To better understand the distribution of soil microorganisms in Populus euphratica forests in Xinjiang, northwestern China, we studied and compared the populations and numbers of bacteria, fungi and actinomycetes in the soil at four different age stages of natural P. euphratica forests, i.e., juvenile forests, middle-aged forests, over-mature forests and degraded forests. Results showed that there were clear differences in the amount of microorganism biomass and composition rates across the four forest stages. Dominant and special microorganisms were present in each of the four different soil layers. The vertical distribution showed that the microorganism biomass decreased with increasing soil depth. The population of microorganisms was the lowest at 31–40 cm of soil depth. The microorganisms consisted of bacteria, actinomycetes, as well as fungi. Bacteria were the chief component of microorganisms and were widely distributed, but fungi were scarce in some soil layers. The distributions of ectomycorrhizas and ectomycorrhizal fungal inoculum with soil depth (0–45 cm) were determined in a 40-year-oldBetula platyphylla var.japonica forest. Mycorrhizal and non-mycorrhizal fine roots were measured in each soil core sample that was collected at soil depths of 0–5, 5–10, 10–15, 15–20, 20–25, 30–35, and 40–45 cm. The ectomycorrhizas were mainly distributed (>50%) in the top soil (0–5 cm) of organic forest floor horizons. Below 5 cm the quantity of ectomycorrhizas decreased sharply. The percentage of fine roots which were ectomycorrhizal gradually declined with the depth of soil. The ectomycorrhizal fungal inoculum was evaluated by a bioassay method, measuring the lengths of the entire root system and of the ectomycorrhizal roots of birch seedlings planted in each soil sample. The soil samples were collected from 0–5, 10–15, 20–25, 30–35, and 40–45 cm depths of the soil profile. Ectomycorrhizal formation on birch seedling roots in the bioassay was high in both the soil depth intervals 0–5 cm and 10–15 cm, while the amount was lower in the soil depth interval from 20–45 cm. The results of these investigations show that the amount of the ectomycorrhizas in soil, and the ectomycorrhizal fungal inoculum potential as determined by bioassay, are not always consistent with each other.  相似文献

The role of tetrapartite associations among Frankia, Gigaspora margarita (an arbuscular mycorrhizal fungus), Pseudomonas putida (rhizobacterium), and Alnus sieboldiana in growth, nitrogen fixation, and mineral acquisition of A. sieboldiana was investigated. Seedlings of A. sieboldiana were inoculated with Frankia isolated from root nodules of alder, followed by inoculation of G. margarita and P. putida, and were grown for 5 months in a greenhouse. The seedlings inoculated with Frankia and G. margarita together produced the highest biomass of shoots and root nodules. Nitrogen-fixation activity, measured by acetylene reduction assay, was observed when Frankia was inoculated. The activity, on a per-nodule gram basis, decreased after G. margarita inoculation, but on a per-plant basis there was no significant difference in the activity among inoculation treatments. The mineral content in the seedlings changed after inoculation with Frankia, but not after inoculation with P. putida and/or G. margarita. The results showed a synergistic interaction among Frankia, the mycorrhizal fungus, and the rhizobacterium on the growth of A. sieboldiana.  相似文献

The effect of inoculation with Frankia, a N-fixing actinomycete, on the growth of Alnus sieboldiana seedlings was studied on unsterilized soil from a nursery and an alder stand (forest of Alnus firma). The seedlings of A. sieboldiana were inoculated with Frankia before or after a 2-month culture on sterilized vermiculite, during which they nodulated, and transplanted to unsterilized soil from the nursery and the alder stand. The control seedlings were also cultured on sterilized vermiculite for about 2 months and transplanted to unsterilized soil without Frankia inoculation. The seedling growth, nodulation and N-fixing activity were measured 3, 10 and 16 weeks after the transplantation. Growth and nodule biomass of the seedlings inoculated with Frankia and those grown on the alder soil were better than those without inoculation with Frankia and grown on the nursery soil, respectively. The seedlings inoculated before spontaneous nodulation grew better than those inoculated at the transplantation. Nitrogen-fixing activity measured by acetylene reduction assay at 16 weeks after the transplantation was higher in the seedlings grown on the soil from the nursery than on the soil from the alder stand.  相似文献

To add to knowledge of ectomycorrhizal (ECM) community structure on the roots of Pinus thunbergii seedlings in a Japanese coastal forest, we sampled naturally regenerated current-year and 1- to 5-year-old seedlings. We classified the 667 root tips on current-year seedlings and the 1,927 root tips on older seedlings into 13 phylotypes based on morphological and genetic analyses. Cenococcum geophilum, members of the families Clavulinaceae, Russulaceae, or Thelephoraceae or the genus Trichophaea, were indicated to be fungi forming P. thunbergii ectomycorrhizas. Among them, C. geophilum and Clavulinaceae sp. 1 were the most or second most dominant species. A species accumulation curve based on the number of samples nearly reached a plateau, with observed species richness equal to 11 species and the Jackknife2 and Chao2 richness estimators indicating 14 and 12 species, respectively. In addition, Simpson’s 1/D was 3.89 and Shannon–Wiener’s H′ was 1.71, indicating a relatively low taxonomic diversity. There was no significant difference in the ectomycorrhizal formation rate, or the occurrence frequency and the number of ECM phylotypes between current-year and older seedlings. These results indicated that less diverse fungi were involved in ectomycorrhizal formation on coastal pine seedlings compared with those in comparable inland forests in the study area.  相似文献

Reforestation is one of the most effective ways to reduce the impacts of desertification. Caragana microphylla Lam. and Pinus sylvestris var. mongolica Litrin have been widely used to stabilize shifting sands in the Horqin sandy land area since the 1980s. However, soil water depletion has been of major concern in C. microphylla and P. ongolica plantations and in many places current soil moisture cannot meet the demand of growing plants. To determine the water budget of C. microphylla and P. mongolica plantations, we studied the effect of plantations on soil moisture and assessed the evapotranspiration in plantations of both species. Investigations were conducted at a fenced plot at Wulanaodu (42°29′N, 119°30′E, 479 m a. s. l), located at the western edge of the Horqin Sandy Land area in Inner Mongolia, northern China. Five year old C. microphylla and seven year old P. mongolica plants were selected from the plantations and transplanted to iron boxes (400 cm×200 cm×120 cm) which can drain extra water. Plant spacing of 1 m×1 m was applied to P. mongolica, and two plant spacings of 1 m×1 m and 1 m×2 m to C. microphylla. The transplanted plants grew for two years in the boxes. Soil moisture from soil surface to a depth 80 cm were measured at 20 cm intervals in boxes every 10 d (2004) or 3 d (2005) during the growing season with a TDR water meter. The evapotranspiration was estimated from a mathematical formula and the characteristics of soil water consumption and evapotranspiration of these two plantations were analyzed. The soil water of P. mongolica was more than that of C. microphylla at the same 1 m×1m spacing. The soil water of C. microphylla with the 1 m×2 m spacing was more than that of the 1 m×1 m spacing. The evapotranspiration ranged from high to low as follows: C. microphylla (1 m×1 m), C. microphylla (1 m×2 m) and P. mongolica (1 m×1 m) during the growing seasons. The evapotranspiration of individual plants ranging from high to low was C. microphylla (1 m×2 m), C. microphylla (1 m×1 m), and P. mongolica (1 m×1 m) during the growing season. This paper reports three newly recorded species, Diphascon modestum Binda, Pilato and Dastych, 1984, Diphascon nobilei Binda, 1969, and Diphascon triodon Maucci, 1996, of the genus Diphascon (Tardigrada; Hypsibiidae) from China. Five specimens of Diphascon modestum were collected from Taibai Mt. (34°18′N, 107°42tpE) at 2,500 m above the sea level (a.s.l.), two specimens of Diphascon nobilei were collected from Tonggu County, Jiangxi Province (28°31’N, 114°26′E) at 900 m a.s.l, and one specimen of Diphascon triodon was collected from Linzhi County, Tibet (29°40′N, 94°23′E) at 3,500 m a.s.l. All specimens are deposited at the College of Life Sciences, Shaanxi Normal University, China. A key to the Chinese species of Diphascon was also given. A pine forest in the south of Sweden was treated with lime and wood ash. In early June, 12 months after the ash treatment and 18 months after the lime treatment, one year old Pinus syhestris L. seedlings were planted. Four months later six différent ectomycorrhizal types had infected the seedlings in all the treatments. A mycorrhizal type designated “pink”; was more than twice as common in the lime treatments as in the control and ash treatments. Piloderma croceum Erikss. & Hjorts. was significantly more abundant in limed soil than in ash treated soil. The results were compared to those from a bioassy performed in the laboratory, where P. syhestris seedlings had been grown in soil from the same forest. Similar soil pH values in the two studies resulted in different relative infection rates of the mycorrhizal types found. One additional mycorrhizal type, designated “white”; was found in the field experiment. This suggests that mycelial connections to the mature host plants may significantly alter the ability of different fungi to colonize host plant roots in competition with each other compared to when the fungi infect from propagules in the soil.  相似文献

Australian acacias like Acacia holosericea are excellent candidates for the revegetation of arid zones in Africa. Their high ability to develop multiple symbioses with soil microorganisms is crucial to their rapid development in adverse climatic and edaphic conditions. These symbioses include nitrogen fixation with rhizobia, vesicular arbuscular mycorrhization and ectomycorrhization. We set up an experiment to test the growth response of A. holosericea to ectomycorrhizal inoculation in 13 different soils from Senegal. After autoclaving the soil, the experiment was conducted in a greenhouse for four weeks. Plants were inoculated with Pisolithus albus strain IR100. The following parameters were measured: plant biomass, N, P, K and Ca foliar composition, spontaneous nodulation rate, and ectomycorrhizal colonization. Data were analyzed in light of the physical, chemical and total microbial characteristics of the soil. The results demonstrated a global promoting effect of P. albus inoculation on plant (shoot and root) growth and on foliar P and K, together with a depressive effect on N, while Ca rates were barely affected. Interestingly, spontaneous nodulation with putative water- or airborne rhizobia was stimulated after P. albus inoculation. However, these nodules seemed poorly effective, as they failed to cause any change in plant growth or in foliar N composition within the nodulated and nonnodulated control plants. These results show that plant growth response to ectomycorrhizal inoculation is greatly dependent on soil characteristics, and that root ectomycorrhizal colonization is influenced by biotic factors such as soil microbiota. From a practical point of view, data from the present study demonstrate that it is possible to optimize the effect of fungi on plant growth by screening soils under nursery conditions.  相似文献

Effects of different forest floor vegetation types in secondary forest and of conversion to plantation on the quality and quantity of ectomycorrhizas are mostly unknown.Betula platyphylla var.japonica seedlings were used for bioassays of ectomycorrhizal fungal inoculum using soils from four 50-year-oldB. platyphylla var.japonica forests that had different types of forest floor vegetation: two with shrub types, one with aSasa type, and one with a grass type. Seedlings were also grown in soil from a nearby monospecific plantation ofChamaecyparis obtusa. Ectomycorrhizas formed 13 to 26% of root length of seedlings grown in soil from the five different sites. The maximum percentage of ectomycorrhizal formation was obtained from the grass-type forest. The dominant type of ectomycorrhiza in the two shrub-type forest soils was the same as that in theSasa-type forest soil. The dominant types of ectomycorrhizas in the grass-type forest soil and in theC. obtusa plantation soil were different from that in the two shrub-type forest soils and in theSasa-type forest soil. The results of this investigation suggest that the type of forest floor vegetation, accompanied with changes in thickness of the A0 horizon, might affect the ectomycorrhizal fungi in the soils ofB. platyphylla var.japonica forests. Establishment of artificial plantations ofC. obtusa might change the ectomycorrhizal fungi that could associate withB. platyphylla var.japonica seedlings in soil.  相似文献

Interactions between two isolates (Avondhu and CBS) of Tricholomopsis rutilans and ectomycorrhizal fungi (Hebeloma crustuliniforme, Laccaria laccata and Paxillus involutus) were examined on agar medium in the presence or absence of woodchips. The CBS isolate showed more competitiveness than the Avondhu isolate, when paired with ectomycorrhizal fungi. There was an inhibition of the growth of mycelium of the ectomycorrhizal fungi ranging from overgrowth to avoidance. The ectomycorrhizal fungi exhibited hyphal abnormalities such as discoloration, excessive vacuolation and curling. Formation of mycorrhizas by H. crustuliniforme in Pinus contorta and Picea abies was unaffected by the presence of the CBS isolate, whereas a reduction occurred in the presence of the Avondhu isolate. In non‐mycorrhizal seedlings of both conifers, the Avondhu isolate reduced root and shoot dry mass and number of root laterals and caused a lower number of short roots in lodgepole pine. The significance of these interactions between T. rutilans and ectomycorrhizal fungi in paired culture and during symbiosis is discussed.  相似文献

在实验室条件下模拟环境因子进行沙地樟子松林下常见3种外生菌根真菌的纯培养试验,探讨培养基、pH值、水分和温度对外生菌根真菌生长的影响,以期得到3种外生菌根真菌的最适生长条件.结果表明:外生菌根真菌在含有松针汁、马铃薯汁、酵母粉和维生素的培养基里生长较好;pH值对外生菌根真菌的生长影响较小,L.deliciosus 和B.sp.的最适生长pH分别为6.0和5.0,L.sp.在较广的pH范围内均可生长得良好,且在中性和弱碱性的培养基中长势均好于另外两种;水分对实验菌株的影响较大,在较低的PEG浓度(100 g PEG·kg-1 H2O)下,三种菌株生长良好,最适得生长PEG浓度为10%;高温容易导致外生菌根真菌死亡,3种外生菌根真菌的最适生长温度范围是25-28℃.  相似文献

木麻黄是我国沿海防护林基干林带中的主栽树种,与土壤中的微生物可以形成共生关系,提高自身耐干旱、抗贫瘠、抗盐碱等适应恶劣环境的能力。土壤中的细菌和真菌能够与木麻黄根系形成稳定的相互作用的微生态系统,其中微生物主导的物质代谢循环是植物获取营养的主要来源之一,反之木麻黄的根系分泌物可以为微生物的生长提供养分。文中综述了木麻黄与根系微生物之间相互作用的相关研究,尤其是接种弗兰克氏菌(Frankia)及菌根菌提高木麻黄抗逆性方面的研究进展,对木麻黄与根系微生物体系的促生、抗逆机制进行了探讨和展望。  相似文献

Root biomass and root distribution were studied in Entisols derived from the thick deposition of volcanic pumice on Hokkaido Island, Japan, to examine the effect of soil conditions on tree root development. The soil had a thin (<10 cm) A horizon and thick coarse pumiceous gravel layers with low levels of available nutrients and water. Two stands were studied: a Picea glehniiAbies sachalinensis stand (PA stand) and a Larix kaempferiBetula platyphylla var. japonica stand (LB stand). The allometric relationships between diameter at breast height (DBH) and aboveground and belowground biomass of these species were obtained to estimate stand biomass. The belowground biomass was small: 30.6 Mg ha−1 for the PA stand and 24.3 Mg ha−1 for the LB stand. The trunk/root ratios of study stands were 4.8 for the PA stand and 4.3 for the LB stand, which were higher than those from previous studies in boreal and temperate forests. All species developed shallow root systems, and fine roots were spread densely in the shallow A horizon, suggesting that physical obstruction by the pumiceous layers and their low levels of available water and nutrients restricted downward root elongation. The high trunk/root ratios of the trees may also have resulted from the limited available rooting space in the study sites.  相似文献

A study was conducted to determine the dynamic of Zn content between soil and plant in the natural protection zone of Leymus chinensis grassland in Changling County (44°30′–44°45′N; 123°31′–124°10′E), Jilin Province, China. Results showed that the total Zn content was lower, available Zn content had a moderate level in the soil, and the plants was not lack of Zn. During the growing season, content of total Zn and available Zn in soil showed a down-trend distribution along the soil profile. Content of total Zn had a significantly positive correlation with that of the organic matter, but it was negatively correlated to soil pH. Monthly dynamic of the average content of total Zn showed a “V” type curve in the growing season from May to August, and July was the nadir. The trend of the average content of available Zn was similar to the content of total Zn, but was down after August; Zn content variation in the organs and litter of L. chinensis was great, with the order of root>rhizome >leaf>stem>litter. The ratio of available Zn content in A layer versus B layer was more than 2 times that of the total Zn, which indicated that the soil of A layer had higher enrichment capacity of available Zn. The enrichment of Zn in the root of L. chinensis was 44.17 times as that in the soil. The absorbing intensity of root had a significantly negative correlation with the activity of Zn in the soil (r=−0.8800, p<0.01). As part of a study on soil carbon flow in forest ecosystems, the biomass of fine roots (2.0mm in diameter) and root-associated fungi, including ectomycorrhizal fungi, were estimated in the summer season in 1998 at a Pinus densiflora (Japanese red pine) stand in western Japan. Fine roots of pine were classified into three categories: class I roots (0.5–2.0mm in diameter), long class II roots (long roots with diameter 0.5mm; IIL), and short class II roots (short roots with diameter 0.5mm; IIS). Total biomass of fine roots (I + IIL + IIS) at this stand was estimated to be 91.0gm–2, about 23% of which was class II roots (IIL + IIS). Ergosterol, which is a component of fungal membranes, was analyzed to estimate the biomass of root-associated fungi in roots. In the upper soil layers (from the surface to 13.4cm in depth), ergosterol contents in the class I, IIL and IIS roots were in the ranges 43.1–82.2, 126.1–196.3 and 271.2–321.0µgg–1 root DW, respectively. The ergosterol content was converted to fungal biomass using the median (minimum–maximum) value of ergosterol concentration reported for ectomycorrhizal fungi. Root-associated fungal biomass in this stand was estimated to be 2.0 (0.5–9.6) gm–2. The data suggest the biomass of ectomycorrhizal fungi in the P. densiflora stand is small compared with that in other forest ecosystems.  相似文献

Steps leading to the biosynthesis of syringyl lignans and tetrahydronaphthalene and naphthalene lignans, especially the formation of the C2–C7′ linkage, have not been elucidated. Lyoniresinol is a typical syringyl lignan, as well as a tetrahydronaphthalene lignan found in Lyonia ovalifolia var. elliptica. To demonstrate the biosynthetic pathway for (+)-lyoniresinol, three putative biosynthetic intermediates of lyoniresinol, syringaresinol, 5,5′-dimethoxylariciresinol, and 5,5′-dimethoxysecoisolariciresinol, were isolated from wood. The identity of the putative intermediates was confirmed by spectroscopic analyses, as well as by comparison of spectral and chromatographic data with those of authentic samples previously synthesized. The stereochemistry (enantiomeric composition and absolute configuration) of the isolated lignans were determined as (±)-syringaresinol, (8S,8′S)-(−)-5,5′-dimethoxylariciresinol [46% enantiomeric excess (e.e.)], (8S,8′S)-(+)-5,5′-dimethoxysecoisolariciresinol (91% e.e.), and (8R,8′R)-(+)-lyoniresinol (42% e.e.). The absolute configurations of (+)-and (-)-5,5′-dimethoxylariciresinols, and (+)-and (-)-5,5′-dimethoxysecoisolariciresinols were determined by their synthesis (catalytic reduction) from (8R,8′R)-(+)-and (8S,8′S)-(-)-syringaresinols and by subsequent chiral high-performance liquid chromatography analysis. Delignified hinoki wood and cellulose as well as hinoki and lauan woods were carbonized at 590°C for 1 h. The dielectric properties of these specimens were measured at 20°C in a frequency range of 20 Hz to 1 MHz. Inflection points in the dielectric constant (ε′) versus the logarithm of frequency (log f) curves as well as in the logarithm of the electric conductivity (log σ) versus log f curves for all specimens prepared were recognized. Peaks in the dielectric loss and the imaginary part of the complex conductivity versus the log f curves were detected in the frequency location corresponding to the inflection point in the ε′ and log σ versus log f curves. It was considered that this relaxation was responsible for the interfacial polarization observed in heterogeneous materials because no permanent dipoles existed in the specimens carbonized above 500°C. The Cole–Cole circular arc law was applied to account for this relaxation. Similar average relaxation times were obtained for all specimens. These results suggested that the observed relaxation was ascribed to interfacial polarization at microscopic levels in the cell walls.  相似文献

To investigate the potential effects of nitrogen (N) deposition on Japanese forests, a chronic N-addition experiment that included three treatments (HNO3, NH4NO3, and control) was carried out in a 20-year-old Japanese cedar (Cryptomeria japonica D. Don) stand in eastern Japan over 7 years. The amount of N applied was 168 kg N ha−1 year−1 on the HNO3 plots and 336 kg N ha−1 year−1 on the NH4NO3 plots. Tree growth, current needle N concentration, and soil solution chemistry were measured. Nitrogen application decreased the pH and increased NO3 , Ca2+, Mg2+, and Al concentrations in the soil solution. The needle N concentration increased in both of the N plots during the first 3 years. Nevertheless, the annual increments in height and in the diameter at breast height of the Japanese cedars were not affected by N application, and no visible signs of stress were detected in the crowns. Our results suggest that young Japanese cedar trees are not deleteriously affected by an excess N load.  相似文献

To explore the effects of plant hormones levels excreted by ectomycorrhizal (ECM) fungi on the growth of poplars, Populus × euramericana cv. NL-895 seedlings were inoculated with nine species of ECM fungi. We investigated the status of ectomycorrhizal formation and the effects of these fungi on poplar growth, and using the HPLC method, we measured the contents of four kinds of plant hormones, indole acetic acid (IAA), zeatin (Z), gibberellin (GA) and abscisic acid (ABA) in both the culture filtrate and the mycelium of these fungi. The results showed that the effects of nine ECM fungi on the growth of poplar NL-895 varied. The inoculated seedlings, whether or not obvious mycorrhizas were developed, grew better than those non-inoculated ones. All nine ectomycorrhizal fungi excreted the four plant hormones, but at different levels. The hormone contents in culture filtrate were higher than that in mycelium, which showed a definite relationship with poplar growth.
