performance (mean fecundity, egg fertilization, egg viability) and egg biochemistry data for this species. Sixteen flame angelfish broodstock pairs were maintained on one of three formulated diets or a raw diet (consisting of squid, shrimp, fish eggs and algae) to determine the effect of diet composition on the fecundity, egg fertilization rates and egg viability over a 7‐month period. Although all the diets supported similar mean spawn frequency and fecundity over the duration of the experiment, the mean fertilization rates and egg viability were significantly greater among pairs receiving the raw diet. Egg fatty acid composition was generally reflective of the maternal diet and similar to the values reported for other tropical species. In particular, dietary factors such as n‐3 highly unsaturated fatty acid and ARA level (which were elevated in the raw diet and eggs from fish receiving that treatment) are correlated with egg quality and are therefore worthy of future investigation. Because of the ease of conditioning, adaptability to varied formulated diets and continuous, sustained reproductive output, flame angelfish are an excellent model for examining the factors affecting marine fish egg and larval quality.  相似文献

A stripspawning methodology was evaluated for tilapia (oreochromid) species. This technique achieved an average hatching success of 68.6 ± 3.6% (N= 31). Female fecundity and spawning frequency were dependant on both genetical and husbandry factors. Egg yields for Oreochromis niloticus, O. mossambicus, and O. niloticus±O. mossambicus hybrids averaged 4.54, 10.86 and 10.36 eggs/g female/spawn, respectively. Female broodstock that were adapted to an intensive spawning regime exhibited a significant increase in fecundity. Additionally, egg survival was not affected by hydration for up to 15 minutes prior to fertilization. Results suggest that the strip spawning of tilapia species may be an efficient method of providing viable gametes for hatchery purposes.  相似文献

The objective of the study was to acclimatise wild-caught meagre (Argyrosomus regius) to captivity to produce viable eggs for aquaculture production. Twelve meagre (3 males and 9 females, mean weight?=?20?±?7?kg) were caught and transported to a land-based facility on 26 October 2006. During, March to June 2007, all three males were spermiating and five of the nine females were in vitellogenesis with mean maximum oocyte diameter ≥550?μm. No spontaneous spawning was observed. Two hormone treatments, either a single injection of gonadotropin-releasing hormone agonist (GnRHa, 20?μg?kg(-1) for females and 10?μg?kg(-1) for males) or a slow-release implant loaded with the same GnRHa (50?μg?kg(-1) for females and 25?μg?kg(-1) for males), were used to induce spawning on three different dates on 26 March 2007, 4 May 2007 and 18 April 2008. From each spawning event, the following parameters were determined: fecundity, number of floating eggs, egg size, fertilisation and hatching success, unfed larval survival, and proximal composition and fatty acid profile of the eggs. In 2007, two females that were injected on 26 March and 4 May spawned a total of 5 times producing 9,019,300 floating eggs and a relative fecundity of 198,200 eggs?kg(-1) and two different females that were implanted on the same dates spawned 14 times producing 12,430,000 floating eggs and a relative fecundity of 276,200 eggs?kg(-1). In 2008, a pair that was implanted spawned five times producing a total of 10,211,900 floating eggs and a relative fecundity of 527,380 eggs?kg(-1). The latency period was 48-72?h. Parameters were compared between hormone treatments, date of hormone induction and parents determined by microsatellites. Australian bass, Macquaria novemacideata (Steindachner),mature but do not spawn in fresh or brackish water ponds. Ovulation and spawning of captive (n = 158)and wild Australian bass (n = 123) was induced in the normal breeding season by single injections of human chorionic gonadotropin (hCG). Doses of 100-4000IU kg-“ hCG induced ovulation and the optimum dosage was 500 IU kg” hCG. The breeding season was from mid-May to late August for wild fish,and extended into September for captive fish. There was a tendency for mean fertilization and hatching success to decline over the breeding season. Greater fertilization and hatching success was obtained from fish which spawned naturally than from stripped fish. Fish spawned after 34,2 ± 0.4 h (mean ± SE, n = 74). Ovulating fish that failed to spawn were stripped after 40.2 ± 0.3 h (n = 76). The timing of stripping and fertilization was an important factor determining hatching success. There was no apparent difference in latency periods or the number of eggs spawned between captive and wild fish. However, the mean number of eggs obtained from naturally spawned fish was higher than for stripped fish. The techniques described in this paper will assist the largescale production of Australian bass by increasing the quality and quantity of larvae from hCG-induced spawnings.  相似文献

Two experiments addressed the spontaneous spawning of red snapper, Lutjanus campechanus , under controlled temperatures and photoperiods and the effect of broodstock diets supplemented with oils rich in polyunsaturated fatty acids. In Experiment 1, broodfish were fed a standard diet (ST1) and one enriched with menhaden oil (ER1) over a 355-d period. ER1-influenced egg fatty acid profile, however, did not positively influence egg production. Both diets produced highly viable eggs and larvae but results varied within treatments. In Experiment 2, broodfish were fed either a standard diet (ST2) or one supplemented with oils (ER2) rich in docosahexaenoic and arachidonic acid using a 203-d cycle. Both treatments produced eggs but fertilization rates ranged 0–10%. There was no clear influence of the diets on egg fatty acid profiles. These results indicate that red snapper can spawn spontaneously in tanks under controlled environmental conditions and produce viable eggs and larvae when fed diets based on squid, shrimp, and fish. The fatty acid composition of the diets was reflected in the eggs to some degree, but the oil enrichments did not further enhance the reproductive performance and egg quality under the conditions of this study.  相似文献

为了解铠平鲉繁殖生物学特征及胚胎发育规律,完善铠平鲉繁殖生物学研究内容,为其种质资源保护和资源可持续利用提供基础资料,实验利用春季于青岛胶州湾海域采集的铠平鲉生殖群体样本,开展了其繁殖生物学研究,并使用显微数码摄像系统,观察了铠平鲉体内胚胎的发育过程,定性和定量描述了各发育阶段胚胎的形态特征。结果显示,实验采集样本的体长范围为62~137 mm,体质量范围为9.4~126.8 g,雌雄性比为1.85∶1,最小性成熟年龄为2龄,生殖群体主要由2~4龄个体组成,初次性成熟体长为70.6 mm。绝对怀卵量F为656~9235粒,平均3176粒,体长相对怀卵量FL为12~67粒/mm,体质量相对怀卵量FW为72~140粒/g。雌鱼体内受精卵呈正圆球形,平均卵径为(1.50±0.03)mm,淡黄色,自心跳期开始,受精卵逐渐由正圆球形变为梨形。受精卵在海水中为沉性,具多个油球,在胚胎发育早期存在油球聚合现象,一般至原肠胚末期时仅剩1个大油球。雌鱼体内初孵仔鱼全长(5.57±0.06)mm。研究表明,虽然卵胎生铠平鲉的胚胎发育规律与大多数海水硬骨鱼类基本相似,但胚胎发育中油球数量、色素分布和受精卵外部形态变化等仍具有其特异性。  相似文献

Effects of maternal age on fecundity, spawning interval, and egg quality of Nile tilapia, Oreochromis niloticus (L.), was examined for four age groups (4, 9, 16, and 24 mo) in circular concrete tanks in a recirculating system. Fecundity (eggs per spawn), weight (g), and egg quality were monitored every 4 d for the 168‐d experimental period. Eggs per spawn correlated with maternal age, while it did not correlate with body weight. Eggs per spawn increased by twofold between 4‐ and 24‐mo‐old females, while eggs per female per d did not differ. Implying that eggs per female per d was influenced by spawn per female as well. Eggs per female per d was 1.8 and 2.4 times higher in 9‐mo‐old females than 16‐ and 24‐mo‐old females, respectively. Eggs per female per d showed a decline concurrent with per spawn per female trend after 18‐mo old. Four‐mo‐old females produced eggs that were more uniform in size and weight than others. While egg size (mm) and weight (mg), and their variations within eggs per spawn increased, percent fertilization and hatchability decreased with age of females. Biological optimum age range for reproducible production of good quality eggs was 6–18 mo. This could be adopted in selecting breeders for better seed production.  相似文献

Hormone Induced Spawning of Summer Flounder Paralichthys dentatus   总被引:2,自引:0,他引:2  
During their first year in captivity, summer flounder Paralicthys denratus were induced to spawn with gonadotropin releasing hormone analogue (GnRHa) implants, injected carp pituitary extract (CPE) or human chorionic gonadotropin (hCG) injections. The percentage of fertile eggs was greatest (69%) in CPE-treated females. CPE, but not GnRHa or hCG, was capable of stimulating oocyte growth (increased follicle diameter during vitellogenesis) followed by ovulation. Fish with maximum ovarian follicle diameters between 180 and 435 μm at the initiation of CPE injections produced the greatest percentage of fertile eggs. For most females, fertilization rate was greatest for the first batch of eggs ovulated. The mean fertilization rate for the first spawn of CPE-treated fish was 42% compared with 14% for the second spawn from the same fish. Fish with maximum initial follicle diameters of 585 40 μm that were implanted with GnRHa ovulated the greatest number of eggs, but fertility was low and variable. Approximately 35% of females injected with hCG ovulated a limited number of eggs, but only one hCG-treated female produced fertile eggs. Only a limited number of spermiating males were available for spawning trials. Hormone treatments used on females were ineffective for inducing or maintaining spermiation in male summer flounder.  相似文献

Successful rearing of larval fish requires culture conditions and feeding strategies matching the ontogenetic status of larvae. This study describes the external morphology and development of organs and structures involved in the feeding process (i.e. sensorial organs, mouthparts and digestive system) from hatching until first feeding in Pacific red snapper. Hatching occurred 26 h after fertilization at 26°C and total length (TL) was 2.45 ± 0.08 mm. The larvae showed an undifferentiated eye and digestive tract. At 48 hah, TL was 3.31 ± 0.12 mm. Yolk and oil globule were still present. The mouth was still closed, but the Meckel's, quadrate, hyoid and hyomandibular cartilages were present. The retina was formed by 5 layers, and a thin layer of pigment epithelium was observed in the outer nuclear layer (ONL). At 70 hah, TL was 3.44 ± 0.22 mm. A remnant of oil globule was still present. The mouth and anus were open. At 93 hah, the number of cones in the ONL of the retina have increased and there was more pigment in the pigment epithelial layer. A joint between Meckel's and the quadrate cartilage and also a joint between the hyomandibular cartilage and the skull were present. The presence of live feed was detected in the digestive tract of these larvae. Based on these observations, the Pacific red snapper larvae is functional to start ingesting live feed between the 3rd and 4th day after hatching.  相似文献

Abstract.— This paper describes current techniques used at our laboratory for the controlled spawning, maturation and larval rearing of the yellowtail snapper. Juvenile yellowtail snapper were obtained from Florida and transferred to the Fisheries and Mariculture Laboratory in July 1990. Temperature and photoperiod manipulation resulted in precocial spawning in 1991, with sustained successive spawning beginning in February 1992. Average weekly spawning from 1992–1994 was 308,000 and 247,000 eggs/tank, with fertilization rates of 46.1 % and 32.5% for fish maintained in two separate spawning tanks. Observations on spawning activity from 1995 to 1996, during which a decreasing trend in egg and larval quality was observed, imply a possible problem with broodstock nutrition. Evaluation of feeding regimes during this period indicated a shift to a high (almost exclusively) usage of fresh squid and a reduction in fish and shrimp. Returning to a feeding regime of alternating feedings of fresh fish, squid. and shrimp have yielded improved spawning and egg quality. Larval rearing techniques using live and prepared feeds has resulted in an overall survival of 3% from egg to advanced juvenile. Growout of first generation (F1) juveniles yielded a marketable size (1 1b) fish in 25 mo. Spawning of F1 fish hegan in 1998. larval rearing and grow out of F2 fish are currently underway. Results demonstrate that yellowtail snapper culture is technically feasible; however. further research to develop species specific culture techniques will be required before the culture potential of this species can be accurately evaluated.  相似文献

Egg and larval quality of red snapper, Lutjanus campechanus from natural spawns of domesticated brooders and hormone-induced spawns of wild fish were compared. Eggs and larvae from natural spawns were found to be more viable in terms of fertilization, hatching and survival rate. Also, eggs from natural spawns were larger, and eggs and recently hatched larvae had larger oil-globule. These findings indicate that natural spawning of red snapper can be a sustainable and reliable source of good quality eggs.  相似文献

The effect of incubation temperature on embryonic development and yolk‐sac larva of the Pacific red snapper Lutjanus peru were evaluated by testing the effect of 26, 28 and 30°C, as this is the natural thermal interval reported during the spawning season of Pacific red snapper in the Gulf of California, Mexico. Sixteen developmental stages were observed. The incubation temperature affected the rate of development and time to hatching, being shorter at 30 than at 26°C, but no significant effect (P < 0.05) on larval length at hatching was registered. The depletion rate of yolk sac and oil globule was affected by incubation temperature particularly during the first 12 h post hatching (hph). At the end of the experiment (48 hph), significantly (P < 0.05) larger larvae were recorded at 26°C (TL = 3.22 ± 0.01 mm) than at 28° (TL = 3.01 ± 0.02 mm) and 30°C (TL = 2.97 ± 0.05 mm). Incubation of newly fertilized eggs at 26°C produces larger larvae, which may help to improve feeding efficiency and survival during first feeding.  相似文献

Interest in the commercial production of cobia Rachycentron canadum continues to rise as additional insight is gained into the hardy and fast growing nature of this species. However, research regarding the biochemical composition of captive-spawned eggs and egg and larval quality remains scarce. Such data is essential as a common bottleneck to production is a steady supply of fingerlings for grow-out. This study quantified the biochemical composition and quality of cobia eggs produced over 2 spawning seasons by broodstock on a traditional ‘trash fish’ diet which is commonly fed to tank spawning cobia. Throughout the study, batch fecundity, proportion of floating eggs and percent hatch averaged > 1 million eggs,  0.8 and 70%, respectively. Batch fecundity was significantly higher during the second spawning season as a result of the increased size of the females which weighed 18/22 kg and 22/26 kg at the beginning of each season. A positive correlation was found between the proportion of floating eggs and hatch rate for both spawning seasons. No correlations were found between egg composition (total lipid (30.0 ± 1.1% dry wt), protein (25.4 ± 2.2% dry wt), carbohydrate (2.4 ± 0.3% dry wt), vitamin E (10.2 ± 0.6 μg/g wet wt) or dry weight (119.1 ± 5.5 μg/egg)) and egg quality (proportion of floating eggs, hatch rate, larval growth and larval survival). Further, no differences in egg composition were noted between seasons or over the course of each season. The fatty acid composition of cobia eggs varied between seasons possibly due to changes in the quality of frozen feed (fish, shrimp, squid) given to the broodstock. The only correlation between the fatty acid profile and egg quality was a decrease in the proportion of floating eggs as the total amount of n-3 highly unsaturated fatty acids increased. No relationship between egg quality and amino acid content was noted with the most prominent amino acids being glutamate, leucine, alanine, proline, lysine and aspartate nor were any differences detected between spawning seasons.  相似文献

The scale‐up of spotted rose snapper, Lutjanus guttatus, larval rearing is described. Fertilized eggs (480,000) were obtained from a 1‐d harvest of a natural spawning captive broodstock acclimatized for 1 yr and 6 mo in two fiberglass tanks (18 m3). Fourteen hours after spawning, 89.6% of the collected eggs were floating, of which 96.2% were transparent with live embryos. Incubation at 25–26 C lasted 21 h, with 90.2 ± 2.1% hatching percentage of normal larvae. The percentage of viable larvae at 48 h after hatching was 79.7 ± 1.9%. Initial stocking density was 10.4 ± 1.0 larvae/L 2 days after hatching (d.p.h.). A total of 22,600 juveniles (1256 ± 170 juveniles/m3) were harvested from six 3‐m3 cylindrical fiberglass tanks. Average survival was 12.1 ± 1.1%. Final mean length and weight were 5.5 ± 0.05 cm and 2.24 ± 0.04 g, respectively. Growth expressed in total length was TL = 2.1476e0.0543t (R2 = 0.9911). Final mean biomass and condition factor were 2.8 kg/m3, 12.3% and 1.346. General length‐weight ratio was W = 0.05460 LT2.2306.  相似文献
