BACKGROUND: The potato psyllid is a serious pest of potatoes. Sampling plans on potatoes for the potato psyllid have yet to be developed, thus the authors' objectives were (1) to determine the most efficient within‐plant sampling unit, (2) to determine the spatial dispersion of potato psyllids in potato fields and (3) to develop a binomial sequential sampling plan for this pest. RESULTS: Significantly more potato plants were infested with potato psyllids on the edges of the field, and significantly more plants were infested with psyllids on the ‘top’ and ‘middle’ of the potato plant. Significantly more psyllids were also found on the undersides of leaves. The potato psyllid has an aggregated distribution in potato fields. Binomial sequential sampling plans were developed for three action thresholds representing 0.5, 1 and 5 psyllids per plant. The average sample numbers for these action thresholds were between 12 and 16 samples, depending on the action thresholds. However, based on the shape of the operating characteristic curve, the 0.5 and 1 sampling plans were more reliable than the 5 psyllids per plant plan. CONCLUSION: The binomial sequential sampling plans are useful for detecting potato psyllids at low levels of infestation, which will be useful for pest management purposes. Studies were conducted to evaluate the effect of a kaolin-based particle film against the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae). The effect of the particle film on development, survival and host plant selection was tested on calabrese seedlings, Brassica oleracea italica Plenck cv. Fiesta F(1), under controlled environmental conditions in the laboratory. Survival to adulthood was significantly reduced and development time increased on kaolin-treated compared with water-treated host plants. The hatch rate of neonate P. xylostella larvae from eggs laid on an artificial substrate was unaffected by the particle film applied after oviposition. Experiments showed that, when given the choice, significantly fewer larvae first made contact with the kaolin-treated compared with the water-treated plant material and that after 24 h there were still significantly fewer larvae present on kaolin-treated leaves. Adult females, however, deposited more eggs on host plants coated with the kaolin particle film. As part of this series of experiments the water control treatment was compared with plant material which had been sprayed with kaolin on the upper surface only and on both leaf surfaces. In general, any observed treatment effect was enhanced with an increase in the particle film coverage. The results warrant the extension of investigations of kaolin-based particle films to field-based P. xylostella management strategies.

Candidatus Liberibacter solanacearum” (Lso), transmitted by the potato psyllid (Bactericera cockerelli), is the causal agent of potato zebra chip, but can also infect other solanaceous plants, including peppers. Studies were conducted to investigate whether Lso could be transmitted to the next generation of plants through seeds from infected pepper plants. In 2014, jalapeno pepper plants were infested with psyllids carrying a mixture of Lso A and B (AB) at the AgriLife Research Station at Bushland. The study was again conducted in 2019 and pepper plants were infested with psyllids carrying Lso B or Lso AB. In each of the studies, noninfested plants served as controls. At harvest, fruits were collected and tested for the presence of Lso using quantitative PCR. Seeds from infected fruits were then tested for Lso. Overall, the percentage of seeds that tested positive for Lso ranged from 33% to 70%. However, Lso detection in embryos ranged only from 0% to 8%. Seed samples from Lso-positive fruits were planted in the greenhouse to determine the impact of Lso on emergence and the incidence of Lso in emerged plants. Although plant emergence differed between some of the seeds obtained from Lso-positive and -negative fruits, the overall impact of Lso on plant emergence was not consistent. However, of the 182 plants that emerged from seeds collected from infected fruits, none was positive for Lso, suggesting that seeds are unlikely to serve as sources for new Lso infections and their impact on disease epidemiology is negligible.

Carrot psyllid Trioza apicalis was recently found to carry the plant pathogenic bacterium ‘Candidatus Liberibacter solanacearum’ (CLs). To confirm the transmission of bacteria by the psyllids and to dissect the symptoms caused in carrot plants by psyllid feeding and CLs infection, a greenhouse experiment with single psyllids feeding on separate plants was performed. A positive correlation was found between the amount of CLs bacteria in the psyllids and in the corresponding plants exposed to feeding, indicating CLs transmission. The female psyllid feeding caused more severe damage than male feeding, and resulted in a substantial decrease in the root weight. Female psyllid feeding also significantly reduced the carrot leaf weight and increased the number of curled leaves. The number of curled leaves was also increased by the nymphs when their number exceeded 10 per plant. A high titre of CLs bacteria significantly reduced root weight, while not affecting the weight or number of the leaves. However, the amount of CLs correlated with the number of leaves showing discolouration symptoms. Microscopy of infected carrot plants revealed that the phloem tubes throughout the whole plant, from leaf veins to the root tip, were colonized by bacteria. The bacterial cells appeared to be long and thin flexible rods with tapering ends and a transversally undulated surface. Microscopy also revealed collapsed phloem cells in the infected carrots. Damage in the phloem vessels is likely to reduce the sucrose transport from source leaves to the root, explaining the observed leaf discolouration and reduction in root weight.

Laboratory and field experiments were conducted to evaluate the efficacy of kaolin particle film on winterform oviposition of the pear psyllacacopsyllapyri (L.) (Hemiptera: Psyllidae). Kaolin particle film (kpf) treatment significantly reduced winterform oviposition rates in both choice and no-choice tests in the laboratory. In choice tests, overwintered females laid 11 times more eggs on water-treated dormant shoots than on shoots treated with particle film, within 21 days after treatment. In no-choice tests, kpf treatment also significantly reduced egg numbers compared with water-treated controls. The results of field trials carried out in southwestern Turkey (Antalya) during the late winter — early spring of 2004 and 2005 showed very good efficacy of kpf treatment in comparison with water-treated control. Kpf treatment (just before egg laying, during the dormant-postdormant period) delayed egg-laying activity in overwintered females for up to 3 weeks from the beginning of the experiment and continued to suppress oviposition significantly in comparison with watertreated control up to 4 weeks. Our overall results suggest that kpf treatment may be useful in pear psylla management programs by decreasing winterform oviposition, particularly in conjunction with other control tactics.

Citrus huanglongbing (HLB), previously called greening, is a serious citrus disease in Asia, eastern and southern Africa. It is caused by Candidatus Liberibacter asiaticus (Las), a phloem-limited, nonculturable bacterium transmitted by the Asian citrus psyllid ( Diaphorina citri ) in Asia. A PCR-based assay was developed for monitoring Las in vector psyllids using a rapid DNA extraction from psyllid bodies and PCR amplification. The entire procedure for Las detection in psyllids can be completed within 5 h. Using this method, Las can be accurately detected in psyllid adults as well as nymphs in different instar stages. The assay is sensitive enough for Las detection in single-psyllid extract from adult, fifth, fourth and third instars. In a transovarial transmission experiment, Las was not detected in eggs or in offspring produced by Las-carrying psyllid females. In a retention test, the Las-carrying psyllids remained Las-positive for 12 weeks after they were moved to common jasmine orange, a Las-immune plant. From these experimental results it was concluded that Las persists in the Asian citrus psyllid vector, but is not transovarially transmitted by the vector. These data help in understanding epidemiological characteristics of Las and psyllids in citrus HLB.

Zebra chip, an emerging disease of potatoes, has recently been associated with ‘Candidatus Liberibacter solanacearum’ in New Zealand. The phloem-limited bacterium is known to be vectored by the tomato potato psyllid (Bactericera cockerelli). In this study, the role of tuber transmission in the spread of Ca. L. solanacearum was investigated by re-planting potato tubers infected with Ca. L. solanacearum in the absence of the psyllid. Nested PCR demonstrated that Ca. L. solanacearum could be transmitted from the mother tubers both to the foliage of growing plants and to progeny tubers, resulting in symptomatic and asymptomatic plants. Of 62 Ca. L. solanacearum-infected tubers four did not sprout symptomatic of zebra chip. A further two plants developed foliar symptoms associated with zebra chip during the growing season and died prematurely. Fifty-six of the infected tubers produced asymptomatic plants, although Ca. L. solanacearum was detected in the foliage of 39 of them indicative of transmission into asymptomatic progeny plants. At harvest, Ca. L. solanacearum was found in the daughter tubers of only five of the 39 asymptomatic plants, and only one of these plants was found to have zebra chip symptoms in the daughter tubers. Our results show that tuber transmission of Ca. L. solanacearum could play a role in the life cycle of this pathogen, providing a source for acquisition by Bactericera cockerelli and for movement of the pathogen to other regions of New Zealand via transport of seed tubers.

ABSTRACT With diseases caused by vector-borne plant pathogens, acquisition and inoculation are two primary stages of the transmission, which can determine vector efficiency in spreading the pathogen. The present study was initiated to quantify acquisition and inoculation successes of 'Candidatus Liberibacter solanacearum', the etiological agent of zebra chip disease of potato, by its psyllid vector, Bactericera cockerelli (Hemiptera: Triozidae). Acquisition success was evaluated in relation to feeding site on the host plant as well as the acquisition access period. Inoculation success was evaluated in relation to vector number (1 and 4) on the plants. Acquisition success was influenced by the feeding site on the plant. The highest acquisition success occurred when insects had access to the whole plant. The results of the inoculation study indicated that the rate of successfully inoculated plants increased with the vector number. Plants inoculated with multiple psyllids had higher bacterial titer at the point of inoculation. Although disease incubation period was significantly shorter in plants inoculated with multiple psyllids, this effect was heterogeneous across experimental blocks, and was independent of pathogen quantity detected in the leaflets 3 days postinoculation. Disease progress was not affected by bacterial quantity injected or psyllid numbers.

BACKGROUND

The tomato potato psyllid, Bactericera cockerelli (Šulc) is a new invasive pest in Western Australia, which may disperse across the whole of Australia within a few years and cause significant economic losses. Chemical control is the most widely used approach to manage B. cockerelli, but insect resistance, chemical residue and effects on non-target species have become an increasing concerned. Therefore, in this study, the biocontrol potential of variegated lady beetle, Hippodamia variegata (Goeze) was investigated. The impact of utilizing B. cockerelli as a food source on the predator's development and reproduction was assessed by formulating age-stage, two sex life tables. The predatory potential of H. variegata on B. cockerelli nymphs was assessed in a closed arena and the effects of releasing H. variegata for the control of B. cockerelli were then evaluated.

RESULTS

H. variegata could successfully develop and oviposit when feeding on B. cockerelli. However, both survival and the rate of development were higher for H. variegata feeding on Myzus persicae (Sulzer) than B. cockerelli or a mixed population of B. cockerelli and M. persicae. A type II functional response was observed for H. variegata. In the greenhouse, the releases of H. variegata larvae reduced the number of B. cockerelli nymphs by up to 66% and adults by up to 59%, which positively influenced the plant chlorophyll content and biomass.

CONCLUSIONS

This study demonstrated the potential of the resident generalist predator, H. variegata as a biocontrol agent for the invasive pest, B. cockerelli, which may help improving current management strategies. © 2022 The Authors. 《EPPO Bulletin》2017,47(3):513-523

Specific scope

This Standard describes a national regulatory control system for Bactericera cockerelli and the bacterial pathogen ‘Candidatus Liberibacter solanacearum’ the cause of zebra chip disease in potato. The scope is as follows:
  • Exclusion from the EPPO region of B. cockerelli an efficient vector of ‘Ca. L. solanacearum’ within solanaceous crops (e.g. potato, tomato)
  • Eradication of incursions of B. cockerelli
  • Exclusion from the EPPO region of ‘Ca. L. solanacearum’ haplotypes A and B. Although reference will only be made to haplotypes A and B, the Standard would also apply to new non‐European haplotypes of ‘Ca. L. solanacearum’ which may have different host ranges, or which may be vectored more efficiently by psyllids which are widespread in the region.
The reduction of the risk of spreading ‘Ca. L. solanacearum’ haplotypes C, D and E to potato production systems and potatoes being moved within the EPPO region may be recommended in future when more information is available but is not covered in this Standard.

The possible transovarial transmission of two phytoplasmas, ' Candidatus Phytoplasma mali' and ' Candidatus Phytoplasma prunorum', through their respective psyllid vectors Cacopsylla melanoneura and Cacopsylla pruni , was investigated. Different life stages of the progeny of infected female psyllids were analysed by PCR detection of phytoplasma DNA. While ' Ca. Phytoplasma mali' could not be detected in any of the C. melanoneura life stages tested, ' Ca. Phytoplasma prunorum' could be detected in eggs, nymphs and newly emerged adults of C. pruni . Infectivity tests using both nymphs and newly emerged adults of C. pruni showed that ' Ca. Phytoplasma prunorum' inherited from infected females can be transmitted to healthy plum plants. Although further validations are required, these findings open up new perspectives on the study of the epidemiology of diseases associated with European stone fruit yellows.

Experiments were carried out in the laboratory to assess the insecticidal effect on Myzus persicae Sulzer of different oils applied alone or combined with imidacloprid or pirimicarb. The oils tested were a horticultural mineral oil, a refined rapeseed oil, a refined soya oil and a raw fish oil. When the oils were sprayed alone on pepper plants infested with M. persicae, mineral oil caused the highest mortality of aphids (over 80%). Applied before aphid infestation of pepper leaves and in mixture with low doses of imidacloprid (at one-fifth of the dose recommended by the manufacturer) and pirimicarb (at one-tenth of the dose recommended by the manufacturer), the oils did not significantly increase the toxicity of the insecticides alone. However, sprayed on aphid-infested pepper plants, the mortality rates achieved by imidacloprid/mineral oil and imidacloprid/rapeseed oil mixtures were significantly higher than those achieved by imidacloprid alone at 16 and 24 h. In a field experiment the effect on the incidence of the potato virus (PVY) of the oils in combination with imidacloprid was determined. Mineral oil, rapeseed oil and soya oil were sprayed eight times onto seed potato plants treated with imidacloprid before sowing. Mineral oil reduced PVY-infected plants by 60% and rapeseed oil by 40% compared with plots treated with imidacloprid. The oils applied as 10 ml litre-1 emulsions in water did not cause symptoms of phytotoxicity on the potato plants, and yield was not reduced.

Intercropping systems are practiced to reducing pest density, enhancing predator's diversity and stabling crop yield. We evaluated the effect of strip-intercropping potato and annual alfalfa on populations of Leptinotarsa decemlineata Say, predator's biodiversity and potato yield over two seasons (2014 and 2015). Our results revealed that the densities of egg masses, eggs, larvae and adults of Colorado potato beetle (CPB) were significantly lower in intercrops than in monoculture. The main CPB predators recorded on potato plants (ladybirds and the green lacewing) showed a significant increase in the density at each of the three intercrops than in monoculture. The percentage of dry tubers weight loss was 40.9%–42.6% in monoculture, 16.3%–18.7% in 6P: 2A and <10% in 2P: 2A and 4P: 2A in two studied years. Our results suggest that strip-intercropping potato with annual alfalfa, particularly in 2P: 2A and 4P: 2A patterns may be an effective way in integrated management of CPB for reducing pest density, enhancing the presence of predators in potato fields and improving potato yield.

Background: The efficacy of aerial electrostatic‐charged sprays was evaluated for spray deposit characteristics and season‐long control of sweet potato whitefly (SWF), Bemisia tabaci Genn. biotype B (aka B. argentifolii Bellows & Perring), in an irrigated 24 ha cotton field. Treatments included electrostatic‐charged sprays at full and half active ingredient (AI) label rate, uncharged sprays and conventional sprays applied with CP nozzles at full label rate with several different insecticides. Results: Spray droplet size was significantly smaller for electrostatic‐charged sprays than for conventional sprays in top‐ and mid‐canopy locations. The seasonal mean numbers of viable eggs and live large nymphs on cotton treated with electrostatic‐charged sprays were comparable with those on cotton treated with conventional applications. Lethal concentration (LC50) for adults for electrostatic‐charged sprays was comparable with that for conventional sprays. Conclusion: The amenability of electrostatic‐charged sprays to a wide array of pesticides with different chemistries should be a useful tool in combating insect resistance. Results reported here suggest that the potential exists for obtaining increased efficacy against whiteflies using an electrostatic spray charging system, and that additional research will be required to improve charge‐to‐mass (Q/M) ratio in order to increase deposition of pest control materials to the lower surfaces of cotton leaves where the whiteflies reside. Three haplotypes of the recently discovered bacterium species “Candidatus Liberibacter solanacearum” are described and related to geographic ranges. The first two are associated with Zebra Chip/Psyllid Yellows of potatoes and other solanaceous plants, vectored by the tomato/potato psyllid Bactericera cockerelli in North and Central America and New Zealand. The third is associated with diseased carrots in Finland and vectored by the carrot psyllid Trioza apicalis. The haplotypes are described by SNPs on the 16s rRNA, 16s/23s ISR and 50s rplJ and rplL ribosomal protein genes. These SNPs are inherited as a package across the three genes. Haplotype “a” has been found primarily from Honduras and Guatemala through western Mexico to Arizona and California, and in New Zealand. Haplotype “b” is currently known from eastern Mexico and northwards through Texas to south central Washington. These haplotypes show some range overlap in Texas, Kansas and Nebraska. The haplotypes are not yet known to elicit biological differences in the plant or insect hosts. These apparently stable haplotypes suggest separate bacterial populations of long standing.

Candidatus Liberibacter solanacearum”, a phloem-limited and Gram-negative bacterium that is spread from infected to healthy plants by psyllid insect vectors, is an economically important pathogen of solanaceous and carrot crops in the Americas, New Zealand and Europe. Three haplotypes of “Ca. L. solanacearum” have previously been described, two (LsoA and LsoB) in relation to solanaceous crops in the Americas and New Zealand and the third (LsoC) to carrots in Finland. Herein, we describe a fourth haplotype of this ‘Candidatus Liberibacter’ species (LsoD), also associated with carrots, but from Spain and the Canary Islands and vectored by the psyllid Bactericera trigonica. In addition, LsoC was confirmed in carrot and psyllid samples recently collected from Sweden and Norway. Phylogenetic analysis of the 16S rRNA gene suggests that two of the haplotypes, one in the Americas and the other in northern Europe are closer to each other in spite of a large geographic separation and host differences. Furthermore, during this study, potatoes with symptoms of zebra chip disease recently observed in potato crops in Idaho, Oregon and Washington states were analyzed for haplotype and were found to be positive for LsoA. This liberibacter haplotype was found in psyllids associated with the diseased potato crops as well. This finding contrasts with an earlier report of LsoB from psyllids in Washington which came from a laboratory colony originally collected in Texas.

The Chinese box orange (Severinia buxifolia) was shown by graft-inoculation and psyllid-transmission tests to be an alternative host of the bacterium causing citrus Huanglongbing (HLB). A PCR-based assay for detection of the HLB bacterium (HLBB) was used to monitor HLBB. In graft-inoculation tests, the Chinese box orange (CBO) grafted with HLBB-infected scions of Luchen sweet orange (LSO) were positive for HLBB, 2–3 months after grafting. The back-grafting test demonstrated that HLBB-infected CBO scions could transmit HLBB back to LSO hosts via grafting. In psyllid-transmission tests, psyllids (insect vectors) transmitted HLBB to CBO plants, in which HLBB could be detected 3–4 months after inoculation. Acquisition-access tests of psyllids revealed that HLBB-free psyllids can acquire HLBB from diseased CBO hosts and can transmit HLBB back to the LSO plants. A field survey verified the presence of HLBB-infected CBO plants in the vicinity of citrus orchards. In this paper, CBO is shown to be a susceptible host plant in which HLBB can exist and replicate. It is also a donor plant from which HLBB can be transmitted to citrus hosts by grafting or by psyllid vectors.
