Summary The ability of the European biotypes A and B of Gremmeniella abietina var. abietina to hybridize in artificial pairings was tested. Using three types of molecular markers progenies with parental markers mixed indicating that the two biotypes can hybridize were observed. However, the resulting progeny had a low rate of successful germination, grew poorly on artificial medium and one was most probably a partial diploid or heterokaryon. Thus, both biotypes are genetically distinct populations and should be described as separate species as soon as sufficient discriminating information is available.  相似文献

Bemisia tabaci is a species complex, of which the B and Q biotypes are globally distributed. B. tabaci feeds on more than 600 plant species including cotton, which is one of the main crops in Pakistan. In this study, the biotypes/haplotypes of B. tabaci populations collected from cotton plants in Pakistan, China, and Egypt were identified, and their phylogenetic relationships were investigated. None of the populations from Pakistan or Egypt belonged to the B or Q biotype: all 16 samples from Pakistan belonged to haplotype “PCG-1” and it is a group of unresolved populations all from Pakistan, while all three populations from Egypt belonged to unresolved haplotype “ECG.” The three populations from China belonged to the B biotype. Phylogenetic analysis indicated that the PCG-1, ECG, and B biotypes clustered into different clades even though they share the same cotton plant species as their host. The association between the outbreaks of cotton leaf curl disease and the occurrence of the PCG haplotype of B. tabaci in Pakistan is discussed.  相似文献

The recent outbreak of Lophodermium needlecast in North America first appeared in 1966 in forest nurseries on red pine (Pinus resinosa) and Scotch pine (P. sylvestris). Dispersal of infected nursery stock throughout North America has created a serious needlecast problem in Scotch pine Christmas tree plantations. Two biotypes of L. pinastri are present. The pathogenic strain may be a recent introduction. The fungicides maneb and chlorothalonil are being used for control.  相似文献

The sweetpotato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a widely distributed and destructive agricultural pest on various host plants. The biology of two biotypes of B. tabaci: the invasive B and a new defined Cv biotype, on a range of host plants (hibiscus, laurel, poinsettia, collard, cucumber and tomato) were studied in the laboratory. Results revealed that the developmental periods of the B biotype immatures were not significantly different on the tested host plants except those between laurel and collard. The Cv biotype immatures developed significantly slower on cucumber and tomato than on the other plants. B. tabaci B biotype had the highest survivorship on collard (68.55%), and the lowest on laurel (33.24%), while the Cv biotype had the highest and lowest survivorships on laurel (61.63%) and tomato (36.74%). Host plants did not significantly affect the pre-ovipostion period regardless of biotype. The longest averaged longevity and highest fecundity of B biotype were both recorded on collard: 25.15 days and 143.0 eggs. The highest fecundity of Cv biotype was 196.49 eggs on laurel and its longest longevity was on hibiscus (19.62 days). The intrinsic rate of natural increase (r m) of B biotype on the three vegetables were all higher than those on the three ornamentals whereas the r m of Cv biotype on the three ornamentals were all higher that those on the three vegetables. Our research indicates that B. tabaci B and Cv biotypes have different host plant suitabilities. The three tested vegetables were more suitable for B biotype while the three tested ornamental plants were more suitable for Cv biotype. The potential mechanism for the different suitability of B and Cv biotypes on various host plants is also discussed.  相似文献

Since the myrtle rust pathogen (Austropuccinia psidii) was first reported (as Puccinia psidii) in Brazil on guava (Psidium guajava) in 1884, it has been found infecting diverse myrtaceous species. Because Apsidii has recently spread rapidly worldwide with an extensive host range, genetic and genotypic diversities were evaluated within and among Apsidii populations in its putative native range and other areas of myrtle rust emergence in the Americas and Hawaii. Microsatellite markers revealed several unique multilocus genotypes (MLGs), which grouped isolates into nine distinct genetic clusters [C1–C9 comprising C1: from diverse hosts from Costa Rica, Jamaica, Mexico, Puerto Rico, and USA‐Hawaii, and USA‐California; C2: from eucalypts (Eucalyptus spp.) in Brazil/Uruguay and rose apple (Syzygium jambos) in Brazil; C3: from eucalypts in Brazil; C4: from diverse hosts in USA‐Florida; C5: from Java plum (Syzygium cumini) in Brazil; C6: from guava and Brazilian guava (Psidium guineense) in Brazil; C7: from pitanga (Eugenia uniflora) in Brazil; C8: from allspice (Pimenta dioica) in Jamaica and sweet flower (Myrrhinium atropurpureum) in Uruguay; C9: from jabuticaba (Myrciaria cauliflora) in Brazil]. The C1 cluster, which included a single MLG infecting diverse host in many geographic regions, and the closely related C4 cluster are considered as a “Pandemic biotype,” associated with myrtle rust emergence in Central America, the Caribbean, USA‐Florida, USA‐Hawaii, Australia, China‐Hainan, New Caledonia, Indonesia and Colombia. Based on 19 bioclimatic variables and documented occurrences of Apsidii contrasted with reduced sets of specific genetic clusters (subnetworks, considered as biotypes), maximum entropy bioclimatic modelling was used to predict geographic locations with suitable climate for A. psidii which are at risk from invasion. The genetic diversity of Apsidii throughout the Americas and Hawaii demonstrates the importance of recognizing biotypes when assessing the invasive threats posed by Apsidii around the globe.  相似文献

The genetic diversity of Bemisia tabaci was investigated in Southern Italy using the mitochondrial cytochrome oxidase I (COI) gene as molecular marker and sampling whiteflies on cultivated plants, weeds and bushes. Phylogenetic analysis of COI sequences and restriction analysis of COI amplicons were used to genotype whitefly populations. A PCR-RFLP method based on digestion with the endonuclease ApoI was set up to identify the B. tabaci genetic variants so far recorded from the Mediterranean region. In general, biotype Q populations were most frequently collected (76.0% of all sampled populations). In greenhouse districts, 87.0% sampled populations were identified as biotype Q, and 13.0% were assigned to biotype B. Outside the greenhouse districts, the biotype B was never collected, whilst biotype Q populations were found on weeds and on plants cultivated in family gardens in different environments, also located in interior plains or in mountain areas distant from intensive cultivations of whitefly host plants. A new genetic variant unrelated to B and Q biotypes, which was named Ru, was collected on Rubus ulmifolius and grapevine. Phylogenetic analysis of COI sequences shown that Ru haplotypes form a well-supported clade sister to the clade including the Asian/Australian major genetic groups and the Italy major genetic group. The closest relative of the Ru clade (10.7% pairwise genetic distance) was the Italy group, with this latter so far including only the haplotypes of the T biotype. These results were discussed in the light of the recent B. tabaci species concept.  相似文献

Despite a large number of investigations on the molecular genetics and population structure of the whitefly Bemisia tabaci (Gennadius) complex, no such study had been conducted in Iran. The genetic variation of B. tabaci was examined using polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) for 18 field collections from cucumber, eggplant, and tomato in four provinces of Iran. PCR amplification and restriction digestion with two enzymes detected 388 RFLP fragments, of which 16 fragments showed polymorphisms. Cluster analysis of these data placed all B. tabaci individuals within a single group, and there was no evidence for between- or within-population genetic variation. Phylogenetic (Clustal W) analysis of 42 B. tabaci mtCOI sequences (n = 21 field collections) from Iran, and a comparison with well-studied haplotype or biotype reference sequences available in public sequence databases, revealed that the Iranian B. tabaci populations were most closely related to the B biotype at 0–1.2% nucleotide identity. The B biotype is a well-known member of a sister clade from the Middle East–North African region of the world, owing to its nearly worldwide distribution and invasive characteristics. This report indicates that a single major haplotype of B biotype is prevalent in Iran and that its closest relative is the B biotype. Also, given the extent of known variation in the Middle East and African continent, data indicate somewhat surprisingly that the B. tabaci collections sampled in Iran had limited genetic variation and population substructure. Knowledge that the B biotype of B. tabaci predominates in Iran is important for designing effective pest management strategies given that biotypes of B. tabaci are known to differ greatly with respect to insecticide resistance, host range, virus–vector interactions, and other key biological characteristics.  相似文献

Hairy fleabane is a weed found increasingly in agronomic crops and rural areas. Although glyphosate resistance is now widespread in this species, the ecological fitness of glyphosate-resistant (GR) and glyphosate-susceptible (GS) populations has not been compared. When seeds of both biotypes were subjected to different alternating temperature, germination occurring and seedling vigor did not differ between the biotypes with maximum germination at 10/20 and 15/25 °C. Under noncompetitive conditions, growth and seed production of the GR and GS biotypes were similar. The competitive ability of the two biotypes, as determined by a replacement series experiment, was also similar. Initially, the relative crowding coefficient (RCC) between the biotypes was close to one. However, the RCC values for each characteristic increased with time indicating that the GR had a small competitive advantage as plants matured. Overall, there was no apparent fitness penalty associated to glyphosate resistance. Therefore, the GR populations are likely to persist unless effective management strategies are adopted.  相似文献

Poplar rusts due to Melampsora larici‐populina (Mlp), M. allii‐populina (Map) and M. medusae f. sp. deltoidae (Mmd) are the most serious disease in Europe on cultivated poplars, that is, Populus × euramericana and P. × interamericana hybrids. These pathogenic species can be identified by the observation of morphological characteristics of urediniospores but this method is not appropriate for high‐throughput analysis and cannot be used on other spore stages, such as aeciospores or teliospores, that are morphologically similar. The aim of this study was to develop a rapid and sensitive molecular method based on PCR amplification that was able to specifically detect these species on various hosts for routine analysis. Three primer pairs ITS‐MLP‐F/ITS‐MLP‐R, ITS‐MAP‐F/ITS‐MAP‐R and ITS‐MMD‐F/ITS‐MMD‐R were designed within the internal transcribed spacer (ITS) sequences of ribosomal DNA to target Mlp, Map and Mmd, respectively, and their specificity were confirmed on a wide range of isolates and species. ITS‐MLP‐F/ITS‐MLP‐R and ITS‐MAP‐F/ITS‐MAP‐R primers proved to be highly specific to Mlp and Map, respectively, whereas ITS‐MMD‐F/ITS‐MMD‐R cross‐reacted with DNA from M. larici‐tremulae and M. pinitorqua. However, these species are not pathogenic on cultivated poplars that all belong to sections Aigeiros and Tacamahaca of the genus Populus. Specific Mmd primers proved to be very sensitive as a positive signal could be obtained with DNA extracts from 6 target urediniospores mixed with 800 000 urediniospores of Mlp. An internal amplification control (IAC) was included to discriminate false negative results due to the potential presence of inhibitory compounds in DNA extracts. ITS‐MMD‐F/ITS‐MMD‐R primers are therefore efficient for the detection of the quarantine pathogen Mmd on samples collected on poplar or larch and are fit for use in official tests. This new PCR assay has been used in routine for ten years, and Mmd has hitherto never been detected in commercial poplar nurseries in France.  相似文献

Using data collected by two different observation networks, it was possible to clarify the distribution of Collybia fusipes in France. It is frequently found in the western and northern parts of France, mainly on Quercus rubra and Quercus robur, and to a somewhat lesser extent on Quercus petraea. When the relative areas covered by these species are compared, it is evident that Q. rubra is much the most susceptible of the three species. The fungus has been less frequently reported in the south‐eastern quarter of France, where Quercus ilex and Quercus pubescens are the dominant species. Complementary observations carried out at the National Arboretum of Les Barres indicate that a large number of oak species are susceptible to infection by this fungus, particularly those in the subgenus Erythrobalanus. These observations are discussed with reference to reports of this fungus throughout Europe. In a Q. rubra stand, fruit‐body clumps were monitored and root damage was assessed on 73 final‐crop trees and 40 codominant trees. The results suggest that C. fusipes does not preferentially infect weakened trees. Both the severity of root damage and the number of fruit‐body clumps increased with increasing depth to signs of waterlogging.  相似文献

Armillaria causes problems of root rot, kill trees and decay wood in the forests of Serbia and Montenegro, but the species involved have not hitherto been identified. The aim of this study was to identify field isolates collected on 25 localities. Identification was based on restriction fragment length polymorphism (RFLP) analysis of intergenic spacer 1 (IGS1) region and comparisons of IGS1 sequence with those available on NCBI database. Phylogenetic analysis was performed on sequence information from selected isolates to determine possible interrelationships between isolates with different banding patterns and previously identified tester isolates of five European Armillaria species. Five Armillaria species were identified in 90 isolates obtained from forests in Serbia and Montenegro. Armillaria gallica was most frequently isolated, followed by A. cepistipes, A. mellea, A. ostoyae and A. tabescens; two isolates remained unidentified. Restriction digestion of IGS1 amplification products with AluI produced 10 RFLP patterns. Patterns G4 (400, 250, 180) for A. gallica and pattern X (400, 180, 140) for isolates 74 and 79 are reported for the first time in European isolates. Eight RFLP patterns were observed after restriction with TaqI. Two patterns each were observed for A. ostoyae and A. gallica, and one each for A. cepistipes, A. mellea, A. tabescens and isolates 74 and 79. Parsimony analyses based on the IGS1 region placed the isolates into four clades: one including A. mellea, the second containing A. gallica–A. cepistipes isolates, while isolates of A. ostoyae and A. borealis were in the third clade. Armillaria tabescens differed from all annulate species. Phylogenetic analysis supported the conclusion that European Armillaria species are closely related and separated from a common ancestor in the near past. According to this survey five European Armillaria species are present in the forests of Serbia and Montenegro, while A. borealis is not present in the studied ecosystems.  相似文献

 The mechanical performance of pine sapwood (pinus sylvestris), impregnated with linseed oil to different take-up levels, is evaluated using several test methods. SEM is used to study morphological changes following the impregnation procedure. The reduction of mechanical properties is attributed to a) localized cell wall damage in the ray region that facilitates longitudinal inter-cell split in L-R plane (macrocrack) initiation and propagation; b) submicroscopical cracking in the S1 sublayer that reduces the resistance to Mode I and Mode II inter-cell splitting at any location where the oil front has passed. Mechanical testing shows the following effect of the impregnation on failure a) the Mode I fracture toughness G Ic in L-T and L-R planes, determined in DCB test, is significantly lowered with no significant difference in fracture resistance reduction in between planes; b) 3-point flexural test for specimen geometry leading to cracking in R-L and T-L planes show that the flexural strength as well as flexural modulus are reduced due to impregnation; c) 3-point flexural tests on longitudinal specimens used to determine the impregnation effect on longitudinal modulus E L and shear moduli G LT and G LR , reveal only minor changes. Fracture surfaces in mechanical tests are analyzed using SEM, and differences are explained by described microdamage mechanisms. Received 10 August 1999  相似文献

Metarhizium anisopliae sensu stricto (ss) (Metsch.) Sorok. isolate ICIPE 07 is being developed as biopesticide for the control of ticks. In addition, leaf extracts of Calpurnia aurea Benth, and the attraction aggregation and attachment pheromone (AAAP) are being used as ticks’ attractant. The three agents are being considered for use in combination in an autodissemination approach, whereby ticks that are attracted to semiochemicals are infected with the inoculum. Experiments were therefore conducted to evaluate in vitro the compatibility between C. aurea, AAAP, and the M. anisopliae on vegetative growth, conidial production, and spore viability. Calpurnia aurea leaf extract was compatible with the fungus at all the concentrations tested, whereas AAAP inhibited all the fungal growth parameters. The virulence of M. anisopliae formulated in emulsifiable extracts of C. aurea was also tested against different developmental stages of Rhipicephalus appendiculatus in laboratory bioassays. No significant differences in virulence were observed between M. anisopliae applied alone and M. anisopliae formulated in different concentrations of C. aurea leaf extracts. These results suggest that C. aurea leaf extracts is compatible with M. anisopliae and could be mixed together for “spot-spray” treatments as low-cost and environmental-friendly technology to control ticks in grazing field, while AAAP should be used separately.  相似文献

Ganoderma root rot is the most serious disease affecting commercially planted Acacia mangium in plantations in Indonesia. Numerous Ganoderma spp. have been recorded from diseased trees of this species and to a lesser extent Eucalyptus, causing confusion regarding the primary cause of the disease. In this study, a large collection of Ganoderma isolates were obtained from the roots of A. mangium showing early signs of root rot in disease centres in South Sumatra plantations. Isolates were also collected from Eucalyptus roots at Lake Toba in North Sumatra showing similar symptoms as well as from sporocarps connected to these samples. Phylogenetic analyses showed that a single Ganoderma sp., identified as G. philippii, is the major causal agent of Ganoderma root rot on A. mangium. Results from this study also showed that the isolates obtained for Eucalyptus trees in North Sumatra belong to G. philippii. Isolates from roots and connected fruiting bodies together with the morphology of the fruiting structures confirmed this identification. Symptoms associated with this pathogen are obvious and it should not be confused with other diseases. Other Ganoderma spp. found in disease centres are considered to be of minor importance and management strategies for root rot should be focused on G. philippii.  相似文献

Phytophthora is considered as an important pathogen on walnut, and severe losses are reported in European as well as in American walnut stands. Though several Phytophthora spp. are known to attack walnut, P. cinnamomi is considered the most virulent and widespread in southern Europe. Up to now, no walnut species or hybrid is known to have a high resistance level towards P. cinnamomi. Efforts are addressed in finding rootstock material graft compatible with English walnut and resistant/tolerant to P. cinnamomi. The extension of P. cinnamomi lesions on five Juglans species was studied to find out sources of resistance/tolerance to this pathogen. Walnut species clustered into two main groups, J. hindsii, J. nigra, and J. mandshurica were the less susceptible to the colonization of P. cinnamomi, while J. regia and J. sieboldiana were the most susceptible. On this account, J. mandshurica represents the best alternative as rootstock because its employment overcomes the risk of the occurrence of black line disease, it has good level of resistance to Agrobacterium temefaciens and Brenneria nigrifluens, and it is tolerant to Xanthomonas arboricola pv. juglandis. J. mandshurica is also compatible in cross‐pollinations with J. regia and J. nigra. Differences in virulence of P. cinnamomi isolates was assessed and a marked interaction between species and isolate emerged. Treatment with fosetyl‐Al by dipping was mainly efficient in reducing the length P. cinnamomi lesions, and an interaction between species and treatment was evident with the highest efficacy on J. regia and J. sieboldiana.  相似文献

Brazilian forest‐based industries are supported by more than 5.5 million hectares of Eucalyptus growing under different climatic conditions with different degrees of favourability for rust Puccinia psidii, including both traditional and expanding areas, where such disease is of major concern for the industry, foresters and scientists. The main objective of this study was to define favourable climatic zones for Eucalyptus rust in Brazil with the following aims: (i) to develop a spatial method for estimating the mean night‐time temperature (Tng); (ii) to assess and validate a Eucalyptus rust model; and (iii) to map Eucalyptus rust favourability zones in Brazil based on the proposed model. A straightforward method, based only on latitude, day of the year, maximum and minimum air temperatures, was developed to estimate Tng, which is a key variable for a proper application of Ruiz rust model. Based on 37 field experiments with natural rust occurrence, it was observed that climatic conditions are determining factors for disease severity. Significant correlations between disease severity observed in the field and climatic conditions were found: a negative correlation with temperature (r = .50, < .01) and a positive correlation with relative humidity (r = .89, < .0001). A significant correlation (r = .81, < .0001) between the normalized infection index, produced from Ruiz model, and the average rust score was also observed. Once the model was validated under field conditions, it was applied using historical average data of air temperature and leaf wetness duration to obtain monthly Eucalyptus rust favourability maps for the whole country. These final maps show that favourable climatic zones for Eucalyptus rust are extremely dynamic, with high temporal and spatial variability in Brazil and that climatic conditions should be considered for expansion of Eucalyptus to new areas, in breeding programmes, and for defining the most suitable seasons for forest establishment in each climate of the country. These results provide forestry managers with practical tools to reduce uncertainty about the expected severity of Eucalyptus rust in Brazil.  相似文献

Species in the Botryosphaeriaceae are important canker pathogens of woody plants, including Eucalyptus spp. The recent discovery of the Eucalyptus pathogen, Chrysoporthe austroafricana, on ornamental Tibouchina trees raised the question as to whether Tibouchina spp. might be alternative hosts for other Eucalyptus pathogens. Therefore, the aim of this study was to consider whether species of the Botryosphaeriaceae occurring on Eucalyptus spp. might also occur on ornamental Tibouchina spp. Isolations were made from Tibouchina trees in South Africa, New Zealand and Australia. Isolates were identified using morphological characteristics and DNA‐based techniques. Neofusicoccum parvum and N. mangiferae were identified from the samples. Pathogenicity trials on T. urvilleana showed that both species are pathogenic to this host.  相似文献

In this study a Sphaeropsis sp. is recorded as causing cankers on cypress in South Africa. These cankers are either found alone or on trees which are infected with the well-known cypress canker pathogen, Seiridium unicorne. When the two fungi occurred together, lesions could not be distinguished from each other, although the greater number of lesions were caused by the Sphaeropsis sp. The fungus appears to be identical to that described as Sphaeropsis sapinea f.sp. cupressi, in Israel, which is a fungus very different from the pine pathogen, Sphaeropsis sapinea. Pathogenicity tests showed that the Sphaeropsis sp. is significantly more pathogenic to Cupressus lusitanica than to Pinus roxburgii and Pinus elliottii. S. sapinea was pathogenic only to the Pinus spp. tested, and not to C. lusitanica. In contrast, the cypress pathogen Seiridium unicorne, was pathogenic to C. lusitanica and also to the two Pinus spp. tested.  相似文献

Data from a nationwide set of Pinus radiata D. Don plots established at a range of conventional stand densities were analysed at age 6 to (i) determine how environment and competition from weeds influence dynamic modulus of elasticity (E) of the lower stem base, (ii) develop a predictive multiple regression model of E for basal stemwood and (iii) identify significant direct and indirect environmental influences (through stem slenderness) on E using path analysis.Site had a highly significant (P < 0.001) influence on E, which exhibited a 3-fold range from 1.6 to 5.3 GPa, across 30 sites. When compared to the weed-free controls, weed competition had a significant (P < 0.0001) and substantial effect on E, increasing values by on average 16% (2.76 GPa vs. 2.38 GPa).The positive linear relationship between stem slenderness (determined as tree height/ground-line tree diameter) and E was by far the strongest relationship (R2 = 0.71; P < 0.001) among the 20 variables that were significantly related to E. A multiple regression model that included stem slenderness, mean minimum air temperature in mid-autumn, Tmin, as positive linear relationships and net nitrogen (N) mineralisation in a negative linear form accounted for 86% of the variance in E. A cross-validation indicated that this model was stable and unbiased, with the validation accounting for 82% of the variance in E. The final path analysis model included Tmin, net N mineralisation, below canopy solar radiation and stem slenderness as significant (P < 0.05) direct influences on E. Below canopy radiation, maximum air temperature during mid-summer, soil total phosphorus and carbon:nitrogen ratio were indirectly associated with E through their significant (P < 0.05) direct relationship with stem slenderness.These results provide considerable insight into how environment regulates E of juvenile P. radiata. Low fertility sites that have warm air temperatures and either a high canopy leaf area index, or high levels of woody weed competition, are most likely to produce trees with high stem slenderness and high E. Conversely, sites that are cool over summer and autumn and high in fertility, with low levels of intra- or inter-specific competition for light are likely to produce trees with low stem slenderness and low E.  相似文献
