Summary Variation for resistance toPuccinia graminis f.sp.tritici, P. recondita f.sp.tritici andP. striiformis was induced in theTriticum aestivum cultivar Lalbahadur using nitrosomethyl urea. Variations were isolated from the M2 population in the post-seedling stage in the field when infected with a mixture of races of each of the three rusts. Plants exhibiting simultaneous resistance to stem rust, leaf rust and yellow rust were indentified. Repeated screening in the subsequent generations confirmed the resistance of the mutant lines that are morphologically similar to the parental cultivar. The rust resistance of 20 mutant lines was also confirmed at the seedling stage using individual races of stem rust and leaf rust. The different patterns observed in the mutant lines tested against a wide range of races show that these lines can be used as components of a multiline. The patterns of variation compared with those of the known genes for resistance against the Indian races of the pathogens suggest that the mutations for rust resistance are due to factor different from those already known in bread wheat, providing a broadened genetic base for future breeding programmes.  相似文献

New sources of resistance to crown rust, Puccinia coronata f. sp. avenae (Eriks.), the major fungal disease of cultivated oat, Avena sativa L. (2n = 6x = 42), are constantly needed due to frequent, rapid shifts in the virulence pattern of the pathogen. Crown rust resistance identified in the diploid oat A. strigosa (Schreb.) (2n = 2x = 14) accession CI6954SP was transferred into cultivated oat using two methods: direct cross of the diploid to a hexaploid cultivar facilitated by embryo rescue, and initial cross of the diploid to a wild tetraploid oat to make a synthetic hexaploid for subsequent crossing to a hexaploid cultivar. Two tetraploids, a crown rust resistant A. murphyi (Ladiz.) accession P12 and a susceptible A. insularis (Ladiz.) accession INS-1, were used in the 2x·4x crosses. Resistant backcross-derived lines were recovered by both methods. Although the 2x·4x synthetic method did not require the laborious discovery and rescue of an infrequent initial hybrid embryo of the direct cross, the direct cross method provided more rapid backcross recovery of plants with high fertility, full transmission of resistance, and desired plant and seed phenotypes. A suppressor effect, present initially but segregating in backcrosses, appeared to come from the CI6954SP donor and is the same as, or analogous to, suppression by an oat line with the crown rust resistance gene Pc38. No resistance from A. murphyi P12 was detected in advanced generations when it was introduced either as a component of a synthetic hexaploid or in direct crosses to A. sativa, indicating suppression of its resistance in interploidy combinations. That the dominant resistance gene transferred from CI6954SP and a gene Pc94 introgressed earlier from a different A. strigosa accession may be the same or quite similar to one another is indicated by their in-common specificity to suppression of resistance expression, susceptibility to a newly recovered rust isolate, and close linkage to the molecular marker SCAR94-2. The introgressed resistance genes from the different sources, even if the same, may have different cultivar genomic introgression sites, which would allow tests of dosage effects on resistance expression.  相似文献

Summary Although wild oats (Avena fatua L.) have been considered a potential source of genes for cultivated oat (Avena sativa L.) improvement, most progenies of A. sativa/A. fatua crosses have weak straw and are very susceptible to crown rust (Puccinia coronata CDA. var. avenae Fraser and Led.). Backcrossing to A. sativa has been suggested as a method of improving progeny lines while introducing new genes from wild oats to cultivated oats. In this study, A. sativa/A. fatua F1 hybrids were backcrossed twice to A. sativa, and lines from three backcross populations were selected on the basis of agronomic performance in segregating generations. The A. sativa recurrent parents were Dal (tall and late) and Stout (short and early).Backcross lines and recurrent parents were evaluated in five performance trails from 1983 through 1985. There was significant variation among backcross lines for most traits, but most backcross lines did not produce higher grain and straw yields than their A. sativa parent. Several backcross lines were higher than their recurrent parent in test weight and groat percentage. A line derived from Stout, 175BC2-6, was considered the most promising backcross line in the study. This line produced more grain, had heavier kernels, and headed 3.3 days earlier than Stout. Although 175BC2-6 does not have sufficient straw strength and crown rust resistance to be released as a cultivar, it is considered to be a new source of high grain yield, high test weight and earliness for oat breeding.  相似文献

Summary To understand how multiline cultivars of wheat develop better protection against leaf rust, seven experimental multilines with 0, 28, 40, 50, 58, 60 and 70% susceptibility were subjected to leaf rust epiphytotics in the field along with their pure line components. A mixture comprising 12 leaf rust races, 10, 11, 12, 17, 20, 63, 77, 106, 107, 108, 162 and 162 A was used.Both the initial inoculum (Xo) and rate of increase (r) of leaf rust were substantially reduced in the multiline cultivars. Xo was reduced by 45–75% and the over-all infection rate (r) by as much as 16% over the average of components.As a result of reduced Xo and r, the intensity of leaf rust in the multilines was also significantly affected at all stages of rust development. It was reduced from 32,10 to 89.54% over the average of components differing from one multiline to another and also from time to time. The susceptible recurrent parent, Kalyansona at the peak period of rust infection exhibited 86.75% severity while in the multilines it ranged from 5.80 to 35%.The rate of increase in the multilines was found to be proportional to the logarithm of the proportion of susceptible plants in the host mixture.Further, it was found that even if as many as 50% susceptible plants are present in a multiline they would not suffer much from leaf rust damage.  相似文献

Summary Two lines of descent were established from an F3 bulk lot of oats (Avena sativa L.) initiated by mixing seeds from approximately 250 crosses. For one line of descent, seeds were radiated with thermal neutrons or X-rays from F3 through F6, followed by five generations of bulk propagation. The second was propagated for 10 generations. No artificial selection was practiced in either line of descent. Grain yield data from 20 random strains from each of four generations from the radiated (F7, F8, F9, and F11) and five from the nonradiated (F3, F6, F7, F8, and F12) line of descent and 20 check cultivars tested in 14 environments were used for estimating regression stability indexes of oat strains.The 14 environments were assigned randomly to two sets of seven, and regression stability indexes were computed for the 180 experimental oat strains for both sets. Intrageneration correlations between regression stability indexes from the two sets of environments ranged from –0.35 to 0.64 (18 d.f.), and only one of nine was significant, indicating poor repeatability for estimates of this statistic computed from different sets of environments.Correlations between regression stability indexes from two sets of environments, one in which the environments varied by soil nitrogen levels and a second in which they varied by soil phosphorus levels, ranged from –0.01 to 0.28, none of which was significant.The relative magnitudes and ranking of the regression stability index values for the oat genotypes were nearly identical when environmental productivity indexes were assessed with any number of check cultivars from 2 to 20.Journal paper No. J-8080 of the Iowa Agriculture and Home Economics Experiment Station. Summary Maintaining maize (Zea mays L.) inbred lines without genetic change is an important concern of maize breeders and seed producers. Long-time inbred lines, however, have been shown to be genetically unstable, and with selection pressure acting on the resulting genetic variation, these lines have evolved into different strains. The objective of this study was to compare maintenance of line integrity under reproduction by sib-mating and self-pollination in ear-to-row progenies. Ten lines, varying from 5 to 35 generations of previous ear-to-row selfing, were maintained for 11 successive generations under each method of reproduction. Alternate generations were compared by using a randomized complete-block design with each family of lines as a separate experiment. Data were collected for 10 plant and yield traits: pollen shed and silk emergence dates, plant and ear heights, tassel branch number, kernel row number, ear length and diameter, grain yield, and 300-kernel weight. Summarizing for F-tests over all traits and experiments, 30% were significant among sib-mated generations and 56% among selfed generations. Also, overall generation means of the two methods were different for 51% of the comparisons, and the selfed lines were less vigorous for 79% of the significant comparisons. It was concluded that sib-mating lessens the effect of genetic instability and that a reproduction system of sib-mating with intermittent generations of selfing may be more beneficial than continuous ear-to-row selfing for certain lines.Joint contribution: USDA-ARS, and Journal Paper No. J-11880 of the Iowa Agric. and Home Econ. Summary The oat line Pc54 was found to be resistant to powdery mildew under both field and glasshouse conditions. The ratio of resistant to susceptible F2 and F2 progeny of a cross between a selection from the Pc54 line (Cc7422) and a susceptible cultivar (Selma) showed that, in addition to carrying the crown rust resistance gene Pc54 and the pg15 gene for stem rust resistance, the mildew resistance of the Pc54 line was conditioned by a single incompletely dominant gene along with additional factors which modified the expression of resistance. Previous results, that there was no linkage between genes Pc54 and Pg15, were confirmed. In addition, there was no evidence of linkage between the mildew resistance gene and gene Pc54. Evaluation of selections from within the Pc54 line showed that the expression of both stem rust and mildew resistance was modified by, or linked to, plant height. The effectiveness of genes Pc54 and Pg15, as measured by virulence frequencies, in central and eastern Europe is described.  相似文献

Summary A partial male sterility system in the soybean (Glycine max (L.) Merr.) germplasm population AP6(SI)CI was found to be controlled monogenically by a recessive allele, msp. Observations of msp msp plants in different environments suggested that environmental conditions significantly affect expressivity of the msp allele with respect to male sterility. We obtained no experimental evidence of cytoplasmic effects on msp expression. Summary Using the cultivar Arina as the recurrent parent, six backcrosses were made with two donor lines carrying the leaf rust resistance genes Lr1 and Lr9, respectively. Selection for leaf rust resistance occurred at the seedling stage in the greenhouse; the first plants transferred to the field were BC6F4s. Frequency distribution of the 332 Lr1/7 × Arina and the 335 Lr9/7 × Arina lines showed continuous variation for yellow rust resistance and heading date in these leaf rust near-isogenic lines (NILs). Similar results were also obtained for plant height, for resistance to powdery mildew and glume blotch, as well as for baking quality characters in another set of more advanced NILs. The available information on the behaviour of one of the parents of cultivar Arina led to the conclusion that the expressed yellow rust resistance is quantitative and might possibly be durable.  相似文献

The genetic bases of leaf rust resistance in wheat (Triticum aestivum L.) line CSP44, selected from the Australian cultivar Condor, and Indian cultivar VL404, were studied. The reaction patterns of CSP44 and VL404 against Indian races 12, 77, 77-1, 77-2, 77-3, 77-4, 77-5 and 108 were different from reaction patterns shown by near-isogenic lines with known adult plant resistance (APR) genes, viz. Lr12, Lr13, Lr22b and Lr34. Although the reaction patterns of CSP44 and VL404 were similar to the near-isogenic line Tc+Lr22a, tests of allelism indicated absence of Lr22a in both CSP44 and VL404. On the basis of genetic studies, their resistances in field tests against race 77-5, the most virulent race from the Indian sub-continent, were each ascribed to two genes. One of the two genes in each wheat was identified to be the non-hypersensitive APR gene Lr34. The second APR genes in CSP44 and VL404 gave hypersensitive reaction types and were recessive and dominant, respectively. The gene in CSP44 was designated Lr48and the gene in VL404, Lr49. Summary A chromosome interchange in soybean (Glycine max (L.) Merr.) was studied agronomically and genetically, and comparisons of seed or abortion position within the pod were related to chromosome structure. Comparisons were among plants heterozygous for a chromosome interchange (N/T), plants homozygous for the interchange (T/T), and homozygous normal-chromosome plants (N/N). The latter two genotypes were male fertile and female fertile. Heterozygous interchange plants, which are about 50% pollen sterile and ovule sterile, are typical of a large number of plants that have equally frequent alternate and adjacent chromosome segregation. Yield, lodging, plant height, and seed oil and protein percentages among all three genotypes were similar even though significant differences existed for seed weight, seed number, and pod number per plant. Seed abortions were more frequent in the basal position of the pod than in either the middle or apical positions in N/N and T/T genotypes. Ovule abortions in N/T plants were equally frequent among all positions both in two- and three-ovule pods. Summary Gene action and heritability of groat protein percentage were determined in F1, F2, and F3 generations of nine crosses between three Avena sativa L. cultivars and three A. fatua L. selections. Relationships among groat protein percentage, grain yield, and 100-seed weight also were evaluated. The three A. sativa parents were Dal (high grain yield and intermediate groat protein percentage), Goodland (low grain yield and high groat protein percentage), and Stout (high grain yield and low groat protein percentage). The three A. fatua parents were chosen for the study on the basis of vigorous plant growth and high groat protein percentage. The study was conducted at Madison, Wisconsin in 1979 and 1980.There was partial dominance towards low groat protein percentage. Narrow sense heritability estimates for groat protein percentage were low in Dal and Goodland crosses and intermediate in Stout crosses. In the F2 generation, groat protein percentage was significantly higher in shattering than in nonshattering plants in 1979, but not in 1980. There were significant, positive correlations between groat protein percentage, 100-seed weight, and grain yield in F1 and F2 generations, but they were not large numerically. Relationships among these traits were either negative or nonsignificant in the F3 generation. Although our results indicated that selection for higher groat protein percentage is possible when a low protein A. sativa cultivar is used, most of our simple cross progenies from A. sativa x A. fatua crosses had weak straw and were susceptible to crown rust (Puccinia coronata Cda. var. avenae Fraser and Led.).  相似文献

Summary Our objective was to determine the average numbers of pollen grains from fertile plants (Ms 1) and the average numbers of coenocytic microspores from genetic male sterile plants (ms 1 ms 1) in soybeans, Glycine max L. Merr. Comparisons were made between the average numbers of pollen grains and the average numbers of coenocytic microspores with respect to environment where plants were grown and to stamen position in the flower. Five male sterile lines were used. They included the North Carolina ms 1 mutant, the cultivar Hark with the ms 1 gene, and mutants identified as the Urbana, Tonica, and Ames male steriles. Three environments used were the Agronomy Farm, University of Georgia, Athens, Georgia; the Agronomy and Agricultural Engineering Research Center, Iowa State University, Ames, Iowa; and the Agronomy Greenhouse, Iowa State University, Ames, Iowa.Pollen production from fertile plants varied from 374 to 760 pollen grains per anther among genetic lines and environments. This variation may be an important consideration in selecting a male parent to use as a pollinator for hybrid seed production.Among fertile plants, the average numbers of pollen grains per anther of the separate stamen and and of the lower whorl of stamens were significantly different only in greenhouse-grown plants. Among male sterile plants, the average numbers of coenocytic microspores per anther of the separate stamen and of the lower whorl of stamens were significantly different in three genotype x environment combinations. These three exceptions did not conform to any genetic or environmental pattern. Summary Molecular mapping is a promising strategy for studying and understanding traits with complex genetic control, such as partial resistance to oat crown rust. The objectives of this research were to develop molecular maps from the progenies of the cross UFRGS7 (susceptible) × UFRGS910906 (partially resistant) and to identify QTLs (quantitative trait loci) associated to partial resistance to oat crown rust in two generations of that population.DNA of 86 genotypes of the F2 and 90 genotypes of the F6 UFRGS7 × UFRGS910906 population were used to generate AFLP markers. Molecular maps were constructed using Mapmaker Exp. 3.0 and QTLs for partial resistance to oat crown rust were identified with Mapmaker/QTL software. Five hundred and fifty seven markers in the F2 and 243 markers in the F6 generations were identified. The F2 map integrated 250 markers in 37 linkage groups. The F6 map integrated 86 markers in 17 linkage groups.Five QTLs were identified for partial resistance to oat crown rust in the F2 generation and three QTLs in the F6. The QTL identified on F6 through the PaaaMctt340 AFLP marker showed consistency across two environments and two generations (F4 and F6), and appear to have potential for marker-assisted selection in oat.  相似文献

Summary The variation among the component lines of the KSML 3 (a multiline based on cultivar Kalyansona, spring bread wheat) was studied for agronomic characteristics. For days to earing and plant height the variation was small. This helped in imparting uniformity to the multiline. The lines had an improved tillering ability and had larger seeds. This partially explained the increased yield potential of the multiline as compared with Kalyansona.All the lines were susceptible only to one or two races of the yellow and brown rust. In no case any race was virulent against all the lines. In the field all the lines were resistant to both rusts.  相似文献

Summary Two sets of near-isogenic lines of wheat carrying single genes for stem rust resistance were grown in yield tests to determine whether the resistance genes were deleterious. One set was based on the cultivar Marquis and the second set on a susceptible, day-length insensitive line, LMPG. The results indicated that the effects of resistance genes vary with different genes and different environments. However, there appeared to be a tendency for resistance genes to reduce yield. In most cases the reductions were too small to be of much concern to wheat breeders.  相似文献

Summary Cluster analysis techniques were used to examine phenotypic variation within the USDA Soybean Germplasm Collection. This study included ten Plant Introductions each of the domesticated soybean, Glycine max (L). Merr., the wild soybean, G. soja Sieb. & Zucc., and a disputed species, G. gracilis Skvortz. G. max and G. soja were found to be morphologically distinct entities and G. gracilis was found to be conspecific with G. max. Summary Investigations of variable expression of msp partial male sterility in soybeans (Glycine max (L.) Merr.) showed that higher temperatures promote male fertility in msp homozygotes and showed that infectious agents are not intrinsic to the sterility system. Exchange grafts failed to modify fertility levels of msp msp rootstocks, Msp Msp scions, and their self-progeny. Tests for soybean mosaic virus and tobacco ringspot virus were negative in partially male-sterile plants, in control fertile plants, and in self-progeny of grafted plants. Growth-chamber experiments and field observations manifested that male fertility of msp msp plants is higher in hot environments than in cooler ones. The unexpected aberrant ratios of fertile to partially male-sterile plants observed in 1977 (Stelly & Palmer, 1980) are explainable on the basis of msp temperature sensitivity.Our observations suggest that homogeneous msp msp populations may be increased in hot environments.Research Geneticist, SEA-AR, USDA, Iowa State University, Ames, Iowa 50011, USA.  相似文献

Summary True-breeding, leaf rust (Puccinia coronata Cda. f. sp. avenae Fraser and Led.) resistant oat lines with somatic chromosome numbers of 42 and 44, were recovered in the advanced generations of a pentaploid hybrid from the cross between C.I. 7232, a leaf rust resistant and dark lemma colored derived tetraploid and Clarion, a susceptible hexaploid oat. Microsporocyte observations and breeding behaviour of these lines and their F1 hybrids with susceptible hexaploid oat cultivars indicated that the 42- and 44-chromosome lines are disomic substitutions (20+1A) and disomic additions (21+1A), respectively, of the same alien chromosome which carries genes for dark colored lemma and leaf rust resistance. The substitution and addition lines possess good field resistance to leaf rust. Plants of substitution lines were less vigorous, shorter, with poor straw and reduced fertility. Addition lines had reduced tillering ability, plant vigour, culm thickness, leaf width and fertility and were late maturing. Progeny tests for resistance indicated that substitution lines are more stable than addition lines. It is suggested that these lines may be of value in a program of radiation-induced gene transfer.  相似文献
