为了探究海湾扇贝南部亚种(Argopecten irradians concentricus)的遗传多样性特征,通过全基因组重测序分析,系统解析了其群体遗传结构以及其与海湾扇贝北部亚种(Argopecten irradians irradians)间的遗传差异,共筛查出15059961个高质量单核苷酸多态性(single nucleotide polymorphisms,SNPs)位点。基于全基因组SNP的群体祖先成分推断、主成分分析和系统发生重构皆表明,南北亚种之间存在较为显著的遗传分化。其中,海湾扇贝南部亚种的近交水平和连锁不平衡程度更高。通过选择性清除分析,在海湾扇贝南部亚种中共鉴定出349个正选择基因,其中GHSR、HSF1、HABP2和Dna J等基因可能与其生长速度较快、耐热性较强以及免疫功能较强等环境适应性特征相关。该研究揭示了海湾扇贝亚种间遗传分化和选择特征,为扇贝的基因组研究和分子育种提供了数据支持。  相似文献

Triploidy was induced in the bay scallop, Argopecten irradians, by treating newly fertilized eggs with cytochalasin B. Treatment for 20 min duration 10 min after fertilization resulted in 66% triploidy of scallops in the 0.05 mg/l cytochalasin treatment group and 94% triploidy of scallops in the 0.1 mg/l cytochalasin treatment group, detected by measuring the relative DNA content of scallop granulocytes using flow cytometry. Individually labelled scallops of known ploidy were grown in the Damariscotta River estuary in south-central Maine, U.S.A. from May 24 to September 15, 1982. Mean adductor muscle weight in triploid scallops was 73% greater than that of diploid controls and mean total body tissue wet weight was 36% greater. Shell inflation was significantly greater in triploids, while shell height and length were unaffected. The indexes of weight and glycogen content of the adductor muscle were significantly greater in triploids at the end of the growing season. The majority of triploid scallops failed to ripen during the summer months, while diploid scallops ripened and spawned.  相似文献

Browning in the adductor muscle of bay scallop (Argopecten irradians) tends to occur during hot air drying due to polyphenoloxidase (PPO) and Maillard reaction and decreases the sensory quality and commercial value of bay scallop adductor muscles (BSAM). In the present study, the effects of glutathione on the browning inhibition of BSAM were evaluated. The PPO activity in BSAM was inhibited by 97.8 with 0.08% glutathione. The browning of BSAM pretreated with glutathione was evaluated during hot air drying. Compared with the control, BSAM pretreated with glutathione resulted in less drying time and browning during hot air drying. The results indicate that pretreatment with glutathione may be a promising method of BSAM browning inhibition during hot air drying.  相似文献

The harvest of bay scallops (Argopecten irradians) from Buzzards Bay, Massachusetts, U.S.A. undergoes large interannual fluctuations, varying by more than an order of magnitude in successive years. To investigate the extent to which these fluctuations may be due to yearly variations in the transport of scallop larvae from spawning areas to suitable juvenile habitat (settlement zones), a high‐resolution hydrodynamic model was used to drive an individual‐based model of scallop larval transport. Model results revealed that scallop spawning in Buzzards Bay occurs during a time when nearshore bay currents were principally directed up‐bay in response to a persistent southwesterly sea breeze. This nearshore flow results in the substantial transport of larvae from lower‐bay spawning areas to settlement zones further up‐bay. Averaged over the entire bay, the spawning‐to‐settlement zone connectivity exhibits little interannual variation. However, connectivities between individual spawning and settlement zones vary by up to an order of magnitude. The model results identified spawning areas that have the greatest probability of transporting larvae to juvenile habitat. Because managers may aim to increase scallop populations either locally or broadly, the high‐connectivity spawning areas were divided into: (i) high larval retention and relatively little larval transport to adjoining settlement areas, (ii) both significant larval retention and transport to more distant settlement areas, and (iii) little larval retention but significant transport to distant settlement areas.  相似文献

In this study, we used microsatellite‐based parentage analysis to assess the mating system of a mass spawning bay scallop, Argopecten irradians irradians. Parental assignment was performed on 237 offspring and 14 candidate parents. Overall, 95% of all offspring were unambiguously allocated to their putative parents based on information from six highly polymorphic loci. Offspring from both the self‐fertilization and cross‐fertilization style were observed, the large variation of self‐fertilization rate among individuals displays a mixed mode of mating. The high variances in reproductive success resulted in a decline in effective breeding numbers (Nb/N = 0.71). The self‐fertilization rate was 32.89% for this mass spawning and the coefficient of inbreeding was estimated at around 16.5% per generation. When selfed larvae were removed, the relationship between relatedness and individual reproductive success was proved insignificant between half‐sibings and unrelated, which suggest that selection pressures other than avoidance of inbreeding may be responsible for the evolution and maintenance of the self‐compatible mating system of A. i. irradians. These results lend insight to A. i. irradians cultivation in present China and provide a genetic basis for the development of more effective spawning techniques.  相似文献

Cross‐breeding has been successfully applied in agriculture and aquaculture industries to improve important production traits. In our previous studies, we produced the F1 hybrids with high heterosis in growth by crossing the bay scallops with the Peruvian scallops and selected a new strain, Bohai Red, from the F1 hybrids. In this study, we selected another new strain from the backcross (BC1) of the F1 hybrids with bay scallops. The base cohort (G0) was size‐selected from the BC1, and the G1, G2, G3 and G4 cohorts were produced with size‐selected brood stocks from the G0, G1, G2 and G3 cohorts respectively. Our results showed that average shell height, shell length, shell width and whole body weight of the G4 cohort were increased by 16.8%, 16.8%, 11.5% and 43.6%, respectively, compared with the unselected control group of bay scallop. This new strain has now become one of the major cultured Argopecten scallops in northern China waters and was later named QN‐2.  相似文献

In the present study, investigated were the preparation of water-soluble chitosan (WSC) by hydrolysis of original chitosan with commercial α-amylase and the suppressive effect of WSC on denaturation of bay scallop adductor muscle myofibrillar protein (Mf) during frozen storage at ?18°C for 12 months. The WSC (2.5- to 10-g dry weight) was added to 100 g of the Mf, and the changes in the Ca-ATPase activity, solubility, and sulfhydryl content were examined during frozen storage. The addition of WSC significantly reduced the denaturation of the Mf Ca-ATPase. Moreover, a significant increase in Mf solubility and sulfhydryl content was observed in the WSC-treated bay scallop adductor muscles compared with the control (p < 0.05) during frozen storage at ?18°C.  相似文献

We investigated strategies to enhance populations of bay scallops,Argopecten irradians irradians (Lamarck, 1819), in a presumablyrecruitment-limited natural habitat. At present, the Niantic Riverestuary supports only a minor bay scallop population that is harvestedrecreationally. Three enhancement strategies were evaluated; (1)collection and redistribution of natural spatfall, (2) introduction andover-wintering of hatchery-reared stock into natural habitat to providenew spawning stock, and (3) over-wintering of hatchery-reared stock insuspension culture for creation of mobile spawner sanctuaries. Anassessment of natural bay scallop recruitment in the Niantic Riverconducted in 1997 indicated that few spat were found, they were widelydispersed within the river, and peak spawning occurred in late July1997. Direct re-seeding was evaluated as an enhancement measure byplanting hatchery-reared scallops ( 38 mm shell height) insmall-scale, 100-m2 plots at different times and densities.Time of planting and the inferred predation intensity were major factorsaffecting survival; whereas, planting density had no significant effect.Approximately 9,000 scallops (35–45 mm shell height), broadcastwithin an eelgrass bed in November 1997, had high over-winter survivaland underwent gametogenesis and spawning during 1998. Of 26,000 bayscallops ( 45 mm shell height) over-wintered in suspensionculture from 1998–1999, approximately 60–80%survived, and these scallops spawned in mobile sanctuaries, during thesummer of 1999. There is good potential for using aquacultural methodsfor enhancement of bay scallop populations when natural recruitment ispoor and habitat and environmental conditions are not limiting.  相似文献

对两个不同地理种群的栉孔扇贝的两个群体———中国种群(C)和日本种群(J)的纯种后代及其正反两个杂交组合(C♀×J♂和J♀×C♂)的杂种个体,早期(10月龄)同地同期生长发育进行了测定和统计分析比较。结果表明,两个种群正、反交杂种一代在壳高、壳长、壳宽、活体重及成活率5个生长发育指标上均表现不同程度的杂种优势,反交杂种F1′除成活率外,其余各项生长发育指标显著优于正交杂种;中国和日本两个亲本群体各个性状的测定性能表现为壳宽差异显著和成活率差异极显著外,其它各项指标均没有显著差异。实验证明杂交是提高扇贝生产性能和抗逆性的重要途径。  相似文献

The bay scallop (Argopecten irradians irradians Lamarck 1819) has become one of the most important aquaculture species in China. Genetic improvement of cultured bay scallop can benefit greatly from a better understanding of its genome. In this study, we developed amplified fragment length polymorphisms (AFLPs) and simple sequence repeat markers from expressed sequence tags (EST‐SSRs) for linkage analysis in bay scallop. Segregation of 390 AFLP and eight SSR markers was analysed in a mapping population of 97 progeny. Of the AFLP markers analysed, 326 segregated in the expected 1:1 Mendelian ratio, while the remaining 74 (or 19.0%) showed significant deviation, with 33 (44.6%) being deficient in heterozygotes (A/a). Among the eight polymorphic EST‐SSR loci, one marker (12.5%) was found skewing from its expected Mendelian ratios. Eighteen per cent of the markers segregating from female parent were distorted compared with 21% of the markers segregating from male parent. The female map included 147 markers in 17 linkage groups (LGs) and covered 1892.4 cM of the genome. In the male map, totally 146 AFLP and SSR markers were grouped in 18 LGs spanning 1937.1 cM. The average inter‐marker spacing in female and male map was 12.9 and 13.3 cM respectively. The AFLP and SSR markers were distributed evenly throughout the genome except for a few large gaps over 20 cM. Although preliminary, the genetic maps presented here provide a starting point for the mapping of the bay scallop genome.  相似文献

We describe herein the discovery of 3905 putative single-nucleotide polymorphisms (SNPs) from the alignment of 4968 sequences in a bay scallop Argopecten irradians irradians expressed sequenced tag (EST) database. The observed frequency of SNPs was estimated at one every 118.2 bp of contig sequences. Thirty of the SNPs were chosen for validation by tetra-primer amplification refractory mutation system-polymerase chain reaction procedure, and 17 of them were polymorphic with minor allele frequency ranging from 0.016 to 0.484. BLASTX gave significant hits for all but one of the 17 genotyped SNP-containing contigs, 11 of which were located in coding regions and all resulted in a synonymous substitution. Parentage simulation demonstrated that SNP markers were more sensitive to the number of parents compared with microsatellites, thus more SNPs were needed to counteract low polymorphism. A table of optimal codons was deduced from the analysis of the A. i. irradians EST dataset to explain the frequency difference for a specific SNP. It seems that the selection of codon usage may be partly responsible for the frequency difference between the two alleles of the coding SNPs. These are the first SNPs developed for the bay scallop and will provide a useful complement to currently available genetic markers.  相似文献

采用双性状选择方法在经群体继代大壳高选育的海湾扇贝(Argopecten irradians concentricus Say)南部亚种F3养殖群体中进行闭壳肌定向选育, 以评价闭壳肌的选择效果和现实遗传力, 为海湾扇贝南部亚种育种提供理论依据。结果表明, 在以壳高为第一选择性状、体质量为第二选择性状、壳高选择压力为10%、体质量选择压力为1%情况下, 正、负向选择组、对照组在产卵量、受精率、孵化率、幼虫期壳长增长率、中培期存活率、成体各形态参数等方面的差异不显著(P>0.05); 在稚贝育成率、养成期存活率、成体各质量性状等方面均存在显著差异(P<0.05), 各指标值均呈现正向选择组>对照组>负向选择组; 正、负向选择组中, 体质量与闭壳肌质量均出现较高的现实遗传力, 且正向选择组高于负向选择组; 性状选择时, 体质量与闭壳肌质量的当代遗传获得明显高于单性状选择结果。壳高和体质量对闭壳肌质量均具有密切的遗传相关, 任何一个性状的选择均可对闭壳肌质量产生显著影响, 双性状同时选择可对闭壳肌质量产生叠加效应, 使闭壳肌质量性状的遗传进展达到最大

Abstract— A complete diallel crossing of three strains, Stirling, Korean and Local, was used to evaluate heterosis of intraspecific crossbreeding on early growth of Nile tilapia Oreochromis niloticus . All possible F-1 hybrid offspring were obtained and their growth rates to 90 d were compared. The specific heterosis for F-1 hybrids in Stirling × Korean, Stirling × Local and Korean × Local crosses were 7.61, 5.63 and 7.70 for weight, and 0.47, 0.26 and 0.55 for length, respectively. The purebred Stirling strain showed better growth than the other two strains (P < 0.05). When growth of F-1 hybrids was compared between sexes, males grew more rapidly than females (P < 0.05). However, no differences were observed in growth of male or female between hybrid groups. Purebred males in the Stirling strain were slightly heavier than those in the Korean and Local strains, and Local × Stirling, Korean × Local and Local × Korean crosses. However, in purebred females Stirling was superior to Korean, but not the Local strain and all F-1 hybrid crosses. Implications to choose the appropriate strains for developing a breeding program of tilapia are also discussed.  相似文献

Growth and survival of the scallop Lyropecten nodosus were studied in 1997 at two sites (inner and outer Turpialito Bay) during a non‐upwelling period normally occurring between August and November. Individuals had an initial shell height of 4.86 cm (SD=1.64 cm). Both experimental groups were held in suspended plastic baskets at the same depth (4 m). Measurements of shell height and dry weights of shell, gonad, digestive gland, remaining tissues and shell biofouling were taken at monthly intervals. Environmental parameters, including temperature, phytoplanktonic biomass, total particulate material (TPM) and associated organic (POM) and inorganic (PIM) fractions, were recorded simultaneously. At the end of the study, significant differences in growth and survival of scallops were observed between the two experimental sites. Scallops maintained inside the bay showed a 22% greater increase in shell height (7.41±0.27 cm) than those placed outside the bay (6.37±0.41 cm). Survival of scallops inside the bay was 31% higher compared with scallops outside. The greater availability of food of phytoplanktonic origin during the first two experimental months (July and August) together with greater POM throughout the whole experimental period except September, at the inner bay site, probably explained survival and growth differences observed between the two locations. Results suggest that, during the non‐upwelling period (characterized by low primary productivity and high water temperatures), POM of sedimentary origin may play an important role as an energy source required for metabolic and reproductive activities of L. nodosus.  相似文献

Two size classes of the surf clam, Spisula solidissima, and the bay scallop, Argopecten irradians, were exposed to different concentrations of gas-supersaturated seawater in a flowing seawater system. Both species tested experienced no mortality when held in the control treatment maintained at 96% oxygen and 109% nitrogen. Mortality, gill tissue damage, gas emboli, membranous tissue blisters, and abnormal secretion of shell material were induced experimentally at elevated levels of gas supersaturation. Results indicate significant mortalities of surf clams and scallops held at 114% O2 and 195% N2, and at higher levels of gas concentration. These values suggest a point of reference for the bivalve culturist in identifying potential problems which can be caused by gas-supersaturated seawater.  相似文献

Serotonin induced spawning in six bivalve species using individual spawning techniques without any additional stimuli. Intragonadal injection of serotonin induced spawning in the bay scallop Argopecten irradians, the American oyster Crassostrea virginica, and the surf clam Spisula solidissima. Injection of serotonin into the anterior adductor muscle of the ocean quahog Arctica islandica, the ribbed mussel Geukensia demissa, and the hard clam Mercenaria mercenaria induced spawning. The dosage of 0.4 ml of 2 mM serotonin solution used in this study stimulated bivalves to spawn within 15 min.  相似文献

Genetic diversity and correlation analysis between single nucleotide polymorphism (SNP) of myostatin (MSTN) gene and the growth traits were performed in Argopecten irradians. Around 279 individuals from three A. irradians populations were screened for the polymorphic sites of the MSTN gene. Two SNPs were found: T910G and G1162A. The observed heterozygosity of the two SNPs ranged from 0.1310 to 0.3299, whereas the expected heterozygosity ranged from 0.1436 to 0.2973. The values of D′ and r2 of linkage disequilibrium (LD) ranged from 0.475 to 1.0 and 0.052 to 0.174 respectively. Association analysis shows that individuals with genotype TT and TG of locus W4 had significantly higher adductor muscle mass than that with genotype GG in population CA (P < 0.05). Individuals with genotype AA of locus W5 had significantly higher body mass, soft‐tissue mass, adductor muscle mass, shell width and shell mass than those of other genotypes in population RZ (P < 0.05). The two SNPs found in our study can be used for molecule marker‐assisted breeding of A. irradians.  相似文献

用放在海水池塘中的8个陆基围隔(5.0m×5.0m×1.8m),研究中国对虾(Penaeuschinensis)与不同密度海湾扇贝(Argopectenirradians)投饵混养的放养方式、生产力和极限放养量.对虾体长(2.85±0.16)cm,扇贝壳长(1.10±0.12)cm,放养密度分别为6.0尾/m2和0,1.5,4.5,7.5粒/m2,用投饵和施肥(鸡粪和化肥)饲养.结果表明,扇贝密度为0和1.5粒/m2时,对虾的成活率无显著差异.混养(扇贝1.5粒/m2)时对虾的出塘体长、体重和产量分别比单养高2.5%,3.8%和6.5%;当扇贝密度高于1.5粒/m2时,对虾的平均体长、体重和产量随扇贝密度的增加而显著减少(P<0.05).扇贝密度为1.5和7.5粒/m2时,其产量由470kg/hm2增至1236kg/hm2;当扇贝密度高时,去壳后湿重占体重的百分数从(42.84±3.44)%降至(37.88±4.26)%.扇贝的极限放养量为600~800kg/hm2,适宜放养密度为1.0~1.5粒/m2.  相似文献

2010年进行了虾夷扇贝吊耳养殖试验研究。吊耳养殖分为一针四贝及一针二贝2种模式,经过6个月的对比试验,结果显示,与笼养方式相比,吊耳养殖扇贝的壳高增长率、单贝质量、贝柱质量、台产量、台产值等指标具有较大优势,存活率、肥满度、出丁率等指标互有优劣。在吊耳养殖模式下,虾夷扇贝壳高的生长优势明显,其中一针四贝模式的生长优势最大。本试验中吊耳养殖扇贝的总体生长情况优于传统的笼养方式;综合对比2种吊耳养殖模式,一针四贝模式的养殖效果好于一针二贝模式。  相似文献

以吉富罗非鱼与奥利亚罗非鱼为繁育亲本,采用完全双列杂交繁育4组F1,将初始规格基本一致的4组罗非鱼饲养100 d后,运用“加性-显性”遗传分析模型,分析了4组F1罗非鱼8个生长相关性状杂种优势、遗传效应以及与性状间的相关性.结果表明:(1)F1群体平均优势为0.129 4~0.368 4.除尾柄长超亲优势较大外,其他性状的群体超亲优势较小或表现出负向超亲优势.(2)8个性状的广义遗传率(HB)为0.714 2~0.995 3,表明加性效应和显性效应对性状的遗传变异影响极显著(P<0.01).除尾柄长外,其他性状的狭义遗传率(HN)介于0.469 4~0.737 9,表明加性遗传方差在表型方差中所占比率较高.(3)体质量、全长、体长、体高、体宽、头长、尾柄长、尾柄高性状之间表型相关在0.776 6~0.999 7范围内,而遗传相关在0.994 1~1.000 0之间,表明这些性状间都存在极显著的正相关.取样结束后,采用3.95×106 CFU/mL的海豚链球菌菌液进行腹腔感染,吉富罗非鱼自繁组F1代12 h后出现死亡,而奥利亚罗非鱼自繁组F1192 h后才出现死亡.384 h后,吉富罗非鱼自繁组F1死亡率为40%,正反交组F1分别为20%和23.3%,奥利亚罗非鱼自繁组F1死亡率最低,为6.67%.研究结果表明,除尾柄长外,杂交F1的其他性状不具备超亲优势,然而杂交可以提高选育后代的抗病力.  相似文献
