Cercospora species cause cercospora leaf blight (CLB) and purple seed stain (PSS) on soybean. Because there are few resistant soybean varieties available, CLB/PSS management relies heavily upon fungicide applications. Sensitivity of 62 Argentinian Cercospora isolates to demethylation inhibitor (DMI), methyl benzimidazole carbamate (MBC), quinone outside inhibitor (QoI), succinate dehydrogenase inhibitor (SDHI) fungicides, and mancozeb was determined in this study. All isolates were sensitive to difenoconazole, epoxiconazole, prothioconazole, tebuconazole, and cyproconazole (EC50 values ranged from 0.006 to 2.4 µg/ml). In contrast, 51% of the tested isolates were sensitive (EC50 values ranged from 0.003 to 0.2 µg/ml), and 49% were highly resistant (EC50 > 100 µg/ml) to carbendazim. Interestingly, all isolates were completely resistant to azoxystrobin, trifloxystrobin, and pyraclostrobin, and insensitive to boscalid, fluxapyroxad, and pydiflumetofen (EC50 > 100 µg/ml). The G143A mutation was detected in 82% (53) of the QoI-resistant isolates and the E198A mutation in 97% (31) of the carbendazim-resistant isolates. No apparent resistance mutations were detected in the succinate dehydrogenase genes (subunits sdhB, sdhC, and sdhD). Mancozeb completely inhibited mycelial growth of the isolates evaluated at a concentration of 100 µg/ml. All Argentinian Cercospora isolates were sensitive to the DMI fungicides tested, but we report for the first time resistance to QoI and MBC fungicides. Mechanism(s) other than fungicide target-site modification may be responsible for resistance of Cercospora to QoI and MBC fungicides. Moreover, based on our results and on the recent introduction of SDHI fungicides on soybean in Argentina, Cercospora species causing CLB/PSS are insensitive (naturally resistant) to SDHI fungicides. Fungicides have not been effective in controlling the wheat blast disease in Brazil. An earlier analysis of 179 isolates of Pyricularia oryzae Triticum lineage (PoTl) sampled from wheat fields across six populations in central-southern Brazil during 2012 discovered a high level of resistance to strobilurin fungicides. Here we analysed azole resistance in the same strains based on EC50 measurements for tebuconazole and epoxiconazole. All six Brazilian populations of PoTl exhibited high resistance to both azoles, with in vitro EC50 values that were at least 35 to 50 times higher than the recommended field doses. We sequenced the CYP51A and CYP51B genes to determine if they were likely to play a role in the observed azole resistance. Although we found five distinct haplotypes in PoTl carrying four nonsynonymous substitutions in CYP51A, none of these substitutions were correlated with elevated EC50. CYP51B was sequenced for nine PoTl isolates, three each representing low, medium, and high tebuconazole EC50. Both PoTl CYP51A and CYP51B could complement yeast CYP51 function. All PoTl CYP51A-expressing yeast transformants were less sensitive to triazoles than the PoTl CYP51B ones. Transformants expressing PoTl CYP51A haplotype H1 carrying the R158K substitution were not more resistant than those expressing PoTl CYP51A haplotype H5, which is synonymous to haplotype H6, found in triazole-sensitive P. oryzae Oryza isolates from rice blast. Therefore, the reduced triazole sensitivity of wheat blast isolates compared to rice blast isolates appears to be associated with a non-target-site related resistance mechanism acquired after higher exposure to triazoles.  相似文献

Quinone outside inhibitors (QoIs) and succinate dehydrogenase inhibitors (SDHIs) are major groups of agricultural fungicides. However, resistance to some of these fungicides has been reported in a Japanese population of Puccinia horiana, the causal agent of chrysanthemum white rust disease. Because their mechanisms are not well understood, we investigated the existence of mutations in QoI and SDHI target protein-encoding genes. Eight out of nine isolates from cultivated chrysanthemum carried L275F and L299F amino acid substitutions in cytochrome b, the target protein of QoIs. These isolates showed 23- and 17-fold higher EC50 values for the QoI fungicides azoxystrobin and kresoxim-methyl, respectively, in basidiospore germination inhibitory tests, while they were hypersensitive to another QoI, famoxadone. All nine isolates were resistant to SDHI oxycarboxin and carried the I88F substitution in SdhC. This substitution was orthologous to the SdhC-I86F substitution found in some Brazilian isolates of the soybean rust fungus, Phakopsora pachyrhizi, showing reduced sensitivity to some SDHIs. Although the rarity of wild-type sensitive isolates, the subsequent limited number of comparisons between wild types and mutants, and a difficulty in applying reverse genetic analysis to this obligate parasite, are obstacles in making definitive conclusions, L275F and L299F in cytochrome b and SdhC-I88F are suspected to be responsible for the different patterns of sensitivity to QoI and for oxycarboxin-resistance in P. horiana, respectively.  相似文献

Since 2007, serious damage to tomato from leaf mould caused by Passalora fulva has frequently been observed in commercial greenhouses in Gifu Prefecture, Japan. One of the factors relating to this damage was suspected to be a decrease in azoxystrobin sensitivity of the pathogen. Biological and molecular studies were conducted to characterize fungicide resistance. In in vitro sensitivity tests using mycelial homogenate placed on fungicide‐amended medium, the minimum inhibitory concentrations (MIC) of azoxystrobin for mycelial growth of the isolates divided into two ranges, 0.031–0.5 mg L?1 and 8–32 mg L?1. Isolates with MICs within the two ranges were considered as sensitive and resistant, respectively, to azoxystrobin because, in in vivo tests, the percentage protection conferred by this fungicide (100 mg a.i. L?1) against these isolates was 89.7–100% and 4.5–31.1%, respectively. Resistant isolates had a replacement of phenylalanine with leucine at codon 129 (F129L) in cytochrome b. Forty‐five percent of the 271 isolates collected from 63 tomato greenhouses from 2007 to 2008 were resistant to azoxystrobin. In many greenhouses where the isolation frequency of resistant isolates was 80% or more, azoxystrobin had been used twice per crop for approximately 6 years. In 2012, 27% of the 405 isolates collected were resistant to azoxystrobin, and there was a marked difference in the frequency of occurrence of resistant isolates in the field populations between the three locations sampled. The occurrence of azoxystrobin‐resistant P. fulva isolates (F129L mutants) inflicted considerable damage on greenhouse tomatoes.  相似文献

Stem rot caused by Lasiodiplodia theobromae is an important postharvest disease of papaya in Brazil, responsible for reducing the quality and quantity of fruits. Fungicide use is one of the main disease management measures. However, there are no estimates available of pathogen sensitivity to commonly employed fungicides. Therefore, the EC50 from 120 isolates of L. theobromae from northeastern Brazil, representative of six populations of the pathogen, was estimated in vitro for fungicides of the methyl benzimidazole carbamates—MBC (benomyl and thiabendazole) and demethylation-inhibiting—DMI (imazalil, prochloraz, tebuconazole) groups. Mycelial growth on fungicide-free media and virulence on papaya fruits of the MBC-sensitive and non-sensitive isolates were compared. For MBCs, 8.4% of isolates were non-sensitive to fungicides. For the remaining 91.6%, the mean EC50 ranged from 0.002 to 0.13 μg ml−1 and 0.36 to 1.27 μg ml−1 for benomyl and thiabendazole, respectively. For DMIs, the mean EC50 range for imazalil was 0.001 to 2.27 μg ml−1, 0.04 to 1.75 μg ml−1 for prochloraz, and 0.14 to 4.05 μg ml−1 for tebuconazole. The EC50 values of non-sensitive isolates were significantly (P≤0.05) higher those for the sensitive isolates for each of the DMI fungicides. Differences (P≤0.05) were found in the levels of sensitivity to DMI fungicides among the isolate populations associated with orchards. The populations from two orchards were less sensitive to DMIs. No solid evidence was found for fitness costs relating to MBC non-sensitive isolates because mycelial growth in fungicide-free media and virulence on papaya fruits were similar to those of sensitive isolates.  相似文献

Two hundred isolates ofBotrytis cinerea were collected from greenhouse vegetables between 2003 and 2006 to determine their baseline sensitivity to triadimefone, penconazole, tebuconazole and fenhexamid. Mean values of 50% effective concentrations (EC50) of inhibiting growth were 4.853±5.102, 0.41±0.215, 0.19±0.099 and 0.36±0.891 mgl −1, respectively (mean±SD). Individuals ofB. cinerea in the population differed by a factor (EC50 of the least sensitive isolate/EC50 of the most sensitive isolate) of 6625, 20, 603 and 1800, respectively. Naturally fenhexamid-resistant isolates were detected with an unexpected high frequency of 10% although the pathogen population had never been exposed to this fungicide. The resistance level (mean EC50 of resistant isolates / mean EC50 of sensitive isolates) was 19.5. These naturally resistant isolates also were resistantin vivo, and there was no significant difference in growth rate, conidial production or pathogenicity ability between naturally resistant and wild sensitive isolates. These results indicated that there was a potential risk of practical resistance if fenhexamid was applied alone. Negative cross-resistance was observed between fenhexamid and tebuconazole in 90% of the naturally resistant isolates. A total of 1511 isolates of Phytophthora capsici were collected from farms with no history of exposure to the carboxylic acid amide (CAA) fungicides in 32 provinces in China during 2006 to 2013. All 1511 isolates were assayed for mating type and 403 were assayed for sensitivity to dimethomorph (DMM) and metalaxyl. The DMM EC50 values ranged from 0·126 to 0·339 μg mL?1. Both A1 and A2 mating types were detected on the same farms in four provinces and with a 1:1 ratio. Most isolates were sensitive to metalaxyl but a few exhibited intermediate resistance or resistance to metalaxyl. The segregation of DMM resistance and sensitivity among 337 progeny obtained from hybridization or self‐crossing in vitro indicated that the resistance of P. capsici to DMM is controlled by two dominant genes. Eighteen progeny that were derived from hybridization differed in DMM sensitivity and in fitness. Some progeny were as fit as parental isolates. Given the distribution of mating types and therefore the potential for sexual reproduction, the control of resistance by two dominant genes, and the fitness of hybrid progeny, the risk of P. capsici populations developing DMM resistance in China is substantial.  相似文献

Early blight caused by Alternaria solani is a highly destructive disease of potatoes. Control of early blight mainly relies on the use of preventive fungicide treatments. Because of their high efficacy, azoxystrobin and other quinone outside inhibitors (QoIs) are commonly used to manage early blight. However, loss of sensitivity to QoIs has previously been reported for A. solani in the United States. Two hundred and three A. solani field isolates collected from 81 locations in Germany between 2005 and 2011 were screened for the presence of the F129L mutation in the cytochrome b gene; of these, 74 contained the F129L mutation. Sequence analysis revealed the occurrence of two structurally different cytb genes, which differed in the presence (genotype I) or absence (genotype II) of an intron, with genotype I being the most prevalent (63% of isolates). The F129L mutation was detected only in genotype II isolates, where it occurred in 97%. Sensitivity to azoxystrobin and pyraclostrobin was determined in conidial germination assays. All isolates possessing the F129L mutation had reduced sensitivity to azoxystrobin and, to a lesser extent, to pyraclostrobin. Early blight disease severity on plants treated with azoxystrobin was significantly higher for A. solani isolates with reduced fungicide sensitivity in the conidial germination assay compared with sensitive isolates. Data suggest an accumulation of F129L isolates in the German A. solani population over the years 2009–2011. It is assumed that the application of QoIs has selected for the occurrence of F129L mutations, which may contribute to loss of fungicide efficacy.  相似文献

Fifty-eight isolates of Phytophthora infestans were evaluated in vitro and on detached leaves of potato for their sensitivities to metalaxyl-M and cymoxanil. The isolates belonged to the clonal lineage, EC-1, which is dominant on potato in Peru and Ecuador. All isolates were collected in Huánuco, Peru, an area of year-round potato production, where the potential for development of fungicide resistance is high. All isolates were resistant to metalaxyl-M, with in vitro EC50 values ranging from 468.30—813.57 mg l−1. In contrast, we found no evidence for resistance to cymoxanil for which in vitro EC50 values ranged from 0.03—1.11 mg l−1. Resistance to each fungicide was also evaluated for five isolates in a detached leaf assay in which the fungicide was sprayed on the leaf surface prior to inoculation. With metalaxyl-M, the range of EC50 values was 158.85—828.29 mg l−1, similar to that for the in vitro assay. For cymoxanil, EC50 values ranged from 1.41 to 2.31 mg l−1, which was higher than in the in vitro assay but still two orders of magnitude lower than the concentration applied by farmers in the field.  相似文献

采用生长速率法测定了采自北京平谷区3个桃园的125株桃褐腐病菌对甲基硫菌灵、戊唑醇和异菌脲3种杀菌剂的敏感性,发现甲基硫菌灵对桃褐腐病菌的EC50主要分布在1.0×10-5~0.2μg/mL,戊唑醇对桃褐腐病菌的EC50主要分布在0.006~0.022μg/mL之间。异菌脲对桃褐腐病菌的EC50主要分布在0.15~0.55μg/mL之间。研究结果表明,北京地区的桃褐腐病菌对这3种杀菌剂都比较敏感,未产生明显的抗药群体。建立了褐腐病菌对异菌脲抗药性的敏感基线。而且,数据分析表明:甲基硫菌灵、戊唑醇和异菌脲之间均不存在交互抗性。  相似文献

Asian soybean rust (ASR), caused by Phakopsora pachyrhizi, is one of the most serious diseases of soybean. The soybean landraces PI 594767A, PI 587905 and PI 416764 previously showed high levels of resistance to a wide range of ASR fungus, while the genetic basis of the resistance has yet to be understood. In this study, the ASR resistance loci were mapped using three independent mapping populations, POP‐1, POP‐2 and POP‐3 derived from crosses BRS184 × PI 594767A, BRS184 ×  PI 587905 and BRS184 × PI 416764, respectively. In each population, the resistance to ASR segregated as a single gene, but the resistance was dominant in PI 594767A and PI 587905 and incompletely dominant in PI 416764. The resistance genes from both PI 594767A and PI 587905 were mapped on chromosome 18 corresponding to the same location as known resistance locus Rpp1. Quantitative trait locus (QTL) analysis performed on POP‐3 identified the putative ASR resistance locus in PI 416764 on the defined region of chromosome 6 where Rpp3 was located. The QTLs detected by the mapping explained about 67–72% of the phenotypic variation in POP‐3. Cluster analysis based on disease reactions to 64 ASR populations demonstrated the presence of at least two types of functional resistant Rpp1 alleles: strong and weak allele(s), e.g. soybean accession PI 594767A and PI 587905 carry the strong resistant Rpp1 allele(s). Introducing or pyramiding strong Rpp1 allele(s) in elite soybean cultivars is expected to be useful against the South American rust population.  相似文献

Fungicide sprays on soybean in Brazil have contributed to the selection of less sensitive isolates of Corynespora cassiicola. We collected 59 isolates of Ccassiicola from three Brazilian states and two isolates from Paraguay. We investigated their EC50 to quinone outside inhibitors (QoI) and methyl benzimidazole carbamate (MBC), any cross-resistance to compounds within QoI and MBC groups, and characterized the polymorphisms in their cytb and β-tubulin genes. Local associations of polymorphisms identified in each gene were statistically correlated with assays results. In total, 79% and 74% of the isolates were classified as resistant to QoI and MBC fungicides, respectively. There was positive cross-resistance to active ingredients within QoI and MBC groups. For QoI, all isolates presented heteroplasmy in G143A of cytb gene; the mutations F129L and G137R were not found. For MBC, 63% of isolates possessed E198A and 21% possessed F200Y mutations, associated with reduced control by MBC fungicides. Heteroplasmy was identified in two and one isolates from Brazil with E198A and F200Y mutations, respectively. The resistance factor for isolates with E198A (10.9) was statistically similar to the isolate with F200Y (8.8) mutation. Genic association analysis of the in vitro assays using discriminatory doses proved them to be accurate. Reduced sensitivity of Ccassiicola to QoI and MBC was also identified in isolates from Paraguay and resistance to QoI and MBC was widely present in Ccassiicola isolates from the main soybean-producing states in Brazil. Thus, integrated management measures should be adopted to manage soybean target spot in these countries.  相似文献

Pristine® (pyraclostrobin + boscalid) is a fungicide registered for the control of alternaria late blight in pistachio. A total of 95 isolates of Alternaria alternata collected from orchards with and without a prior history of Pristine® sprays were tested for their sensitivity towards pyraclostrobin, boscalid and Pristine® in conidial germination assays. The EC50 values for 35 isolates from orchards without Pristine® sprays ranged from 0·09 to 3·14 µg mL?1 and < 0·01 to 2·04 µg mL?1 for boscalid and Pristine®, respectively. For pyraclostrobin, 27 isolates had EC50 < 0·01 µg mL?1 and six had low resistance (mean EC50 value = 4·71 µg mL?1). Only one isolate was resistant to all three fungicides tested, with EC50 > 100 µg mL?1. Among 59 isolates from the orchard with a history of Pristine® sprays, 56 were resistant to pyraclostrobin; only two were sensitive (EC50 < 0·01 µg mL?1) and one was weakly resistant (EC50 = 10 µg mL?1). For the majority of these isolates EC50 values ranged from 0·06 to 4·22 µg mL?1 for boscalid and from 0·22 to 7·74 µg mL?1 for Pristine®. However, seven isolates resistant to pyraclostrobin were also highly resistant to boscalid and Pristine® and remained pathogenic on pistachio treated with Pristine®. Whereas strobilurin resistance is a common occurrence in Alternaria of pistachio, this is the first report of resistance to boscalid in field isolates of phytopathogenic fungi. No cross resistance between pyraclostrobin and boscalid was detected, suggesting that Pristine® resistance appears as a case of multiple resistance.  相似文献

Sugar beet is a major crop in Morocco and Cercospora leaf spot is one of its most important fungal diseases. In Morocco, thiophanate methyl (benzimidazole) and difenoconazole (demethylation inhibitor, DMI) have been used extensively in the management of Cercospora leaf spot. In this study, samples of Cercospora beticola Sacc were collected from four major production areas. The identification of all isolates was confirmed using a PCR test with specific primers. Radial mycelia growth of each isolate in unmodified potato dextrose agar medium was compared to mycelia growth in the same medium modified with thiophanate methyl (1, 5, 10 and 50 ppm) or the DMIs difenoconazole, epoxiconazole and tetraconazole (0.1, 0.5, 1, 5, 10 and 50 ppm) or the quinone outside inhibitors (QOIs) azoxystrobin and trifloxystrobin (1, 5, 10 and 50 ppm). The percentage of inhibition obtained was used for the half maximal effective concentration (EC50) calculation. All the isolates showed resistance to the thiophanate methyl molecule to different degrees. Three groups were identified: low resistance with EC50 less than 100 ppm, moderate resistance with EC50 between 100 and 1000 ppm, and very resistant with EC50 more than 1000 ppm. For difenoconazole, 41% of isolates were sensitive (EC50 < 0.05 ppm) and 59% were resistant, while for tetraconazole and epoxiconazole 6% were sensitive (EC50 < 0.01 ppm), 66.7 % had medium resistance (0.01 < EC50 < 1 ppm) and 27.3% were resistant (EC50 > 1 ppm). For QOIs, azoxystrobin was less effective for mycelial inhibition than trifloxystrobin, with 54.5% of isolates with resistance to azoxystrobin (EC50 > 100 ppm).  相似文献

The ascomycete fungus Microdochium nivale is a major pathogen of many species of the gramineae. Control measures rely heavily on chemical fungicides, making alternative means of disease reduction desirable. Phosphite (PO33?), has proven efficacy in reducing susceptibility of different species of gramineae to oomycetes, and has adverse effects on the in vitro growth of numerous other pathogens. The effect of phosphorous acid (H3PO3), phosphoric acid (H3PO4), dihydrogen potassium phosphite (KH2PO3), dihydrogen potassium phosphate (KH2PO4) and potassium hydroxide (KOH) on the in vitro mycelial growth and development of M. nivale was determined. Radial growth on amended potato dextrose agar (PDA) was used to calculate mean daily growth and percentage inhibition. PO33? had a significant inhibitory effect on mycelial growth, with EC50 values ranging between 35.9 and 40.99 μg mL?1, whilst PO43? and KOH had no significant inhibitory effect. Microscopic examination of mycelia showed morphological deformities in hyphae growing on PO33? amended PDA, whilst hyphal growth was normal on PO43? and KOH amended PDA. Conidial germination of M. nivale was significantly reduced following immersion in solutions of 50, 100 and 250 μg mL?1 of PO33?, while PO43? and KOH at the same concentrations induced no inhibitory affect. These results show that PO33? is a significant inhibitor of the growth of M. nivale and may have the potential to be used as a chemical control agent in the field.  相似文献

BACKGROUND: Isolates of Didymella bryoniae (Auersw.) Rehm, causal agent of gummy stem blight on cucurbits, developed insensitivity to azoxystrobin in the eastern United States 2 years after first commercial use in 1998. Baseline sensitivity of this fungus to azoxystrobin has never been reported. The objectives were to compare baseline sensitivities of D. bryoniae from South Carolina and other locations to sensitivities of isolates exposed to azoxystrobin for one or more seasons, and to compare sensitivity in vitro and in vivo. RESULTS: Sixty‐one isolates of D. bryoniae collected before 1998 were sensitive. Median EC50 was 0.055 mg L?1 azoxystrobin (range 0.005 to 0.81). Forty isolates collected after exposure during 1998 also were sensitive. Fifty‐three of 64 isolates collected in South and North Carolina between 2000 and 2006 were insensitive to 10 mg L?1 azoxystrobin. Sensitive and insensitive isolates were distinguished by disease severity on Cucumis melo L. seedlings treated with azoxystrobin (20 or 200 mg L?1). CONCLUSIONS: An azoxystrobin baseline sensitivity distribution was established in vitro for isolates of D. bryoniae never exposed to strobilurins. Baseline values were comparable with those of other ascomycetes. Insensitive isolates were found in fields with a history of strobilurin applications. Sensitivity of field isolates (121) ofBotrytis cinerea from France (1992), Germany (1979–1992), Israel (1990) and the Netherlands (1970–1989) to the triazoles tebuconazole and triadimenol, the benzimidazole benomyl and the dicarboximide vinclozolin were tested in radial growth experiments. Resistance to benomyl (in 21 to 100% of isolates tested) and vinclozolin (in 25 to 71% of isolates tested) was common in most countries. EC50s (concentrations of fungicides inhibiting radial mycelial growth ofB. cinerea on B5-agar by 50%) for tebuconazole and triadimenol ranged between 0.01–1.64 and 0.4–32.6g ml–1, respectively, and were log-normally distributed. The variation factor (ratio between EC50s of the least and most sensitive isolate tested) amounts 164 and 82 for tebuconazole and triadimenol, respectively. These values are comparable to those for azole fungicides applied in control of other pathogens. Hence, variation in sensitivity to triazoles can probably not explain limited field performance of triazoles towardsB. cinerea. Isolates from south west Germany (1992) were significantly less sensitive to tebuconazole than isolates collected earlier in Germany, Israel and the Netherlands. Such less sensitive populations may contribute to the limited field performance of DMI fungicides towardsB. cinerea. The sensitivity of isolates from south west Germany to tebuconazole was similar to that of DMI-resistant mutants generated in the laboratory. These mutants displayed stable resistance with Q-values (ratio between EC50 of resistant mutant and wild type isolate) between 5 and 20. Although fungicide resistance in crop pathogens is a global threat to food production, surprisingly little is known about the evolutionary processes associated with the emergence and spread of fungicide resistance. Early stages in the evolution of fungicide resistance were evaluated using the wheat pathogen Zymoseptoria tritici, taking advantage of an isolate collection spanning 20 years in Oregon, USA, and including two sites with differing intensity of fungicide use. Sequences of the mitochondrial cytb protein conferring single‐mutation resistance to QoI fungicides and the nuclear CYP51 gene implicated in multiple‐mutation resistance to azole fungicides were analysed. Mutations associated with resistance to both fungicides were absent in the 1992 isolates, but frequent in the 2012 collection, with higher frequencies of resistance alleles found at the field site with more intensive fungicide use. Results suggest that the QoI resistance evolved independently in several lineages, and resulted in significant mitochondrial genome bottlenecks. In contrast, the CYP51 gene showed signatures of diversifying selection and intragenic recombination among three phylogenetic clades. The findings support a recent emergence of resistance to the two fungicide classes in Oregon, facilitated by selection for mutations in the associated resistance genes.  相似文献
