The prophylactic effect in parturient paresis of an intramuscular injection of 10 million i.u. vitamin D3 one week prior to the expected calving date, was investigated in 84 cows which had previously suffered from parturient paresis. Clinical observations in connection with the development of parturient paresis after injection of vitamin D3 were compared with corresponding observations made on cows within the same population at the previous calving when no vitamin D3 was administered. The incidence of clinical parturient paresis in cows given vitamin D3 was 44.3 per cent. There were no significant differences in the incidence of parturient paresis in cows injected with vitamin D3 during the periods less than 2, 2--4, 5--8 and greater than 8 days before parturition. Average plasma calcium levels (mg/100 ml) before first calcium treatment, results of treatment, and the incidence of retained placenta in vitamin D3 injected cows did not differ significantly from corresponding data for cows with clinical parturient paresis which had not been given vitamin D3 before calving. It is concluded that vitamin D3 does not have any prophylactic effect in parturient paresis in cows in Eastern Norway.  相似文献

为了阐明日粮添加过瘤胃维生素D对围产期奶牛低钙血症的调控机制,在某集约化奶牛场选取40头奶牛,随机分为4组,对照组(C)饲喂基础日粮,试验Ⅰ、Ⅱ、Ⅲ组分别于产前10d至分娩后10d在日粮中每天添加1.5、3.0、6.0g/头过瘤胃维生素D。对4组试验奶牛围产期(分别于产前10、7、3、0d和产后3、7、10d)血清维生素D3(1,25-(OH)2D3)、钙、羟脯氨酸、甲状旁腺素和降钙素水平进行检测。结果表明,围产期奶牛血钙水平分娩时达最低,产后逐渐上升,且产前7d至产后10d对照组奶牛血钙浓度显著低于试验Ⅰ组(P<0.05);分娩当天试验Ⅰ组奶牛血浆中1,25-(OH)2D3、羟脯氨酸和降钙素水平显著高于对照组(P<0.05),甲状旁腺素水平显著低于对照组(P<0.05)。提示,日粮中每天添加1.5g/头过瘤胃维生素D能调节甲状旁腺素和降钙素等激素的分泌,促进肠道对钙的吸收,动员骨钙,从而防止奶牛产后低钙血的发生,且围产期奶牛低血钙症与机体血钙稳态调节作用密切关联。  相似文献

Vitamin D3 was orally supplemented to determine the supplemental dose that improved beef tenderness in different cattle breed types. Feedlot steers (n = 142) were arranged in a 4 x 3 factorial arrangement consisting of four levels of supplemental vitamin D3 (0, 0.5, 1, and 5 million IU/steer daily) administered for eight consecutive days antemortem using three biological types (Bos indicus, Bos Taurus-Continental, and Bos Taurus-English). Warner-Bratzler shear force (WBSF) was measured at 3, 7, 10, 14, and 21 d postmortem, and trained sensory analysis was conducted at 7 d postmortem on LM, semimembranosus, gluteus medius, and supraspinatus steaks. Concentrations of vitamin D3 and the metabolites 25-hydroxyvitamin D3, and 1,25-dihydroxyvitamin D3 were determined in the LM, liver, kidney, and plasma. Biological type of cattle did not interact (P > 0.10) with vitamin D3 supplementation for sensory or tenderness traits, suggesting that feeding vitamin D3 for 8 d before slaughter affected the different biological types of cattle similarly. Supplementing steers with 0.5, 1, or 5 million IU/(steer(d) decreased (P < 0.05) LM WBSF at 7, 10, 14, and 21 d postmortem compared with controls, and vitamin D3 treatments of 0.5, 1, and 5 million IU decreased (P < 0.05) semimembranosus WBSF at 3, 7, and 14 d postmortem. In general, vitamin D3-induced improvements in WBSF were most consistent and intense in LM steaks. Sensory panel tenderness was improved (P < 0.05) by all vitamin D3 treatments in LM steaks. Sensory traits ofjuiciness, flavor, connective tissue, and off-flavor were not (P > 0.05) affected by vitamin D3 treatments. All vitamin D3 treatments decreased micro-calpain activity and increased muscle Ca concentrations (P < 0.05). Vitamin D3 concentrations were increased (P < 0.05) by supplementation in all tissues tested (liver, kidney, LM, and plasma); however, cooking steaks to 71 degrees C decreased (P < 0.05) treatment residue effects. The vitamin D metabolite 1,25-dihydroxyvitamin D3 was increased (P < 0.05) only in plasma samples as a result of the vitamin D3 treatments. These results indicate that supplementation with vitamin D3 at 0.5 million IU/steer daily for eight consecutive days before slaughter improved tenderness in steaks from different subprimal cuts by affecting muscle Ca concentrations, micro-calpain activities, and muscle proteolysis, with only a small effect on tissue residues of vitamin D3.  相似文献

Plasma levels of vitamin D3 or 25-hydroxyvitamin D3 in ewes after administration of a single massive intravenous dose of vitamin D3 (2 X 10(6) IU) or 25-hydroxy vitamin D3 (5 mg) were determined at zero, one, two, three, five, ten and 20 days postinjection. In six ewes injected with vitamin D3 conversion of vitamin D3 to 25-hydroxy vitamin D3 resulted in a six-fold increase in the plasma 25-hydroxy vitamin D3 level within one day. Elevated levels were maintained until day 10 but by day 20 a substantial decline in the plasma 25-hydroxy vitamin D3 level had occurred. Peak levels of vitamin D3 were reached one day after injection and then continuously declined until day 20. Administration of 25-hydroxy vitamin D3 increased plasma concentrations of 25-hydroxy vitamin D3 to fivefold higher levels than those observed when vitamin D3 was injected, with approximately threefold higher levels of 25-hydroxy vitamin D3 maintained for five days. On day 10 and day 20 ewes which were injected with 25-hydroxy vitamin D3 still maintained plasma levels of 25-hydroxy vitamin D3 which were twice as high as those of ewes injected with vitamin D3. In six ewes injected with vitamin D3, a sharp increase in vitamin D3 level in milk occurred within one day and more than a tenfold elevation of milk vitamin D3 concentrations were maintained for ten days. By 20 days the milk vitamin D3 level had returned to preinjection levels. These observations suggest that indirect supplementation of the suckling ruminant with vitamin D3 may be achieved through maternal injection and subsequent mammary transfer.  相似文献

OBJECTIVE: To determine the effect of vitamin E supplementation on urokinase-plasminogen activator (u-PA) receptor (u-PAR) expression by neutrophils of dairy cows. ANIMALS: 16 healthy Holstein dairy cows. PROCEDURE: 16 cows were assigned to 1 of 2 experimental groups: control (no vitamin E supplementation) and vitamin E supplementation. Supplementation of vitamin E started 4 weeks prior to and continued up to 4 weeks after parturition and included oral administration of vitamin E at 3,000 U/cow per day; these cows also received 1 injection of vitamin E (5,000 units), 1 week prior to the expected date of parturition. Blood samples were collected, and neutrophils were isolated weekly throughout the experimental period. The following variables were measured: u-PA (mRNA), total cell-associated u-PA activity, membrane-bound u-PA activity, and free unoccupied u-PA binding sites on the cell membrane of neutrophils. RESULTS: Stimulated neutrophils isolated from cows that received vitamin E supplementation had significantly higher u-PA mRNA and total cell-associated and membrane-bound u-PA activity at postpartum week 1, compared with those of stimulated neutrophils isolated from control cows.There were no differences between groups throughout the whole experimental period in u-PA binding sites of neutrophils. CONCLUSIONS AND CLINICAL RELEVANCE: The increased total cell-associated and membrane-bound u-PA activity in neutrophils isolated from cows that received vitamin E may facilitate the ability of neutrophils to extravasate and reach the mammary gland at postpartum week 1. Rapid recruitment of neutrophils is critical for proper defense of the gland.  相似文献

Pregnant ewes were injected intramuscularly with 300,000 iu of vitamin D3 in a water miscible vehicle either 10, seven or four weeks before the expected lambing date and the effects on plasma concentrations of 25-hydroxyvitamin D3 were monitored. The concentrations increased quickly and remained high at parturition but at no time were they outside the normal physiological range. The concentrations in the plasma of the newborn lambs were higher than in uninjected controls and were well correlated with the concentrations in their mothers. Dosing pregnant ewes with 300,000 iu of vitamin D3 in a rapidly available form, approximately two months before lambing, provided a safe means of increasing the vitamin D status of the ewe and the newborn lamb by preventing the seasonally low concentrations of 25-hydroxyvitamin D3.  相似文献

本试验旨在研究饲粮维生素D添加形式对种用公猪繁殖性能的影响。选取16头18月龄的约克公猪,随机分成2组,2组饲粮中分别含有50μg/kg维生素D3(VD3)和25-羟基维生素D3(25-OHD_3),每组8个重复,每个重复1头猪。试验期16周。结果表明,与VD3组相比:1~16周,25-OHD_3组公猪精子活力和每次射精的有效精子数显著提高(P0.05),而精子畸形率显著降低(P0.05);第112天,25-OHD_3组血浆钙离子(Ca2+)和雌二醇含量及芳香化酶活性显著提高(P0.05),精清25-OHD_3、Ca2+、果糖含量和酸性磷酸酶的活性显著提高(P0.05);25-OHD_3组芳香化酶、维生素D 25-羟化酶、维生素D 24-羟化酶和维生素D受体基因的表达量显著升高(P0.05)。综上所述,与同等水平VD3相比,种公猪饲粮中添加25-OHD_3能更有效增加血浆维生素D含量,从而改善精子的形态和运动能力,提高公猪的繁殖性能。  相似文献

The effect of one single oral or parenterally administered dose of 1 million IU vitamin A on the vitamin A depot in the liver and on blood plasma vitamin A concentrations was investigated in 3 individual feeding experiments with involvement of 18 and 24 calves or 24 fattening bulls. 50% of all animals in each of the 3 experiments received feed without any vitamin A through 108 or 112 or 209 days, prior to vitamin A administration, or received 10.000 IU/100 kg live weight and day. Parenteral vitamin A administration in either group yielded rise in blood plasma from 0.06--0.35 to 26.2--30.2 mumol/l, after 1 or 2 days. The maximum value measured after oral administration was 1.9 mumol/l. Most of the plasma values had returned to normal (0.6--12. mumol/l) within 14 days from administration. Oral and parenteral vitamin A doses, after 14 days, caused significant rise in vitamin A concentrations in the liver (from 15.5 to 82.5), with the increase resulting from parenteral administration (from 13.7 to 99.1) being clearly higher than that resulting from oral administration (from 17.3 to 65.9 mumol/kg fresh liver tissue). The same trends were recorded from recovery of vitamin A from the liver (26.8% after parenteral administration versus 15.0% in the wake of oral doses). Storage in and recovery from vitamin-A depleted animals were below values recorded from young cattle with sufficient vitamin A supply. These findings are likely to confirm that one single parenteral vitamin A administration was of clearly higher effectiveness, as compared to oral application.  相似文献

The effects of vitamin D (VD) deficiency on calcium (Ca) metabolism during pregnancy were evaluated in rats fed VD-repleted (VD-repleted rats) and VD-depleted (VD-depleted rats) diets. In the VD-depleted rats, the plasma concentrations of 1,25-dihydroxyvitamin D and Ca decreased severely, whereas the parathyroid hormone concentrations increased. The Ca contents of the feces of the VD-depleted rats were higher than those of the VD-repleted rats. The fetal Ca contents of the VD-repleted and VD-depleted rats increased continuously, but that of the VD-depleted rats was lower. These data reveal that VD deficiency during pregnancy induces severe hypocalcemia due to reduced intestinal absorption of Ca and elevated fetal demand for Ca.  相似文献

The effect of supplementing diets with various levels of vitamin D3 to provide 0, 0.5, 1, and 5 million IU/(steer x d) for 8 d before slaughter on the mineral content and localization of Ca in LM and muscle fragments was studied during the postmortem aging process. Twelve feedlot steers of three biological types were given access to the four levels of vitamin D for 8 d before slaughter. Differential centrifugation techniques were used to determine the concentrations of minerals relative to protein in different muscle fragments on d 3 and 21 postmortem. Electron microscopy visualization of bound Ca indicated that vitamin D3 mobilized Ca from the sarcoplasmic reticulum and transverse tubule system into the myofibrils. Bound Ca was concentrated near the Z-line at the A-band/I-band juncture within the sarcomere. Supplementing steers with 1 and 5 million IU/(steer x d) of vitamin D3 increased (P < 0.05) Ca, P, and Mg concentrations per unit of protein in the cytosol. Soluble cytosolic Ca concentrations were greater (P < 0.05) on d 21 than on d 3 postmortem only when steers were supplemented with 5 million IU/d. Concentrations of Ca, P, and Mg in isolated tissues were increased (P < 0.05) in nuclei and myofibrilar proteins by supplementing steers with 1 and 5 million IU/ (steer x d) of vitamin D3. All supplemental vitamin D3 treatments also increased (P < 0.001) Mg concentrations in the cytosol, regardless of aging treatment, and increased Mg concentrations (P < 0.04) within the mitochondria at d 3 postmortem. Thus, supplementation of feedlot steers with vitamin D3 at levels of 0.5 to 5 million IU/(steer x d) increased Ca concentrations within respiring muscle, resulting in increased bound tissue Ca concentrations. When the respiring muscle was converted to meat, the increased bound tissue Ca resulting from vitamin D3 treatment released Ca concentrations into the cytosol during aging (P < 0.05). Results of this study indicate that vitamin D3 supplementation increased total cytosolic Ca, P, and Mg concentrations in meat.  相似文献

OBJECTIVE: To determine effects of breed and supplemental administration of vitamin E and selenium (Se) during late gestation on circulating concentrations of these micronutrients in periparturient Jerseys and Holsteins. DESIGN: Randomized controlled clinical study. ANIMALS: 16 Jersey and 36 Holstein cows. PROCEDURE: Cows were allotted to blocks on the basis of breed and expected parturition date. Cows within blocks were randomly assigned to be given vitamin E or Se parenterally 3 to 4 weeks prior to anticipated parturition in a 2 x 2 factorial design. RESULTS: Results of ANOVA indicated Jerseys had higher blood concentrations of Se and lower serum concentrations of vitamin E than Holsteins at the end of lactation. Jerseys had higher blood concentrations of Se than Holsteins 3 to 4 weeks prior to parturition and at parturition. Selenium administration increased blood concentrations of Se at parturition. Administration of nutrients did not affect serum concentrations of vitamin E at parturition or 2 to 3 weeks after parturition or blood concentrations of Se 2 to 3 weeks after parturition. CONCLUSIONS AND CLINICAL RELEVANCE: Jerseys and Holsteins consuming rations of comparable Se content differ in blood concentrations of Se during the nonlactating period, suggesting breed-related differences in Se metabolism during late lactation and the nonlactating period. Parenteral administration of Se 3 to 4 weeks prior to anticipated parturition increased blood concentrations of Se at parturition; however, Se concentrations of both groups at parturition were considered within the reference range for clinically normal cattle.  相似文献

Serial vitamin D3 (D3) and 25-hydroxyvitamin D3 (25 OH D3) concentrations of plasma were measured in confined, shorn sheep that had either been supplemented with vitamin D3 (50 micrograms/d) or exposed daily to ultraviolet irradiation (UVI). In the sheep administered D3 orally, plasma D3 increased continuously until d 35. This was followed by small fluctuations of the plasma D3 concentrations until a plateau was reached after 56 d of supplementation (.94 ng/ml plasma). Plasma 25 OH D3 concentrations increased continuously and plateaued between d 65 to 75 at about 21 ng/ml plasma. In the UVI sheep, plasma D3 and 25 OH D3 concentrations increased continuously for the first 49 d, then plateaued at 2.03 ng D3 and 29.6 ng 25 OH D3/ml. When a plateau was reached in plasma 25 OH D3 concentrations in both treatment groups, a 3H-labeled tracer dose of 25 OH D3 was given i.v., and disappearance of the 3H-labeled 25 OH D3 was followed. The UVI group had a faster decline in specific activities during the first exponential phase but a slower decline during the prolonged terminal elimination phase. These differences are reflected in the intercompartmental transfer rates. Our data indicate that UVI is as effective as oral vitamin D3 supplementation for improving vitamin D status of confined sheep.  相似文献

Because of the Ca dependency of the calpains, oral supplementation of vitamin D3 (VITD) can increase the Ca content of muscle to activate the calpains and improve tenderness. Feedlot steers (n = 142) were arranged in a 4 x 3 factorial arrangement consisting of four levels of VITD (0, 0.5, 1, and 5 million IU/[steer x d]) for eight consecutive days antemortem using three biological types (Bos indicus, Bos taurus-Continental, and Bos taurus-English). Feedlot performance factors of ADG, DMI, and G:F were measured, and carcass quality, yield, and color data were collected. Plasma Ca and P concentrations were measured during d 4 to 6 of supplementation and at exsanguination, and carcass pH and temperature were measured in the LM at 3 and 24 h postmortem. Vitamin D3 treatment at 5 million IU/(steer x d) decreased ADG (P < 0.05) over the supplementation and feed intake for the last 2 d of feeding compared with untreated control steers. Likewise, G:F was decreased (P = 0.03) in steers supplemented with 5 million IU/d compared with controls. Overall, there was a linear decrease (P < 0.01) in ADG and G:F as a result of VITD supplementation. Plasma concentrations of Ca and P were increased (P < 0.05) by VITD concentrations of 1 and 5 million IU/(steer x d). All VITD treatments increased (P < 0.05) LM temperature at 3 h postmortem and pH at 24 h postmortem. Vitamin D3 treatments did not affect (P = 0.07) any other carcass measurements, including USDA yield and quality grade; thus, any improvements in meat tenderness as a result of VITD supplementation can be made without adversely affecting economically important carcass factors. Biological type of cattle did not interact with VITD treatment for any carcass or feedlot performance trait. Although feeding 5 million IU/(steer x d) of VITD for eight consecutive days had negative effects on performance, supplementing VITD at 0.5 million IU/ (steer x d) did not significantly alter feedlot performance.  相似文献

Eight pregnant heifers (primiparous cows) and seven pregnant cows in their second, third and forth pregnancies (multiparous cows) were assigned to two groups and fed either a low calcium (Ca) diet (Ca, 0.46%) or a high Ca diet (Ca, 0.86%) ad libitum from 3 weeks before the expected calving date to 3 days after parturition. All cows were examined for a change in dry matter intake (DMI), plasma minerals and bone turnover around parturition. The dietary Ca level did not affect the DMI in both primiparous and multiparous cows. The DMI of primiparous cows was significantly lower than that of multiparous cows (P < 0.05) in both the low and high dietary Ca groups. The dietary Ca level did not affect the concentrations of plasma Ca, phosphorus, magnesium and parathyroid hormone throughout the experimental period. Plasma phosphorus in primiparous cows was significantly higher (P < 0.05) than that of multiparous cows around parturition. Plasma Ca and magnesium tended to be higher (P < 0.10) in primiparous cows. The dietary Ca level did not affect the plasma osteocalcin (OC) level measured as bone formation or the urinary deoxypyridinoline (DPD) excretion measured as bone resorption before parturition in both primiparous and multiparous cows. After parturition, the plasma OC level was lower than it was before parturition in multiparous cows fed a low Ca diet, and in primiparous cows. There were no significant differences in urinary DPD excretion between each group before and after parturition. Both the plasma OC level and urinary DPD excretion of primiparous cows were significantly higher than those of multiparous cows in both the low and high dietary Ca groups.  相似文献

Feedlot steers (n = 36) from three biological types (Bos indicus, Bos taurus-Continental, and Bos taurus-English) were used to determine the Ca, P, and vitamin D3 status of feedlot cattle. The USDA yield and quality grade traits were measured at slaughter, and the concentrations of vitamin D3 (VITD) and the metabolites 25-hydroxyvitamin D3 (25-OH D) and 1,25-dihydroxyvitamin D3 (1,25-(OH)2 D) were determined in LM, liver, kidney, and plasma. Plasma and muscle Ca and P concentrations also were determined. Biological type of cattle affected a number of carcass traits. Carcasses from Bos taurus-English cattle had more marbling, resulting in higher quality grades (P < 0.05). Carcasses from Bos taurus-Continental cattle had lower calculated yield grades (P < 0.05) than did carcasses from cattle in the other biological types. In general, differences in carcass traits resulting from biological type were consistent with other reports. Plasma and LM Ca and P concentrations were not affected (P = 0.06) by biological type of cattle, indicating that Ca and P homeostasis is a conserved trait across the different types of cattle. Plasma VITD and 25-OH D concentrations were not affected (P = 0.41) by biological type, whereas plasma 1,25-(OH)2 D concentration was lower (P < 0.05) in Bos taurus-English cattle than in Bos taurus-Continental and Bos indicus cattle. Liver VITD and 25-OH D were not affected by biological type (P = 0.76), but liver 1,25-(OH)2 D concentration was greater (P < 0.05) in Bos indicus cattle than in Bos taurus-Continental cattle. Kidney vitamin D metabolite concentrations were not affected by biological type of cattle (P = 0.21). Muscle VITD concentration was greater (P < 0.05) in Bos taurus-English cattle than in the other two biological types, and muscle 25-OH D concentrations were greater (P < 0.05) in Bos taurus-English cattle than in Bos indicus cattle. Muscle 1,25-(OH)2 D concentration was less (P < 0.05) in the Bos taurus-Continental cattle than in the other two biological types. Cooking eliminated vitamin D metabolite differences among the biological types. Our results suggest that Bos indicus cattle had greater 1,25-(OH)2 D (the biologically active form) in tissues, and greater 1,25-(OH)2 D plasma concentrations than Bos taurus cattle. Thus, the need for VITD supplementation and optimal levels of Ca and P in feedlot diets might differ between Bos indicus and Bos taurus cattle.  相似文献

Vitamin D3 ranging in total amount from 10(5) iu (2.5 mg) to 9 X 10(5) iu (22.5 mg) was given intramuscularly either as a single injection or in three aliquots at three-weekly intervals to housed nonpregnant ewes on a vitamin D deficient diet. The effects on plasma concentrations of 25-hydroxyvitamin D3 (25-OHD3), the major metabolite of vitamin D, were monitored. The increase in plasma 25-OHD3 showed a large variation between animals and was related to, but not proportional to, the dose. None of the treatments produced 25-OHD3 concentrations greater than those in grazing sheep in summer. Repeated dosing provided for more efficient use of the injected vitamin D3.  相似文献

Three experiments were conducted to determine whether feeding 25-hydroxyvitamin D3 (25-OH D3) or 1,25-dihydroxyvitamin D3 (1,25-(OH)2 D3) improves the tenderness of longissimus dorsi (LD), semimembranosus (SM), and infraspinatus (IF) muscles similar to supplemental vitamin D3 without leaving residual vitamin D3 and its metabolites in muscle. In the first two experiments, 24 crossbred steers were used to determine the effects of different oral amounts of 1,25-(OH)2 D3 (Exp. 1; n = 12) and 25-OH D3 (Exp. 2; n = 12) on plasma Ca2+ concentrations. In the third experiment, crossbred steers were allotted randomly to one of four treatments: 1) control placebo (n = 7); 2) 5 x 10(6) IU of vitamin D3/d (n = 9) for 9 d and harvested 2 d after last treatment; 3) single, 125-mg dose of 25-OH D3 (n = 8) 4 d before harvest; or 4) single, 500-microg dose of 1,25-(OH)2 D3 (n = 9) 3 d before harvest. The LD and SM steaks from each animal were aged for 8, 14, or 21 d, whereas steaks from the IF were aged for 14 or 21 d. All steaks were analyzed for tenderness by Warner-Bratzler shear force and for troponin-T degradation by Western blot analysis. Supplementing steers with vitamin D3 increased (P < 0.01) the concentration of vitamin D3 and 25-OH D3 in all muscles sampled. Feeding steers 25-OH D3 increased (P < 0.05) the concentration of 25-OH D3 in meat, but to an amount less than half that of cattle treated with vitamin D3. Supplemental 1,25-(OH)2 D3 did not affect (P < 0.10) shear force values; however, there was a trend (P < 0.10) for supplemental vitamin D3 and 25-OH D3 to produce LD steaks with lower shear values after 8 and 14 d of aging, and lower (P < 0.10) shear force values for the SM aged for 21 d. Analysis of Western blots indicated that LD steaks from cattle supplemented with vitamin D3 and 25-OH D3 had greater (P < 0.05) troponin-T degradation. Antemortem supplementation of 25-OH D3 seems to increase postmortem proteolysis and tenderness in the LD and SM without depositing large concentrations of residual vitamin D3 and its metabolite 25-OH D3.  相似文献
