To determine if a transient increase in uterine blood flow (BF) and estradiol-17 beta (E2 beta) secretion occurs during maternal recognition of pregnancy in ewes (as previously observed for sows and cows), 40 nonpregnant (NP) ewes were assigned in equal numbers to surgery on d 9, 11, 13 or 15 postestrus (d 0 = day of estrus). For 20 NP ewes (five/day), each uterine horn (UH) was flushed with saline and uterine flushings (UF) collected. For the remaining 20 ewes, BF was determined for each UH using electromagnetic transducers, and samples of uterine arterial (UA) and uterine venous (UV) blood were obtained from each UH. After an intervening cycle, each ewe was mated, subjected to surgery on the same day postmating as during her previous nonmated cycle, and BF measurements and UA and UV samples were obtained. In addition, each UH of pregnant (P) ewes was flushed and the location of conceptuses was determined. Concentrations of E2 beta and progesterone (P4) in UA and E2 beta in UV and UF were determined by radioimmunoassay. For NP ewes, BF (ml/min) was not different for UH ipsilateral or contralateral to the ovary bearing the corpus luteum (CL), and did not differ across days, averaging 6.5 +/- .4. For P ewes, BF to UH contralateral to the ovary bearing the CL on all days and BF to UH ipsilateral to ovaries bearing CL on d 9 was similar to BF of either UH of NP ewes, averaging 6.8 +/- .6. Sixteen primiparous sows were bred and fed either a control ration (n = 8) or a diet containing purified zearalenone (n = 8; 1 mg/kg of body weight) from days 7 to 10 after breeding. On day 7 after breeding, the jugular vein of each sow was cannulated and blood was collected at 20-minute intervals for 4 hours before feeding and 4 hours after feeding. On day 10 after breeding, blood samples were collected from 4 control sows and 4 zearalenone-fed sows at 20-minute intervals for 4 hours before collection of blastocysts. A similar blood sampling schedule was followed for the remaining 4 control and 4 zearalenone sows on day 14 after breeding. On day 10 after breeding, spherical blastocysts were recovered from all control sows and from 3 of 4 zearalenone-treated sows. Average diameter of blastocysts from zearalenone-treated sows were similar to that of control sows. On day 14 after breeding, blastocysts were recovered from all control sows and 3 of 4 zearalenone-treated sows. Blastocysts from the control sows were filamentous, whereas blastocysts from zearalenone-treated sows were fragmented and contained foci of necrosis. Incidence of luteinizing hormone (LH) secretory spikes per sow was less (P less than 0.01) in zearalenone-treated sows (0.25 +/- 0.25/4 h) than control sows (1.75 +/- 0.25/4 h) on day 10 after breeding. First litter sows in naturally occurring post-weaning estrus were hand mated to proven boars and were fed a diet supplemented with zearalenone, an estrogenic mycotoxin (1 mg zearalenone/kg body weight), or a control diet on days 7 through 10 after mating. Embryos (blastocysts) and endometrial biopsies were collected from control and treated sows on days 9, 11, and 13 after mating. All blastocysts harvested on day 9 were spherical; treatment of sows with zearalenone had no effect on blastocyst development. Blastocysts collected from treated sows on day 11 were in stages of elongation comparable to those of blastocysts from control sows but had mild degenerative changes in the embryonic disks, characterized by slightly retarded development and an increase in the number of necrotic cells. Blastocysts collected from treated sows on day 13 were in an advanced stage of degeneration, characterized by circumferential constrictive division, fragmentation, and degeneration and disorganization of the embryonic disk. Feeding zearalenone to pregnant sows had no effect on the normal decrease in height of the endometrial luminal epithelium on days 9 through 13 after mating and no effect on morphologic appearance of secretory vesicles in the endometrial glandular epithelium. The dosage scheme of zearalenone used in this study did not cause any morphologic changes in the endometrium that could be associated with hyperestrogenism.  相似文献

A RIA was developed for porcine intrauterine folate binding protein (FBP). Displacement of [125I]FBP caused by increasing dilutions of uterine flushings collected from either d-15 pregnant or nonpregnant gilts or media from culture of endometrial tissue from d-15 pregnant or nonpregnant gilts was parallel to the displacement caused by the standard curve. Addition of known amounts of purified allantoic fluid FBP to dilutions of either intrauterine flushings or endometrial culture medium were measured accurately with the RIA. To test specificity, 2-mL samples of uterine flushings collected from d-15 pregnant and nonpregnant gilts were preincubated with 10 microCi of [3H]folic acid and then chromatographed using Sephadex G-100 (Sigma Chemical Co., St. Louis, MO). The fractions were subsequently assayed for radioactivity by liquid scintillation spectrophotometry and for FBP by RIA. The [3H]folic acid and FBP peaks coincided, indicating that the RIA is specific for FBP. Uterine flushings were collected on d 10, 11, 12, 13, 14, and 15 of the cycle or pregnancy from 1) White crossbred, 2) progesterone-treated White crossbred (200 mg of progesterone at 48 and 72 h after estrus), and 3) Meishan gilts and assayed for FBP. Total FBP increased 140-fold from d 10 to 15, and the pattern of change across day did not differ between pregnant and nonpregnant gilts. Progesterone treatment increased intrauterine FBP content on d 10 and 11. No difference in FBP concentrations was detected between White crossbred and Meishan gilts. These results indicate that the RIA for FBP is valid, allowing measurement of this protein in uterine flushings and endometrial culture medium. The onset of FBP secretion by the uterus between d 10 and 15 of the cycle or pregnancy is influenced by the timing of onset of progesterone influence in a manner similar to the endometrial proteins uteroferrin and retinol binding protein. In contrast to these endometrial proteins, FBP concentrations are similar in Meishan and White crossbred gilts.  相似文献

Experiments were performed to determine the effects of components of the GH-IGF axis on conceptus development and postnatal growth in sheep. In Exp. 1, ewes received one of the following treatments: 1) sustained release GH at breeding, 2) sustained release GH at breeding and estradiol-17beta at d 5 and 6, 3) only estradiol-17beta at d 5 and 6, or 4) no treatment. Uteri were flushed on d 7, and flushings were analyzed for content of IGF-I. A single injection of sustained-release bovine GH at breeding increased IGF-I content in uterine luminal flushings compared with control ewes (P < 0.05). Treatment with estradiol-17beta on d 5 and 6 after breeding did not alter IGF-I content compared with control ewes, and it blocked the effect of GH on uterine luminal IGF-I content. In Exp. 2, sustained release GH or no treatment was administered at breeding, and gravid uteri were collected at d 25, 80, or 140 of gestation. On d 80, GH-treated ewes had smaller chorioallantoic weights (P < 0.05) and tended to have more efficient placentae (fetal weight/total placental weight; P = 0.052), with a higher percentage of placental weight as cotyledons (P = 0.068) compared with control ewes. In Exp. 3, ewes were treated with or without sustained release GH at progesterone withdrawal. Lambs from GH-treated ewes were heavier at birth (P < 0.05). Lambs from GH-treated ewes reared as singles, but not lambs reared as multiples, were heavier at 30, 60 (P < 0.05), and 75 d (P = 0.075) of age than lambs from control ewes. In conclusion, ewes treated with sustained-release GH at breeding developed smaller, more efficient placentas, and had larger lambs at birth.  相似文献

Reproductive components were compared between a line of sows selected (S) for improved sow productivity index (SPI = 6.5 x number born alive + adjusted 21-d litter weight) and sows from an unselected control (C) line. Generation 9 and 10, second-parity, Landrace sows were chosen from both the S (n = 35) and C (n = 33) line. Sows were slaughtered at a commercial slaughter plant at approximately 75 d of gestation and their reproductive tracts were recovered. Reproductive tracts were evaluated for uterine weight (UTWT), uterine horn length (UTLN), ovulation rate (OR), number of fully formed fetuses (NF), number of mummified fetuses (NM), percentage of fetal survival (FS = NF/OR), fetal space (FSPACE = UTLN/[NF + NM]), and fetal position, sex, and weight. Select-line sows had greater NF (P less than .10) and higher FS (P less than .10) than C-line sows. Select-line sows had longer (P less than .05), and heavier (P less than .01) uteri than C-line sows. However, uterine length adjusted for NF was not different between the two lines. Uterine weight adjusted for NF was greater in S-line sows (P less than .05). Select-line sows had greater total fetal weight (TFWT) (P less than .05) than did C-line sows. Female fetuses positioned between two male fetuses were lighter in weight than all other female fetuses (P less than .01). An experiment was conducted to study the effects of major dietary energy source fed from weaning to ovulation or from ovulation to d 35 of pregnancy on reproductive traits in primiparous sows. Dietary energy sources were used to manipulate the plasma insulin concentration. One hundred thirteen sows were used in a split-plot design. From weaning to ovulation sows were fed at two times maintenance either a diet with tallow (Fat) or maize starch plus dextrose (Starch) as the major energy source. From ovulation onward, sows within each dietary group were alternately reassigned to either the Fat or the Starch diet and were fed at 1.25 times maintenance. Estrus detection was performed three times a day from d 3 to 9 after weaning and sows were inseminated each day of standing estrus. On d 35 of pregnancy, the sows were slaughtered and their reproductive tracts were removed. Plasma insulin concentration was higher in sows fed the Starch-rich diet than in sows fed the Fat-rich diet on d 4 after weaning (1.30 vs 0.97 ng/mL, P = 0.08) and on d 32 of pregnancy (1.20 vs 0.51 ng/mL, P < 0.001). Plasma glucose and IGF-I concentration on d 4 after weaning and d 32 of pregnancy did not differ between sows on the two dietary energy sources. The percentage of sows exhibiting estrus within 9 d after weaning was 52 and 67% for the Fat and Starch diet before ovulation, respectively (P = 0.11), whereas the weaning-to-estrus interval was 134 vs 123 h, respectively (P = 0.12). Survival analysis showed that sows fed the Fat-rich diet had a 1.6 times higher risk to remain anestrous until d 9 after weaning than sows fed the Starch-rich diet (P = 0.04). No effect of dietary energy source, either before or after ovulation, on uterine, placental, or embryonal development on d 35 of pregnancy was found. It can be concluded that the dietary energy source provided after weaning can affect the risk of sows to remain anestrous but does not affect uterine, placental, or embryonic traits.  相似文献

It has been shown that zearalenone disrupts early pregnancy in swine without altering intrauterine content of estradiol 17 beta or progesterone, embryo migration, or estradiol-17 beta synthesis by blastocysts. However, serum concentrations of progesterone were reduced 2 to 3 weeks after mating in gilts that ingested zearalenone. Therefore, progesterone was administered to gilts during early pregnancy to determine whether it could counteract the detrimental actions of zearalenone on embryonic development. Thirty-two crossbred gilts (Hampshire x Chester White x Yorkshire x Duroc) were assigned randomly to a 2 x 2 factorial arrangement of treatments: zearalenone (Z); zearalenone plus progesterone (ZP); progesterone (P); or control (C). From postmating days 4 to 15, Z- and ZP-treated gilts were fed 1 mg of Z/kg of body weight, and P-treated and C gilts were fed ethanol as vehicle in a corn-soybean diet. On postmating days 3 to 15, P- and ZP-treated gilts were injected IM with 100 mg of progesterone, and C and Z-treated gilts were injected with progesterone carrier (15% ethanol, 15% benzyl alcohol, 70% propylene glycol). Blood was collected from gilts by puncture of the jugular vein daily from days 3 to 15, on alternate days from days 17 to 31, and then twice weekly until the end of the experiment. Fetal development was assessed in Z- and ZP-treated gilts on postmating day 47.6 +/- 2.9 by cesarean section and in P-treated and C gilts at slaughter on postmating days 51.2 +/- 3.2. Serum concentrations of progesterone in P-treated gilts were greater on days 7 to 8, 10 to 15, 17, and 19 than in C gilts. Serum concentrations of progesterone were greater on days 8, 10, and 12 in ZP-treated than in C gilts. The release of opsonins into the uterine lumen of mares susceptible or resistant to endometritis was examined after intrauterine inoculation of a filtrate of Streptococcus culture fluid or vehicle. Uterine flushings were collected at 0.5 hour before and 2, 4, 6, 8, and 24 hours after inoculation on day 2 or 3 of estrus and on day 7 or 8 after ovulation. Amounts of opsonins in flushings were quantified as the H2O2 produced by leukocytes incubated with flushings-opsonized bacteria, compared with H2O2 produced by leukocytes incubated with nonopsonized bacteria. Opsonin values in flushings increased (P less than 0.025) in all mares after inoculation of filtrate or vehicle. For mares resistant to endometritis, opsonin values were greater at diestrus than at estrus. The opposite was true for mares susceptible to endometritis, resulting in a status (susceptible vs resistant) X stage of cycle interaction (P less than 0.025). Overall, opsonins were higher (P less than 0.05) in flushings of mares susceptible to endometritis than in flushings of mares resistant to endometritis, but this difference was only apparent at estrus. Preliminary characterization of opsonins in uterine secretions by ammonium sulfate fractionation and gel filtration indicated that opsonins were mainly associated with an ammonium sulfate-soluble fraction of high molecular weight (greater than 4 X 10(6] and an ammonium sulfate-precipitable fraction that was associated with immunoglobulin G.  相似文献

Sixteen crossbred sows (Yorkshire x Duroc) were used to determine the effect of recombinantly derived porcine somatotropin (pST) on lactational performance and the pattern of plasma metabolites and growth rate of nursing pigs. Daily s.c. injections of either pST (8.22 mg.sow-1.d-1) or excipient were administered at 1000 on d 12 through d 29 of lactation. Jugular cannulas were inserted in three sows/treatment and hourly blood samples were collected on d 11 to 13 and d 28 to 29 of lactation to determine the effect of treatment on plasma concentrations of somatotropin, glucose and nonesterified fatty acids in plasma. Milk production and weight of nursing pigs were determined pretreatment (d 9 and 10) and on d 16, 22 and 28. Milk production of sows receiving pST progressively increased above that of control sows and was 22% greater on d 28. Milk composition was not affected by treatment with pST (P greater than .10), so that the increase in yields of milk fat, lactose and solids paralleled the increases in milk yield. Total milk protein yield tended to be higher in sows receiving pST, but protein yield was greater (P less than .10) only on d 28. Pigs suckling sows treated with pST weighed .34 kg more at the end of the lactation period (P less than .05). Sows receiving pST consumed less feed (P less than .05) during the treatment period, and, as a result, lost more weight (P less than .10) and backfat (P less than .05) than control sows. Average concentrations of plasma somatotropin were elevated approximately 2.5-fold above baseline levels by exogenous pST. No acute alterations in plasma glucose or nonesterified fatty acids were observed in response to pST treatment, however, sows receiving pST had a chronic elevation of plasma glucose on d 29 of lactation.  相似文献

A cooperative regional research study using 194 sows, from which data were collected from 381 litters, was conducted at 3 research stations to determine the effects of added psyllium (a concentrated fiber source) or soybean hulls to gestation diets on reproductive performance of sows and preweaning performance of their pigs. Primiparous and multiparous sows were allotted to the 3 treatments of control (corn and soybean meal-based), 0.30% psyllium, or 20% soybean hulls. Sows fed the control and 0.30% psyllium diets were provided 1.82 kg/d, and sows fed the 20% soybean hulls diet were provided 2.0 kg/d to equalize ME, Lys, Ca, P, and vitamin and trace mineral intake. Treatments 1 to 3 had 130, 130, and 121 litters per treatment from 64, 64, and 63 sows, respectively. Gestating sows fed psyllium had a greater (P < 0.01 to 0.10) d 110 gestation, farrowing, weaning, and 17 d postpartum BW and gestation ADG compared with sows fed soybean hulls. Sows fed psyllium also had a greater (P < 0.10 and 0.08) d 110 gestation BW and gestation ADG than the control sows. Sows fed soybean hulls had a reduced (P < 0.06) farrowing BW compared with the control sows. Sows fed psyllium weaned lighter (P < 0.09) pigs than sows fed the control diet. Litter size was not affected (P > 0.10) by diet. Sows fed psyllium had a reduced (P < 0.03) feed intake compared with sows fed soybean hulls for d 5 to 7 postpartum, and sows fed the control diet were intermediate. Fecal scores (1 to 5 with 1 = dry and 5 = watery) were greater (P < 0.001) and DM content was less (P < 0.001 to 0.01) in the feces of sows fed soybean hulls compared with sows fed psyllium or the control diet on d 112 of gestation and d 4 postpartum. Fecal scores were greater (P < 0.10) and fecal DM content was less (P < 0.02) in sows fed psyllium compared with sows fed the control diet only on d 4 postpartum. In summary, sows fed soybean hulls during gestation had reduced BW compared with sows fed the control diets. In contrast, sows fed psyllium had an increased BW.  相似文献

Exudate and uterine flushings were collected at either 30, 60, 120 or 240 mins after intrauterine infusions of Streptococcus zooepidemicus in genitally normal mares during oestrus. Uteri were also flushed without prior induction of endometritis. Protein concentrations in exudate and flushings increased with time and exudate pH decreased with time; the pH of flushings did not alter. Lysozyme and lactate dehydrogenase were present in flushings from non-infected uteri, but concentrations increased with time after infection. Immunoreactive prostaglandin E2 was undetectable before infection, but concentrations rose after infection. No neutrophils were present in non-infected flushings but, by 30 mins, there were significant (P less than 0.01) neutrophil numbers in exudate and flushings; thereafter numbers increased, particularly in exudate. Acute endometritis resembled acute inflammation at other sites in the horse and a significant response had occurred by 30 mins after experimental infection.  相似文献

Background: Zearalenone(ZEN) is an estrogenic mycotoxin that is primarily produced by Fusarium fungi and has been proven to affect the reproductive capacity of many species to varying degrees. The present experiment was designed to study the maternal persistent effects of zearalenone toxicity in gestating sows on growth and muscle development of their offsprings, and the alleviation of zearalenone toxicity by modified halloysite nanotubes(MHNTs).Methods: Eighteen sows were fed with one of three dietary treatments that included the following:(1) a control diet,(2) a contaminated grain diet(with 50 % moldy corn, 2.77 mg/kg ZEN), and(3) a contaminated grain diet(with 50 %moldy corn, 2.76 mg/kg ZEN) + 1 % MHNTs. Each sow was exclusively fed its experimental diets from 35 to 70 d of gestation at a total of 2 kg daily. Muscle samples were collected from six piglets per treatment at birth, weaning and finishing.Results: The results showed that feeding the sows with the ZEN-contaminated diets from 35 to 70 d of gestation decreased the ADG, ADFI and G:F of their offsprings(P 0.05). The muscle fiber numbers in the newborn, weaning and growing-finishing pigs and the muscle fiber diameters at birth and weaning were also decreased by maternal ZEN exposure(P 0.05). The expressions of IGF-I, IGF-II, Myf-5 and Mstn at birth and IGF-II, Pax7, Myf-5 and Myo D1 at weaning were altered by feeding gestating sows with ZEN-contaminated diets(P 0.05). The MHNTs reduced most of the ZEN-induced toxic effects: the ADG and ADFI on growth performance, the muscle fiber numbers at weaning and finishing and the muscle fiber diameters at weaning(P 0.05). The expression levels of IGF-II and Mstn in newborn piglets and IGF-II and Myf-5 in weaning piglets were also prevented by adding 1 % MHNTs(P 0.05).Conclusions: The present study demonstrated that the offsprings of sows fed with ZEN-contaminated diets from 35 to 70 day of gestation exhibited weakening on growth performance, physiological changes in their muscle fibers and alterations of m RNA expression in their muscle tissues, and also indicated that MHNTs prevented most of the ZENinduced weakening in the muscle tissues.  相似文献

Twenty-five crossbred ewes were utilized to examine the timing of embryonic migration relative to maternal recognition of pregnancy. These ewes also were utilized to examine whether ovine embryos synthesized estradiol-17 beta in association with embryonic elongation and intrauterine migration. Embryos were flushed on d 11 through 15 from hemiovariectomized ewes. Recovery of embryos from the uterine horn contralateral to the remaining ovary indicated that migration had occurred. Ewes subsequently were returned with rams to determine their interestrous interval. Recovered embryos were classified morphologically, their length determined and individually incubated. Changes in estradiol within the medium were determined after a 6-h incubation. Embryo migration began on d 14 (P less than .05); on consecutive days from 11 through 15, 0, 0, 0, 60 and 100% of ewes examined, respectively, had an embryo in the contralateral horn. Extended estrous cycles (greater than 20 d) were observed in 0, 0, 40, 80 and 100% of ewes examined (P less than .05) following removal of embryos on d 11 through 15 of the cycle. Ovine embryos were longer (P less than .05) on d 14 (4.8 +/- 1.1 cm length, mean +/- SE) compared with d 13 (.2 +/- .1 cm) and increased further (P less than .05) on d 15 (7.8 +/- 1.1 cm). 本试验旨在研究饲粮中添加玉米赤霉烯酮(ZEN)对母猪繁殖性能和胎盘免疫相关基因表达量的影响。选择胎次相近、体重200 kg、妊娠第30天的长×大二元杂交母猪40头,随机分为2组,每组20个重复,每个重复1头。对照组饲喂基础饲粮,试验组饲粮在基础饲粮中添加1.5 mg/kg的ZEN。试验期74 d。结果表明,与对照组相比:1)饲粮中添加ZEN显著提高了妊娠期母猪死胎数和弱仔猪数(P0.05),显著降低了母猪总产仔数(P0.05);2)饲粮中添加ZEN显著提高了妊娠期母猪血清孕酮含量(P0.05);3)饲粮中添加ZEN显著提高了妊娠期母猪胎盘中Toll样受体-2(TLR-2)和孕酮受体(PGR)基因表达量(P0.05)。由此可见,母猪妊娠期饲粮中添加1.5 mg/kg ZEN可显著降低母猪总产仔数,并显著提高死胎数和弱仔猪数。饲粮中低水平的ZEN对母猪繁殖性能仍产生不利影响。  相似文献

The objective of this experiment was to determine the reproductive performance and factors that affect on-farm application of low-dose intrauterine insemination (IUI) in sows. Four hundred twenty-two sows were used in a simple arrangement of four treatments to determine the effect of spermatozoa per dose (0.5 x 10(9), 1 x 10(9), or 4 x 10(9) IUI, and 4 x 10(9) with a conventional catheter) on the main effects of conception, litter size, and farrowing rate. Following weaning at approximately 18 d after parturition, estrus detection was performed daily in the presence of a mature boar. At the time of estrus detection, sows were blocked for parity (1, 2, or 3+), weaning-to-estrus interval (WEI; 3, 4, or 5 d), and assigned randomly to be serviced twice with semen from the same boar(s). Treatment services were equally divided among three technicians. Delivery of acceptable numbers of spermatozoa per dose with either device (IUI or conventional) produced similar reproductive performances; however, farrowing rate, total pigs born, and total born alive decreased (P < 0.05) when suboptimal numbers (< or = 1 x 10(9)) of spermatozoa were used with IUI. Treatment interactions with parity were not detected and were removed from the final model. Treatment interactions with WEI on farrowing rate were detected (P < 0.05), and sows with WEI of 3 d had a markedly lower (P < 0.05) farrowing rate than all other treatment groups. The results from this experiment suggest that placement of semen at the beginning of the uterine horn with conventional volumes and spermatozoa numbers produces results similar to placement of semen in the cervical cavity with a conventional AI catheter. Although there is little published evidence of reproductive performances in a commercial setting with suboptimal numbers of spermatozoa, these results suggest that insemination beyond the cervix does not offset effects of suboptimal numbers of spermatozoa.  相似文献

A 2-year comparative study was carried out to evaluate the effect of ovary size, follicle size and oocyte quality of 3-month-old Simmental calves and the efficiency of using calf ovaries in an in vitro fertilization (IVF) programme. We evaluated the effects of different concentrations of follicle-stimulating hormone (FSH) and oestradiol-17beta (E-17beta) in the maturation medium on the in vitro development of calf oocytes into morula and blastocysts. The proportion of recovered oocytes (62.1%; 42.8%; 25.3%) and the percentage of good quality cumulus oocyte complexes (84.2%: 59.8%; 45.9%) decreased significantly (P < 0.01) with decreasing ovary size (L, M and S). The rates of two or more cells on Day 2 and of blastocysts on Day 7 and Day 9 were significantly lower (P < 0.01) for calf oocytes (61.5%; 18.9%: 15.9%) compared with those from sexually matured females (70.1%: 32.3%; 22.2%). Calf oocytes. matured in medium supplemented with 20 microg/ml or 10 microg/ml FSH plus 2 microg/ml E-17beta had higher rates of cleavage on Day 2 (64.1% and 64.7%) and blastocysts on Day 7 (24.5% and 22.4%) than the control supplemented with 10 microg/ml FSH (55.6% and 19.2%, respectively). Groups supplemented with 20 microg/ml FSH plus 2 microg/ml E-17beta and 10 mg/ml plus 4 mg/ml E-17beta showed a significantly lower developmental rate of blastocysts on Day 7 (14.6% and 14.5%). High concentrations of E-17beta (4 microg/ml) resulted in a significantly lower development of blastocysts on Day 9 (8.1%) and hatched blastocysts on Day 13 (3.5%) (P < 0.01). We conclude that the proportion of calf oocytes obtained from immature animals and their suitability for IVF are lower than those of cows. Thus, the use of oocytcs from sexually immature females would decrease the relative efficiency of IVF programmes. Supplementation with high concentrations of FSH can improve the maturation and developmental capacity of oocytes from prepubertal calves.  相似文献

Two experiments were conducted to evaluate the effects of soluble (SF) and insoluble (ISF) dietary fiber during gestation on embryo survival and sow performance. In Exp. 1, 43 gilts were assigned randomly to 1 of 4 experimental diets: a corn-soybean meal control (C; 1.16% SF, 9.98% ISF); a 30% oat bran high in SF (HS; 3.02% SF, 10.06% ISF); a 12% wheat straw diet high in ISF (HIS; 1.08% SF, 18.09% ISF); and a 21% soybean hull diet (HS + HIS; 2.46% SF, 24.55% ISF). Gilts were fed the experimental diets based on their initial BW to meet their daily nutrient requirements. At estrus, gilts were inseminated artificially 3 times using pooled semen. Reproductive tracts were harvested 32 d postmating (range = 28 to 35 d). Statistical analysis of data included the effects of diet with days of gestation as a covariate. There were no differences in ovulation rate among gilts fed the experimental diets (avg. = 14.1). Number of live embryos was less for HIS and HS + HIS gilts compared with C and HS (9.9 and 9.1 vs. 11.9 and 10.6, respectively; P < 0.05). Total embryo survival rate (P < 0.05) was less for gilts fed HS + HIS compared with those fed the C and HS diets. These results suggest that high dietary ISF might decrease the total embryo survival rate without affecting ovulation rate. In Exp. 2, 716 sows were used in 3 concurrent trials. In trial 1, diets included a corn-soybean meal control (C; 0.43% SF, 10.50% ISF; n = 122) or a 31% oat bran diet (HS; 1.93% SF, 8.87% ISF; n = 124). In trial 2, diets included a C (n = 97) or a 13% wheat straw diet (HIS; 1.10% SF, 17.67% ISF; n = 119), and in trial 3 sows were fed a C (n = 123) or a 21% soybean hull diet (HS + HIS; 1.50% SF, 17.77% ISF; n = 131). All diets were offered to sows beginning 2 d postmating. All sows had ad libitum access to a standard lactation diet. Statistical analysis included the effects of diet, parity group, genetic line, and season as well as their interactions. The inclusion of SF and ISF in gestation diets did not affect litter size. Sows fed the HS + HIS diet had a greater ADFI and lost less BW during lactation (P < 0.01) than sows fed C. Under the conditions of this study, feeding gestating sows increased levels of SF and ISF from d 2 after breeding to d 109 of gestation did not increase litter size.  相似文献

Multiparous Angus and crossbred Angus cows were used to determine the effect of induced endometritis on plasma concentrations of 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) and progesterone (P4) and on duration of the estrous cycle of treatment. Beginning on the day of calving (d 0), blood samples were collected on alternate days. On three consecutive days, ranging from d 8 to 14 of the first postpartum estrous cycle, uterine horns were inoculated transcervically with either 3 x 10(9) colony forming units (cfu) of Actinomyces pyogenes and 1.5 x 10(9) cfu of beta-hemolytic Escherichia coli (treated; n = 9) in sterile PBS or with sterile PBS alone (control; n = 9). Samples of uterine fluid were collected by transcervical aspiration twice weekly from just before the start of each series of inoculations until the end of the experiment. Endometrial biopsies were collected transcervically between d 4 to 6 and 11 to 13 after inoculation. Based on clinical observations and results of bacterial cultures, all treated cows developed acute uterine infections. Controls did not develop uterine infections. Endometrial biopsies indicated that there were no significant diffuse or focal cellular reactions in response to the infection. The interestrous interval was greater (P less than .0003) for treated (27.7 +/- 1.0 d) than for control (20.6 +/- 1.0 d) cows, but P4 concentrations were similar between the two groups. Mean PGFM concentration and PGFM profiles were similar (P greater than .10) between treated and control cows before bacterial infusions. Bacterial infusions increased mean PGFM concentration (P less than .0001) and changed the shape of the PGFM profile (P less than .02).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
