The transfer factor-values feedstuff/meat so as the value of biological half life time for caesium are not constant but are depending on external and internal conditions. The most important external condition is the level of feedstuff contamination which reduces proportional the transfer factor's value of 0.5 +/- 0.1 asymptotically to 0.3 +/- 0.1. These values are then decreasing till to two thirds by the increasing part of uncontaminated feedstuff in the daily diet. The internal conditions, namely type of gastrointestinal tract, metabolic individuality and actual animal health, may cause a variability of transfer factor's values up to +/- 35% of the mean resulting from external conditions. The value of biological half life time of caesium may be considered as a constant for each respective tissue or organ in balanced conditions, but the changes of external conditions, especially the beginning of uncontaminated feeding, results in immediate gradually shortening of this half life time till to 50-30% of its original value.  相似文献

A radioactive contamination monitoring system of the Czechoslovak Federal Ministry of Agriculture and Food is described, including measuring and calibration methods and the monitoring results. Cesium contamination higher than the international derived intervention level was found only in game. The reasons were as follows: uncultivated green fields, forest herbage and needle-leaves are sufficient sources of game contamination; there exist evident differences between continuous ingestion of contaminated or mixed feed with respect to the cesium contamination of tissues.  相似文献

Broiler meat is regarded as the most common source of Campylobacter infection and risk management measures are required to reduce broiler meat contamination. Among the quantitative risk assessments for Campylobacter in broiler meat, evaluation of the poultry processing stage is particularly important for predicting the contamination level of broiler meat and the effects of control measures. In this study, we built a simulation model for cross‐contamination during poultry processing focusing on Campylobacter contamination at the individual carcass level. Using this model, we examined changes in the prevalence of contaminated carcasses and the number of Campylobacter per carcass after processing. As a result, it was found that the prevalence and number of Campylobacter after processing were largely influenced by the number of Campylobacter on the contaminated carcasses before processing. In the baseline model, where it was assumed that the mean number of Campylobacter on contaminated carcasses before processing was 4.7 log10 cfu per carcass, the prevalence after processing was less than that before processing. Although the median value of Campylobacter on contaminated carcasses was reduced after processing, the distributions after processing became wider and the upper limit of the 95% credible interval of Campylobacter on contaminated carcasses remained elevated. The individual‐based simulation model can trace individual level changes considering discrete interactions, while models tracing mean values cannot handle these interactions in detail. The individual‐based approach is considered useful for modelling cross‐contamination among individual carcasses during poultry processing.  相似文献

The effects of pickle-curing on a decrease in the cesium-nuclides amount in red-deer meat were investigated. This meat was contaminated through ingestion of natural feed in seasons after the fall-out from Chernobyl accident. Two pickles were studied: vinegar solution and the same one with added vegetables and spices. The pickle curing was carried out at temperatures of 5 degrees C and 11 degrees C after cutting the meat into little pieces of about 1.5 cm. Adecrease in cesium activity was measured by means of gamma-spectrometry at given time intervals after the process was started. It was proved that the efficiency of this process is mostly influenced by replacement of the pickle solution and by the duration of the process. A decrease in the cesium activity of about 80% was achieved without the change of the pickle after seven hours of curing. But after the same time and one pickle solution change, the activity decrease reached more than 90%. No important influence of added vegetables and spices into the pickle nor of the temperature between 5 degrees C and 11 degrees C was observed.  相似文献

A vestibulocerebellar disorder was observed in 2- to 4-month-old swine after consumption of broken rice contaminated with 13% Aeschynomene indica seeds. Affected animals recovered in 2-14 days after removal of the contaminated food. To reproduce the syndrome, 10 pigs were fed 13% A. indica seeds in commercial pig ration or rice. They showed clinical signs similar to those observed in the spontaneous cases, 1 hour to 6 days after ingestion. Three pigs recovered after the withdrawal of the contaminated food. The others were killed. Two pigs that were fed for 25 days with food containing 3% and 6% seeds and two control pigs did not show clinical signs. Histologic lesions were characterized by symmetric focal degeneration in the cerebellar and vestibular nuclei. The initial lesion, observed 24 hours after the first ingestion, was characterized by vacuolation of the neuropil. Subsequently, there was progressive loss of parenchyma, vascular reaction, a few small spheroids, astrocytosis, and accumulation of gitter cells. The brains from four pigs were perfused with a buffered solution of glutaraldehyde-paraformaldehyde 12-48 hours after they started to ingest the seeds. No ultrastructural lesions were observed in the cerebellar nuclei after 12 hours of seed consumption. At 24 hours, endothelial cells and pericytes were separated by empty spaces from astrocyte foot processes, suggesting perivascular edema. The astrocytes were enlarged, consistent with intracellular fluid accumulation. These results suggest that A. indica seeds are toxic, causing functional derangement of the vestibulocerebellar system, followed by alterations in the microvasculature in the cerebellar and vestibular nuclei.  相似文献

Radioactivity inspection of slaughtered cattle is generally conducted using a portion of the neck muscle; however, there is limited information about the distribution of radioactive cesium in cattle. In this study, therefore, we measured not only radioactive cesium but also stable cesium in various tissues of 19 cattle that had been kept in the area highly contaminated by the Fukushima nuclear accident. Skeletal muscles showed approximately 1.5‐3.0 times higher concentration of radioactive cesium than internal organs. Radioactive cesium concentration in the tenderloin and top round was about 1.2 times as high as that in the neck muscle. The kidney showed the highest concentration of radioactive cesium among internal organs, whereas the liver was lowest. Radioactive cesium concentration in the blood was about 8% of that in the neck muscle. Characteristics of stable cesium distribution were almost the same as those of radioactive cesium. Correlation coefficient between radioactive cesium and stable cesium in tissues of individual cattle was 0.981 ± 0.012. When a suspicious level near 100 Bq/kg is detected in the neck of slaughtered cattle, re‐inspection should be conducted using a different region of muscle, for example top round, to prevent marketing of beef that violates the Food Sanitation Act.  相似文献

Growing pigs were fed freshly milled ergot sclerotia (Claviceps purpurea Fr (Tul)) from North American wheat containing 0-31 per cent of total alkaloids of which 30 per cent was ergotoxine, 30 per cent ergosine, 20 per cent ergotamine and 20 per cent other ergot alkaloids. Pigs were induced to ingest a diet heavily contaminated with ergot (10 per cent w/w). The presence of ergot reduced feed intake and growth of the pigs, but there was no evidence of peripheral necrosis, of internal or external haemorrhage, or of changes in haemotological characteristics. Balance experiments in which the diet was contaminated with 4 per cent (w/w) of ergot showed that 90 per cent of the alkaloids were absorbed by the pigs, but none could be detected in tissues or urine. The ingestion of ergot did not affect the digestibility of the diet, but the urinary excretion of nitrogen was increased and the retention of nitrogen reduced. The presence of ricinoleic acid was shown to be a useful diagnostic feature of ergot contamination of diets, digesta and faeces; it was not found in body tissues.  相似文献

On farms where rats and mice are present, unprotected livestock feed may become contaminated with rodent faeces, thereby creating a possible source of infection for cattle and sheep. Livestock unable to avoid contaminated feed may choose to eat it entirely, reject it completely or attempt to reject faeces selectively while consuming some of the feed. Two experiments were conducted to investigate which of these three responses were demonstrated by livestock. Ten cattle and ten sheep were presented individually with three repeats of ten feed treatments. Treatments were based on two feed types (meal and pelleted compound), with three levels of contamination (none, 'low' and 'high'), from one of two rodent species (rat and mouse).Avoidance behaviour was greater for feed contaminated with rat faeces compared to feed contaminated with mouse faeces. At low levels of rat contamination there was evidence that livestock actively rejected faeces whilst consuming feed. At higher levels of contamination animals rejected faeces and feed. Livestock could not actively discriminate against mouse faeces and thus rejection of feed was used to avoid faeces ingestion.Despite rejection of contaminated feed and some discrimination against faeces, significant numbers of rodent faeces were ingested illustrating that livestock feeding behaviour cannot prevent ingestion of rodent faeces. Feed contaminated with rodent faeces therefore poses a significant risk of disease infection to livestock.  相似文献

Qualitative and quantitative contamination of ready-to-eat food-stuffs with the pathogen Listeria monocytogenes was studied in 1586 samples collected from 103 supermarkets (n = 946) and 61 households (n = 640) in Vienna, Austria. Seventeen groups of ready-to-eat foods were classified into three risk categories for contamination (CP1-CP3). Three to four samples were randomly collected at the retail level from each CP. Regarding the households, the sampling procedure was started with food items of CP1, and if not available, was continued with sampling of food items of CP2 and finally of CP3. Additionally, 184 environmental samples (swabs from the kitchen area, dust samples from the vacuum cleaner) and faecal samples (household members and pet animals) were included. One-hundred and twenty-four (13.1%) and 45 (4.8%) samples out of 946 food samples collected from food retailers tested positive for Listeria spp. and L. monocytogenes, respectively, with five smoked fish samples exceeding the tolerated limit of 100 CFU/g food. Food-stuffs associated with the highest risk of contamination were twice as frequently contaminated with L. monocytogenes as food-stuffs associated with a medium risk of contamination. Products showing the highest contamination rate were fish and seafood (19.4%), followed by raw meat sausages (6.3%), soft cheese (5.5%) and cooked meat products/patés (4.5%). The overall contamination rate of foods collected at the household level was more than two times lower. Only 5.6% and 1.7% of 640 food-stuffs analysed tested positive for Listeria spp. and L. monocytogenes, respectively. However, CP1 foods were rarely collected. Pulsed-field gel electrophoresis (PFGE) typing of the collected L. monocytogenes isolates revealed a high degree of diversity between the isolates, with some exceptions. PFGE typing of isolates harvested from green-veined cheese revealed a match among strains, although the manufacturer seemed to be distinguishable. Typing of household strains revealed an epidemiological link within one family. In this case, food-stuffs and the kitchen environment were contaminated by an indistinguishable isolate. In addition, the same isolate was collected from a pooled faecal sample of the household members suggesting that consumption of even low contaminated food items (<100 CFU/g) results in Listeria shedding after the passage through the gut.  相似文献

1. Experiments were carried out in two poultry slaughtering plants to estimate cross‐contamination occurring during the scalding and plucking of broilers.

2. To simulate the external (dust and feather) and internal (intestinal) contamination of broilers the carcasses were artificially contaminated with a strain of Escherichia coli K12.

3. Cross‐contamination occurred at both stages in the processing when the carcasses had been contaminated externally; when the broilers had been contaminated internally slight cross‐contamination occurred only during plucking.

4. Broilers which were contaminated externally before scalding resulted in more numerous carcasses that were contaminated after the whole slaughtering procedure than those contaminated internally.

5. In one processing plant there were fewer contaminated carcasses after cooling than after plucking, while in the other plant no differences were found in the number of positive carcasses after these two stages in processing.  相似文献

Pork has been traditionally considered an important source of human Toxoplasma gondii infection. Pigs, as other meat‐producing animals, can become infected by the ingestion of oocysts that are shed in the environment by infected cats or by the consumption of cysts present in tissues of infected mammals, commonly small rodents. The objective of this study was to investigate the level of T. gondii infection in swine from southern Chile that can be associated with the ingestion of oocysts and therefore exposure to a contaminated environment. A total of 340 serum samples from swine were obtained from three commercial slaughterhouses located in the Araucania and Los Rios Regions from southern Chile. Study animals originated from local farms, mainly small commercial producers, and the meat is sold locally. Overall, 8.8% (30/340) of the samples showed Tgondii‐specific IgG antibodies. Of these sero‐positive animals, 80% (24/30) were also positive for antibodies specific against the oocyst stage of the parasite, indicating that animals had been infected recently by the ingestion of oocysts. The observed results suggest a high level of environmental contamination with oocysts on the farms of origin. In addition to the food safety problems associated with the consumption of meat from infected animals, the high level of environmental contamination on the farm represents a direct health risk for people living and/or working on these farms. Consequently, there is a need to develop on‐farm monitoring programmes and identify risk reduction strategies (food storage, water purification, rodent control and contact with cats) that are appropriate and cost‐effective for informal and outdoor type of farms.  相似文献

Background: Peritoneal D-Dimer concentration can be determined to assess peritoneal fibrinolysis activity in horses with gastrointestinal disorders. However, blood contamination of peritoneal fluid may occur during collection and could alter peritoneal D-Dimer concentration.
Hypothesis/Objectives: Blood contamination in peritoneal fluid does not affect interpretation of peritoneal D-Dimer concentration in horses with colic.
Animals: Thirty-four horses with colic and 4 healthy horses.
Methods: Peritoneal fluid and blood samples were simultaneously collected upon admission. Then, peritoneal fluid was serially contaminated with the horse's own blood; final contaminations corresponded to 1, 5, 10, and 20% of blood in peritoneal fluid. D-Dimer concentration was determined in blood, peritoneal fluid, and contaminated peritoneal fluid samples. Data were analyzed using a longitudinal linear model and a generalized estimating equations analysis to assess the quantitative and qualitative variations of the effect of blood contamination on peritoneal D-Dimer concentration.
Results: Peritoneal D-Dimer concentration was only quantitatively affected when peritoneal fluid was contaminated at 20% of blood. However, when using increasing cut-off values of peritoneal D-Dimer concentration (100, 2,000, 8,000, and 16,000 ng/mL), this effect disappeared at the highest cut-off values (8,000 and 16,000 ng/mL). When peritoneal fluid contamination was grouped as "minimally contaminated" (≤1% of blood) and "highly contaminated" (≥5% of blood), no significant differences on D-Dimer concentration between both groups at each cut-off value were observed.
Conclusions and Clinical Importance: Although quantitative results of peritoneal D-Dimer concentration could be affected by high levels of blood contamination (≥20%), interpretation of increased peritoneal fibrinolytic activity was not significantly affected.  相似文献

Ochratoxin A (OTA) is a nephrotoxic and carcinogenic mycotoxin, a secondary metabolite produced by mould fungi belonging to several Aspergillus and Penicillium species. It is formed during the storage of cereal grains and other plant-derived products. OTA ingested by humans and animals with the food or feed may exert deleterious effects on health. The purpose of this study was to investigate the ochratoxin contamination of the most important potential sources of OTA. The OTA content of cereal samples for human consumption (36 baking wheat, 16 wheat flour and 6 maize coarse meal samples) and feed grain samples (30 feeding wheat, 32 feeding maize and 20 feeding barley samples) collected in the mid-phase or at the end of the storage period and of 50 commercial coffee samples was determined. The analyses were performed by immunoaffinity column--high-performance liquid chromatography (IAC-HPLC). The limit of detection of the method was 0.1 ng/g. Of the wheat samples intended for human consumption, 8.3% contained OTA at 0.29 ng/g on the average (OTA ranges: 0.12-0.5 ng/g; Table 2). The OTA contamination of wheat flour and maize meal samples for human consumption was similar to that of the baking wheat samples. OTA contamination was found in 26.7% of the feeding wheat, 15.6% of the feeding maize and 35% of the feeding barley samples. The average values and the ranges of OTA levels found in the above samples were 12.2 and 0.3-62.8 ng/g, 4.9 and 1.9-8.3 ng/g, and 72 and 0.14-212 ng/g, respectively (Table 3). Sixty-six percent of the coffee samples were contaminated with OA (average level: 0.57 ng/g, ranges: 0.17-1.3 ng/g; Table 4). OTA contamination of baking wheat samples was found to be relatively low, presumably as a result of the favourable weather at harvest and the optimal storage conditions. Calculations made on the basis of the obtained results show that the daily OTA intake of an adult human from edible cereals is only 6.7 ng, while the amount taken up with coffee is 4.1 ng daily. The high prevalence and high levels of OTA contamination in feed grains can be explained by the unfavourable storage conditions, and this finding suggests that OA-related health problems may arise in animals, and that foods of animal origin may be contaminated with this mycotoxin.  相似文献

The accident at the Fukushima Daiichi Nuclear Power Plant (FNPP) released a large amount of radioactive substances into the environment. Furthermore, beef contaminated with radioactive cesium above the 500 Bq/kg safety standard was circulated in the food chain in 2011. Japanese consumers remain concerned about the safety of radioactively contaminated food. In our previous study, we detected a linear correlation between radioactive cesium (137Cs) activity in blood and muscle around 500 to 2500 Bq/kg in cattle. However, it was unclear whether the correlation was maintained at a lower radioactivity close to the current safety standard of 100 Bq/kg. In this study, we evaluated 17 cattle in the FNPP evacuation zone that had a 137Cs blood level less than 10 Bq/kg. The results showed a linear correlation between blood 137Cs and muscle 137Cs (Y = 28.0X, R2 = 0.590) at low radioactivity concentration, indicating that cesium radioactivity in the muscle can be estimated from blood radioactivity. This technique would be useful in detecting high‐risk cattle before they enter the market, and will contribute to food safety.  相似文献

Observation was made of the influence of natural zeolite (clinoptilolite) supplement in food on 134Cs excretion and distribution after oral internal contamination of laboratory brown rats. After diet administration with 2.5, 5, and 10% zeolite supplement the 134Cs elimination in droppings increased and the radionuclide deposition in liver, kidneys and femoral musculature decreased. The zeolite decontamination effects were observed with preventive administration, as well as with sorbent administration from the 24th hour after a single internal contamination.  相似文献

ABSTRACT: Cyprinid herpesvirus 3 (CyHV-3), also known as Koi herpesvirus, is the etiological agent of a mortal disease in common and koi carp. Recently, we investigated the entry of CyHV-3 in carp using bioluminescence imaging and a CyHV-3 recombinant strain expressing luciferase (LUC). We demonstrated that the skin is the major portal of entry after inoculation of carp by immersion in water containing CyHV-3. While this model of infection mimics some natural conditions in which infection takes place, other epidemiological conditions could favour entry of virus through the digestive tract. Here, we investigated whether ingestion of infectious materials mediates CyHV-3 entry through the digestive tract. Carp were fed with materials contaminated with the CyHV-3 LUC recombinant (oral contamination) or immersed in water containing the virus (contamination by immersion). Bioluminescence imaging analyses performed at different times post-infection led to the following observations: (i) the pharyngeal periodontal mucosa is the major portal of entry after oral contamination, while the skin is the major portal of entry after contamination by immersion. (ii) Both modes of inoculation led to the spreading of the infection to the various organs tested. However, the timing and the sequence in which some of the organs turned positive were different between the two modes of inoculation. Finally, we compared the disease induced by the two inoculation modes. They led to comparable clinical signs and mortality rate. The results of the present study suggest that, based on epidemiological conditions, CyHV-3 can enter carp either by skin or periodontal pharyngeal mucosal infection.  相似文献

OBJECTIVE: To evaluate the effectiveness of four methods of povidone-iodine preparation on skin bacterial flora of arthrocentesis sites, in horses, with and without evidence of skin contamination. STUDY DESIGN: Prospective randomized study. ANIMALS: Twenty-four adult horses. METHODS: Horses were assigned to either the clean or contaminated group based on housing environment and visual evidence of contamination. Using a moist sterile swab, microbial culture samples were obtained from the skin over the distal interphalangeal joints immediately before and after preparation. Each site was aseptically prepared with 1 of 4 povidone-iodine techniques: 10-minutes scrub, 5-minutes scrub, three 30-second scrubs, or commercial one-step iodophor surgical solution. Colony forming units (CFUs) were determined for each sample, 24 hours after inoculation, on blood agar plates. RESULTS: Mean (+/-SD) pre-scrub CFUs/mL was significantly higher in the contaminated group (9588.33+/-1223.65) compared with the clean group (4489.00+/-3842.03) (P<.01). After preparation of the arthrocentesis sites, there were no significant differences in post-scrub CFUs/mL among the 10 minutes (mean clean, 46.00+/-64.36; mean contaminated, 28.67+/-18.04), 5 minutes (mean clean, 84.17+/-109.80; mean contaminated, 40.33+/-44.52), three 30 seconds povidone-iodine scrubs (mean clean, 95.50+/-172.29; mean contaminated, 46.67+/-56.94), or application of a commercial one-step iodophor surgical solution (mean clean, 102.17+/-161.78; mean contaminated 117.67+/-143.78); or between the clean (81.96+/-131.69) and contaminated groups (58.33+/-85.90) (P<.01). CONCLUSIONS: Preparation of the distal interphalangeal joint arthrocentesis site with each of these techniques significantly reduces the bacterial flora to a similar level for arthrocentesis in horses with and without evidence of skin contamination. Clinical Relevance- Aseptic preparation of the skin over the distal interphalangeal joint may be accomplished with any of these techniques.  相似文献

Dioxins represent a group of undesired substances that may contaminate feedstuffs. Due to their persistency in the environment it can be expected that feed contamination by dioxins will occur in the future, too. Therefore efforts in agriculture and in the feed industry have to be focussed on measures that minimize the dioxin contamination of feedstuffs, especially under the aspects of food safety. Several dioxin scandals in the past have contributed to a higher awareness in feed production as well as on the side of food consumers and administration resulting in changes of feed legislation (implementation of maximum levels of dioxins in feed and food). Some of the dioxin scandals enlarged the knowledge concerning the potential ways dioxins entering the food chain (for example dioxin contamination during the drying process of grass meal, recycling of contaminated oils, use of contaminated kaolinit a.s.o.). In difference to those accidents in feed producing agriculture there is a main influence of potential soil contamination on the dioxin contents in feedstuffs (like roughage). Producing roughage (green fodder, silage, hay) on areas with grass land by grazing animals can result in corresponding consumption of soil that contaminates the green fodder. Intending a lowest feed contamination by dioxins implies that the soil contamination is known. Depending on the rate of soil contamination farmers should decide on the kind of feed production and on the techniques of harvesting the fodder. For example it is possible to produce maize (whole plant in high cut technique) without higher dioxin contents in maize silage, on the other hand it can not be recommended to use contaminated fields for growing beets or by grazing ruminants, pigs or poultry (outdoor systems). In general during the harvesting process technical measures have to be chosen that reduce/minimize the soil contamination because the dioxin concentration in the soil of exposed areas is much more higher than the dioxin concentration within the feedstuffs. The high costs of dioxin analyses limit an implementation of a monitoring program including samples of soil and feedstuffs of each feed producing unit or individual farms. Therefore it is necessary to focus the monitoring efforts on areas and samples that let expect a higher risk of dioxin contamination. New screening tests should be developed and implemented that allow a higher frequency and density of control measures concerning dioxin. In the case of suspicious findings only those samples could be analysed by the classic chemical procedure (time consuming, high costs). Especially in cases of food contamination (for example detected in monitoring programs of the food industry) it is necessary to find out the way and cause, not neglecting the potential role of the contaminated soil. Last but not least it is recommended to inform the public continuously about levels of dioxins that are actually found in food as well as in feed (and soils), especially on behalf of the consumers confidence in food and feed control and its related administration. Continuous information will increase the consumers understanding that there is (and will be for further years) a background contamination of feed and food in general (that decreased markedly in the last decades), and that cases of detected higher dioxin levels are exceptional findings that result immediately in efforts of the food and feed industry to obstruct the way that enabled an entering to the food chain.  相似文献

BackgroundDue to multiple similarities in the structure and physiology of human and pig skin, the pig model is extremely useful for biological drug testing after subcutaneous administration. Knowledge of the differences between subcutaneous injection sites could have a significant impact on the absorption phase and pharmacokinetic profiles of biological drugs.ObjectivesThis study aimed to analyze the impact of administration site on pharmacokinetics and selected biochemical and hematological parameters after a single subcutaneous administration of ustekinumab in pigs. Drug concentrations in blood plasma were analyzed by enzyme-linked immunosorbent assay. Pharmacokinetic analyses were performed based on raw data using Phoenix WinNonlin 8.1 software and ThothPro v 4.1.MethodsThe study included 12 healthy, female, large white piglets. Each group received a single dose of ustekinumab given as a 1 mg/kg subcutaneous injection into the internal part of the inguinal fold or the external part of the inguinal fold.ResultsThe differences in absorption rate between the internal and external parts of the inguinal fold were not significant. However, the time of maximal concentration, clearance, area under the curve calculated between zero and mean residence time and mean residence time between groups were substantially different (p > 0.05).
