This experiment deals with the effects of pre‐slaughter stress and storage temperature on muscle pH, fillet contraction, colour and texture in pre‐rigor filleted farmed cod fillets. The fish were either sedated with a low dose of MS‐222 (14.3 mg L?1) (unstressed groups) or exposed to the air for 3 min (stressed groups) before being submerged in a benzocaine bath (150 g L?1). The fish were then killed by a blow to the head, their gills cut, filleted and finally stored at either 4 or 20°C. The stressed groups had significantly lower pH values after slaughter (pH=7.0) than the unstressed groups (pH=7.3). This difference was maintained until post rigor for the fish stored at 4°C, but at 20°C it was immediately overshadowed by a decrease in pH caused by temperature‐dependent processes. The length contraction and changes in registered colour values were more pronounced at both the higher temperature and the higher level of pre‐slaughter stress. Again temperature dominated, but significant and consistent effects were registered from stress. No significant effects of stress on texture post rigor were observed. It is concluded that high storage temperature masks the majority of effects caused by pre‐slaughter stress on the measured variables. Stress management protocols, however, are important when the fillets are kept at the common storage temperature of 4°C.  相似文献

Improved slaughtering procedures of farmed fish may provide sufficient time so that filleting can be performed pre‐rigor while the muscle pH is still high. Such filleting not only reduces fillet gaping but also lowers the transportation costs and makes fresh fillets available to the markets at an earlier stage. The aim of our work was to determine the weight reduction of the fillets due to liquid loss and to study the proteins and enzymes in the drip. After 11 days of cold storage, fillets of farmed Atlantic cod produced pre‐rigor had a weight loss of 10% and a fillet contraction of 19% while for fillets produced post‐rigor the values were 5% and 4% respectively. At the same time, approximately twice the amount of proteins had been lost from the pre‐rigor‐produced fillets. Sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis showed that the proteins in the drip were similar to the sarcoplasmatic proteins extracted from the muscle. Specific analysis of proteolytic enzymes indicated that they are less stable in the expelled liquid than in the flesh during storage. The extensive loss of weight and proteins from pre‐rigor‐produced fillets during subsequent storage must be taken into account if such processing is considered for farmed cod.  相似文献

The effects of dietary α‐tocopheryl acetate supplementation and different slaughtering methods were investigated on the flesh quality of farmed market‐size turbot Scophthalmus maximus (L). Turbot were divided into three groups and fed commercial diets, supplemented with different levels of α‐tocopheryl acetate at the following dietary inclusion levels: 72 (100), 547 (500), 969 (1000) (mg of α‐tocopheryl acetate kg?1 diet, analytical values with diet codes in brackets). After 5 months, fish (mean weight 1056 ± 19.7 g) from each dietary treatment were sampled, applying three different slaughtering methods: (A) bleeding in ice water; (B) thermal shock, no bleeding; (C) percussion followed by bleeding in ice water. The time course of rigor mortis was evaluated, using pH, rigor index and mechanical compression tests. The results showed that the three parameters corresponded very well. Percussive stunning resulted in higher initial post‐mortem pH (P < 0.01) and a significantly delayed onset of rigor mortis (P < 0.05). Diet significantly affected shelf‐life, with fillets from fish fed diets 500 and 1000 having lower TBARS (thiobarbituric acid reactive substances) numbers from day 2 (P < 0.001) and less colour deterioration from day 7 of storage on ice onwards (P < 0.05). These results suggest that an increase in dietary α‐tocopheryl acetate before slaughter as well as careful selection of the slaughtering method may greatly enhance the flesh quality of market‐size turbot.  相似文献

Food quality aspects of farmed sea bass (Dicentrarchus labrax) were compared following two methods of slaughter: the normal commercial method of killing, by immersion in an ice slurry, or by first electrically stunning the fish, before immersion in an ice slurry. Quality was assessed for up to 10 days of storage on ice after slaughter. No differences were found between the slaughter methods in terms of an overall sensory evaluation of cooked fillets, or in terms of overall carcass quality: overall appearance, internal and external haemorrhage, fin damage, burst gall bladder, staining of the body cavity by leakage from the gut or damage to the spine. Using objective measurements of colour, no differences between fish from either treatment were found in terms of external colour or colour of the fillets. A chemical analysis of flesh nucleotide breakdown products as well as the freshness indicator Ki value did not differentiate the two treatments nor did the industry standard freshness scoring technique (QIM, quality index method), over 10 days of storage on ice. Flesh pH was marginally lower in electrically stunned fish at 4 h post mortem (6.42 cf 6.56) but by 24 h, pH in fish from both treatments had decreased to a similar level (6.22). Humane electrical stunning of sea bass at slaughter neither measurably improved nor decreased product quality for between 1 and 10 days of storage on ice. Electrical stunning accelerated the pattern of onset and resolution of rigor mortis. If electrical stunning were to be widely adopted, re‐education of buyers would be necessary as rigor mortis is currently used by buyers as a proxy measure of fish freshness.  相似文献

Abstract The objective was to evaluate industrial and research slaughter methods for Atlantic salmon (Salmo salar), gilt‐head seabream (Sparus auratus) and eel (Anguilla anguilla) with respect to welfare and quality. As a general term of reference, an optimal slaughter method should render fish unconscious until death without avoidable excitement, pain or suffering prior to killing. For Atlantic salmon, commercial slaughter methods (carbon dioxide stunning followed by gill cutting, and gill cutting alone) are not in conformity with the general term of reference, as the fish are not rendered unconscious immediately and possibly experience stress. Evaluation of automated percussive stunning remained unconclusive. More research should enable us to ascertain whether loss of consciousness is instantaneous. Electrical stunning can be humane if applied properly. However, because flesh of electrostunned fish was characterized by occasional bloodspots, optimization of the electrical parameters is required. Prototypes for percussive and electrical stunning of salmon have been recently developed. This implies that humane slaughter of salmon is feasible for industry. For gilt‐head seabream, neither aphyxia in air nor transfer of the fish to an ice slurry were considered to be humane: the methods did not induce immediate brain dysfunction and vigorous attempts to escape occurred. Percussive and electrical stunning can be in conformity with the general term of reference. However, conditions for stunning whole batches of seabream have not been established. Quality of the fish slaughtered by percussive stunning was similar to that obtained by the industrial method, i.e. immersion in an ice slurry. Further work is required to establish optimal stunning conditions and to develop prototypes. For eel, desliming in a salt‐bath followed by evisceration, electrical stunning performed under the conditions prescribed by the German legislation, and live chilling and freezing were not considered to be humane. In contrast, it was established that a 10–20 kg batch of eels in fresh water could be rendered unconscious immediately and until death by applying electricity in combination with nitrogen gas. The conditions used were 0.64 A dm?2 for 1 s, followed by 0.17 A dm?2 combined with nitrogen flushing for 5 min. A preliminary assessment of flesh quality suggests that it may be improved by application of the latter method, compared with the salt bath. The results clearly indicated that humane slaughter of eels is possible in practice.  相似文献

Fresh Atlantic cod is available in large amounts in Norway during the first 5 months of the year. Live-storage of cod may extend the marketing period of fresh cod products throughout the year. In addition, this concept makes pre-rigor processing possible. The main problem of keeping wild cod in captivity is that it does not easily accept formulated dry feed. The purpose of this study was, therefore, to investigate how long-term live-storage of mature Atlantic cod in the absence of feed (79 days) affects the onset and development of rigor mortis, as well as fillet quality by assessing hardness and water-holding capacity of the muscle, pH, protein, and water content. The results showed that starvation of Atlantic cod for 23 days reduces the pre-rigor time from 29 to 17 h. Further starvation did not decrease this period significantly, suggesting that live-stored cod deprived of feed for 79 days may still be industrially processed before the onset of rigor mortis. More than 51 days of starvation reduced protein concentration and increased water content of the muscle. After 51 days, the muscle texture was softer than in fish starved for a shorter period.  相似文献

In order to test seasonal, rearing, maturing and anaerobic muscle activity effect on the flesh quality of turbot (Scophthalmus maximus) a total of 80 farmed turbot from three different strains from reared under natural or continuous light were killed by a percussive blow to the head in November (winter, Icelandic strain), March (spring, Portuguese strain) and June (summer, domesticated strain (France turbot)). To test the effect of anaerobic muscle activity, 10 fish were on each occasion pre rigor filleted, where one fillet was used as a control, while the other fillet was electrically stimulated using a squared 5 Hz, 10 V pulsed DC for 3 min. All pre rigor fillets were measured for pH, weighed, wrapped in aluminum foil and stored in polystyrene boxes with ice. After 7 days of storage the fillets were measured instrumentally for pH, drip loss, colour (CIE L* a* b*) and texture properties such as hardness and shear force, while fillet shrinkage and colour (RBG) were evaluated with computer imaging on photographs from a standard lightbox. Results showed that softness of the flesh was mainly influenced by factors associated with growth, such as season, photoperiod and maturation. Anaerobic muscle activity simulated with electrical stimulation caused an increase in drip loss (<1%) and loss of shear force (<4%), but had no effect on hardness or fillet shrinkage. Computer imaging revealed that muscle contractions related to the electrical stimulus forced out blood from the fillet causing less reddishness for the entire storage period. We conclude that a pH drop upon slaughter associated with anaerobic muscle activity has a minor effect on the flesh quality in the short run, while seasonal/alternatively genetic effects are predominant.  相似文献

Fish fillet quality may be influenced by the antioxidant level in preslaughter diet. Thus, the effects of dietary α‐tocopheryl acetate supplementation and feeding time on the flesh quality of farmed Atlantic halibut Hippoglossus hippoglossus L. were investigated. Halibut of mean initial weight of 312 ± 12.3 g were divided into two groups and fed commercial diets, supplemented with different levels of α‐tocopheryl acetate at the dietary inclusion levels of 189 and 613 mg kg?1 diet. Fish were sampled after 6,9,12 and 24 weeks. Over the experimental period, they reached a final mean weight of 1320 ± 108.4 g. Tissue α‐tocopherol of fillet and liver was significantly affected by the levels of α‐tocopheryl acetate given with the diets (P < 0.001). In storage on ice, fillets of fish fed the diets high in α‐tocopheryl acetate exhibited significantly lower (P < 0.001) levels of lipid oxidation. The colour of fillets in all groups deteriorated slightly, but diet did not affect this process. Halibut fed the supplemented diets for longer periods were better protected against lipid oxidation (P < 0.001) and colour deterioration (P < 0.01) than those fed for shorter periods. However, after 9 days of storage, lipid oxidation levels were still extremely low [< 0.6 µg malondialdehyde (MDA) g?1 fillet], even in fillets of fish fed the low α‐tocopheryl acetate diet for a short period preslaughter. Different slaughtering methods tested at the end of the trial showed that percussive stunning can delay the onset of rigor mortis by 8–12 h compared with bleeding of the fish. These results suggest that halibut fillets have enhanced shelf‐life stability even at low doses of dietary α‐tocopheryl acetate, and that other factors in the antioxidant defence mechanisms of the species might play a major role in the prevention of lipid oxidation.  相似文献

This study evaluated the effects of AQUI‐S®20E (10% eugenol) sedation on the survival and behaviour of yellow perch Perca flavescens (Mitchill) and Nile tilapia Oreochromis niloticus L. held in high loading densities. Fish were held in 0–300 mg L?1 AQUI‐S®20E (0–30 mg L?1 eugenol) for up to 10 h in static tanks. At 17°C, yellow perch held in 200 and 300 mg L?1 AQUI‐S®20E were lightly sedated for up to 7 h. Yellow perch at 200 and 300 mg L?1 AQUI‐S®20E also had >95% mean survival 7‐days post exposure using loading densities up to 360 g L?1. Nile tilapia were only sedated for ≤3 h in concentrations up to 300 mg L?1 at 22°C and had >90% mean survival at loading densities ≤480 g L?1. Ammonia in tanks increased significantly as loading density increased, but treatment with AQUI–S®20E did not reduce ammonia accumulation. Results suggest that AQUI–S®20E was effective to sedate yellow perch and Nile tilapia at high loading densities, but sedation varied with loading density and species.  相似文献

The aim of this study was to investigate the effect of prehandling stress on the flesh quality of Atlantic cod (Gadus morhua). In order to stress the animal, water was reduced in the tank before a total of 30 fish were caught five at a time using a dipnet and held for 3 min, causing stress by hypoxia. This fish was compared with a control group (n=30) of fish exposed to anaesthetics directly in the tank. All fish were killed by a percussive blow to the head and exsanguinated and stored on ice before flesh quality was measured. Immediately after death and after ice storage for 3 and 8 days, 10 fish per group were measured for muscle pH, texture, fillet gaping, colour, drip loss and cathepsin D. Handling stress resulted in an initial increased fillet lightness, drip loss and decreased fillet shear force, although these differences did level off during ice storage. Stress caused by handling resulted in earlier onset of rigour mortis as compared with the control group, which reached maximum rigour tensions within 26 and 36 h postmortem respectively. We conclude that handling before harvest results in reduced time before entering rigour. After 8 days of ice storage, no effect of handling stress was seen on the muscle pH, flesh colour, fillet shear force, gaping score, drip loss or cathepsin D activity.  相似文献

Anaesthetic agents are very useful for reducing the stress caused by handling, sorting, transportation, artificial reproduction, tagging, administration of vaccines and surgical procedures in fish. The efficacy of two anaesthetics: MS‐222 and AQUI‐S® were tested on rohu, Labeo rohita advanced size fry. The lowest effective doses that produced induction in 3 min or less and recovery times 5 min or less and meet the most criteria of good anaesthetic characteristics were 125 mg L?1 of MS‐222, and 30 mg L?1 of AQUI‐S® in rohu, Labeo rohita advanced size fry. Induction times were significantly decreased with increased in the concentrations of any of the two tested anaesthetic agents. The lowest doses suitable for transportation of rohu advanced size fry observed were: 10–15 mg L?1 of MS‐222 and 2.5 mg L?1 AQUI‐S®. Both anaesthetics showed promising to be used as anaesthetics for handling and transportation in rohu (Labeo rohita) advanced fry.  相似文献

Growth performance and white muscle cellularity were investigated in Atlantic cod (Gadus morhua L.) (500–600 g) to determine if texture and thereby quality is affected by exercise. Three exercise regimes [low flow, 0.5 body lengths per second (BL s?1) and 1.0 BLs?1] were applied, and the experiment was run for 9 months, at which point the fish had reached around 2 kg body weight. There were no significant differences in fork length or condition factors between groups that could be attributed to the exercise treatment, but the 0.5 BL s?1 group had significantly lower body weight performance than the control group. The average specific growth rate ranged from 0.37% to 0.42% per day. Muscle fibre diameter was positively correlated with fish weight. However, there was no difference in the distribution of muscle fibre diameters between treatment groups. A texture analyser, model TA‐XT2, equipped with a Warner–Bratzler shear blade was used to measure the texture of the fillet. No correlation was found between the shear force required to cut through the sample and the muscle fibre diameter, but the shear force decreased with increasing growth rate. A tristimulus colorimeter was used to measure the colour composition of the fillet instrumentally. Red/green chromaticity (a*) was positively correlated with the shear force. It was concluded that the exercise regimes tested were insufficient to influence either growth performance or muscle growth patterns. Individual variations in fibre diameter did not influence the colour or texture of the fillet.  相似文献

Threonine is the third‐limiting essential amino acid in diets based on cereal ingredients. A 4‐week trial was conducted to determine the threonine requirement of large Nile tilapia based on fish growth, feed efficiency, body composition, protein and amino acid retention. Six hundred fish (563.3 ± 15.1 g) were distributed into twenty 1.2‐m3 cages. Five diets containing 288 g kg?1 of crude protein, 12.7 MJ kg?1 of digestible energy and 8.9, 10.5, 12.2, 13.7 and 15.4 g kg?1 of threonine were elaborated. Fish were hand‐fed five times a day to extruded diets. Significantly, differences in growth performance and amino acids retention among the treatments were observed. Fish fed 10.5 g kg?1 of threonine showed higher daily weight gain, gutted weight and fillet weight (P < 0.05) compared to fish fed with other experimental diets. Diets containing 8.9–15.4 g kg?1 of threonine did not affect whole body and muscle proximate composition. Based on second‐order regression analysis, the dietary threonine requirement estimated based on final gain, fillet weight and fillet yield was 12, 12.1 and 11.5 g kg?1 diet, respectively. The dietary threonine requirement for maximum fillet yield of Nile tilapia was estimated to be 11.5 g kg?1.  相似文献

Farmed Atlantic cod with a mean weight of 4.8 kg were maintained for 9 weeks in sea cages and fed diets where the dietary fishmeal component was substituted with increasing proportions (0%, 22%, 63% and 100%) of meal from Antarctic krill, Euphausia superba. Wild‐caught cod were included in the study as external control. At termination of feeding, all fish were slaughtered and muscle pH was immediately recorded. The fish were then stored on ice for three days and assessed for muscle pH and objective (skin and fillet colour, and fillet texture) and subjective (sensory evaluation) quality criteria. Replacement of fishmeal with krillmeal in the diets resulted in the skin colour above and below the lateral line to be more red than the control group without krillmeal substitutions, even though this difference was not significant, and with more yellow hue. Additions of krillmeal increased the muscle whiteness and yellow hue compared with the control group and wild fish. There was no difference in red hue between the groups. Muscle pH, texture or sensory attributes were unaffected by dietary inclusion level of krillmeal. Wild‐caught cod deviated in several aspects from the farmed cod. It is concluded that the replacement of fishmeal with Antarctic krillmeal in the diets two months before slaughter did not move the sensory attributes more towards wild fish.  相似文献

Different stunning methods (carbon dioxide [CO2] and hypothermia) used in industrial fish processing were compared with asphyxia in air, which is used in traditional fishing, to evaluate the effects of these methods on the stress responses and the meat quality of the Amazon hybrid surubim, Pseudoplatystoma fasciatum female × Leiarius marmoratus male. After the application, blood and behavioral indicators were evaluated. Following death by gill cutting, the fish were gutted, and analyses of muscle pH, rigor mortis, the exudation of the meat, and coloration were performed. Significantly higher levels of cortisol and glucose were exhibited by the fish subjected to asphyxia than by the fish stunned by either CO2 or hypothermia. The fish subjected to asphyxia displayed decreasing muscle pH during the first 2 h after death and showed higher rates of rigor mortis after 3 h than the fish that were stunned by the other two methods. The asphyxia is a practice that exposes fish to suffering by causing an increase in stress responses, which affects meat quality. The immersion of fish in water and ice (hypothermia) for 5 min was more effective stunning method than the use of CO2, resulting in higher loss of sensibility and greater welfare.  相似文献

The objective was to assess neural, behavioural responses and product quality in farmed sea bass (Dicentrarchus labrax) upon electrical stunning in seawater. The electrical sinusoidal 50 Hz or pulse square wave alternating 133 Hz current induced a general epileptiform insult with a current of 3.3±0.2 or 3 A dm?2, respectively, for 1 s head to tail in seawater. The total duration of the insult was 48±34 and 23±11 s. After stunning, the electro‐cardiogram revealed fibrillation and ceased or showed malfunction. Product quality was assessed in a group electrically stunned, followed by chilling in ice water and the controls were only chilled in ice water. The pH of the fillets was 0.1–0.2 lower (P<0.01) when stunned electrically at days 1, 2, 8 and 10 postmortem, where the colour did not differ. The percentage of sea bass effectively stunned using an electrical sinusoidal or pulse square wave current was above 85% within a confidence level of 95%. A combination of electrical stunning for 10 s, followed by chilling in seawater with ice flakes resulted in the death of all fish. The former method is recommended to be adapted for implementation in practice.  相似文献

Atlantic salmon (Salmo salar) were fed graded levels of dioxins (polychlorinated dibenzo‐p‐dioxins and polychlorinated dibenzofurans) and dioxin‐like polychlorinated biphenyls (DLPCBs) in their diets for 7 months. The dioxin and DLPCB concentrations in both fillet and whole body of salmon increased with increasing dietary exposure. DLPCBs transferred more efficiently from the feed to edible flesh of salmon than dioxins, and contributed a higher proportion to the total toxic equivalents (TEQ). At the end of the trial, the maximum concentrations of dioxins in fillet and whole fish were 1.9 and 2.3 pg WHO‐TEQ g?1 fresh weight, respectively. Hence with this feeding period even with the most contaminated feed (4.9 pg WHO‐TEQ g?1 dw) the dioxin concentrations in salmon did not exceed the maximum level set by the European Commission [4 pg WHO‐TEQ g?1 (EC 2375/2001)]. The inclusion of DLPCBs in this study provides valuable information for forthcoming risk assessments and the future establishment of maximum limits for these compounds in feed and fish.  相似文献

Supranutritional dietary selenium (Se) has been demonstrated beneficial for fish health, however, its effects on fish flesh quality remain unclear. This study investigated the effects of supranutritional dietary Se on the flesh quality of rainbow trout (Oncorhynchus mykiss) and their potential mechanism. Fish were fed a basal diet supplemented with or without graded Se yeast for 10 weeks. Results showed that Se supplementation significantly increased fillet crude protein and enhanced fillet water‐holding capacity as well as fillet firmness. Supplementing with both 2 and 4 mg/kg Se significantly downregulated the expressions of two autophagy–lysosome‐related genes (autophagy‐related 12‐like and gamma‐aminobutyric acid type A receptor‐associated protein‐like 1) in fish muscle, while supplementing with 4 mg/kg Se also significantly downregulated the expression of two ubiquitin–proteasome‐related genes (muscle RING finger 2 and F‐box protein 25). Correlation analysis indicated that the improved fillet quality parameters were closely correlated with the expressions of these differentially expressed genes. This study revealed that dietary Se was effective for the improvement of rainbow trout flesh quality, and the improved fish flesh quality was associated with the inhibited protein degradation in fish muscle.  相似文献

The study was designed to investigate the effect of four cycles of 5 weeks starvation followed by 10‐week refeeding compared with daily feeding under either natural photoperiod or continuous light (LL) regime on body composition and flesh quality in Atlantic cod in sea cages, northern Norway. The fish were sampled for body composition and flesh quality parameters at the start of the trial, twice at the end of a 10‐week feeding period and twice at the end of a 5‐week starvation period. There was effect of both feeding and light regime on growth, the two starving groups losing weight during starvation and regaining weight during refeeding, and the group under LL being heavier. But, the mean overall growth did not vary between groups. Starvation/refeeding regime resulted in higher slaughter yield, but no overall effect was seen on hepatosomatic index, water content, water holding capacity (WHC), muscle pH, hardness or flesh colour compared with control groups. Continuous light increased gutted weight and slaughter yield, lowered WHC and depressed maturation compared with fish under natural light regime. Increased growth rate resulted in softer fillets and lower muscle pH.  相似文献

Food quality aspects of farmed turbot (Psetta maxima) were compared following two methods of slaughter: the current commercial method, by immersion in an ice slurry, which is then dewatered after approximately 20 min, or by first humanely, electrically stunning the fish using a prototype commercial stunner, before immersion in an ice slurry, which is dewatered after 20 min. Quality was assessed for up to 10 days of storage on ice following slaughter. No differences were found between the slaughter methods in terms of an overall carcass quality: overall appearance, haemorrhage, damage, burst gall bladder, staining of the body cavity by leakage from the gut or damage to the spine. No detectable difference was found between the treatments using the industry standard freshness scoring system, the Quality Index Method. Both groups of fish were classified as ‘Fresh’ after 10 days of storage on ice. Using objective measurements of colour, no differences between fish from either treatment were found in fillet colour. Changes in flesh pH were similar in electrically stunned and traditionally killed fish with a mean pH (±SE) at 2 h post‐mortem of 6.80±0.027 declining to 6.44±0.032 at 24 h post‐mortem. Humane electrical stunning of turbot at slaughter neither detectably improved nor decreased product quality as measured between 1 and 10 days of storage on ice.  相似文献
