Summary Forty F6 lines, the two parental lines, and a susceptible check cultivar of wheat (Triticum aestivum L.) were inoculated in the young flag leaf stage with leaf rust (Puccinia recondita f.sp. tritici) and evaluated for latent period, receptivity, and uredinium size in a greenhouse experiment. Genotypic (rg) and phenotypic (rp) correlations between latent period and uredinium size were –0.81 and –0.62, respectively. A negative correlation (rg=–0.50, rp=–0.41) was found between latent period and receptivity and a positive correlation (rg=0.28, rp=0.26) between uredinium size and receptivity was found. Area under the disease progress curve (AUDPC) and final rust severity (FRS) obtained from a subsequent field study with common entries were negatively correlated with latent period and positively correlated with uredinium size. Correlations of receptivity with both AUDPC and FRS were not significant. The distributions of F6 family mean uredinia size and latent period were continuous between slow rusting and fast rusting parents: however, the distribution for receptivity was discrete. Narrow-sense heritability estimates were 63%, 57%, and 47% for uredinium size, latent period, and receptivity, respectively. Estimates of the minimum number of effective factors were three for latent period and three or four for the uredinium size and receptivity. Summary Different inoculum densities had negligible effects on latent period, uredinium density and uredinium size measured on flag leaves of adult RL6058 (Thatcher*6/PI58548[Lr34]) plants kept at low (17.5°C) post-infection temperatures in a glasshouse. In a qualitative assessment of rust severity at higher (24.6°C) temperatures, all three components of resistance indicated a susceptible flag leaf response on RL6058. In the latter environment, precise estimations of receptivity to different inoculum densities showed that adult RL6058 plants supported significantly less pustules than the leaf rust-susceptible cultivar Thatcher. In tillering plants, statistically equal numbers of uredinia developed on RL6058 and Thatcher in all paired temperature-inoculum density combinations. Growth stage-related susceptibility, and higher temperatures conducive to a shorter latent period and larger uredinia, could result in high terminal severities of Puccinia recondita f. sp. tritici on wheat genotypes containing Lr34. The reduction in receptivity associated with this gene may contribute, however, to delayed disease increase on cultivars or lines with monogenic Lr34 resistance.  相似文献

An inventory of 481 lines derived from 12 Ethiopian barley (Hordeum vulgare L.) landraces and the checks was made for partial resistance to Puccinia hordei under greenhouse and field conditions at Adet, Ambo and Sinana Agricultural Research Centers in 2003 and 2004 cropping seasons in Ethiopia. The experiments were laid out in a triple lattice design. Each plot consisted of two rows of 1–m long with spacing of 0.20 m between rows. The overall mean leaf rust epidemics varied from area under disease progress curve (AUDPC) of 86 to 1,835. The disease was as high as AUDPC 1,378 on the susceptible check L94. Highly significant variations were recorded between and within the landraces/lines in leaf rust incidence, severity, days to heading, plant height, thousand seed weight and yield. Similarly, the variations between and within barley groups from three altitude areas and three ear-types were significant. Landraces 1686, 3255, 3262 and 3783 had the least and landraces 219900, 3975 and 3980 had the highest leaf rust severity. Of the 481 lines tested, 413 (86%) had significantly lower disease than the susceptible check, but not than the partial resistant check Vada. In contrast, the yields were more for lines with less disease than for those with high. The frequency of resistant landraces/lines was more in altitude 2,301–2,500 m, and irregular and two rows ear-types than in lower altitude areas and six rows ear-type. Nevertheless, the correlation and regression analysis revealed the adverse effect of the disease in the yields of barley. The 413 lines with high infection types at seedling stage and lower AUDPC under field conditions possess partial resistance to leaf rust.  相似文献

The aim of this work was to evaluate the leaf rust resistance introduced into introgressive triticale lines with Triticum monococcum genes, and to study the expression of these genes at the hexaploid level. The introgressive lines were developed by incorporating diploid wheat (T. monococcum s.s.) genes into hexaploid triticale LT 522/6 using the synthetic allotetraploid T. monococcum/Secale cereale (AmAmRR) as a bridging form. A group of 44 those lines, parental stocks and check cultivars were inoculated at the seedling stage (in a greenhouse) and at the adult‐plant stage (in the field) with four pathotypes of Puccinia triticina. At the seedling stage the assessment of infection type showed that four lines had resistance to all pathotypes as high as in the T. monococcum donor. Adult plant examinations showed some introgressive lines with complete resistance and also lines with partial resistance, expressed in area under the disease progress curve (AUDPC) calculations as slow rusting. Some lines comprise low AUDPC with complete resistance at seedling stage.  相似文献

Diploid wheat, Triticum monococcum s.l., is a host for the wheat leaf rust fungus, Puccinia triticina. Some accessions have been reported to show a high degree of prehaustorial resistance. This is non-hypersensitivity resistance, which acts before the formation of haustoria by the pathogen. To assess the frequency of prehaustorial resistance 598 accessions of diploid wheat were inoculated with the wheat leaf rust isolate Felix. Most T. monococcum s.s. accessions (84%) were resistant whereas all T. urartu and all but three T. boeoticumaccessions were susceptible. Histological components analysis revealed that a high percentage of prehaustorial resistance to P. triticina was found in only three T. monococcum accessions. No haustoria were observed in such infection units confirming the prehaustorial nature of the resistance. Prehaustorial abortion of certain infection units in an accession always coincided with posthaustorial abortion of the other infection units. Four components of resistance to Uromyces viciae-fabae, namely, latent period (LP), infection efficiency (IE), pustule size (PS) and spore production (SP) were evaluated on lentil (Lens culinaris) in glasshouse experiments. Four cultivars, Gudo, R-186, FLIP-87-66L and FLIP-89-60L, with different levels of resistance, and the susceptible check EL-142, were included in this study. The cultivars were also compared for area under the disease progress curve (AUDPC), area under the pustule density curve (APDC), apparent infection rate (rG), disease severity (DS), pustule density (PD), LP and PS under field conditions. Gudo and R-186 had significantly smaller and fewer pustules, lower spore yield and longer latent period than EL-142. FLIP-87-66L was intermediate for infection efficiency and pustule size. In addition, significant differences were found between cultivars for AUDPC, APDC, DS and rG. Estimates of AUDPC, APDC, DS and rG were reduced in Gudo, R-186 and FLIP-87-66L compared with the susceptible check EL-142. FLIP-89-60L also showed low AUDPC, APDC and DS. Some of the components obtained in the field were significantly correlated with each other (r = 0.92–0.99, P 0.05) and those measured in the glasshouse. Most of the components studied in the glasshouse were significantly (P 0.05) correlated with AUDPC and DS. Data indicated the existence of incomplete [partial (PR)] resistance in the test cultivars, and the possibility of using IE, LP, SP and PS as selection criteria in the evaluation of PR to rust in lentil. Since there was an interdependence of the components, selection based on more than one component should help obtain lines with higher levels of PR. The AUDPC, DS and rG could also be used for selecting lines with PR in the field.  相似文献

Three barley cultivars, Shyri, Clipper and Terán, with different levels of partial resistance to barley leaf rust, caused by Puccinia hordei, were exposed to six levels of the pathogen. These levels were obtained by 5, 4, 3, 2, 1 and 0 fungicide (Propiconazol) applications respectively and occurred every 15 days starting at 66 days after sowing. No application served as the control treatment. There were three replicates. The six-row plots were surrounded by a row of a highly susceptible barley cultivar in which the barley leaf rust arrived naturally in a fairly early plant stage. The disease severity was assessed five times with 15 days intervals starting at 66 days after sowing. The yields were determined by harvesting the inner four rows of the plots. An analysis of variance was carried out on the area under the disease progress curve (AUDPC) data and on the yield data. ‘Shyri’ was the least susceptible cultivar, ‘Terán’ the most. The percentage yield losses varied with the six levels of pathogen from 0 to 31.5% (‘Shyri’), from 0 to 46% (‘Clipper’) and from 0 to 63.5% (‘Terán’). The yield losses correlated strongly with the AUDPC, the linear correlation coefficient being 0.97. The yield losses indicate the importance of the pathogen in Ecuador. It also means that high levels of partial resistance are needed to prevent significant yield damage.  相似文献

Summary Spot blotch caused by Cochliobolus sativusis considered a major disease problem of wheat(Triticum aestivumL.) in the warm areas of South Asia. This study estimated heritability (h 2) of resistance to spot blotch and its correlation with days to heading DH) and maturity (DM), one-hundred-kernel weight (HKW), and plant height (PHT) in 14 crosses involving four resistant (‘Attila’, ‘Chirya 7’, ‘G 162’, and ‘SW89.5422’) and two susceptible (‘Sonalika’ and ‘HD2329’) wheat genotypes. Data were recorded on F5and F6lines in fields under natural epidemics of spot blotch in 2003 and 2004, respectively. Heritability was estimated for area under disease progress curve (AUDPC), AUDPC/day, and the highest disease score (HDS) using offspring-parent regression (h op 2) and realized heritability (h 2 R) procedures. Heritability estimates were low to high in terms of AUDPC (0.21 < h op 2< 0.64; 0.32 < h R 2< 0.70), AUDPC/day (0.40 < h op 2< 0.96; 0.42 < h R 2< 0.99), and HDS (0.29 < h op 2< 0.92; 0.32 < h R 2< 0.95). The h 2estimates for AUDPC/day were higher than for AUDPC and HDS. Estimates of h R 2were by and large higher than h op 2in the same cross. A weak negative or nonsignificant correlation of spot blotch score with HKW, DH, DM, and PHT indicated that independent selection for resistance and these agronomic traits is possible.  相似文献

One thousand four hundred and seven spring wheat germplasm lines belonging to Indian and CIMMYT wheat programs were evaluated for their leaf angle and resistance to spot blotch caused by Bipolaris sorokiniana during three consecutive crop seasons, 1994–95, 1995–96 and 1996–97.Disease severity was recorded at six different growth stages beginning from tillering to late milk stage. Three crosses (M 3109 × Sonalika, HP 1808 × K 9006 and HD 2662 × K 9006) were made between genotypes with erect and drooping leaves. M 3109 was resistant, Sonalika susceptible while the other three lines possessed moderate resistance to spot blotch. Individually threshed F2 plants were used to advance the generations. Leaf angle and spot blotch resistance were recorded in parents, F1, F3, F4and F5 generations. Leaf erectness in F1 generation showed partial dominance. Evaluation of F3, F4 and F5 progenies(120–150) revealed that leaf angle was under the control of few genes that appeared to be close to three. Germplasm lines with erect and semi-erect leaves displayed lower spot blotch severity than those having drooping and semi-drooping leaves. Lines homozygous for erect leaf posture in F3,F4 and F5 generations showed significantly lower mean AUDPC than those with drooping leaves. A positive correlation (0.58) between leaf angle and AUDPC further indicated a positive influence of leaf erectness on severity to spot blotch disease. The protection provided by the leaf rust resistance gene Lr34 against Puccinia recondita f. sp. tritici was studied in the field over two seasons. In leaf-rust inoculated and fungicide-sprayed control plots, yield of RL6058, the ‘Thatcher’ backcross line with Lr34, was compared to that of the susceptible cultivar ‘Thatcher’. Leaf rust severity remained low on RL6058 in both seasons, but was high on ‘Thatcher’. The latent period of wheat leaf rust isolate 3SA132 in flag leaves of RL6058 was 256 h longer than in ‘Thatcher’. The uredinium density on ‘Thatcher’ was 14.4/cm2, compared to 3.7/cm2 flag leaf surface on RL6058. Leaf rust infection of ‘Thatcher’ reduced the total grain yield per plot by 25.4% and 1,000 kernel mass by 15.6%. Leaf rust caused little or no damage on RL6058 and rusted plots outyielded the control plots by 0.3 %. Seed weight of RL6058 was reduced by 0.7%. Compared to previous greenhouse studies, the adult-plant resistance conferred by Lr34 is more clearly expressed in the field. Evaluation of milling and baking quality characteristics revealed that compared to ‘Thatcher’, RL6058 had a higher flour protein content, but that its milling, dough development and baking properties were inferior.  相似文献

Genetic analysis of common wheat cultivar PBW343 confirmed temperature-sensitive leaf rust resistance and adult plant stripe rust resistance. At low temperatures, PBW343 was resistant to P. triticina (Ptr) pathotype (pt.) 121R63-1, and at high temperature it was resistant to Ptr pt. 121R127. The low temperature resistance to pt. 121R63-1 was attributed to interaction between dominant and recessive genes. The dominant gene involved in low-temperature resistance to pt. 121R63-1 also conferred resistance to pt. 45R35. The high-temperature resistance to Ptr pt. 121R127 was governed by a different single partially dominant gene. Agra Local (a commonly used susceptible check) and IWP94 (a leaf rust differential used in India) are also resistant to pt. 121R127 at high temperatures. An allelism test indicated that PBW343 and IWP94 possessed a common gene for high temperature resistance to this pathotype. The adult plant stripe rust resistance against P. striiformis (Pst) was possibly conferred by one gene in addition to Yr27.  相似文献

Sixteen barley cultivars with a susceptible infection type (IT = 7–8) in the seedling stage to an isolate of race 24 of Puccinia striiformis f. sp. hordei were planted at two locations in México. Disease severity (DS) parameters were assessed for the flag leaf and for the upper three leaves. The cultivars represented at least five levels of quantitative resistance ranging from very susceptible to quite resistant. “Granado”, “Gloria/Copal” and “Calicuchima-92” represented the most resistant group and had an IT of 7 or 8. The cultivar × environment interaction variance, although significant, was very small compared with the cultivar variance. The disease severity parameters were highly correlated. The monocyclic parameter DSm, measured when the most susceptible cultivar had reached its maximum DS, was very highly correlated with the area under the disease progress curve (AUDPC), r being 0.98. Components of quantitative resistance were evaluated in two plant stages. In the seedling stage small cultivar effects for the latency period were observed, which were not correlated with the quantitative resistance measured in the field. In the adult plant stage the latency period (LP), infection frequency (IF) and colonization rate (CR) were measured in the upper two leaves. The LP was much longer than in the seedling stage and differed strongly between cultivars. The differences in IF were too large, those in CR varied much less. Leaf rust caused by the fungus Puccinia triticina is one of the most important diseases of wheat (Triticum aestivum) worldwide. The use of resistant wheat cultivars is considered the most economical and environment-friendly approach in controlling the disease. The Lr38 gene, introgressed from Agropyron intermedium, confers a stable seedling and adult plant resistance against multiple isolates tested in Europe. In the present study, 94 F2 plants resulting from a cross made between the resistant Thatcher-derived near-isogenic line (NIL) RL6097, and the susceptible Ethiopian wheat cultivar Kubsa were used to map the Thatcher Lr38 locus in wheat using simple sequence repeat (SSR) markers. Out of 54 markers tested, 15 SSRs were polymorphic between the two parents and subsequently genotyped in the population. The P. triticina isolate DZ7-24 (race FGJTJ), discriminating Lr38 resistant and susceptible plants, was used to inoculate seedlings of the two parents and the segregating population. The SSR markers Xwmc773 and Xbarc273 flanked the Lr38 locus at a distance of 6.1 and 7.9 cM, respectively, to the proximal end of wheat chromosome arm 6DL. The SSR markers Xcfd5 and Xcfd60 both flanked the locus at a distance of 22.1 cM to the distal end of 6DL. In future, these SSR markers can be used by wheat breeders and pathologists for marker assisted selection (MAS) of Lr38-mediated leaf rust resistance in wheat.  相似文献

Summary A set of 105 European wheat cultivars, comprising 68 cultivars with known seedling resistance genes and 37 cultivars that had not been tested previously, was tested for resistance to selected Australian pathotypes of P. triticina in seedling greenhouse tests and adult plant field tests. Only 4% of the cultivars were susceptible at all growth stages. Twelve cultivars lacked detectable seedling resistance to leaf rust, and among the remaining cultivars, 10 designated genes were present either singly or in combination. Lr13 was the most frequently detected gene, present in 67 cultivars, followed by the rye-derived gene Lr26, present in 19 cultivars. Other genes present were Lr1, Lr3a, Lr3ka, Lr10, Lr14a, Lr17b, Lr20 and Lr37. There was evidence for unidentified seedling resistance in addition to known resistance genes in 11 cultivars. Field tests with known pathotypes of P. triticina demonstrated that 57% of the cultivars carried adult plant resistance (APR) to P. triticina. The genetic identity of the APR is largely unknown. Genetic studies on selected cultivars with unidentified seedling resistances as well as all of those identified to carry APR are required to determine the number and inheritance of the genes involved, to determine their relationships with previously designated rust resistance genes, and to assess their potential value in breeding for resistance to leaf rust.  相似文献

Summary Common and durum wheat populations obtained from Sweden and originally collected in Ethiopia were screened for resistance to steum rust and leaf rust. Resistant selections of common wheat were crossed and backcrossed with either stem rust susceptible RL6071, or leaf rust susceptible Thatcher. Genetic studies, based largely on tests of backcross F2 families, showed that four of the selections had in common a recessive gene SrA. Plants with this gene were resistant (1+ infection type) to all stem rust races tested. This gene was neither Sr26 nor Sr29. The resistance of other selections, based on tests with an array of rust isolates, was due to various combinations of Sr6, 8a, 9a, 9d, 9c, 11, 13, 30, and 36. One of the selections had linked genes, Lr19/Sr25. Another selection had a dominant gene for resistance (;1 infection type) to all the races of leaf rust. With the possible exception of this gene for leaf rust resistance and SrA, no obviously new resistance was found.  相似文献

聚合兼抗白粉病、条锈病和叶锈病的慢病性基因,是培育持久多抗小麦品种的重要措施。百农64和鲁麦21均为慢白粉病品种,分别含有4个和3个慢白粉病抗性QTL。将百农64与鲁麦21杂交,获得21个聚合2~5个慢白粉病抗性QTL的F6株系,于2012-2013年度分别在四川郫县和甘肃天水进行条锈病田间抗性鉴定,在河北保定和河南周口进行叶锈病田间抗性鉴定。分析21个株系条锈和叶锈病的最大严重度和病程曲线下面积,检测单个QTL和QTL聚合体对条锈病和叶锈病的抗性效应。结果表明,QPm.caas-4DL、QPm.caas-6BS和QPm.caas-2BL对条锈病均有显著的抗性,分别解释表型变异的16.9%、14.1%和17.3%;QPm.caas-4DL对叶锈病也有显著抗性,可解释表型变异的35.3%;QPm.caas-1A/QPm.caas-4DL/ QPm.caas-2DL/QPm.caas-2BS/QPm.caas-2BL和QPm.caas-1A/QPm.caas-4DL/QPm.caas-2BS/QPm.caas-2BL聚合体对条锈病和叶锈病的抗性显著高于两亲本,它们均含有来自百农64的QPm.caas-4DL以及来自鲁麦21的QPm.caas-2BL和QPm.caas-2BS,表明这些QTL具有明显的兼抗性效应。在小麦抗病育种中,聚合慢病性QTL越多,慢病性越强,聚合4~5个慢病性QTL时,株系可达到高抗甚至接近免疫的水平,是选育持久抗性小麦品种的重要手段。  相似文献

The objective of this study was to identify polymorphic molecular markers associated with partial resistance to coffee leaf rust, Hemileia vastarix. A segregating F 2 population derived from a cross between the susceptible Coffea arabica cv. Caturra and a C. canephora-introgressed Arabica line exhibiting high partial resistance was analyzed. Rust resistance measured as rust incidence (RI) and defoliation (DEF) was evaluated in field conditions in three consecutive years (2003–2005). During the 2003 season, which was characterized by favorable conditions for a rust epidemic, the F 2 plants exhibited different levels of resistance ranging from very susceptible (50.1% for DEF and 49.5% for RI) to highly partial resistance (9.1% for DEF and 3.7% for RI). Molecular analysis enabled identification of seven polymorphic markers (5 AFLP and 2 SSR) exhibiting significant association with partial resistance. Coexistence of resistance homozygous alleles (RR) at codominant SSR loci was correlated with high resistance. This study is the first attempt to develop PCR-based sequence specific markers linked to partial rust resistance in coffee.  相似文献

Summary The inheritance of the slow rusting character was studied on F5 progenies from seven spring wheat cultivars (Triticum aestivum) crossed in all possible combinations without reciprocals. The cultivars and their progenies were evaluated for slow rusting in 1974 and 1975 in epidemics of Puccinia graminis f. sp. tritici, races 15 and 151, and traces of other races. Slow rusting varied significantly among the parents and among the F5 progeny of each cross. Transgressive segregation occurred in each cross, i.e. some progeny rusted more slowly than the parents and some faster. In crosses with both Idaed 59 and Kenya 58 the progeny distributions were skewed towards slow rust development but the distributions in the other crosses were normal. The genetic control of slow rusting was predominantly additive, and narrow sense heritability was approximately 80 percent. The number of segregating genes having an effect on slow rusting was estimated to be 2 to 12 pairs depending on the cross. Over 540 accessions of wild Lycopersicon species or their crosses with L. esculentum were screened for resistance in a series of trials. Forty-six accessions were selected for the final screening trial based on lower disease ratings in previous trials. Of these, L. hirsutum had the greatest number of resistant accessions, followed by L. esculentum and L. peruvianum. Twenty accessions were quantified for their levels of resistance based on leaf area infected, area under disease progress curve (AUDPC), and the degree of sporulation. There was a significant positive correlation between the AUDPC calculated from 20 accessions evaluated under growth room and field conditions. Five L. hirsutum accessions had no sporulation associated with leaf lesions, whereas L. esculentum accessions had an average of 1.6×104 conidia/cm2 of leaf tissue. There was significant positive correlation between the AUDPC values and the number of conidia per cm2 of leaf tissue.  相似文献

The Indian groundnut cultivars have a narrow genetic base. Hence, it was of interest to investigate the genetic variability among wild Arachis species and their accessions for tolerance to thermal stress. A wide variation was observed in leaf morphological characters such as colour, shape, hairiness, length and width and thickness (SLA). The temperature and time required for 50% leaf injury was worked out with limited number of genotypes and was found to be 54°C for 50 min. Among 36 genotypes (having SLA in the range of 66 and 161 cm2 g−1) screened, the inherent potential for cold as well as heat tolerance in terms of relative leaf injury (RI) was observed. Thus, based on RI-values, A. glabrata 11824 and A. paraguariensis 12042 were identified as heat-tolerant and cold-tolerant genotypes, respectively while A. appresipila 11786 was found to be susceptible to both heat and cold. Correlation between SLA and RI values for heat (r = 0.38, P < 0.05) and cold (r = 0.52, P < 0.05) tolerance was positive, indicating that thicker the leaf the lower the injury or higher the tolerance. Among six species and 13 accessions, comprising both heat-tolerant and heat- susceptible genotypes, the concentrations of various leaf chemical constituents such as total protein, phenols, sugars, reducing sugar, amino acids, proline, epicuticular wax load and chlorophyll varied significantly. The epicuticular wax load ranged between 1.1 and 2.5 mg dm2 among 13 A.glabrata accessions. These accessions were categorized into two groups, i.e. high-wax (range: 2.0–2.5 mg dm2) and low-wax types (range: 1.1–1.6 mg dm2). The high-wax type showed a higher diffusion resistance (dr) as compared to low-wax type; though the transpiration rate (tr) in high-wax type was moderate (between 9.5 and 11.6 μg cm−2 s−1). Genetic variability in parameters such as canopy temperature, dr and tr was also distinct. The fully turgid leaves with relative water content ≥91%, showed leaf water potential (ψleaf) between −0.7 and −1.2 MPa. Results indicated that the plants with thicker leaves are better protected from heat injuries. Further, epicuticular wax load seems to help in maintaining stomatal regulation and leaf water relations, thus affording adaptation to wild Arachis species to thrive under water-limited environments. The sources of tolerance, as identified in this study, could be utilized to improve thermal tolerance of the groundnut cultivars by intra-specific hybridization, following either conventional breeding using embryo rescue techniques, if required or utilizing biotechnological tools.  相似文献
