The aim of the present study was to investigate whether temporal changes in polyamine concentration and synthesis could be found in the luminal content and wall tissue of the rumen and abomasum, two organs which have entirely different growth patterns during the first month of life. In the abomasal mucosa there was a marked gradual decrease in the ornithine decarboxylase (ODC) activity during the first month of life, while the ODC activity in the ruminal mucosa was low during the whole experimental period. However, injury of the rumen wall was followed by increased ODC activity. The ODC activity in duodenal mucosa was about 10 times higher than in the ileal mucosa and the ruminal epithelium. In ruminal liquid a clear peak in ODC activity was observed during the period 51-70 days after birth. The polyamine concentration did not parallel the ODC activity, in either the ruminal epithelium or the ruminal liquid. Of the polyamines, the spermine concentration was always highest, and with the exception of duodenal mucosa, the putrescine concentration was lowest. In liver a clear decrease in spermidine concentration from day 1 to about day 60 after birth was observed. Otherwise no marked temporal changes in tissue polyamine concentrations were observed. Two and a half hours after oral administration of 14C-labelled spermine, nearly all of the radioactivity was found in the lumen of the gastrointestinal tract. On the other hand, 1 h after intravenous injection of polyamines the walls of the gastrointestinal tract were strongly labelled. In conclusion, the polyamines needed for ruminal epithelial development seem to come from sources other than the ruminal epithelium itself or the ruminal lumen.  相似文献

Ornithine decarboxylase (E.G. 4.1.1.17) and S-adenosylmethionine decarboxylase (E.G. 4.1.1.50) and their products putrescine, spermidine and spermine were estimated in the rumen liquid from 3 groups of growing kids and 23 adult goats. Polyamines were also estimated in the feedstuff used. Marked differences in polyamine synthesis in rumen liquid were observed between the different groups of kids. Two groups of kids growing up together with adult goats had at an age of 2–4 months a peak of a few days duration in enzyme activity as well as in polyamine concentration. In these groups ornithine decarboxylase activity reached maximal values of 158±79 s (n = 4) and 100 (66–117) (n = 3) nmol[14CO2]/ml rumen liquid/h at an age of 120 and 77 days, respectively. The corresponding activity in rumen liquid from kids who were isolated from other animals was only about 1/10 of this value. By comparison ornithine decarboxylase activity in adult goats was 30.7±20 (n = 43) nmol[14CO]/ml/h.In rumen liquid from kids grown up together with adults, concentrations of the polyamines reached maximum at about the same time as ornithine decarboxylase activity. The mean maximal concentration of putrescine in the 2 groups was about 350 and 500 nmol/ml, while the corresponding value for spermidine was about 200 nmol/ml in both groups.Relatively constant and high concentration of polyamines were present in the feedstuff used. However, in growing kids the ruminai putrescine and spermidine concentration at times far exceeded those that could be accounted for by the estimated intake of polyamines by the food. The results therefore strongly indicate that polyamines are formed in considerable amounts in rumen content of kids during the phase of rapid growth. Background: Administration of exogenous progesterone(P4) to ewes during the pre-implantation period advances conceptus development and implantation. This study determined effects of exogenous P4 on transport of select nutrients and pathways that enhance conceptus development. Pregnant ewes(n = 38) were treated with either 25 mg P4 in 1 mL corn oil(P4, n = 18) or 1 mL corn oil alone(CO, n = 20) from day 1.5 through day 8 of pregnancy and hysterectomized on either day 9 or day 12 of pregnancy. Endometrial expression of genes encoding enzymes for synthesis of polyamines,transporters of glucose, arginine, and glycine, as wel as progestamedins was determined by RT-qP CR.Results: On day 12 of pregnancy, conceptuses from P4-treated ewes had elongated while those from CO-treated ewes were spherical. The mR NA expression of AZIN2, an arginine decarboxylase, was lower in endometria of P4-treated than CO-treated ewes on day 9 of pregnancy. Expression of FGF10, a progestamedin, was greater in endometria of CO and P4-treated ewes on day 12 of gestation in addition to P4-treated ewes necropsied on day 9 of gestation. Treatment with P4 down-regulated endometrial expression of amino acid transporter SLC1 A4 on day 12 of pregnancy.Conclusions: Results indicated that administration of exogenous P4 during the pre-implantation period advanced the expression of FGF10, which may accelerate proliferation of trophectoderm cel s, but also was correlated with decreased expression of glycine and serine transporters and polyamine synthesis enzyme AZIN2. Further research with increased sample sizes may determine how differential expression affects endometrial functions and potentially embryonic loss.  相似文献

Arginine catabolism in lactating porcine mammary tissue   总被引:3,自引:0,他引:3  
In vivo studies have shown that the uptake of plasma arginine by the lactating porcine mammary gland greatly exceeds the output of arginine in milk, but little is known about the metabolic fate of arginine in this organ. The objective of this study was to quantify arginine catabolism via arginase and nitric oxide synthase pathways in the mammary tissue of sows on d 28 of lactation. Mammary tissue slices (approximately 60 mg) were incubated at 37 degrees C for 1 h in 2 mL of Krebs bicarbonate buffer containing 0.5 or 2 mM L-[U-14C]arginine, and arginine metabolites were measured using HPLC and radiochemical techniques. Rates of arginine utilization were similar to rates of urea production. Proline, ornithine, urea, glutamate, glutamine, CO2 and polyamines (putrescine + spermidine + spermine) were formed from arginine, accounting for 46, 31, 17, 2.3, 1.5, 0.22, and 0.30%, respectively, of the metabolized arginine carbons. Relatively small amounts of arginine were utilized for nitric oxide and citrulline synthesis, with citrulline accounting for 2% of the metabolized arginine carbons. Production of all arginine metabolites increased with increasing extracellular arginine concentrations from 0.5 to 2 mM, indicating a high capacity for arginine degradation. Consistent with the metabolic findings, the activities of arginases, ornithine aminotransferase, and pyrroline-5-carboxylate reductase were high, whereas those of pyrroline-5-carboxylate dehydrogenase, ornithine decarboxylase, and nitric oxide synthases were relatively low, and there was no proline oxidase, ornithine carbamoyltransferase or pyrroline-5-carboxylase synthase activity in the mammary tissue. Our results demonstrate for the first time that proline, ornithine, and urea were the major products of arginine catabolism via the arginase pathway in lactating porcine mammary tissue and provide a biochemical basis to explain a relative enrichment of proline but a relative deficiency of arginine in sow's milk.  相似文献

Interferon tau (IFNT), a type I IFN produced by the conceptus trophectoderm, is the signal for maternal pregnancy recognition in ruminants. The purpose of this study was to investigate whether IFNT effected on the proliferation of ovine trophectoderm cells in an autocrine manner. Elongated ovine conceptuses (Days 15, Day 0 = day of mating) were collected for isolation of mononuclear ovine trophectoderm (oTr‐1) cells, and conceptuses (Days 15 and 20, n = 4 and 3, respectively) were collected for RNA extraction. We demonstrated that the IFNT receptor, IFNAR1, was expressed in trophectoderm of day 15 and 20 conceptuses. Interestingly, the ovine trophectoderm cell line oTr‐1 cultured in the presence of recombinant bovine IFNT (rbIFNT) displayed increased expressions of IFN‐stimulated genes (ISGs), such as IFN‐stimulated gene 15 (ISG15), 2‐5‐oligoadenylate synthetase 1 (OAS1) and bone marrow stromal cell antigen 2 (BST2). Meanwhile, the presence of rbIFNT in the culture media could promote the cell proliferation in a dose‐dependent manner. Furthermore, the connective tissue growth factor, which has diverse functions in cell proliferation and is involved in conceptus elongation, was upregulated in oTr‐1 cell by rbIFNT treatment in vitro. These data indicated that IFNT could act as an autocrine factor to regulate trophectoderm cell proliferation.  相似文献

The aim of the study was to explore production of seven biogenic amines (phenylethylamine, histamine, cadaverine, tyramine, putrescine, spermine and/or spermidine) by selected staphylococci and enterococci. Thirty three enterococcal strains isolated from rabbit meat (Oryctolagus cuniculus f. domesticus) and 21 staphylococcal strains isolated from intestinal content of trout (Salmo trutta morpha fario) were tested. Production of biogenic amines was evaluated after cultivation of the tested microorganisms in the de Man, Rogosa and Sharpe Broth (enterococci) or in the Brain Heart Infusion Broth (staphylococci). Both the above cultivation media were enriched with selected amino acids (histidine, tyrosine, arginine, ornithine and lysine; 2g/L each) serving as precursors of biogenic amines. After cultivation, levels of the monitored biogenic amines in broths were analysed by a high performance liquid chromatography equipped with a UV/VIS DAD detector. Among 21 staphylococci, 18 strains produced tyramine or cadaverine, 13 strains formed putrescine or phenylethylamine and only one strain generated histamine. Two staphylococcal strains produced cadaverine levels above 1000mg/L. Among 33 enterococcal strains, 27 formed cadaverine, 18 strains produced tyramine, 10 strains generated phenylethylamine, and 2 strains gave putrescine. Most of the tyramine producing enterococci generated more than 1000mg/L of this biogenic amine. Production of spermine or spermidine by the studied strains was not proved.  相似文献

Maternal recognition of pregnancy refers to the requirement for the conceptus (embryo and its associated extra-embryonic membranes) to produce a hormone that acts on the uterus and/or corpus luteum (CL) to ensure maintenance of a functional CL for production of progesterone; the hormone required for pregnancy in most mammals. The pregnancy recognition signal in primates is chorionic gonadotrophin which acts directly on the CL via luteinizing hormone receptors to ensure maintenance of functional CL during pregnancy. In ruminants, interferon tau (IFNT) is the pregnancy recognition signal. IFNT is secreted during the peri-implantation period of pregnancy and acts on uterine epithelia to silence expression of estrogen receptor alpha and oxytocin receptor which abrogates the oxytocin-dependent release of luteolytic pulses of prostaglandin F2-alpha (PGF) by uterine epithelia; therefore, the CL continues to produce progesterone required for pregnancy. Pig conceptuses secrete interferon delta and interferon gamma during the peri-implantation period of pregnancy, but there is no evidence that they are involved in pregnancy recognition signaling. Rather, pig conceptuses secrete abundant amounts of estrogens between Days 11 to 15 of pregnancy required for maternal recognition of pregnancy. Estrogen, likely in concert with prolactin, prevents secretion of PGF into the uterine venous drainage (endocrine secretion), but maintains secretion of PGF into the uterine lumen (exocrine secretion) where it is metabolized to a form that is not luteolytic. Since PGF is sequestered within the uterine lumen and unavailable to induce luteolysis, functional CL are maintained for production of progesterone. In addition to effects of chorionic gonadotrophin, IFNT and estrogens to signal pregnancy recognition, these hormones act on uterine epithelia to enhance expression of genes critical for growth and development of the conceptus.  相似文献

Mechanisms of intrauterine migration were examined in 55 ewes. In the first experiment, corpora lutea were removed from unilaterally ovariectomized ewes on d 4 (d 0 = estrus) and pregnancy was maintained by giving exogenous progesterone. In Exp. 2, the reproductive tract was altered surgically such that embryos initially entered the uterine horn contralateral to the site of ovulation. In Exp. 3, ewes received beads of silastic polydimethylsiloxane that released either cholesterol or estradiol-17 beta in an attempt to mimic embryonic synthesis of estradiol. In the fourth experiment, unilaterally ovariectomized ewes were superovulated and spacing of embryos within the uterus was then examined. In all experiments, ewes were slaughtered on d 15 and recovery of embryos or beads from each uterine horn indicated that migration had occurred. All ewes in Exp. 1 and 2 that had two conceptuses experienced embryonic migration. Beads impregnated with estradiol migrated farther (P less than .01) than cholesterol-containing beads (27.6 +/- 4.3 vs 12.5 +/- 1.6 cm, respectively). In Exp. 4, only one conceptus had migrated into the contralateral horn in all ewes. These results demonstrated that 1) embryonic migration was not affected by local vs systemic exposure to progesterone, 2) embryos migrated into the unoccupied horn, regardless of the initial horn of entry, 3) estradiol may stimulate embryonic migration, and 4) conceptuses were not equally distributed between horns.  相似文献

采用盆栽试验方法研究了紫花苜蓿(品种:陇东) 叶片和根系中游离态腐胺(Put)、亚精胺(Spd)、精胺(Spm)及精氨酸脱羧酶(ADC)、鸟氨酸脱羧酶(ODC)、多胺氧化酶(PAO)活性的动态变化。结果表明,干旱胁迫下紫花苜蓿叶片和根系中的多胺含量增加,随着胁迫时间的延长, Put、Spd、Spm及ADC、ODC、PAO活性均呈先上升后下降的趋势,且随胁迫程度加重而增大。ADC、ODC活性与Put含量呈极显著正相关,参与了紫花苜蓿Put的合成代谢。PAO活性与Spd、Spm含量变化趋势相似且呈极显著相关关系,叶和根中多胺代谢显著相关,表明多胺代谢与紫花苜蓿抗旱性关系密切。  相似文献

OBJECTIVE: To determine the incidence, ultrasonographic characteristics, and risk factors associated with embryonic development characterized by formation of an embryonic vesicle without an embryo in mares. DESIGN: Prevalence survey. ANIMALS: 159 pregnant mares. PROCEDURES: From 1994 to 1998, mares between 11 and 40 days after ovulation with normal and abnormal embryonic development were examined ultrasonographically, and characteristics of each conceptus were recorded. RESULTS: The incidence of abnormal embryonic development in mares characterized by formation of an embryonic vesicle without an embryo was 7/159 (4.4%) during the 5 breeding seasons. Age and breed of mare or type of semen used did not differ for mares with normal and abnormal embryonic development. The percentage of mares in which the conceptus was undersized during > or = 1 examination was significantly higher for mares with abnormal conceptuses (5/7), compared with mares with normal conceptuses (2/147; 1.4%). The percentage of examinations during which the conceptus was undersized was significantly higher for abnormal conceptuses (12/27; 44.4%), compared with normal conceptuses (4/448; 0.9%). CONCLUSIONS AND CLINICAL RELEVANCE: To diagnose an embryonic vesicle without an embryo, mares should be examined by use of transrectal ultrasonography on day 25 after ovulation. When an embryo cannot be identified at that time, mares should be reexamined at intervals of 1 to 3 days until day 30. Because undersized conceptuses are more likely to be abnormal, development of undersized conceptuses should be monitored closely.  相似文献

Silages from five ripened varieties of silage maize with dry matter contents ranging between 275 and 410 g/kg were prepared in five laboratory experiments. Whole-plant maize was fermented at 22 degrees C and silages were then stored at the same temperature for 4 months. Spontaneously fermented silages were prepared as control variants and compared with silages inoculated with commercial strains of Lactobacillus plantarum, Lactobzcillus buchneri and a mixed preparation Microsil containing L. plantarum, Lactobacillus casei, Enterococcus faecium and Pediococcus pentosaceus. The starter cultures were applied at doses 5 x 10(5) and 5 x 10(6) CFU/g of chopped maize. Seven biogenic amines and polyamines were extracted from silages with perchloric acid and determined as N-benzamides by micellar electrokinetic capillary chromatography. Common chemical criteria of silage quality were also determined. All three inoculants, mainly at the higher dose, decreased significantly contents of tyramine, putrescine and cadaverine, three undesirable amines occurring at the highest levels. L. plantarum was the most effective. Contents of histamine and tryptamine were low in all experimental silages. Also relatively low were levels of polyamines spermidine and mainly of spermine.  相似文献

l-proline (Pro) is a precursor of ornithine, which is converted into polyamines via ornithine decarboxylase (ODC). Polyamines plays a key role in the proliferation of intestinal epithelial cells. The study investigated the effect of Pro on polyamine metabolism and cell proliferation on porcine enterocytes in vivo and in vitro. Twenty-four Huanjiang mini-pigs were randomly assigned into 1 of 3 groups and fed a basal diet that contained 0.77% alanine (Ala, iso-nitrogenous control), 1% Pro or 1% Pro + 0.0167% α-difluoromethylornithine (DFMO) from d 15 to 70 of gestation. The fetal body weight and number of fetuses per litter were determined, and the small and large intestines were obtained on d 70 ± 1.78 of gestation. The in vitro study was performed in intestinal porcine epithelial (IPEC-J2) cells cultured in Dulbecco''s modified Eagle medium-high glucose (DMEM-H) containing 0 μmol/L Pro, 400 μmol/L Pro, or 400 μmol/L Pro + 10 mmol/L DFMO for 4 d. The results showed that maternal dietary supplementation with 1% Pro increased fetal weight; the protein and DNA concentrations of the fetal small intestine; and mRNA levels for potassium voltage-gated channel, shaker-related subfamily, member 1 (Kv1.1) in the fetal small and large intestines (P < 0.05). Supplementing Pro to either gilts or IPEC-J2 cells increased ODC protein abundances and polyamine concentrations in the fetal intestines and IPEC-J2 cells (P < 0.05). In comparison with the Pro group, the combined administration of Pro and DFMO reduced the expression of ODC protein and spermine concentration in the fetal intestine, as well as the concentrations of putrescine, spermidine and spermine in IPEC-J2 cells (P < 0.05). Meanwhile, the percentage of cells in the S-phase and the mRNA levels of proto-oncogenes c-fos and c-myc were increased in response to Pro supplementation, whereas depletion of cellular polyamines with DFMO increased tumor protein p53 (p53) mRNA levels (P < 0.05). Taken together, dietary supplementation with Pro improved fetal pig growth and intestinal epithelial cell proliferation via enhancing polyamine synthesis.  相似文献

【目的】通过研究柑橘提取物对肥育猪后肠食糜蛋白质代谢产物的影响,旨在为柑橘提取物从源头调控臭气的产生提供依据。【方法】选取108头56日龄(19.73±0.39)kg杜×长×大猪,公母各半,随机分为3组,每组分6栏,每栏6头猪。对照组饲喂基础饲粮,抗生素组在基础饲粮中添加75 g/t金霉素,柑橘提取物组在基础饲粮中添加柑橘提取物(56~112日龄添加250 mL/t柑橘提取物,113~194日龄添加200 mL/t柑橘提取物)。194日龄时收集肥育猪盲肠和结肠食糜,测定食糜中游离氨基酸、酚和吲哚类物质、生物胺的含量。【结果】①在盲肠食糜中,与对照组相比,柑橘提取物组和抗生素组牛磺酸、天冬氨酸、丙氨酸、缬氨酸、亮氨酸、赖氨酸、组氨酸、精氨酸、吲哚和甲胺含量均显著降低(P<0.05)。与对照组相比,柑橘提取物有降低苏氨酸、丝氨酸、谷氨酸、甘氨酸、瓜氨酸、异亮氨酸、脯氨酸和粪臭素含量的趋势(0.05≤P<0.10),苯酚、腐胺、酪胺和精胺含量均显著降低(P<0.05)。与抗生素组相比,柑橘提取物组腐胺和酪胺含量均显著减少(P<0.05)。②在结肠食糜中,与对照组相比,柑橘提取物组和抗生素组天冬氨酸、丝氨酸、异亮氨酸、胱硫醚、亮氨酸、γ-氨基丁酸、总氨基酸、苯酚、对甲酚、粪臭素和甲胺含量均显著降低(P<0.05),脯氨酸和吲哚含量均有降低的趋势(0.05≤P<0.10)。柑橘提取物组丝氨酸、异亮氨酸、胱硫醚、亮氨酸、总氨基酸和对甲酚含量均显著低于抗生素组(P<0.05);与对照组相比,柑橘提取物组苏氨酸、谷氨酸、甘氨酸、鸟氨酸和酪胺含量均显著降低(P<0.05),亚精胺含量有降低的趋势(0.05≤P<0.10);与对照组和抗生素组相比,柑橘提取物组苯丙氨酸、色氨酸和赖氨酸含量均显著降低(P<0.05)。【结论】柑橘提取物降低了肥育猪后肠食糜蛋白质代谢产物的含量,可减少臭气的产生。  相似文献
