After the disastrous incident of the Fukushima Daiichi Nuclear Power Station, various agricultural, livestock and fishery products have been inspected for radioactive contamination with cesium in Japan. In this study, radioactive cesium was measured in various edible parts of cattle to verify the current inspection method for cattle, in which the neck tissues are generally used as samples. Radioactive cesium concentration in the short plate, diaphragm, liver, lung, omasum, abomasum and small intestine were lower and sirloin, tenderloin, top round meat and tongue were higher than that in the neck. There was no significant difference between the other organs (heart, kidney, lumen and reticulum) and the neck. Ninety‐five percent upper tolerance limits of the relative concentration to the neck were 1.88 for sirloin, 1.74 for tenderloin, 1.87 for top round and 1.45 for tongue. These results suggest that a safety factor of 2 is recommended for the radioactivity inspection of cattle to prevent a marketing of meat with higher cesium than the legal limit. Re‐inspection should be conducted using another part of muscle, for example, top round, when suspicious levels of 50–100 Bq/kg are detected in the neck.  相似文献

To contribute to the reconstruction of livestock industry in Fukushima, radioactive cesium (134Cs, 137Cs) and potassium (4°K) were measured in various tissues of beef cattle living in an area where the evacuation order will be lifted in the near future. Radioactive cesium concentration was less than 100 Bq/kg in most of the samples. Skeletal muscles and kidney had the highest concentrations of radioactive cesium, whereas the liver was lowest among samples, excepting blood. Radioactive cesium concentration in the sirloin, tenderloin and top round was significantly higher than that in the neck muscle. Radioactive cesium concentration in the urine was not correlated with that in the blood, but the relationship became proportional when corrected with urinary 4°K. Distribution of 4°K was similar but not identical to that of radioactive cesium. These results suggest that it will be possible to resume livestock production in this area after the decontamination measures are completed and the evacuation order is lifted. Contamination level of living cattle can be estimated not only by blood samples but also by urine samples. If 50–100 Bq/kg of cesium is detected in the neck muscle at a slaughterhouse, the cattle should be reinspected using a sample from different muscles.  相似文献

The 2011 earthquake severely damaged the Fukushima Daiichi Nuclear Power Plant (FNPP), resulting in the release of large quantities of radioactive material into the environment. The deposition of these radionuclides in rice straw as livestock feed led to the circulation of contaminated beef in the market. Based on the safety concern of the consumers, a reliable method for estimating concentrations of radioactive cesium in muscle tissue is needed. In this study, we analyzed the concentrations of radioactive cesium in the blood and skeletal muscle of 88 cattle, and detected a linear correlation between them. We then developed software that can be used to estimate radioactive cesium concentrations in muscle tissue from blood samples. Distribution of this software to the livestock production field would allow us to easily identify high‐risk cattle, which would be beyond the safety regulation, before shipping out to the market. This software is planned to be released as freeware. This software would contribute to food safety, and aid the recovery of the livestock industry from the damage creacted by the 2011 Tohoku earthquake and tsunami.  相似文献

The damage caused by the earthquake on 11 March, 2011 resulted in a serious nuclear accident in Japan. Due to the damage to the Fukushima Daiichi Nuclear Power Plant (FNPP), large amounts of radioactive substances were released into the environment. In particular, one of the largest safety concerns is radioactive cesium (134Cs and 137Cs). Due to the FNPP nuclear accident, a 20 km area was restricted from human activity, and various types of domestic animals were left in the zone. We collected the organs and tissues from sacrificed animals to obtain scientific data to evaluate the internal deposition of radioactive compounds. At first, we found there is a strong correlation between blood 137Cs and organ 137Cs with data from 44 cattle, indicating that skeletal muscle is the target organ of deposition of radioactive cesium. Second, we analyzed the relationship between blood 137Cs and muscle 137Cs within relatively lower radioactive concentration, suggesting that estimation of concentration of 137Cs is possible from blood concentration of 137Cs. Finally, we developed computer software to estimate the muscle 137Cs concentration from blood samples. Our study contributes to the food safety of livestock products.  相似文献

The accident at the Fukushima Daiichi Nuclear Power Plant (FNPP) released a large amount of radioactive substances into the environment. Furthermore, beef contaminated with radioactive cesium above the 500 Bq/kg safety standard was circulated in the food chain in 2011. Japanese consumers remain concerned about the safety of radioactively contaminated food. In our previous study, we detected a linear correlation between radioactive cesium (137Cs) activity in blood and muscle around 500 to 2500 Bq/kg in cattle. However, it was unclear whether the correlation was maintained at a lower radioactivity close to the current safety standard of 100 Bq/kg. In this study, we evaluated 17 cattle in the FNPP evacuation zone that had a 137Cs blood level less than 10 Bq/kg. The results showed a linear correlation between blood 137Cs and muscle 137Cs (Y = 28.0X, R2 = 0.590) at low radioactivity concentration, indicating that cesium radioactivity in the muscle can be estimated from blood radioactivity. This technique would be useful in detecting high‐risk cattle before they enter the market, and will contribute to food safety.  相似文献

Blood and urine were compared to clarify which is a better sample for estimating the concentration of radioactive cesium in muscles of live cattle. The most probable concentration of 137Cs in muscles was 21.0 times that in blood, and the error of this estimation was 28%. The concentration of 137Cs in blood was estimated accurately using urine samples. The estimation error did not increase so much (33%), even when the concentration of 137Cs in muscles was estimated using urine samples. On the other hand, the maximum volume of blood that can be collected with one syringe is 50 ml, whereas it is easy to collect 1,000 ml of urine. It took at least 360 min to confirm that a cow meets the legal standard by inspecting 50 ml of blood. However, with 1,000 ml of urine, a 20 min measurement time was sufficient for this purpose. This difference in the required measurement time is critically important for practical use. In addition, urine can be collected by farmers themselves, whereas the blood collection requires a veterinarian. Therefore, urine is a more convenient and practical sample for estimating the contamination level of live cattle with radioactive cesium.  相似文献

In the region contaminated by the Fukushima nuclear accident, radioactive contamination of live cattle should be checked before slaughter. In this study, we establish a precise method for estimating radioactive cesium concentrations in cattle blood using urine samples. Blood and urine samples were collected from a total of 71 cattle on two farms in the ‘difficult‐to‐return zone’. Urine 137Cs, specific gravity, electrical conductivity, pH , sodium, potassium, calcium, and creatinine were measured and various estimation methods for blood 137Cs were tested. The average error rate of the estimation was 54.2% without correction. Correcting for urine creatinine, specific gravity, electrical conductivity, or potassium improved the precision of the estimation. Correcting for specific gravity using the following formula gave the most precise estimate (average error rate = 16.9%): [blood 137Cs] = [urinary 137Cs]/([specific gravity] ? 1)/329. Urine samples are faster to measure than blood samples because urine can be obtained in larger quantities and has a higher 137Cs concentration than blood. These advantages of urine and the estimation precision demonstrated in our study, indicate that estimation of blood 137Cs using urine samples is a practical means of monitoring radioactive contamination in live cattle.  相似文献

An experiment was designed to test the hypothesis that short-term oral administration of dietary vitamin D3 to beef cattle before slaughter would increase beef tenderness through greater calcium-activated calpain activity in postmortem aged skeletal muscle. Thirty continental crossbred steers were allotted randomly to three treatment groups housed in one pen. One group served as a control; two other groups were administered boluses with either 5 x 10(6) or 7.5 x 10(6) IU of vitamin D3 daily for 9 d. Cattle were slaughtered 1 d later. The longissimus lumborum was excised from each carcass 72 h postmortem and steaks removed at 3, 7, 14, and 21 d postmortem. The semimembranosus muscle (top round) was excised from each carcass 72 h postmortem and steaks removed at 7, 14, and 21 d postmortem. Blood plasma calcium concentration of cattle treated with 5 or 7.5 x 10(6) IU of vitamin D3 was higher (P < .05) than that of controls. Strip loin and top loin steaks from cattle fed supplemental doses of vitamin D3 had lower (P < .05) Warner-Bratzler (W-B) shear values at 14 d postmortem but were not significantly different from controls at 3, 7, or 21 d (strip loins) or 7 or 21 d (top rounds). No significant difference in strip loin steak tenderness was observed by sensory panel at 14 d postmortem (P < .17) between steaks from control and vitamin D3-treated steers. At 14 d postmortem, strip loin and top round steaks from cattle fed 5 x 10(6) IU of vitamin D3, but not from those given 7.5 x 10(6) IU, showed more proteolysis (P < .05) than did steaks from control cattle, based on Western blotting analysis. Therefore, the use of supplemental dietary vitamin D3 given daily for 9 d before slaughter did improve tenderness (lower W-B shear values) of 14-d postmortem aged beef. Increased proteolysis seems to be the mechanism of tenderization.  相似文献

Nine barrows (23.8 +/- 0.9 kg) and 9 gilts (23.1 +/- 0.9 kg) were used to determine the disposition of radiocarbon after oral [14C]clenbuterol (4-amino-alpha-[t-butylaminomethyl]-3,5-dichlorobenzyl [7-(14)C]alcohol hydrochloride) administration and to determine total and parent residues in edible tissues. Three barrows and three gilts, housed in metabolism crates, were fed 1 ppm [14C]clenbuterol HCl for seven consecutive days in three separate trials; a single barrow and gilt from each trial was slaughtered after 0-, 3-, or 7-d preslaughter withdrawal periods. Urine and feces were collected during the dosing and the withdrawal period; edible and inedible tissues were collected at slaughter. Total recovery of radiocarbon was 94.2 +/- 6.5%. Total clenbuterol absorption was greater than 75% for barrows and 60% for gilts. Total radioactive residues in tissues were not different (P > 0.05) between barrows and gilts. Concentrations of parent clenbuterol in liver, kidney, skeletal muscle, adipose tissue, and lung did not differ between barrows and gilts (P > 0.05). Total radioactive and parent residues declined in tissues as withdrawal period increased. After the 0-d withdrawal period, total liver residues (286 ppb) were approximately equal to lung residues, twice those of the kidney, and about 15 times those of adipose tissue and skeletal muscle. After a 7-d withdrawal period, total radioactive residues in liver (15 ppb) were roughly three times greater than lung, kidney, and adipose tissue total residues and about 13 times those of skeletal muscle total residues. Parent clenbuterol represented 79, 63, 42, 67, and 100% of the total radioactive residue in adipose tissue, kidney, liver, lung, and skeletal muscle, respectively, in hogs slaughtered with a 0-d withdrawal period. With increasing withdrawal period, the percentage of total radioactive residue present as parent clenbuterol within edible tissues (including lung) decreased, so that after a 7-d withdrawal period, 7, 16, and 29% of the total residue was composed of parent clenbuterol in kidney, liver, and lung, respectively. After a 7-d withdrawal period, parent clenbuterol exceeded the European maximum residue limit (0.5 ppb) 4.6-fold in liver and 2.4-fold in lung. In muscle, clenbuterol was approximately 40 times the limit after a 0-d withdrawal period but had dropped below 0.5 ppb after a 3-d withdrawal period. Results from this study indicate that clenbuterol HCl is well absorbed in swine and that the use of clenbuterol in this species in an off-label manner is inconsistent with human food safety standards used in developed countries.  相似文献

Glycation (Maillard reaction) starts from non‐enzymatic amino‐carbonyl reaction binding carbonyl group of reducing sugars to amino group of amino acids. The amino‐carbonyl reaction leads to the formation of the stable Amadori product, which ultimately forms advanced glycation end products (AGE). Acceleration of glycation during hyperglycemia increases the production and accumulation of AGE, which is implicated in the gradual development of diabetic complications in diabetes mellitus. Avian species offers the great advantage of providing animal models for diabetology because high blood glucose concentration should accelerate to generate high concentration of AGE. In the present study, radioactive AGE was prepared from chicken serum albumin and 14C‐glucose. Radioactive AGE was administrated to examine the tissue distribution of AGE in chickens. Administration of radioactive AGE into the circulation of chickens revealed that AGE accumulated into specific tissues, liver, spleen and kidney, which have the function for clearing exogenous substances and endogenous metabolic waste products.  相似文献

A radioimmunoassay (RIA) method for hexoestrol using an antiserum against hexoestrol-carboxypropyl ether-BSA and H3-hexoestrol was used to measure the concentrations of residues of hexoestrol in 0.1 ml biological fluids and 1 g edible tissues of implanted cattle and sheep. A preliminary ether extraction of biological fluids was necessary before RIA. The ether extract from tissues was further purified by solvent partition and silica gel column chromatography before RIA. Conjugates of hexoestrol were measured after enzymatic hydrolysis to free hexoestrol. In untreated animals residues were either not detected or very low in all tissues except urine from sheep. The method has a lower limit of detection of approximately 0–10 pg/ml for biological fluids in cattle and 20–100 pg/g for tissues in both sheep and cattle but the lower limit of detection in sheep urine was 70–294 pg/ml urine. In two heifers implanted with 60 mg hexoestrol and slaughtered 2 and 7 days after implantation, residues of hexoestrol were detected in all tissues except muscle with highest concentrations between 2 - 17 ng/g in urine, bile and kidney. The concentration of residues in steers which had been implanted with 45 mg or 60 mg hexoestrol and slaughtered at 90 days after implantation were 0, < 50, 46–96 and 200 pg/ml or g of plasma, muscle, liver and urine, respectively. The concentrations of hexoestrol in sheep implanted with 15 ml hexoestrol and slaughtered after 60 days were 70, 0, 964, 3100 and 4074 pg/g or ml of muscle, fat, liver, kidney and urine, respectively. No hexoestrol was found in control untreated cattle and sheep. It was concluded that some residues of hexoestrol were present in the excretory fluids and tissues of cattle and sheep which had been implanted with hexoestrol at the recommended dose and slaughtered after the recommended withdrawal periods. However, the concentrations of hexoestrol in muscle and fat were extremely low or not detectable. The method could be used for the routine screening of animals for treatment with hexoestrol.  相似文献

The occurrence of cysticercosis due to Taenia saginata in cattle slaughtered for meat in Amhara National Regional State, northwestern Ethiopia between September 2005 and February 2007 was investigated. Routine meat inspection of various organs of 4456 cattle in eight abattoirs of this region showed that 824 (18.49%) were infected with Cysticercus bovis. The occurrence rate did not vary significantly from abattoir to abattoir (P>0.5). The tongue, masseter muscles, heart muscles, triceps muscles and thigh muscles were the main predilection sites of the cysts. Of 4102 male cattle, examined, 768 (18.72%) had cysts of C. bovis while 56 (15.82%) of the 354 female animals investigated were infected. The animals slaughtered were all adults. No significant difference in occurrence was recorded between the sexes. Monthly occurrence of the cysts in the animals revealed a rise of infected animals during the dry season.  相似文献

A retrospective study covering a period of 10 years (1990--1999) was conducted using post mortem meat inspection records of the Veterinary Department Information Management Unit at Harare to determine the prevalence and seasonal variation of bovine fasciolosis in Zimbabwe. Records of monthly and annual returns from five major abattoirs were examined in regard to total cattle slaughtered and the corresponding number of livers condemned due to Fasciola gigantica infection. Prevalence of fasciolosis was calculated as the number of cattle found to be infected with F gigantica, expressed as a percentage of the number of cattle slaughtered. Seasonal variations in the prevalence were examined by pooling respective monthly condemnation data over a 10-year (1990--1999) period. A total of 2,474,232 cattle were slaughtered during this period and 917,565 (37.1%) of these cattle were infected with F. gigantica. The pattern of distribution of F. gigantica was significantly higher in cattle originating from catchment areas of high rainfall than in those of relatively low rainfall, and in those slaughtered during the wet season than those slaughtered during the dry season (P< 0.05). Based on the study findings a control programme for the disease in Zimbabwe is suggested.  相似文献

A cross-sectional serological study on cattle less than 2 years old, using an antigen ELISA for the detection of bovine cysticercosis was carried out between November 2009 and February 2010 in 10 slaughterhouses from the Catalonia region (North-Eastern Spain). Circulating antigen was detected in 23 of 2073 animals, i.e. a sero-prevalence of 1.11% (CI95%: 0.76-1.75%). The determined sero-prevalence was about 50 times higher than the prevalence obtained by visual inspection within the same period: 19 positive animals of 90,891 slaughtered animals (0.02%) in the same slaughterhouses. None of the animals with positive result in the Ag-ELISA was detected by meat inspection.  相似文献

Retail cutting tests were conducted on subprimals from cattle fed zilpaterol hydrochloride (ZH) to determine if the improved carcass composition and red meat yield resulting from ZH feeding would translate into increased retail yields of ready-to-cook products. As part of a 3-phase study, selection of carcasses from Holstein steers was done once (fall 2008), followed by the collection of carcasses from beef-type steers on 2 separate occasions (beef study I: summer 2009; beef study II: spring 2010). Each of the 3 groups of steers was assigned previously to 1 of 2 treatments, treated (fed 8.3 mg/kg of ZH for 20 d) or control (not fed ZH). All steers were slaughtered and carcasses were fabricated in commercial beef-processing establishments. Only those carcasses grading USDA Choice or higher were used. Five subprimals were used for both the calf-fed Holstein study (n = 546 subprimals) and beef study I (n = 576 subprimals): beef chuck, chuck roll; beef chuck, shoulder clod; beef round, sirloin tip (knuckle), peeled; beef round, top round; and beef round, outside round (flat). Seven subprimals were used in beef study II (n = 138 subprimals): beef chuck, chuck roll; beef round, sirloin tip (knuckle), peeled; beef round, top round; beef round, eye of round; beef loin, strip loin, boneless; beef loin, top sirloin butt, boneless; and beef loin, tenderloin. A simulated retail market environment was created, and 3 retail meat merchandisers prepared retail cuts from each subprimal so salable yields and processing times could be obtained. Differences in salable yields were found for the calf-fed Holstein steer chuck rolls (96.54% for ZH vs. 95.71% for control; P = 0.0045) and calf-fed Holstein steer top rounds (91.30% for ZH vs. 90.18% for control; P = 0.0469). However, other than heavier subprimals and an increased number of retail cuts obtained, total salable yields measured on a percentage basis and processing times were mostly unaffected by ZH. Cutability advantages of feeding ZH are achieved primarily in the carcass-to-subprimal conversion rather than in the subprimal-to-retail conversion.  相似文献

Reproductive organs from mature Small East African zebu (SEAZ) heifers and cows slaughtered at the Morogoro abattoir were collected twice a month and evaluated over a period of 12 months. Out of the 402 animals from which reproductive organs were taken, 54% were pregnant, 24% were actively cycling and 22% were non-cycling. Various gross abnormalities were observed in the reproductive organs of about 16% of the cattle, and the major reproductive abnormality in both total and the non-cycling animals was various degrees of fibrous adhesion between the ovary and the infundibulum and mesosalpinx. It is concluded that, contrary to common belief, a majority of the female SEAZ cattle that are slaughtered are fertile.  相似文献

The study was conducted from May 2005 to December 2006 in Bahir Dar Abattoir to assess the current status of hydatidosis in cattle and sheep. Hydatid cyst count and characterization were conducted based on routine meat inspection. Of the total 420 cattle and 340 sheep slaughtered in Bahir Dar Abattoir 143 (34.05%) and 36 (10.6%) animals were found harboring hydatid cysts respectively. Thorough meat inspection in the abattoir revealed that 202 and 54 visceral organs were found harboring one or more hydatid cysts in cattle and sheep respectively. Differences in prevalence rates between the two species of animals were highly significant (P < 0.001). The infection of the lung, liver, kidney, spleen and heart were found to be 57.9% , 36.6% , 3% , 1.5% , 1% in cattle and 50%, 48.1% and 1.9% in sheep respectively. From the total of 864 in cattle and 138 in sheep hydatid cysts counted 315 (36.4%), 268 (31.0%), 65 (7.5%), 216 (25.0%) in cattle and 92 (66.7%), 20 (14.5%), 1 (0.7%), 25 (18.1%) in sheep were found to be small, medium, large and calcified cysts respectively and 484 (56.0%), 164 (18.9%), 216 (25%) in cattle and 35 (25.4%), 78 (56.5%), 25 (18.1%) in sheep were sterile, fertile and calcified cysts respectively. Viability rates of 62.2% in cattle and 78.2% in sheep were observed. The rate of cyst calcification was higher in the liver than in the lung while fertility rate was higher among the cysts of the lung for both cattle and sheep.  相似文献

The prevalence of parasitic infections responsible for the condemnation of carcasses and viscera during meat inspection, and their economic implication, was estimated in a year long abattoir survey of 10 277 slaughtered farm animals in the region of Trikala, Greece. The organs examined for the presence of parasitic lesions during meat inspection were: liver and lungs of all animals, rumen of cattle, small intestine of lambs and kids, and muscles of cattle and swine. The parasitic lesions observed in the lungs of cattle, sheep and goats were caused only by hydatid cysts. No hydatid cysts were observed in the lungs of swine. The parasitic lesions observed in the liver of cattle, sheep and goats were as a result of hydatid cysts and flukes of Fasciola hepatica and Dicrocoelium dendriticum, while those of swine were due to milk spots only. Moniezia sp. proglottids were found in the small intestine of lambs only. The prevalence of parasites responsible for the condemnation of marketable organs was low (0.26%). Parasites were responsible for 22% of the total of condemned organs, and their annual cost was 99, 00 GDR (approximately 292 Euros). The parasites most contributing to marketable organ condemnation were hydatid cysts (26%) and D. dendriticum flukes (26%).  相似文献

A cross-sectional study to determine the prevalence of bovine cysticercosis and hydatidosis was conducted from October 2007 to March 2008 in cattle slaughtered at the Jimma municipal abattoir. Cyst distribution and viability of bovine cysticercosis and hydatidosis were also determined. A total of 512 carcasses were inspected of which 15 (2.93%) and 161 (31.44%) were infected with Taenia saginata metacestodes and hydatid cysts, respectively. From a total of 109 cysticerci collected from infected carcasses, 47 (43.12%) were viable. The anatomical distribution of the cysticerci was, shoulder muscle (39.5%), heart (33.9%), neck muscle (13.8%), tongue (10.1%), masseter muscles (1.8%) and diaphragm (0.9%). Of the 1171 hydatid cysts collected 223 (19.0%) were fertile, 505 (43.1%) sterile, 49 (29.8%) calcified and 94 (8.0%) contained pus. A greater proportion of fertile cysts were found in the lungs than in other organs. It was concluded that these zoonotic cestodes deserve due attention to safeguard public health, and that further studies are needed on genotyping, epidemiology and public health importance of Echinococcus granulosus in the study area.  相似文献

Experiments were conducted to determine the total residues remaining in ocular tissues of cattle and turkeys after oral administration of [14C]ractopamine HCl. Twelve cattle were intraruminally dosed with 0.9 mg x kg(-1) x d(-1) of [14C]ractopamine HCl for 7 d. Four cattle each were slaughtered with withdrawal periods of 48,96, and 144 h. Radioactive residues were not detectable in whole-eye homogenates from the cattle. Eight male and eight female turkeys per treatment received either 7.5, 22.5, or 30 ppm dietary [14C]ractopamine HCl (0.33, 1.02, and 1.36 mg x kg(-1) x d(-1); treatment groups 1, 2, and 3, respectively) for 7 d, and the birds were slaughtered with a 0-d withdrawal period. Eyes were dissected into retina/choroid/schlera (RCS), cornea/iris (CI), and aqueous humor (AH) fractions. Residues in RCS, CI, and AH of treatment 1 turkeys were not detectable. Residues in AH were < 0.02 ppm in treatment groups 2 and 3. Mean residues in RCS ranged from 0.15 to 0.26 ppm, and mean CI residues ranged from <0.09 to 0.17 ppm for treatment groups 2 and 3, respectively. The propensities of ractopamine and synthetic ractopamine metabolites to bind to melanin were studied in vitro using radiolabeled ligands with centrifugal filtration to separate melanin from unbound ligand. In vitro studies showed that [14C]ractopamine HCl binds to melanin rapidly and was displaced from melanin by other beta-agonists. Glucuronidation of ractopamine, which produced the major biotransformation product of ractopamine in all species studied to date, prevented binding to melanin. These studies demonstrate that the propensity for the in vivo binding of ractopamine HCl to pigmented ocular tissues is less than that reported for clenbuterol.  相似文献
