A field experiment was conducted in 2001 at Holetta and Kulumsa, Ethiopia, to study genetic diversity in Ethiopian faba bean
(Vicia faba L.) landraces. One hundred sixty random germplasm accessions were grown in an alpha lattice design with two replications. Data
on 12 traits were collected and analyzed. Significant differences were observed among the accessions for most of the traits
(except number of pods/podding nodes) at each location even though differences pooled over location were mostly non-significant.
Cluster analysis distinguished seven diversity classes of different sizes. Accessions from the northern half of the country
(North and South Wello, North Gonder and North Shewa) were closely related while those from the southern part of the country
(Arsi) were highly diverse. Cumulative effects of a number of characters dictated differentiation of the accessions into clusters.
Some overlapping were encountered between accessions from the northern and those from the southern parts of the country. The
study revealed that accessions from different regions might have similar genetic background and those from the same origin
might also have different genetic background. Therefore, geographic diversity should not necessarily be used as an index of
genetic diversity and parental selection should be based on a systematic study of genetic diversity in a specific population.
Genetic distances between most of the clusters were significant that crosses between parents selected out of them are expected
to generate desirable progenies. Future germplasm collection, conservation and utilization strategies should put more focus
not only on inter-regional diversity in the country as a whole but also on intra-regional diversity in Arsi. 相似文献

One hundred and forty-four heterogeneous tef germplasm accessions collected from 10 major tef growing areas in Ethiopia were
evaluated for 18 quantitative traits in a simple lattice design, at Holetta and Ginchi, in 2001 main growing season to study
the genetic diversity in tef. The combined analysis of variance revealed the presence of significant variation among germplasm
accessions for all traits studied. Cluster analysis revealed the overlapping of the germplasm accessions from different origin
and the accessions were grouped into eight distinct clusters of 1–78. Generalized distance square confirmed the presence of
significant genetic distance between clusters. In principal component (Prin) analysis, the first four principal components
with latent root values above one accounted for about 80.6% of the variability existing among the germplasm accessions. Prin1
constituted about 55% of the variability mainly from almost equal contribution of 10 quantitative traits, indicating that
most of the traits are equally important in tef diversity. In this study, the regions and altitudes of origin were not found
to have a substantial effect on the genetic diversity in tef germplasm. Diversity within the regions was found to be significant
and, hence, an opportunity for exploitation in tef improvement. 相似文献

Pigeonpea is an important pulse crop grown by smallholder farmers in the semi-arid tropics. Most of the pigeonpea cultivars
grown to date are selections from the landraces, with a narrow genetic base. With the expansion of the crop to newer areas,
problems of local importance are to be addressed. Hence, an economically feasible and faster germplasm evaluation mechanism,
such as a core collection, is required. This article describes the development of core collection from 12,153 pigeonpea accessions
collected from 56 countries and maintained at ICRISAT, Patancheru, India. The germplasm accessions from 56 countries were
placed under 14 clusters based primarily on geographic origin. Data on 14 qualitative morphological traits were used for cluster
formation by Ward’s method. From each cluster ≈10% accessions were randomly selected to constitute a core collection comprising
1290 accessions. Mean comparisons using Newman–Keuls test, variances’ comparisons by Levene’s test, and comparison of frequency
distribution by χ2-test indicated that the core collection was similar to that of the entire collection for various traits and the genetic variability
available in the entire collection is preserved in the core collection. The Shannon–Weaver diversity index for different traits
was also similar for both entire and core collection. All the important phenotypic associations between different traits available
in the entire collection were preserved in the core collection. The core collection constituted in the present study facilitates
identification of useful traits economically and expeditiously for use in pigeonpea improvement. 相似文献

Patterns of geographical diversity, and the relationship between agro-morphological traits and fatty acid composition were
assessed for 193 safflower (Carthamus tinctorius) accessions representing forty countries. Accessions were assigned to eight groups based on geographical proximity. Cluster
and Principal Component analyses were performed to assess patterns of diversity among the accessions and to select the most
distant accessions from each of eight groups for analysis of randomly amplified polymorphic DNA (RAPD) markers. There was
a large amount of diversity for agro-morphological traits, fatty acid composition, and RAPD markers. Most correlations among
different traits were rather low. Plant height showed a positive correlation with days to flowering (r = 0.63**). Palmitic acid was positively correlated with stearic acid and oleic acid values, and negatively correlated with
linoleic acid (P < 0.01). Oleic acid and linoleic acid showed a strong negative correlation (r = −0.89**). The first three principal components together explained 59% of the variation, however, neither principal component
analysis (PCA) nor marker analysis revealed a clear relationship between diversity pattern and geographical origin. Accessions
from some geographical regions tended to group together, such as accessions from South Western Asia, Central Western Europe,
and the Mediterranean region. The correlation between the morphological matrix and the genetic matrix based on RAPD markers
was not significant (r = 0.027). Wide diversity in safflower germplasm indicates a considerable potential for improving this crop for both agronomic

Diversity among 36 snapmelon landraces, collected from 2 agro-ecological regions of India (9 agro-climatic sub-regions), was
assayed using RAPD primers, morphological traits of plant habit and fruit, 2 yield-associated traits, pest and disease resistance
and biochemical composition (TSS, ascorbic acid, titrable acidity). Typical differences among accessions were observed in
plant and fruit characteristics and snapmelon germplasm with high titrable acidity and possessing resistance to downy mildew,
Cucumber mosaic virus, Zucchini yellow mosaic virus, Papaya ringspot virus, Aphis gossypii and Meloidogyne incognita was noticed in the collection. RAPD based grouping analysis revealed that Indian snapmelon was rich in genetic variation
and region and sub-region approach should be followed across India for acquisition of additional melon landraces. Accessions
of var. agrestis and var. momordica clustered together and there was a separate cluster of the accessions of var. reticulatus. Comparative analysis of the genetic variability among Indian snapmelons and an array of previously characterized reference
accessions of melon from Spain, Israel, Korea, Japan, Maldives, Iraq, Pakistan and India using SSRs showed that Indian snapmelon
germplasm contained a high degree of unique genetic variability which was needed to be preserved to broaden the genetic base
of melon germplasm available with the scientific community.

Guar [Cyamopsis tetragonoloba (L.) Taub.] is an important commercial crop of India, mainly cultivated in the northwestern part and neighboring areas of Pakistan. High morphological diversity is reported in guar. The present study attempts to analyze the genetic diversity in cultivated guar using allozyme polymorphism and compare it with reported morphological diversity. Accessions for the study were selected from a set of guar germplasm conserved at the National Bureau of Plant Genetic Resources, New Delhi. The morphological data of 108 selected accessions was subjected to UPGMA clustering. Fifty-five accessions were then picked randomly from the clusters generated. Tris-borate system (pH 8.3) was found to be most effective for detection of allozyme polymorphism in guar. Ten enzymes were selected for the study out of 18 systems tried, namely, malate dehydrogenase, malic enzyme, phosphogluco isomerase, glutamate oxaloacetate transaminase, esterase, peroxidase, alcohol dehydrogenase, glutamate dehydrogenase, glucose 6-phosphate dehydrogenase and shikimate dehydrogenase. A total of 20 loci were detected with 34 alleles. The number of polymorphic loci in different accessions was observed to be highly variable (0–9). An average of 1.27 allozyme per polymorphic locus was observed. The allele frequency data of the 55 accessions was used to determine genetic diversity parameters. The dendrogram generated on genetic distance showed three clusters in the distance range of 0–0.28. Most of the accessions were in the first clusters in a narrow distance range of 0–0.08. The second cluster consisted of improved varieties and accession from Pakistan. The third cluster of accessions was genetically more distant but there was no definite segregation of accessions according to the place of collection. The accessions from south west Rajasthan were scattered in all the three clusters showing higher variability in this region, also supported by higher morphological variation. Knowledge of the genetic diversity in the germplasm accessions is important for the efficient germplasm management. We studied
45 colored rice accessions, which had been earlier collected from peninsular India. The accessions were evaluated at two diverse
locations for 12 morpho-agronomic traits and genotyped using 50 simple sequence repeat (SSR) markers. Significant genotypic
variability was observed for all morpho-agronomic traits studied. Dendrogram and principal component analysis based on morpho-agronomic
traits separated the accessions into three clusters. The first two principal components accounted for 82% of the total variation.
The most discriminatory traits were number of grains per panicle, biomass yield, and days to flowering and maturity. The SSR
analysis revealed high polymorphic information content value of 0.84. Though, Mantel test did not show a significant correlation,
yet the classification based on phenotyping and genotyping data showed good agreement for the expression of morpho-agronomic
traits in the formation of clusters. Five accessions had high concentration (> 100 μg/g) of Fe and one of Zn. Our study revealed
the presence of large genetic variation among the colored rice accessions evaluated and moderate agreement between morpho-agronomic
and SSR–based classifications and of these with geographic diversity. 相似文献

Target region amplification polymorphism (TRAP) markers were used to assess genetic variability among 38 germplasm accessions
and 10 commercial hybrids of spinach (Spinacia oleracea L.), an economically important leafy vegetable crop in many countries. Germplasm accessions with different geographic origins
and 10 commercial hybrids were examined. For assessing genetic diversity within accessions, DNA was extracted from 12 individual
seedlings from six germplasm accessions and two hybrids. A relatively high level of polymorphism was found within accessions
based on 59 polymorphic TRAP markers generated from one fixed primer derived from the Arabidopsis-like telomere repeat sequence and two arbitrary primers. For evaluating interaccession variability, DNA was extracted from
a bulk of six to 13 seedlings of each accession. Of the 492 fragments amplified by 12 primer combinations, 96 (19.5%) were
polymorphic and discriminated the 48 accessions from each other. The average pair-wise genetic similarity coefficient (Dice)
was 57.5% with a range from 23.2 to 85.3%. A dendrogram indicated that the genetic relationships among the accessions were
not highly associated with the geographic locations in which the germplasms were collected. The seven commercial hybrids were
grouped in three separate clusters, suggesting that the phenotype-based breeding activities tended to reduce the genetic variability.
This preliminary study demonstrated that TRAP markers are effective for fingerprinting and evaluating genetic variability
among spinach germplasms.

Wild species of safflower, Carthamus oxyacanthus Bieb., is highly crossable with cultivated species, C. tinctorius L. and could be directly exploited in broadening safflower gene pool and improving the crop for biotic and abiotic stress
environments. In this study, genetic diversity among accessions of C. oxyacanthus and their relationships with cultivated safflower were evaluated using agro-morphological traits and polymorphic inter-simple
sequence repeats (ISSR) markers. Significant variation was observed among accessions particularly for seeds per capitulum,
seed yield per plant, harvest index and capitula per plant. Cluster analysis based on agro-morphological traits classified
the wild accessions in two groups according to their geographical regions, and separated them from the cultivated genotypes.
ISSR marker also revealed a high genetic variation among the accessions, and cluster analysis based on this marker divided
genotypes into four groups, with cultivated ones in a separate clade. Genetic variation observed among the wild safflower
germplasm at the DNA level was higher than the agro-morphological traits, indicating that ISSR is an effective marker system
for detecting diversity among safflower genotypes and their genetic relationships. Accessions of C. oxyacanthus with high genetic relationship to cultivated species could be used for interspecific hybridization in breeding programs of

Genetic erosion in cultivated wheat provides a good reason for studying genetic diversity in crop wild relatives. In the present study, genetic diversity of 32 accessions belonging to T. boeoticum species collected from different parts of Iran were evaluated using 13 morphological traits as well as ten inter-simple sequence repeat primers. Statistical analysis for morphological traits showed significant differences among accessions (except number of fertile tillers and total tillers per plant). In principal component analysis, the first three PCA showed 82.65 % of the total morphological variation. Based on the morphological traits, accessions were separated into two main groups by cluster analysis. In molecular analysis, polymerase chain reactions amplified 105 DNA fragments, out of which, 95 (90.47 %) were polymorphic. From geographic perspective, the accessions sampled from western and southwestern of Iran showed the highest and lowest polymorphism, respectively. However, the maximum values of effective number of alleles (Ne), Nei’s gene diversity (He) and Shannon’s information index (I) was related to accessions collected from NW regions. Also, according to cluster analysis and PCoA plot genetic diversity was not related to geographical distribution. Overall, our results revealed a remarkable level of genetic diversity among studied Iranian T. boeoticum accessions; especially accessions collected from Kermanshah and Lorestan provinces, which can be of interest for future breeding programs. So, conservation of germplasm of these areas is recommended. 相似文献

Cowpea is an important grain legume and hay crop of many tropical and subtropical regions, especially in the dry savanna region
of West Africa. The cowpea gene pool may be narrow because of a genetic bottleneck during domestication. Genetic variation
within specific breeding programs may be further restricted due to breeding methods, ‘founder effects’ and limited exchange
of germplasm between breeding programs. Genetic relationships among 60 advanced breeding lines from six breeding programs
in West Africa and USA, and 27 landrace accessions from Africa, Asia, and South America were examined using amplified fragment
length polymorphism (AFLP) markers with six near infrared fluorescence labeled EcoRI + 3/1bases/MseI + 3/1bases primer sets. A total of 382 bands were scored among the accessions with 207 polymorphic bands (54.2%). Despite
a diverse origin, the 87 cowpea accessions shared a minimum 86% genetic similarity. Principal coordinates analysis showed
clustering of breeding lines by program origin, indicating lack of genetic diversity compared to potential diversity. Accessions
from Asia and the Americas overlapped and were distinct from West African breeding lines, indicating that germplasm from Asia
and the Americas have common origins outside West Africa. US and Asian breeding programs could increase genetic variability
in their programs substantially by incorporating germplasm from West Africa, while national programs in West Africa should
consider introgression of Asian germplasm and germplasm from other parts of Africa into their programs to ensure long-term

Phenotypic diversity for agronomic characteristics was determined in an experiment with 29 landraces and 14 improved varieties
of rye from the Nordic area, Germany and Poland. The accessions were scored for 12 characters. Effects of the location, year,
type, and country of origin for landraces were investigated with analysis of variance. Phenotypic variations for the traits
were estimated using the Shannon–Weaver diversity index. The genetic variation was high, with an average of H 0 = 0.566. The landraces from Norway, Sweden and Finland showed the highest variation, whereas the improved varieties had the
lowest. The German material also had low variation but the status of this material is uncertain. The genetic diversity showed
that 70% of the variation was found within the accessions. A cluster analysis was carried out to identify the relationship
between the accessions. The material grouped into eight clusters, where clusters I to V included landraces from Sweden, Finland
and Norway, except for cluster III, which included one improved variety from Denmark. Cluster VI comprised a single Swedish
landrace from Gotland. Most of the improved varieties were in cluster VII and the last cluster contained accessions from Germany

Taro [Colocasia esculenta (L.) Schott] germplasm accessions collected from different parts of India were subjected to RAPD (Random Amplified Polymorphic DNA) analysis to assess the genetic diversity prevalent and also to test the genetic basis of morphotypic classification. Thirteen random decamer primers out of the 22 tested were used to analyse 32 taro accessions belonging to 28 morphotypes. Three out of these thirteen primers analysed showed 100 per cent polymorphism. Per cent polymorphism varied from 60 to 100 among the polymorphic primers. High genetic diversity was revealed as the similarity coefficient values ranged from 0.50 to 0.98. No two accessions analysed in the present study showed a similarity coefficient value of one thereby indicating their distinctness and presence of high genetic diversity in Indian taro germplasm. Dendrogram obtained from UPGMA analysis grouped 32 accessions in four clusters and three accessions were placed as outliers. Clustering pattern did not show any strict relationship with geographical distribution, morphotype classification and genotypic diversity. Further, accessions classified, as belonging to the same morphotypic group did not always cluster together. Presence of a very close genepool of the wild, weedy and cultivated forms with extreme levels of phenotypic and genotypic variation is suggested as the reason for high genetic diversity reported. Usefulness of DNA markers such as RAPD in characterising and assessing the genetic diversity in Indian taro germplasm is hereby demonstrated. 相似文献

Jute mallow rich in proteins, vitamins and essential amino acids is an important leafy vegetable in Africa. Ethno-botanical knowledge on the crop in Ghana has been only incompletely documented and little is known about variation and diversity of local accessions. In order to document available ethnobotanical knowledge and investigate diversity of local accessions, 4000 farmers in twenty districts were surveyed using participatory rapid appraisal tools and techniques. 50 accessions of local germplasm were collected from these districts. Using randomized complete block design with three replications, the 50 accessions collected were characterized based on 12 quantitative and 6 qualitative traits. Results of the survey recorded several vernacular names of the crop and their meaning referring to the slimy nature of the leaves. Some local cultivars which were known in the past are no longer cultivated in farmers’ field suggesting genetic erosion of jute mallow in Ghana. Apart from food, respondents use jute mallow to treat fever, waist pain, stomach problems and loss of appetite indicating nutraceutical potential of the crop. Hierarchical cluster analysis grouped the accessions into four distinct clusters and individuals from the same geographical origin were separately classified. Quantitative traits such as leaf length, number of branches per plant, and number of leaves/plant defined the first principal component. Accessions Cagric 26, Cagric 28, Cagric 41, Cagric 08 and Cagric 01 recorded high yields. These accessions could serve as parents for breeding of improved cultivars. 相似文献

The amphidiploid species Brassica carinata A. Braun is believed to have originated in the plateaus of Ethiopia and has been cultivated there as an oilseed crop since antiquity. The species possesses agronomically important genes of rare occurrence. Although there is a large number of collections in Ethiopia, information on the extent of their genetic diversity is very limited. Thirty-six accessions of ecologically diverse regions were tested at three locations and multivariate analyses on 13 morphological and seed characters were performed. There was generally a large amount of divergence in all characters. Geographic isolation of genes, however, was not observed. Both principal component and cluster analyses disclosed complex relationships among the accessions and characters. Accessions with potential genes of interest to improve earliness, yield components and oil and protein contents have been identified. Length of growing period and yield components contributed most for divergence and clustering pattern. Oil, glucosinolate and protein contents also varied among the accessions but not so much between clusters. Increasing protein in high-glucosinolate genotypes reduced oil content and should be more efficient in genotypes of low-glucosinolate genetic background. 相似文献

Genetic diversity of the Turkish watermelon genetic resources was evaluated using different Citrullus species, wild relatives, foreign landraces, open pollinated (OP) and commercial hybrid cultivars by RAPD markers. The germplasm
was consisted of 303 accessions collected from various geographical regions. Twenty-two of 35 RAPD primers generated a total
of 241 reproducible bands, 146 (60.6%) of which were polymorphic. Based on the RAPD data the genetic similarity coefficients
were calculated and the dendrogram was constructed using UPGMA (Unweighted pair-group method with arithmetic average). Cluster
analysis of the 303 accessions employing RAPD data resulted in a multi-branched dendrogram indicating that most of the Turkish
accessions belonging to var. lanatus of Citrullus lanatus (Thunb.) Matsum et Nakai were grouped together. Accessions of different Citrullus species and Praecitrullus fistulosus (Stocks) Pangalo formed distant clusters from C. lanatus var. lanatus. Among 303 accessions, a subset of 56 accessions was selected representing different groups and a second dendrogram was constructed.
The genetic similarity coefficients (GS) within the Turkish accessions were ranged from 0.76 to 1.00 with 0.94 average indicating
that they are closely related. Taken together, our results indicated that low genetic variability exist among the watermelon
genetic resources collected from Turkey contrary to their remarkable phenotypic diversity. 相似文献
