Exophiala sp. LHL08, a gibberellin-producing strain, was investigated to assess its effects on cucumber plant growth and heat (40°C) stress tolerance. The results reveal that Exophiala sp. associated plants had significantly higher plant growth attributes (shoot length, plant biomass, chlorophyll contents, and leaf area) than control under heat stress. Endophytic association helped the plants to obtain adequate water to reduce the leaf electrolytic leakage under stress. High-temperature-induced oxidative stress was less pronounced in Exophiala sp. associated plants as shown by enhanced levels of total polyphenol and reduced activities of glutathione, superoxide anion, and lipid peroxidation. To tolerate heat stress and rescue plant growth, the endophyte association significantly increased catalase and peroxidase activities of the host plants as compared to control plants. Contents of palmitic, stearic, oleic, and α-linolenic were significantly decreased in the Exophiala sp.-inoculated plants than control plants under heat stress. Contents of flavonoids like genistein and daidzein were produced in higher quantities, while glycitein content was almost same in endophyte-associated plants under heat stress than control plants. Contrarily, stress-responsive endogenous abscisic acid and jasmonic acid were significantly activated in non-inoculated control treatments as compared to endophyte-inoculated plants under heat stress. The findings of the study reveal that association of Exophiala sp. with cucumber host plants can modulate heat stress by influencing physiological and biochemical contents of plants under heat stress.  相似文献

Cadmium(Cd) is highly toxic to plants, animals, and humans. Limited information is available on the role of nitric oxide(NO)and/or 24-epibrassinolide(EBR) in response of plants to Cd stress. In this study, a hydroponic experiment was performed to investigate the effects of NO and/or EBR on peanut plants subjected to Cd stress(200 μmol L~(-1)) with sodium nitroprusside(SNP, an exogenous NO donor)(250 μmol L~(-1)) and/or EBR(0.1 μmol L~(-1)) addition. The results showed that Cd exposure inhibited plant growth, and this stress was alleviated by exogenous NO or EBR, and especially the combination of the two. Treatment with Cd inhibited the growth of peanut seedlings, decreased chlorophyll content, and significantly increased the Cd concentration in plants. Furthermore, the concentration of reactive oxygen species(ROS) markedly increased in peanut seedlings under Cd stress, resulting in the accumulation of malondialdehyde(MDA) and proline in leaves and roots. Under Cd stress, applications of SNP, EBR, and especially the two in combination significantly reduced the translocation of Cd from roots to leaves, increased the chlorophyll content, decreased the concentrations of ROS, MDA, and proline, and significantly enhanced the activities of superoxide dismutase(SOD), peroxidase(POD), and catalase(CAT) in peanut seedlings. Exogenous NO and/or EBR also stimulated the activities of nitrate reductase(NR)and nitric oxide synthase(NOS) and increased the contents of antioxidants, such as ascorbic acid(AsA) and reduced glutathione(GSH). Furthermore, exogenous NO and/or EBR enhanced Cd accumulation in the cell wall and thus decreased Cd distribution in the organelles in the roots. The concentrations of calcium(Ca), iron(Fe), magnesium(Mg), and zinc(Zn) were also regulated by exogenous NO or EBR, and especially by the two in combination. These results indicated that SNP and EBR, alone and particularly in combination, can mitigate the negative effects of Cd stress in peanut plants.  相似文献

Effect of high NaCl concentration in the nutrient medium on transpiration, abscisic acid, cytokinin and proline content of two soybean varieties With the differentially salt-sensitive soybean varieties ?Lee”? and ?Jackson”? the effect of salinization on transpiration, Cl? and Na+ accumulation, and on abscisic acid (ABA), cytokinin and proline content was investigated. Salinization with 75 mM NaCl in the nutrient medium drastically inhibited the transpiration (about 40%) of both varieties but more so with the variety ?Jackson”?. Nevertheless this variety translocated substantially more Cl? into the shoot than ?Lee”?. However, ?Lee”? accumulated more Cl? into the roots and thus was able to effectively protect the shoot against a toxic Cl? concentration. The Na+ distribution in the roots and shoots was nearly the same in both varieties. The ABA content of the leaves of both varieties increased 5-fold to 1200 ng × g? dry weight after 48 h of salt stress. About the same time transpiration of the salt-stressed plants reached a minimum. Between 48 and 168 h the ABA content of ?Lee”? dropped to about half. The ABA level in ?Jackson”? remained higher which indicated that the shoot was stressed more intensely and/or longer. The results do not imply a causal relationship between the ABA concentration in the leaves and the exclusion of C1? from the shoot of ?Lee”?. The cytokinin concentration of the two soybean varieties was not significantly affected by salinization. The proline content in the leaves increased markedly with salt stress in both varieties but much more so in ?Jackson”?. Proline content in the leaves increased from about 1.8 μmoles × g?1 dr. w. before salt stress to 24.7 μmoles × g?1 dr. w. after 168 h of stress. However, the proline concentration dropped to nearly the initial level within 48 h after a 120 h salt stress treatment was discontinued and the plants were returned to a control solution. In ?Lee”? salinization only doubled the amount of proline found initially. The highest value was observed after 120 h of salinization.  相似文献

Selection of a phytoextraction plant with high Cd accumulation potential based on compatibility with mechanized cultivation practice and local environmental conditions may provide more benefits than selection based mainly on high Cd tolerance plants. In this hydroponics study, the potential of Cd accumulation by three plant species; arum (Colocasia antiquorum), radish (Raphanus sativus L.) and water spinach (Ipomoea aquatica) were investigated. Arum (Colocasia antiquorum L.) plants were grown for 60 days in a nutrient solution with 0, 10 or 50 μM Cd, while radish and water spinach plants grew only 12 days in 0, 1.5, 2.5, 5 or 10 μM Cd. Growth of radish and water spinach plants decreased under all Cd treatments (1.5 to 10 μM), while arum growth decreased only at 50 μM Cd. At 10 μM Cd treatment, the growth of arum was similar to the control treatment indicating higher tolerance of arum for Cd than radish and water spinach. Cadmium concentrations in different plant parts of all plant species increased significantly with Cd application in the nutrient solution. Arum and water spinach retained greater proportions of Cd in their roots, while in radish, Cd concentration in leaves was higher than in other plant parts. Cadmium concentrations in arum increased from 158 to 1,060 in the dead leaves, 37 to 280 in the normal leaves, 108 to 715 in the stems, 42 to 290 in the bulbs and 1,195 to 3,840 mg kg?1 in the roots, when the Cd level in the solution was raised from 10 μM Cd to 50 μM Cd. Arum accumulated (dry weight?×?concentration) 25 mg plant?1 at 10 μM, while the corresponding values for radish and water spinach were 0.23 and 0.44 mg plant?1, respectively. With no growth retardation at Cd concentrations as high as 166 mg kg?1 measured in entire plant (including root) of arum at 10 μM Cd in the nutrient solution, arum could be a potential Cd accumulator plant species and could be used for phytoremediation.  相似文献

Dark septate endophytic (DSE) fungi are ubiquitous and cosmopolitan, and occur widely in association with plants in heavy metal stress environment. However, little is known about the effect of inoculation with DSE fungi on the host plant under heavy metal stress. In this study,Gaeumannomyces cylindrosporus, which was isolated from Pb-Zn mine tailings in China and had been proven to have high Pb tolerance, was inoculated onto the roots of maize (Zea mays L.) seedlings to study the effect of DSE on plant growth, photosynthesis, and the translocation and accumulation of Pb in plant under stress of different Pb concentrations. The growth indicators (height, basal diameter, root length, and biomass) of maize were detected.Chlorophyll content, photosynthetic characteristics (net photosynthetic rate, transpiration rate, stomatal conductance, and intercellular CO2 concentration),and chlorophyll fluorescence parameters in leaves of the inoculated and non-inoculated maize were also determined.Inoculation with G.cylindrosporus significantly increased height, basal diameter, root length, and biomass of maize seedlings under Pb stress.Colonization of G.cylindrosporus improved the efficiency of photosynthesis and altered the translocation and accumulation of Pb in the plants.Although inoculation with G.cylindrosporus increased Pb accumulation in host plants in comparison to non-inoculated plants, the translocation factor of Pb in plant body was significantly decreased. The results indicated that Pb was accumulated mainly in the root system of maize and the phytotoxicity of Pb to the aerial part of the plant was alleviated. The improvement of efficiency of photosynthesis and the decrease of translocation factor of Pb,caused by DSE fungal colonization, were efficient strategies to improve Pb tolerance of host plants.  相似文献

Effect of abscisic acid in the root medium on Cl? translocation in soybeans under low and high salt conditions With soybeans of the varieties ?Lee”? and ?Jackson”? the effect of abscisic acid (ABA) applied to the root medium in different concentrations (10?4, 10?5 and 10?6M) on Cl? translocation was investigated. Under low salt conditions (0,5 mM NaCl) ABA, depending on its concentration, strongly decreased Cl? translocation to the shoot. At the highest ABA concentration, within 24 h the Cl? content of the shoots was reduced to about 40 % of the control, the Cl? accumulation within the roots was reduced about 25 %. However, the 10?5 and the 10?6M ABA treatment increased Cl? accumulation in the roots but decreased Cl? translocation to the shoot. Under high salt conditions (75 mM NaCI) ABA had no measureable effect on CI- translocation to the shoot. It was found that the genetic mechanism responsible for high CI- accumulation within the roots of ?Lee”? and high CI- translocation to the shoot of ?Jackson”? was not affected by ABA. ABA strongly inhibited transpiration. High ABA concentration in combination with low salt treatment decreased the transpiration rate up to 50 % of the control. ABA also inhibited transpiration under high salt conditions. This result is discussed considering the fact, that ABA added to the highly saline nutrient solution did not decrease the CI- translocation to the shoot. An investigation using 14C-ABA showed, the radioactive substance was taken up by the roots and translocated to the shoot independently from the salt treatment. The metabolism of the radioactive substance was different for roots and leaves. However, no difference could be observed by comparing the two soybeans varieties with or without salt treatment.  相似文献

The effects of selenium (Se) cadmium (Cd) interactions on plant growth and metabolism are not fully clear. In the present study, we assessed whether Se could alleviate the toxic effects of Cd on growth and metabolism of maize. Seeds of maize variety FH-985 were sown in pots filled with sand treated with CdCl2 (0, 50 and 100 µM) and Se (0, 2 and 4 mg L?1) through Hoagland’s nutrient solution. Low Se (2 mg L?1) increased germination percentage and rate, while high Se (4 mg L?1) increased fresh and dry biomass under Cd stress. Interestingly, all Se concentrations were effective in alleviating the toxic effects of Cd on photosynthetic pigments, whereas higher Se mitigated the Cd-induced oxidative stress and increased flavonoids both in the shoots and roots while phenolics in the roots. The results demonstrated that root zone Se altered tissue-specific primary metabolism in maize. Furthermore, low Se mitigated the Cd-induced decrease in total proteins in the root. Overall, Se-mediated decrease in the oxidative stress in the shoots while increase of secondary metabolites in the roots helped the plants to grow faster at early growth stage and caused increase in the biomass under different Cd regimes.  相似文献

‘Lvbao-701’ is a cultivar of Chinese flowering cabbage(Brassica parachinensis) that exhibits low cadmium(Cd) accumulation and high Cd tolerance. In this study, this cultivar was compared with a high-Cd accumulating cultivar, ‘Chixin-4’, to characterize the mechanisms influencing Cd accumulation in B. parachinensis. Root cell walls were isolated by dissolving the cytoplasm with an organic solvent, and root Cd and phytochelatin(PC) contents were analyzed. In addition, a PC synthase gene fragment was cloned and expressed under Cd stress conditions. The proportions of Cd bound to root cell walls were higher in the ‘Lvbao-701’ plants(68.32%–76.80%) than in the ‘Chixin-4’ plants(35.36%–54.18%) after exposure to Cd stress. The proportions of Cd bound to root cell walls measured using cell walls isolated with a non-destructive method were higher than those obtained using a conventional method that required grinding and centrifugation. Exposure to Cd stress induced the PC production and resulted in higher PC contents in the ‘Lvbao-701’ plants than in the ‘Chixin-4’ plants. Cloning and expression analysis of a B. parachinensis PC synthase cDNA fragment indicated that PC synthase gene expression was induced by Cd and occurred mainly in the roots of both ‘Lvbao-701’ and ‘Chixin-4’ plants. However, the PC synthase gene expression level was higher in the‘Lvbao-701’ roots than in the ‘Chixin-4’ roots. Therefore, a higher abundance of Cd in the root cell walls of ‘Lvbao-701’ and up-regulated PC production in the roots are probably the main reasons why ‘Lvbao-701’ exhibits lower Cd translocation to the shoots and higher tolerance to Cd than ‘Chixin-4’.  相似文献

The aim of this in situ study was to investigate the fatty acid (FA) composition and content in roots and shoots of Lolium perenne and Trifolium repens, grown under heavy metal stress (Cd, Pb, Zn). The composition of FA was quite similar for the two plants and the two organs; main FA were palmitic acid (C16:0), oleic acid (C18:1), linoleic acid (C18:2) and linolenic acid (C18:3). For both plants, the major FA that characterized the roots was C18:2 whereas C18:3 was the prominent FA in shoots. For the first sampling (S1), in the roots of L. perenne and T. repens, polyunsaturated fatty acids (PUFA) were affected by metal contamination while, in the second sampling (S2), PUFA were affected in the shoots of the two plants. This alteration of PUFA was well correlated with the bioaccumulation factor of metals which decreased in roots and increased in shoots with the time. Moreover, a positive correlation was found between the PUFA decrease and the malondialdehyde (MDA) content, indicating the occurrence of a lipid peroxidation induced by the metal stress.  相似文献

The use of plants for ecological remediation is an important method of controlling heavy metals in polluted land. Cotinus coggygria is a landscape plant that is used extensively in landscaping and afforestation. In this study, the cadmium tolerance level of C. coggygria was evaluated using electrical impedance spectroscopy (EIS) to lay a theoretical foundation for broad applications of this species in Cd-polluted areas and provide theoretical support to broaden the application range of the EIS technique. Two-year-old potted seedlings of C. coggygria were placed in a greenhouse to analyse the changes in the growth, water content and EIS parameters of the roots following treatment with different Cd concentrations (50, 100, 200, 500, 1000 and 1500 mg kg?1), and soil without added Cd was used as the control. The roots grew well following Cd treatments of 50 and 100 mg kg?1. The Cd contents increased with the increase in Cd concentration in the soil. However, the lowest root Cd content was found at 4 months of treatment. The extracellular resistance re and the intracellular resistance ri increased first overall and then decreased with the increasing Cd concentration, and both parameters increased with a longer treatment duration. The water content had a significant negative correlation with the Cd content (P?<?0.01) and the re (P?<?0.05). C. coggygria could tolerate a soil Cd concentration of 100 mg kg?1. There was a turning point in the growth, water content and EIS parameters of the C. coggygria roots when the soil Cd concentration reached 200 mg kg?1. The root water content and re could reflect the level of Cd tolerance in C. coggygria.  相似文献

Infection with Neotyphodium spp. endophytes increases resistance to drought stress and soil mineral imbalances in tall fescue (Festuca arundinacea Schreb. = Lolium arundinaceum (Schreb.) S. J. Darbysh.) and meadow fescue (Festuca pratensis Huds. = Lolium pratense (Huds.) Darbysh.). We hypothesized that resistance of these grasses to salinity stress may also be attributed to endophyte infection. Two tall fescue genotypes, Fa75 and Fa83, and one meadow fescue genotype, Fp60, infected (E+) with their endophytic fungi, Neotyphodium coenophialum (Glenn, Bacon and Hanlin) and N. uncinatum (Glenn, Bacon and Hanlin), respectively, and their noninfected counterparts (E–) were cultured in nutrient solution at three salinity levels of 0, 85, and 170 mM NaCl. Except for genotype Fa75, E+ plants exhibited higher leaf survival rates than E– clones at a high salinity level (170 mM). Root dry matter was higher in E+ than in E– plants, but shoot dry matter was not affected by endophyte infection. This resulted in a lower shoot‐to‐root ratio in E+ plants (1.63) compared with E– plants (2.40). Sodium (Na+) and chloride (Cl) concentrations were greater in roots of E– than in E+ clones. In shoots, Na+ and Cl concentrations were not affected by the endophyte. In contrast, E+ plants accumulated more potassium (K+), which resulted in a greater K+ : Na+ ratio in shoots of E+ than in those of E– plants. Our results show that endophyte infection reduced Na+ and Cl concentrations in tall fescue and meadow fescue roots but increased K+ concentrations in the shoots. Based on these results, we conclude that endophyte‐infected grasses may thrive better in salinity‐stress environments.  相似文献

ABSTRACT

Effect of cadmium (Cd) on biomass accumulation and physiological activity and alleviation of Cd-toxicity by application of zinc (Zn) and ascorbic acid in barley was studied, using semisolid medium culture including 15 treatments [four Cd concentration treatments: 0.1, 1, 5, 50?µmol?L?1, four treatments with addition of 300?µmol?L?1 Zn or 250?mg?L?1 ascorbic acid (ASA) based on these four Cd concentrations, respectively, and three controls: basic nutrient medium, and with Zn or ASA, respectively]. Cadmium addition to semisolid medium, at a concentration of 1, 5, and 50?µmol?L?1, inhibited biomass accumulation and increased malondialdehyde (MDA) content of barley plants, while the addition of 0.1?µmol?L?1 Cd increased slightly dry mass. There was a tendency to a decrease in Zn, copper (Cu) concentrations both in shoots and roots and iron (Fe) in shoots of barley plants exposed to 1 to 50?µmol?L?1 Cd. In addition, there were indications of a stress repose characterized by increased superoxide dismutase (SOD) and peroxidase (POD) activities relative to plants not subjected to Cd. The physiological changes caused by Cd toxicity could be alleviated to different extent by application of 300?µmol?L?1 Zn or 250?mg?L?1 ASA in Cd stressed plants. The most pronounced effects of adding Zn or ASA in Cd stressed medium were expressed in the decreased MDA and increased biomass accumulation, e.g., MDA contents were reduced (p≤0.01) by 4.8%–17.8% in shoots and 0.5%–19.7% in roots by adding 300?µmol?L?1 Zn, in 50?µmol?L?1 Cd stressed plants, and by 1.3%–7.4% in shoots and 2.6%–4.5% in roots by application of 250?µmol?L?1 ASA, respectively. However, ASA addition may enhance Cd translation from root to shoot, accordingly, ASA would be unsuitable for the edible crops grown in Cd contaminated soils to alleviate phytotoxicity of Cd.  相似文献

Chicory (Cichorium intybus L.) and dandelion (Taraxacum officinale Web.) were demonstrated to be potential indicator plants for heavy metal contaminated sites. Chicory, grown with 0.5–50 μM cadmium (Cd) in nutrient solution, accumulated 10–300 μM Cd g?1 in shoots and 10–890 μg Cd μg?1 in roots and rhizomes. With dandelion, 20–410 μg Cd μg?1 was found in shoots and 20–1360 μg Cd μg?1 in roots and rhizomes. An inverse correlation existed between chlorophyll and Cd concentrations in shoots of both species. Accumulation of Cd from nutrient solution was similar with the counter-anions SO4 2?, Cl1? and NO3 ? in chicory. In chicory grown in Cd-amended (11.2 kg Cd ha?1 applied five years previously) soils, Cd concentrations were substantially higher than in controls in all plant parts following the order: leaf > caudex > stem > root and rhizome. The above trend was the opposite of that observed in solution culture, where Cd accumulation was higher in roots and rhizomes than in shoots. Higher cadmium accumulation was found from a Cd-treated sand (Grossarenic Paleudult) than from a loamy sand (Typic Kandiudult) soil type. Chicory and dandelion are proposed indicator plants of cadmium contamination, and both have the potential to be an international standard heavy phytomonitor species of heavy metal contaminantion.  相似文献

Silicon(Si) offers beneficial effect on plants under cadmium(Cd) stress such as promoting plant growth and increasing resistance to heavy metal toxicity. In this study, a pot experiment was performed to study the role of Si in alleviating Cd toxicity in tobacco(Nicotiana tabacum L.) plants on a yellow soil taken from Guiyang, China. Nine treatments consisting of three concentrations of Cd(0, 1, and 5 mg kg~(-1)) together with three Si levels(0, 1, and 4 g kg~(-1)) were established. Plant growth parameters, Cd concentration, and the malondialdehyde(MDA), chlorophyll, and carotenoid contents were determined 100 d after transplanting of tobacco seedlings.Application of exogenous Si enhanced the growth of tobacco plants under Cd stress. When 5 mg kg~(-1) Cd was added, Si addition at 1 and 4 g kg~(-1) increased root, stem, and leaf biomass by 26.1%–43.3%, 33.7%–43.8%, and 50.8%–69.9%, respectively, compared to Si addition at 0 g kg~(-1). With Si application, the transfer factor of Cd in tobacco from root to shoot under both 1 and 5 mg kg~(-1) Cd treatments decreased by 21%. The MDA contents in the Si-treated tobacco plants declined by 5.5%–17.1% compared to those in the non-Si-treated plants, indicating a higher Cd tolerance. Silicon application also increased the chlorophyll and carotenoid contents by 33.9%–41% and 25.8%–47.3% compared to the Cd only treatments. Therefore, it could be concluded that Si application can alleviate Cd toxicity to tobacco by decreasing Cd partitioning in the shoots and MDA levels and by increasing chlorophyll and carotenoid contents, thereby contributing to lowering the potential health risks of Cd contamination.  相似文献

The phytotoxicity imposed by cadmium (Cd) in sunflower was investigated on biomass, Cd accumulation, superoxide and lipid peroxidation product as well as the activities of superoxide dismutase, catalase and peroxidase. Plants accumulate substantial amount of Cd in different parts, the maximum being in roots, i.e., up to 820 μg g?1 dry matter. Cadmium induced oxidative stress, indicated by increase in lipid peroxidation and superoxide content with increase in metal supply. Under Cd stress, the activities of superoxide dismutase and catalase declined to a greater extent in roots than in leaves. Even though the peroxidase activity increased in leaves, a decreasing trend was observed in root due to Cd stress. The threshold of toxicity (10% growth reduction) and toxicity (33% growth reduction) values of Cd in sunflower were 14 and 72 μg g?1 in leaves, 19 and 90 μg g?1 in stem and 65 and 250 μg g?1 Cd in roots, respectively.  相似文献

ABSTRACT

Salinity stress alleviation through arbuscular mycorrhizal fungi (AMF) application and sodium (Na) localization in strawberry plants were investigated. A greenhouse experiment in a completely randomized design with three replications revealed AMF (Gigaspora margarita) association alleviated salinity stress (200 mM NaCl). AMF inoculated plants had greater dry weight, maintained chlorophyll content, and decreased leaves browning compared to the control under salinity stress. The Na+ concentration and Na+/K+ ratio were found lower in the following organs, young and old leaflets and petioles, main roots and lateral roots of mycorrhizal plants than the control. The scanning electron microscope and energy dispersive x-ray spectroscopy (SEM-EDX) analysis of Na in old petiole and main root tissues revealed, excess Na localized in the vascular bundle margin of old petioles and main roots of both the control and mycorrhizal plants. So, suppression of Na absorption through roots might be the mechanism of salt stress alleviation in mycorrhizal plants than to the control Na localization. The higher cellulose and lignin contents in the cell wall of mycorrhizal roots act as the apoplastic barrier which might be suppressing Na influx.  相似文献

Tomato plants (Lycopersicum esculentum Mill, ‘Red Cherry Small’), grown in 15 cm diameter plastic pots with a standard greenhouse medium (1:1:1, by volume, soil:peat:sand) were irrigated for 15 weeks with liquid sewage sludge containing a liquid cationic conditioner (Petroset SB, Phillips Petroleum Company, Bartlesville, Oklahoma) to determine the effect of the conditioner on Cd and Zn availability. Half of the plants received 50 ml week?1 liquid digested sludge with no conditioner and half of the plants received 50 ml week?1 sludge containing 0.25 ml conditioner (200:1, by volume, sludge:conditioner). Plant height was measured weekly. Plants were harvested 3, 6, 9, 12, and 15 weeks after sludge treatments began and separated into roots, shoots, and fruits for dry weight determination and Cd and Zn analyses. Soil and sludge crusts were sampled at the same times and analyzed for extractable concentrations of Cd and Zn. Dry weights of plants grown with conditioned sludge were similar to those grown with nonconditioned sludge. Plants with conditioner flowered and fruited one and two weeks earlier, respectively, than plants without conditioned. Six weeks after treatments began, when the plants had grown to their greatest height, Cd concentrations in sludge crusts, soil, and roots receiving conditioner were 2.0, 1.5, and 2.1 times greater, respectively, than crusts, soil and roots not receiving conditioner. After the six weeks sampling time, Cd concentrations in crusts, soil, and roots receiving conditioned sludge were similar to those in crusts, soil, and roots receiving nonconditioned sludge. At the third-week sampling time, shoots of plants grown with conditioner had 2.6 times more Cd than shoots of plants grown without conditioner. Cadmium concentrations in shoots from both treatments were similar at later sampling dates. Cadmium content of fruits was the same for both treatments all sampling times. Zinc content of roots, shoots, fruits soil, and sludge crusts was no affected by the conditioner. Results showed that a cationic conditioner, added to sludge, increased the availability of Cd, but not of zn, for tomato plants until maximum height was reached.  相似文献

Barley (Hordeum vulgare L. cv. Martin) plants grown in solution culture, were exposed to increasing cadmium (Cd) concentration (0, 5, 10, 25, 50, and 100 μM) for a duration of 12 days. The sequence of important biochemical steps of nitrate (NO3) assimilation were studied in roots and shoots as a function of external Cd concentration. Cadmium uptake in roots and shoots increased gradually with Cd concentration in the medium. This Cd accumulation lowered substantially root and shoot biomass. The nitrate reductase (NR, EC 1.6.6.1) and nitrite reductase (NiR, EC 1.6.6.4) activities declined under Cd stress. Concurrently, tissue NO3 contents and xylem sap NO3 concentration were also decreased in Cd‐treated plants. These results suggest that Cd could exert an inhibitory effect on the assimilatory NO3 reducing system (NR and NiR) through a restriction of NO3 availability in the tissues. We therefore examined, in short‐term experiments (12 h), the impact of Cd on NO3 uptake and the two reductases in nitrogen (N)‐starved plants that were pretreated or not with Cd. It was found that Cd induced inhibition of both NO3 uptake and activities of NR and NiR, during NO3 induction period. The possible mechanisms of Cd action on NO3 uptake are proposed. Further, in Cd‐grown plants, the glutamine synthetase (GS, EC 6.3.1.2) showed a decreasing activity both in shoots and roots. However, increasing external Cd concentration resulted in a marked enhancement of glutamate dehydrogenase (NADH‐GDH, EC 1.4.1.2) activity, coupled with elevated levels of ammonium (NH4 in tissues. On the other hand, the total protein content in Cd‐treated plants declined with a progressive and substantial increase of protease activity in the tissues. These findings indicate that under Cd stress the usual pathway of NH4 assimilation (glutamine synthetase/glutamate synthase) can switch to an alternative one (glutamate dehydrogenase). The changes in all parameters investigated were concentration‐dependent and more marked in roots than shoots. The regulation of N absorption and assimilation by Cd in relation to growth and adaptation to stress conditions are discussed.
