ABSTRACT Biocontrol activity of Candida saitoana and its interaction with Botrytis cinerea in apple wounds were investigated. When cultured together, yeast attached to Botrytis sp. hyphal walls. In wounded apple tissue, C. saitoana restricted the proliferation of B. cinerea, multiplied, and suppressed disease caused by either B. cinerea or Penicillium expansum. In inoculated apple tissue without the yeast, fungal colonization caused an extensive degradation of host walls and altered cellulose labeling patterns. Hyphae in close proximity to the antagonistic yeast exhibited severe cytological injury, such as cell wall swelling and protoplasm degeneration. Colonization of the wound site by C. saitoana did not cause degradation of host cell walls. Host cell walls in close contact with C. saitoana cells and B. cinerea hyphae were well preserved and displayed an intense and regular cellulose labeling pattern. In addition to restricting fungal colonization, C. saitoana induced the formation of structural defense responses in apple tissue. The ability of C. saitoana to prevent the necrotrophic growth of the pathogen and stimulate structural defense responses may be the basis of its biocontrol activity.  相似文献

 产几丁质酶枯草芽孢杆菌G3菌株的固体培养物在黄瓜灰霉病菌和番茄叶霉病菌抑菌试验中证实,抑菌活性物质存在于过滤上清液中,它们是从酸沉淀物中提取出的伊枯草菌素、生物表面活性素和存在于盐析粗蛋白中的几丁质酶。在叶霉孢子萌发试验中,伊枯草菌素微弱地抑制孢子萌发但强烈破坏芽管和新生菌丝;生物表面活性素和几丁质酶则强烈抑制孢子萌发并长久性地抑制芽管伸长。在PDA平板上的灰霉菌丝抑菌试验中,伊枯草菌素抑制菌丝生长,引发菌丝顶端膨大,形成泡囊,泡囊破裂后原生质外泄;几丁质酶抑制菌丝生长,引发产生不规则的菌丝团;生物表面活性素在平皿上对菌丝则不显示出抑菌活性。  相似文献

Infested wooden boxes, previously used for carrot storage, were sampled in four commercial carrot production farms in Bradford Marsh, Ontario, and screened for fungal occurrence. At least 128 and 465 fungal isolates were recovered from these boxes in 2001 and 2002, respectively, and were classified into 10 taxonomic groups, including Alternaria spp., Aspergillus spp., Botrytis cinerea , Fusarium spp., Mucor spp., Penicillium spp., Rhizoctonia carotae , Rhizopus spp., Sclerotinia sclerotiorum and Trichoderma spp. A subsample of 27 putative pathogenic isolates was further tested for the ability to cause disease on carrots and to colonize wood surfaces under growth room and cold storage conditions. Approximately 60% of the taxa growing on wood caused lesions upon contact with intact carrots in cold storage. Isolates of S. sclerotiorum , B. cinerea and R. carotae caused the most severe diseases, developed most extensively on wooden surfaces in cold storage, and represented 12% of the recovered fungi. Isolates of Alternaria spp., Aspergillus spp., Fusarium spp., Mucor spp., Penicillium spp., Rhizopus spp. and Trichoderma spp. caused negligible or no disease on carrots and represented 88% of recovered fungi. Several of these fungi, however, showed potential to colonize wooden surfaces and cause disease on sliced carrots. This study suggests that pathogenic inocula occurring on used wooden boxes can initiate disease upon contact with healthy carrots and reusing infested boxes can affect carrots in storage.  相似文献

ABSTRACT Ulocladium atrum (strain 385) consistently reduced Botrytis cinerea sporulation on necrotic fragments of strawberry leaves. On these tissues, two strains of U. atrum (isolates 18558 and 18559) showed lower antagonistic activities than the reference strain 385. Colonization of strawberry leaflets by the three U. atrum strains appeared similar in the absence of B. cinerea, whether quantified by chitin or immunological assays. The second method (based on anti-U. atrum antibodies) revealed that strawberry leaflet colonization by U. atrum 385 was better than by the other U. atrum strains in the presence of B. cinerea. An immunoassay using anti-B. cinerea antibodies revealed that the colonization of B. cinerea in tissues was lower in the presence of U. atrum 385 than with the two other U. atrum strains. The enzymatic activities produced by U. atrum 385 during the colonization phases of necrotic tissues were compared to B. cinerea and U. atrum strains 18558 and 18559. U. atrum 385 had the highest lipase, pectate lyase, and cellobiase activities while B. cinerea had the highest endo-beta-1,4-glucanase activity. The study of lytic activities hydrolyzing the fungal cell wall revealed higher beta-1,3-glucanase activity with U. atrum 385, which was stimulated by B. cinerea on necrotic strawberry leaflets. These results suggest that plant and fungal cell wall-degrading enzymes produced by U. atrum 385 may play a complementary role in the competitive colonization of dead strawberry leaves against B. cinerea.  相似文献

丁香酚是从植物中提取的天然化合物。采用菌丝生长速率法测定了其对多种植物病原真菌的抑制作用以及田间分离得到的7株灰葡萄孢Botrytis cinerea对丁香酚的敏感性,同时测定了丁香酚对B.cinerea 03 孢子和菌核的抑制活性。结果显示,丁香酚对多种植物病原真菌均有不同程度的抑制作用;灰葡萄孢不同菌株对丁香酚的敏感性不同,EC50值在29.97 ~83.62 μg/mL之间;丁香酚可抑制灰葡萄孢产孢,但对孢子萌发无影响,可抑制其菌核的产生和萌发。荧光染色结果表明丁香酚能破坏灰葡萄孢菌丝细胞膜。利用原子吸收光谱法和紫外分光光度法分别测定丁香酚处理前后菌丝培养液中K+浓度和OD260值变化,结果显示处理后K+浓度和OD260值均升高。综上所述,细胞膜可能是丁香酚对灰葡萄孢菌丝的作用位点。  相似文献

ABSTRACT A spatially explicit model describing saprophytic colonization of dead cyclamen leaf tissue by the plant-pathogenic fungus Botrytis cinerea and the saprophytic fungal antagonist Ulocladium atrum was constructed. Both fungi explore the leaf and utilize the resources it provides. Leaf tissue is represented by a two-dimensional grid of square grid cells. Fungal competition within grid cells is modeled using Lotka-Volterra equations. Spatial expansion into neighboring grid cells is assumed proportional to the mycelial density gradient between donor and receptor cell. Established fungal biomass is immobile. Radial growth rates of B. cinerea and U. atrum in dead cyclamen leaf tissue were measured to determine parameters describing the spatial dynamics of the fungi. At temperatures from 5 to 25 degrees C, B. cinerea colonies expanded twice as rapidly as U. atrum colonies. In practical biological control, the slower colonization of space by U. atrum thus needs to be compensated by a sufficiently dense and even distribution of conidia on the leaf. Simulation results confirm the importance of spatial expansion to the outcome of the competitive interaction between B. cinerea and U. atrum at leaf scale. A sensitivity analysis further emphasized the importance of a uniform high density cover of vital U. atrum conidia on target leaves.  相似文献

ABSTRACT The exo-beta-1,3-glucanase (EC 3.2.1.58) activity of Pichia anomala strain K, an antagonistic yeast of Botrytis cinerea on postharvest apples, was studied in a synthetic medium supplemented with laminarin, a cell wall preparation (CWP) of B. cinerea, or glucose. The highest enzyme activity was detected in culture media containing a CWP of B. cinerea as the sole carbon source, whereas the lowest activity was observed in culture media supplemented with glucose. Exoglc1, an exo-beta-1,3-glucanase, was purified to homogeneity from culture filtrates of strain K containing a CWP. The molecular mass of exoglc1 was estimated to be under 15 kDa. Optimum activity of exoglc1 was recorded at 50 degrees C and pH 5.5. The exoglc1 K(m) value was estimated at 22.4 mg/ml. Exoglc1 showed in vitro a stronger inhibitory effect on germ tube growth of B. cinerea than on conidia germination and caused morphological changes such as leakage of cytoplasm and cell swelling. Exo-beta-1,3-glucanase activity was detected on apples treated with strain K and was similar to exoglc1 on the basis of activity on native gel. Moreover, the addition of a CWP to a suspension of P. anomala stimulated both in situ exo-beta-1,3-glucanase activity and protective activity against the pathogen, strengthening the hypothesis that exo-beta-1,3-glucanase activity is one of the mechanisms of action involved in the suppression of B. cinerea by P. anomala strain K.  相似文献

Fungal development and plant responses were examined in detached leaves and mid-bulb scales of Allum cepa. Following inoculation with suspensions of 105 conidia/ml distilled water Botrytis squamosa consistently produced spreading lesions in leaves and bulb scales. B. allii produced spreading lesions at most sites in bulbs but was very inconsistent in its infection of leaves; lesions were often confined to inoculation sites. Limited lesions were usually produced by B. cinerea but R. fabae failed to produce symptoms at most sites. Extensive colonization by B. allii and B. tauamosa required rapid penetration and totally necrotrophic fungal growth. During development of a spreading lesion, plant cell walls became very swollen around intramural hyphae and wall swelling appeared to precede epidermal cell death. Resistance to colonization was due to poor germination, failure to produce distinct infection hyphae (associated with accumulation of deposits of granular reaction material [RM] in underlying live cells) or restriction of infection ryphae amongst small groups of dead cells (limited lesion formation). Only B. fabae germinated poorly, and germ-tubes produced often failed to attempt penetration but grew over the leaf or bulb scale surface. Reducing numbers of conidia increased the frequency of sites associated with RM accumulation; granular deposits being particularly common at sites inoculated with low numbers of B. allii conidia. Electron microscopy revealed that RM granules were osmiophilic aggregates formed between the plasma membrane and epidermal cell wall. In the absence of RM, growth of avirulent species was restricted within the swollen walls of dead epidermal cells. Results ae compared with those from studies on tulip and broad bean leaves.  相似文献

Tomato cultivar Moneymaker was independently inoculated with Alternaria alternata, Cunninghamella elegans, Fusarium culmorum, F. oxysporum f.sp. lycopersici, F. oxysporum f.sp. pisi and Stromatinia gladioli and analysed ultrastructurally. The extent and amount of superficial fungal growth on tomato roots was similar but C. elegans , a saprophyte, was exceptional in that hyphae were not closely appressed to plant surfaces and did not adhere to plant cell walls.
In general, the type of plant responses to fungal colonization and infection were similar in all of the interactions studied, with the exception of C. elegans which did not infect tomato root tissue. The failure to penetrate tomato roots by C. elegans may have been associated with the lack of hyphal adhesion to plant cell walls. Migration of cytoplasm and wall apposition/penetration papilla formation were regularly observed in tomato root tissue beneath appressed hyphae and at sites of fungal infection. Specific cellular reactions in the exodermis, namely the formation of wall 'inclusions' and appearance of 'sensitive' cells, indicated that exodermal cells were particularly responsive to fungal challenge.
Fusarium oxysporum f.sp. lycopersici , a pathogen of tomato, invaded tomato root tissue more extensively than the other fungi inoculated onto tomato roots. Infection of tomato by the other fungi studied was variable, and the extent and success of fungal invasion was tentatively associated with their necrotrophic capability and typical host range.  相似文献

研究了新型环烷基磺酰胺类化合物N-(2,4,5-三氯苯基)-2-氧代环己烷基磺酰胺(简称化合物108)对灰葡萄孢菌丝生长、孢子形成和萌发以及菌核产生等不同生育阶段的抑制作用及其对菌丝致病力和形态结构的影响。结果表明:化合物108对灰葡萄孢菌丝生长和孢子形成及孢子萌发具有明显的抑制作用,其EC50值分别为6.90、4.70和4.11μg/mL;菌核形成受到明显抑制,当药剂质量浓度达20μg/mL时,无菌核产生。经化合物108处理后的灰葡萄孢菌丝致病力下降,40μg/mL处理的菌丝致病力显著低于对照。超微结构观察结果表明,化合物108能导致灰葡萄孢菌丝萎缩、塌陷和变形,菌体细胞壁增厚、皱缩及分层。  相似文献

The distribution of N-acetylglucosamine residues in the cell wall of the white-rot pathogenic fungus, Rigidoporus lignosus, was studied by using gold labelled wheatgerm agglutinin bound to ovomucoid-colloidal gold. Ultrastructural investigation of R. lignosus-infected root tissues of Hevea brasiliensis showed a modification of the fungal cell wall throughout the infection process. Gold particles were found to occur on both thick- and thin-walled hyphae of R. lignosus rhizomorphs at the root surface. Walls of hyphae that had penetrated the roots were only labelled when they were out of the host cell, suggesting that modification of chitin molecules may be related to the excretion of host cell wall degrading enzymes. Variation in the distribution of gold particles was observed over hyphal walls of both colonized phellem and xylem cells. The observation that N-acetylglucosamine residues were released in the host cell cytoplasm suggests that lytic enzymes alter the fungal cell walls. Released chitin oligosaccharides may play a role in the induction of the root's defence system against fungal attack.  相似文献

A serological test has been developed that allows the early detection of infection of young petals by Sclerotinia sclerotiorum , an important pathogen of rapeseed. Two steps were required to obtain an antiserum sufficiently specific for S. sclerotiorum. Soluble mycelial extracts of S. sclerotiorum were used to produce the first generation polyclonal antiserum. This was not specific for S. sclerotiorum in double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) and allowed the screening of cross-reacting fungal species such as Botrytis cinerea , a pathogen commonly present on rapeseed petals. Using a polyclonal anti- B. cinerea serum enabled the absorption, by serial cycles, of S. sclerotiorum antigens common to B. cinerea. Residual antigens were then used as immunogens for the production of two second generation antisera (S1 and S2) which were then tested by DAS-ELISA. Cross-reactions with B. cinerea decreased with purification cycles of the immunogen whereas Cross-reactions with some unrelated fungi slightly increased. S. sclerotiorum and B. cinerea were distinguishable using antiserum S2.  相似文献

发展虫生真菌的抗药性是协调生防与化防,稳定真菌杀虫剂应用效果的一个重要策略。本研究从代表性地区采集植物病原样本,分离得到35株菌株,鉴定为3属5种,即灰葡萄孢霉Botrytis cinerea、扩展青霉Penicillium expansum、指状青霉Penicillium digitatum、长柄链格孢菌Alternaria longipes和茄链格孢菌Alternaria solani。应用菌落直径法测定其对苯菌灵的抗性,结果表明:供试的11株灰葡萄孢霉菌株中,最高EC50为936.2670μg·mL^-1;12株青霉属菌株中,最高EC50为0.0384μg·mL^-1;12株链格孢菌株中,最高EC50为703.5557μg·mL^-1。其中灰霉和链格孢属菌株中有4株EC50大于600μg·mL^-1的高抗菌株,即灰葡萄孢霉的菌株SD2和SD1以及茄链格孢的菌株CC2-2和LF1-2,这4株菌株可作为遗传转化抗性基因克隆的基础材料。  相似文献

植物内生真菌能代谢产生多种生物活性物质,本研究从浙江建德千年古树罗汉松 Podocarpus macrophyllus中分离获得1株具有抗菌活性的菌株ZJDH255。通过形态和ITS rDNA序列分析,鉴定为稻黑孢霉Nigrospora oryzae。抑菌活性测试表明,菌株ZJDH255发酵粗提物对植物病原真菌灰葡萄孢Botrytis cinerea和瓜类炭疽菌Colletotrichum orbiculare具有较强抑制活性。采用正相硅胶和葡聚糖凝胶Sephadex LH-20进行柱层析从发酵粗提物中分离获得3个具抗真菌活性的化合物,通过质谱和核磁共振波谱技术将其结构鉴定为蜂蜜曲菌素mellein、灰黄霉素griseofulvin和去氯灰黄霉素dechlorogriseofulvin。  相似文献

Most potential fungal antagonists have been identified only after exhaustive non-selective screening. Bacteria antagonistic to Botrytis cinerea were selected using a simple method based on techniques developed for trace enrichment studies, where a crude cell wall preparation from B. cinerea was used as a selective substrate and bacteria were isolated based on growth on cell wall agar media. Most of the 52 bacterial isolates that were obtained from the surface of organically grown strawberry fruit and could grow on B. cinerea wall media in culture showed some ability to inhibit the growth of the fungus. Potato dextrose agar seeded with B. cinerea spores (104ml–1) were challenged with each of these bacterial isolates at concentrations of 109 and 106 colony forming units ml–1. Eleven of the 52 isolates initially recovered, subsequently demonstrated strong antagonism in vitro and were selected for additional screening tests on strawberry fruit. All 11 isolates reduced grey mold rot incidence on fruit in storage. Three of the best isolates were tested in limited field trials, and also reduced grey mold rot on fruit under field conditions.  相似文献

为探究美国进境苜蓿滑刃线虫对真菌的取食特性以及对植物的寄生性,通过接种试验观察该线虫在链格孢、灰葡萄孢、禾谷镰刀菌、拟盘多毛孢和核盘菌等5种真菌,以及在菊花、大豆、苜蓿和三叶草等4种植物8个品种上的繁殖情况。结果表明,苜蓿滑刃线虫能取食5种真菌的菌丝体,但是在各真菌上的繁殖系数(Rf)有明显差异,在链格孢和拟盘多毛孢上能快速繁殖且数量很高,在灰葡萄孢上繁殖速度稍慢,但存活时间较长。接种苜蓿滑刃线虫后,大豆品种新叶出现轻微畸形或褐色病斑,苜蓿品种新叶出现畸形萎缩,而菊花和三叶草叶片没有症状;该线虫在各植物上的Rf值均低于1,表明这些植物不是合适的寄主。研究结果可为我国口岸实施外来植物线虫的检疫措施提供参考。  相似文献

本文采用单孢分离法对四川汉源和山东烟台等地采集的樱桃果实进行了采后灰霉病的病原菌分离和鉴定;采用区分剂量法分别测定了菌株对苯并咪唑类杀菌剂甲基硫菌灵、乙霉威和二甲酰亚胺类杀菌剂腐霉利的敏感性,并进一步分析了抗药性菌株的分子机制。结果表明,分离得到的54株樱桃采后灰霉病菌均为灰葡萄孢Botrytis cinerea,对甲基硫菌灵的总抗性频率高达79.6%,其中甲基硫菌灵抗性-乙霉威敏感 (BEN R1) 菌株频率为 25.9%;甲基硫菌灵-乙霉威双重抗性菌株 (BEN R2) 频率为53.7%;检测到腐霉利抗性菌株 (DCF R) 9 株,频率为16.7%。甲基硫菌灵抗性菌株在β-tubulin基因上的突变共有2种类型: BEN R1抗性菌株中,第198位密码子发生点突变 (GAG→GCG),编码氨基酸由Glu (E)突变成缬氨酸Ala (A);在BEN R2抗性菌株中,第198位密码子发生点突变 (GAG→GTG),编码氨基酸由Glu (E)突变成缬氨酸Val (V)。DCF R菌株在BcOS1的第365位密码子由ATC突变成AAC或AGC,导致编码的氨基酸由异亮氨酸Ile (I)突变成天冬酰胺Asn (N)或丝氨酸Ser (S)。本研究表明樱桃采后灰霉病菌对甲基硫菌灵和腐霉利存在不同程度抗性,应在加强抗药性监测的同时与其他类型杀菌剂交替使用,延缓抗药性发展。  相似文献

The present study assayed the effect of six isothiocyanates (ITCs), produced by the enzymatic hydrolysis of glucosinolates, on fungal pathogens of pear. Sample pear fruits were artificially inoculated through induced wounds with conidial suspensions of Botrytis cinerea Rhizopus stolonifer Monilinia laxa Mucor piriformis or Penicillium expansum and were then treated with ITCs. Of the six ITCs tested, the ITC from glucoraphenin showed the highest effectiveness after 6 days at 20°C, against M. laxa B. cinerea and M. piriformis . The effectiveness of the ITC from glucoraphenin against M. laxa was assayed in two further trials to test the effect of ITC concentration on different concentrations of inoculum and to determine the duration of the curative effect of this ITC. ITC concentration directly affected fungus control capacity. The highest ITC concentration (3.6 mg mL−1) afforded pathogen control at the highest level of pathogen concentration (106 conidia mL−1) after 6 days at 20°C. Its curative effect was evident up to 40 h after inoculation.  相似文献

Bacterial antagonists against Botrytis cinerea were isolated from different Brassica spp. and identified. All isolates showing in vitro antagonism at 4°C were shown to be either fluorescent pseudomonads or Serratia spp. In vitro antagonism against B. cinerea and Alternaria brassicicola was found to depend on the temperature and concentration of nutrients in the medium.
Bacterial strains which showed in vitro antagonism were tested for in vivo antagonism at 4°C against B. cinerea and A. brassicicola using a leaf disc bioassay. Pseudomonas fluorescens isolates CL42, CL66, CL82 and Serratia plymuthica strain CL43 showed inhibition of Botrytis growth on leaf discs; P. fluorescens isolate CL74 and all Serratia liquefaciens isolates exhibited intermediate control. All other fluorescent pseudomonad isolates showed poor control or caused rotting of the cabbage tissue.  相似文献

ABSTRACT The potential of the endophytic bacterium Serratia plymuthica strain R1GC4 in stimulating defense reactions in cucumber (Cucumis sativus) seedlings inoculated with the soilborne pathogen Pythium ultimum was explored at the cellular level. Bacterial treatment prior to Pythium inoculation resulted in less seedling disease development as compared with that in nontreated control plants, in which typical root symptoms were visible by 3 days after inoculation with the pathogen. Histological investigations of root samples revealed striking differences in the extent of plant defense reactions between bacterized and nonbacterized plants. These observations were further confirmed at the ultrastructural level with the demonstration that restriction of fungal colonization to the outermost root tissues of bacterized seedlings correlated with the deposition of enlarged callose-enriched wall appositions at sites of potential pathogen penetration and the accumulation of an osmiophilic material in the colonized areas. Hyphae of the pathogen, surrounded by this electron-opaque material, exhibited considerable changes including cytoplasm disorganization and, in many cases, loss of the protoplasm. However, labeling with the beta-1,4-exoglucanase resulted in a regular labeling of Pythium cell walls, even at a time when these walls were entirely coated by the osmiophilic material. This material was also found to infiltrate into the invading hyphae to form either an internal coating of the cell wall or a network of polymorphic droplets in the area previously occupied by the cytoplasm. Cytochemical investigations revealed that callose, pectin, and cellulose appeared in the wall appositions. In addition, glucosides, lipids, and phenolics were detected in the electron-dense aggregates forming the core of most wall appositions. Finally, galactose residues were among the minor polysaccharidic compounds detected in the wall appositions. Evidence is provided in this study showing that treatment with S. plymuthica sensitizes susceptible cucumber plants to react more rapidly and more efficiently to Pythium attack through the formation of physical and chemical barriers at sites of potential fungal entry.  相似文献
