Pigeonpea is an important pulse crop grown by smallholder farmers in the semi-arid tropics. Most of the pigeonpea cultivars grown to date are selections from the landraces, with a narrow genetic base. With the expansion of the crop to newer areas, problems of local importance are to be addressed. Hence, an economically feasible and faster germplasm evaluation mechanism, such as a core collection, is required. This article describes the development of core collection from 12,153 pigeonpea accessions collected from 56 countries and maintained at ICRISAT, Patancheru, India. The germplasm accessions from 56 countries were placed under 14 clusters based primarily on geographic origin. Data on 14 qualitative morphological traits were used for cluster formation by Ward’s method. From each cluster ≈10% accessions were randomly selected to constitute a core collection comprising 1290 accessions. Mean comparisons using Newman–Keuls test, variances’ comparisons by Levene’s test, and comparison of frequency distribution by χ2-test indicated that the core collection was similar to that of the entire collection for various traits and the genetic variability available in the entire collection is preserved in the core collection. The Shannon–Weaver diversity index for different traits was also similar for both entire and core collection. All the important phenotypic associations between different traits available in the entire collection were preserved in the core collection. The core collection constituted in the present study facilitates identification of useful traits economically and expeditiously for use in pigeonpea improvement.  相似文献

Finger millet [Eleusine coracana (L.) Gaertn.] is an important cereal food crop in Africa and South Asia. It is a hardy crop that can be grown in very diverse environments from almost at sea level to about 2400 m.a.s.l. Finger millet has an excellent food value as its seeds contain protein ranging from 7 to 14% and are particularly rich in methionine amino acid, iron, and calcium. Despite all these merits, this crop has been neglected from the main stream of crop improvement research. One of the means to boost its production and productivity is to enhance utilization of finger millet germplasm to breed superior varieties. Keeping this objective in view, a core subset of finger millet germplasm (622 accessions) based on origin and data on 14 quantitative traits was developed from the entire global collection of 5940 accessions held in the genebank at ICRISAT, Patancheru, India. The comparison of means, variances, frequency distribution, Shannon-Weaver diversity index (H′) and phenotypic correlations indicated that the core subset represents the entire collection. These tests indicated that sampling was optimal and the diversity has been captured very well in the core subset. The correlation analysis indicated that panicle exsertion and longest finger length could be given lower priority in the future germplasm evaluation work of finger millet.  相似文献

Lupinus angustifolius L. is a Mediterranean species, domesticated in the 20th century, representing an important grain legume crop in Australia and other countries. This work is focused on the collection of wild germplasm and on the characterisation of morphological and molecular diversity of germplasm accessions. It reports the collection of 81 wild L. angustifolius accessions from the South and Centre of Portugal, available at the ‘Instituto Superior de Agronomia Gene Bank’, with subsequent morphological and molecular characterisation of a selection of these and other accessions. A multivariate analysis of morphological traits on 88 L. angustifolius accessions (including 59 wild Portuguese accessions, 15 cultivars and 14 breeding lines) showed a cline of variation on wild germplasm, with plants from Southern Portugal characterised by earlier flowering, higher vegetative development and larger seeds. AFLP and ISSR molecular markers grouped modern cultivars as sub-clusters within the wider diversity of wild germplasm, revealing the narrow pool of genetic diversity on which domesticated accessions are based. The importance of preserving, characterising and using wild genetic resources for L. angustifolius crop improvement is outlined by the results obtained.  相似文献

The development of a core collection, one which represents the genetic diversity of a crop with minimal redundancy and increases utility of the collection as a whole, is especially important as the funding for germplasm collections decreases. With limited resources, it is difficult to manage large germplasm collections and disperse genetically diverse germplasm to plant breeders. An algorithm was developed to assist in selection of core collections based on estimates of genetic distance. The criteria for selection of the maximum genetically diverse set were based on rankings of genetic distance between an accession with respect to all other accessions. Depending on the size core which a user wished, a zone around each selected accession was determined and no other accession within these limits was selected. The premise for the algorithm was that the genetic variability represented in the core must be representative of the distribution of genetic distances within the population of interest. In the present study, the algorithm was used with RAPD-marker-based estimates of genetic distance for 270 Theobroma cacao L. accessions and 134 Capsicum accessions that chose a set representing 18.5% of the population and representing the breadth of RAPD-based variation.  相似文献

A field experiment was conducted in Tanzania for two seasons to assess the genetic diversity of bambara groundnut landraces based on morphological characters. One hundred accessions collected from a wide range of agricultural zones in the country were evaluated in a 10 × 10 triple lattice block design at Maruku station in Bukoba, Tanzania. For the qualitative characters evaluated, considerable morphological variations were observed for growth habit, pod shape and pod colour. Quantitative morphological characters such as peduncle length, number of leaves per plant, terminal leaflet width, terminal leaflet length, petiole length, plant spread, plant length, pod width, seed length, seed width, number of pods per plant, shelling percentage and days to 50% flowering showed significant variation among accessions for the two test seasons. Multivariate analysis for 15 quantitative morphological traits that showed significant variation indicated that the first four PCs with eigenvalues ≥1 accounted for 63.0 and 65.0% of the total variance among the accessions during the two test seasons, respectively. The most important loadings for PC1 and PC2 for the two seasons were terminal leaflet width, terminal leaflet length, petiole length, plant spread, plant height and pod length, pod width, seed length, seed width, numbers of pods per plant. Cluster analysis grouped bambara groundnut accessions into three major groups with respect to their geographic origins. Based on this present study, bambara groundnut landraces from Tanzania displayed a considerable diversity for morphological and agronomic traits useful for germplasm management and utilization into crop improvement.  相似文献

Collections of plant genetic resources managed by genebanks function to conserve the range of genetic diversity present in crop genepools. They can facilitate access to valuable allelic variation for both plant breeders and researchers who are able to request germplasm for use in crop improvement and both basic and applied scientific research. The direct impact of genebank collections is often unclear as downstream uses of germplasm samples may not be reported back to the genebank of origin. This study aims to systematically review scientific use of germplasm using the UK Vegetable Genebank (UKVGB) as a model. Between the years of 1980–2016, a total of 271 publications were identified as using UKVGB material. The frequency of publications and the international nature of use increased significantly over the time period studied. Accessions directly sourced from the UKVGB made up the majority of material used by researchers, but material from research-derived resources such as differential sets and core collections or diversity sets have also been used. Resistance to pests and diseases and genetic diversity were the main topics of study although germplasm was used to address a wide range of other research questions. Genebanks such as UKVGB provide an essential resource of allelic diversity in crop genepools which supports a diverse range of research projects. The utilisation of these plant genetic resources has increased over time, contributing to a substantial number of publications. Developments in sequencing technologies have no doubt played a part as larger numbers of accessions can be utilized in a single experiment, but the increase also no doubt reflects a greater interest in the use of allelic diversity to overcome challenges in crop improvement and research.  相似文献

Cleome gynandra L. is a leafy vegetable native to sub-Saharan Africa and Asia and cultivated on dry areas. The plant plays an important role in the food and nutrition security of local communities. The objective of this study was to illustrate variation and diversity in the Cleome collection held at the World Vegetable Center. In total, 242 accessions were cultivated and morphologically characterized and analyzed. The gynophore and filament, both important organs of the flower, played a key role in taxonomical identification. High morphological variation was detected in traits including plant height, pod length, leaf size, flower color, and earliness. Distinct morphological differentiation was detected between Asian and African accessions. On average, the African accessions were larger, less uniform and later in flowering and seed maturation than the Asian accessions. The results were used to establish a core collection of 49 Cleome accessions. Gaps in the current collection were detected. High within-accessions diversity challenges the existing ex situ conservation system; to maintain diversity, seed should be collected from a large number of plants, both during the collection mission and in ex situ regeneration. The results are relevant for germplasm collection strategies and regeneration protocols for good genebank practices.  相似文献

The potato crop originated in the Andean highlands where numerous farmer's varieties and non-cultivated wild species exist. An Andean potato collection is held in trust at the International Potato Center (CIP) to preserve the biodiversity of this crop and ensure the supply of germplasm for potato improvement worldwide. A core collection representing the biodiversity of the Andean potato germplasm is under construction using morphological, molecular, and geographic data. One of the eight cultivated potato species, Solanum phureja, has been genotyped using the RAPD technique. A protocol suitable for large germplasm collection genotyping has been developed to process numerous samples at reasonable costs. From 106 RAPD primers evaluated, we have selected 12 primers yielding 102 polymorphic markers, which unambiguously discriminated all 128 accessions but 2 that are possible duplicates. The S. phureja germplasm collected throughout the Andean countries appears to have a homogeneous genetic constitution. There was no clear geographic pattern as indicated by cluster analysis of the RAPD data. A sub-group of 20 accessions has been identified on the basis of the marker data and selected to maximize molecular (RAPD) variance and polymorphism. The probability of capturing equal amounts of marker polymorphism in this sub-group of 20 accessions by random sampling is less than 40%. This set accessions represents our first group of accessions that may constitute a core of the S. phureja collection. This tentative core will be challenged for diversity content by alternate markers and agronomic traits. Hence, the methodology for sampling less than 10% of the base collection, proposed for core collections by Brown (1989), can be based on molecular marker data provided cost-efficient fingerprints are developed.  相似文献

Forty-two accessions of Arachis duranensis, a wild groundnut species that has been reported as a source of resistance to several groundnut diseases, were studied for 30 quantitative traits including total protein content, oil content, and reaction to groundnut rust. Protein profiles were also investigated for variation at the molecular level. Principal component analysis was applied to 28 traits that showed significant variation. Of these, only five characters, namely, height of the main stem, length of apical leaflet on the main stem, length of isthmus between pods, width of seed, and reaction to groundnut rust, accounted for more than 61.4% of the total variation. Protein profiles of these accessions were broadly similar, except some accessions which differed in few bands. At the genebank of IPK (Leibniz Institute of Plant Genetics and Crop Plant Research) in Gatersleben, Germany, three long-term conservation methods (field genebank, in vitro slow-growth and cryopreservation) are used for mint germplasm. The plant material of the field genebank was the source to establish the slow-growth in vitro culture collection, from which the cryopreserved collection was set up, using a droplet-vitrification protocol. The genetic identity of 161 samples of three mint accessions (MEN 198, MEN 166 and MEN 186), stored for several years using those three methods, was studied using RAPD markers. Accession ‘MEN 198’ was the only one with a unique RAPD fragment pattern for all its samples, independently of the conservation procedure employed. The field collections of accessions ‘MEN 166’ and ‘MEN 186’ were made up of different genotypes. None of the genotypes detected in the plots of these accessions was represented in the in vitro and cryopreserved samples analyzed, which showed a unique genotype for each accession. From this work we conclude that, for an appropriate management of a germplasm collection of vegetatively propagated species, the determination of the genetic composition of the donor material and a periodical assessment of the preserved material should be carried out.  相似文献

A genetic model, including effects of environments, genotypes, and genotype by environment interaction, was employed to analyze five fiber traits of Island cotton (Gossypium barbadense L.). Genotypic values of 304 accessions were predicted by the adjusted unbiased prediction (AUP). Genetic similarities between different accessions were measured by Mahalanobis distances based on genotypic values. Appropriate sampling strategies, linkage rules in stepwise clustering, and sampling proportion were evaluated. To form a core collection of Island cotton, 60 accessions were sampled by the deviation sampling strategy combined with single linkage rule of hierarchical clustering. The genetic variation and structure captured by the core collection were examined in means, variances, ranges and coefficients of variation, correlation coefficients of quantitative traits, and the accessions distribution plotted by first two principal components between two collections. It was showed that the initial collection was well represented by the core collection for exploiting the Island cotton germplasm. These authors contributed equally to this work.  相似文献

The pigeonpea core collection developed at ICRISAT genebank consists of 1,290 accessions from 53 countries. The core collection, which includes selected lines in extra early, early, medium and late maturity groups was evaluated for 18 qualitative and 16 quantitative characters during the 2004 rainy season, to assess the phenotypic diversity and determine the relative importance of different characters in evaluating pigeonpea germplasm accessions. The four maturity groups differed significantly for all characters under study. The medium maturity group showed significantly higher mean number of primary, secondary and tertiary branches, number of racemes, pod bearing length, pods per plant, shelling percent and plot seed yield. Late maturity group showed significantly higher mean for leaf size, plant height, pod length, seeds per pod and 100-seed weight, indicating this group as a good source of vegetable pigeonpea. Significant positive correlations were found between number of secondary branches and pods per plant in extra early group (r = 0.756), late maturity group (r = 0.776) and entire core (r = 0.728) and between number of racemes and pods per plant in all maturity groups and entire core. Principal coordinate and principal component analysis showed that seven qualitative and nine quantitative traits were important in explaining multivariate polymorphism. The Shannon–Weaver diversity index (H′) varied for different maturity groups and traits. Phenotypic diversity, averaged over all the 16 characters, increases from extra early group (0.36 ± 0.04) to late maturity group (0.42 ± 0.04) suggesting that medium and late maturity groups have greater diversity compared to extra early and early maturity groups.  相似文献

The Lakshadweep is a tiny landmass of 32 km2 area (8o–12oN, 71o–74oE) constituted by a group of 27 small atolls located in the Indian territory of Arabian Sea of Indian Ocean. Studies on crop genetic diversity including wild relatives of crop plants of the region are little known except for coconut palm. This report deals with 106 collections assembled in two exploration and collection missions comprising 46 plant species under 35 genera belonging to 24 families. The plant germplasm collected during the trips are mostly useful as vegetable, fibre, medicinal plant, dye, edible tuber, timber and ecosystem service provider. The crop wild relatives collected during the study include that of sunnhemp, melon, yam, jute, pigeon pea, sugarcane, etc. A set of germplasm accessions was sent to national research institutes for regeneration. Out of the total collection, 64 accessions were assigned with national identity (Indigenous Collection or IC numbers: IC 0572014 to IC 0572077), and conserved in long term storage at National Gene Bank or in the field gene bank of ICAR-NBPGR, New Delhi. The important germplasm assembled from the islands include highly pungent betel leaf and a large fruited noni. Plants of Cajanus scarabaeoides (L.) Thouars, bitter less Cucumis melo L. var. agrestis Naudin saline tolerant Canavalia cathartica Thouars, Vigna marina (Burm.) Merr. and Corchorus trilocularis L. were the unique wild relatives collected. The current report is the first of its kind on the exploration and collection of plant genetic resources (other than coconut) from Lakshadweep Islands. Wild melon accessions collected are currently employed in distant hybridization programs. During the survey, ethnobotanic information of 23 plants for medicinal and other uses was also documented. Novel uses of the native plant species in the islands indicate their potential for exploitation in other coastal ecosystems. This report aims to highlight the potential plant resources of the region for direct human use and in future breeding programs.  相似文献

Momordica cochinchinensis (Cucurbitaceae) is native to South East Asia and is important for its nutritional and medicinal properties. There is little information available on morphological and genetic diversity, crop selection and strategic germplasm collection for M. cochinchinensis. In this study, 42 accessions of M. cochinchinensis collected from Vietnam, Thailand and Australia were analysed for diversity study both at a morphological and molecular level. Both morphological and molecular analyses were in agreement and clustered the accessions based on the country of origin. The markers investigated in this study could be used for future applications with samples from unknown origins or for gene tracking for important cultivars. The Momordica accessions were highly polymorphic based on the markers suggesting high genetic diversity. Therefore the results from this study will be important in future strategic germplasm collection, conservation, horticultural development and commercial propagation of M. cochinchinensis.  相似文献

Bambara groundnut an indigenous crop of African origin is drought tolerant and the third most important leguminous crop in Ghana and Sub-Saharan Africa. In order to assess the level of genetic diversity within a small collection of Ghanaian landraces, molecular analysis was performed using microsatellite markers alongside characterization of morphological features. Genetic distance based on Jaccard’s similarity coefficient from the SSR marker analysis ranged from 0.48 to 0.9 among the landraces. 80 individual genotypes were clustered into 17 units with substantial levels of inter- and intra-landrace polymorphism. The analyses of variance from the morphological characterization for all quantitative traits were statistically significant (p < 0.05) except for terminal leaflet width. The first 4 principal components accounted for 41.97, 20.15, 13.39 and 9.81 % respectively of the morphological variations among the landraces. Qualitative traits however accounted for less of these variations. The results of the present study support the availability of high level of polymorphism within the collection of bambara groundnut analysed. This report is useful to crop improvement and germplasm conservation of bambara groundnut in Ghana.  相似文献

A sesame core collection containing 453 accessions was established by assessing genetic diversity of 4251 accessions collected in China. The accessions in the whole collection were grouped based on their origin, varietal and agro-ecological type. A computer program was used to cluster the data of 14 traits from the random small sample. Ward's method was chosen as the desirable method for clustering sesame germplasm resources. A set of technical methods for establishing core collection has been developed through determining the number of selected accessions based on the diversity distribution. The agronomic traits of the pre-selected core collection were investigated and tested in three ecological locations in China for two years. Among six qualitative traits, including plant type and flower color in a total of 5304 accessions, 97.85% was completely the same as the original data, and three quantitative traits, including growth period and plant height, were significantly and positively related with the original data. The representativeness assessment of the final core collection indicated that 35 phenotypes of 10 qualitative traits in the core collection were basically similar to those of the pre-selected core collection. Six feature values such as mean, standard deviation etc. of four quantitative traits including oil content and 1000-seed weight were close to those of the pre-selected core collection. The final core collection possessed a fair representativeness of the pre-selected core collection.  相似文献

The genetic resources of Allium in India are potential source of genes for widening the crop genetic base. Despite their high economic value, limited number of germplasm accessions of wild species have been collected and conserved mainly due to difficult access to areas of occurrence. The present work mainly highlights information on species diversity distribution and utilization of wild Allium species to assess the value of Indian germplasm and prioritization for future collection and conservation programmes.  相似文献

Cornus officinalis is plant species that is widely used in traditional Chinese medicine. To help the efficient utilization and conservation of this species, the genetic diversity of 73 germplasm resources collected from Zhejiang, Henan and Shaanxi provinces in China were used in a primary core collection. The germplasm resources were classified nine types and the analysis of morphological traits and chemical components showed wide variations among the germplasm types. The primary core collection of C. officinalis was evaluated by using 18 inter-simple sequence repeat (ISSR) markers and a mini core collection was developed based on three strategies on construction of a core germplasm resources. The ISSR results showed that a total of 233 alleles were identified in these 73 accessions, the genetic diversity (P?=?91.02%, H?=?0.293, I?=?0.446) revealed with an average of 2 alleles per locus. The genetic diversity index analysis suggested that there is a great richness and uniqueness of genetic variation in this primary core collection. Cluster analysis by UPGMA and STRUCTURE placed the 73-germplasm resources into four linkage clusters. Almost all samples from one sampling location were clustered together. Compared to the RS and SW strategy, the LDSS strategy proved to be more representative in the core collection of C. officinalis. Finally, a mini core collections by LDSS, composed of 18 accessions (24.66% of the primary core collection), let more than 94.85% of polymorphic loci remained, 97.55% of the observed number of alleles and 99.28% of the effective number of alleles of the mini core collection, respectively. The Nei’s gene diversity and Shannon’s Information index of core collection were 0.294 and 0.443, respectively. Taken all together, the results indicated that mini core collections constructed by the LDSS strategy represented their primary core collections and provided a rational framework for intensive surveys of natural variation in C. officinalis genetic resources.  相似文献
