Summary Fusarium solani (Mart.) Sacc. f. sp.eumartii (Carp.) Snyder & Hansen is the causative agent of two important types of disease in potato (Solanum tuberosum) plants: wilt in stems and leaves and dry rot in tubers. The aim of this work was to study the molecular response of the potato to fungal attack. Micropropagated plantlets were tested, using a biological assay to detect antifungal proteins. An inhibitory activity of spore germination was detected in protein fractions from infected plantlets 7 days after inoculation with the pathogen. In addition, β-1,3-glucanase activity (glucanase III) was measured in protein fractions with antifungal activity. Induction of hydrolytic enzymes and antifungal activity in infected plantlets ofSolanum tuberosum subp.andigena suggests that it could be a suitable source of resistance genes.  相似文献

A survey of selected potato soils in Maine was made to determine the species and populations ofFusarium spp. present.Fusarium solani ‘Coeruleum’ was most often isolated butFusarium roseum ‘Sambucinum’ was not recovered from any soil. Crops or cropping sequences in general showed no direct relationship to observed Fusaria populations or tuber dry rot. Field studies were conducted to determine the effect ofFusarium contaminated seed on soil and daughter tuber contamination. Whole and cut seed were inoculated withFusarium roseum ‘Sambucinum’ and half of each lot treated with thiabendazole (1500 ppm) to controlFusarium seed decay. Soil populations ofF. roseum ‘Sambucinum’ were higher near plants produced from non-treated, cut seed tubers. No increase inFusarium populations was found between plants or near plants produced from whole or thiabendazole treated seed. Daughter tuber contamination was greatest from plants produced from non-treated contaminated cut seed, and lowest from whole or treated seed. Whole non-treated seed produced daughter tubers with contamination equal to treated whole seed. This indicates that the use of whole seed could be a method to reduce daughter tuber contamination and reduce reliance on chemical treatments. Cut seed contaminated withFusarium spp. is an important source of daughter tuber contamination which can result in a high amount of tuber dry rot in storage.  相似文献

The effects of the dormancy releasing chemicals, Rindite and bromoethane, on subsequent tuber response to dry rot were evaluated. Seed tubers of the cultivars Sebago and Acadia Russet were treated with Rindite or bromoethane, and subsequently inoculated with sterile distilled water or spore suspensions ofFusarium sambucinum Fuckel f. 6 Wollenw. After storage for 1 or 11/2 months, tubers were rated for dry rot and dormancy release. Development of seed tuber breakdown byFusarium was not significantly affected by pretreatment with Rindite or bromoethane regardless of any effect on tuber dormancy.  相似文献

The St. Johns potato variety is high-yielding and late-maturing with attractive, round to oblong, white-skinned, white-fleshed tubers with mediumshallow eyes. Its major use is expected to be as a maincrop tablestock variety. Taste panels rated St. Johns better than or equal to Katahdin in texture, color, mealiness, and flavor. St. Johns tubers do not show the net necrosis caused by potato leafroll virus, and are resistant to golden nematode, corky ringspot, hollow heart, and blackspot bruising. St. Johns is also moderately resistant to greening, shatter bruise, verticillium wilt, early blight, common scab, the common race of late blight, leafroll,Fusarium sambucinum (dry rot) andErwinia carotovora (soft rot), although some breakdown has been reported in commercial storages. Symptoms of leafroll virus infection are somewhat difficult to detect.  相似文献

Dry rot is a serious potato disease and causes significant losses in China. Research efforts on potato dry rot have been very limited as well as the attempts to characterize the pathogen in the major potato production regions of China. A total of 260 Fusarium isolates were identified in 698 potato tubers collected in six important potato production regions of northern China, out of which five different Fusarium species, Fusarium sambucinum, F. avenaceum, F. oxysporum, F. equiseti, and F. acuminatum, were isolated. The identification of each species was confirmed by sequencing analysis of ∼700 bp DNA fragment derived from the translation elongation factor-1 alpha gene. F. sambucinum was found to be the predominant species accounting for 56% of the isolates. Different pathogenicity was found to be associated with five most common Fusarium species. Sixty-seven clones used in China were identified as susceptible to F. sambucinum, indicating little scope for developing resistant cultivars using the currently available potato germplasm.  相似文献

Potato virus X (PVX)-free and mild PVX-infected tuber lines originating from the same Katahdin clone were compared for susceptibility to tuber rot incited byFusarium roseum (Lk.) Synd. and Hans. ‘Avenaceum’. The effect of interval between top-kill and harvest on development ofFusarium tuber rot was also determined. A series of tubers from both PVX-free and PVX-infected tuber lines was either (i) mechanically bruised and inoculated by immersion in a spore suspension ofF. roseum ‘Avenaceum’, (ii) mechanically bruised but not inoculated, (iii) inoculated only or (iv) neither bruised nor inoculated. Tubers from all treatments were then stored at temperatures of 18–21 C (65–70 F) for a 3-week period prior to being placed at 4 C (39 F) for the remainder of the storage period. The amount of rot was determined after a 5–6 month storage period at the latter temperature. In a 4-year study PVX-free tubers were significantly (1% level) more susceptible to infection byF. roseum ‘Avenaceum’ than tubers infected with mild PVX. PVX-free tubers left in the ground for 2 or 3 weeks after top-kill were significantly (1% level) more susceptible toFusarium dry rot than those infected with the virus; however, there was no significant difference in disease incidence between PVX-free and PVX-infected tubers when harvested 4–5 weeks after top-kill. Tubers, both PVX-free and PVX-infected, remaining in the ground for a 2 week period following top-kill were significantly (1% level) more susceptible toFusarium tuber rot than those harvested 3 weeks after top-kill; similarly, both PVX-free and PVX-infected tubers harvested 3 weeks after top-kill were significantly (1% level) more susceptible than those left in the ground for a 4–5 week interval.  相似文献

Fusarium sambucinum is one of the most important causal agents that not only cause the dry rot disease of potato tubers in fields and stores worldwide but also capable of producing secondary metabolites toxic for people and animals. Here we present the first draft genome sequence of the strain (F-4) estimated to be around appx. 42.0 Mb. The genome has 12,845 protein coding genes with more than 35,900 exons and gene density of 3.13 per 10Kb. F. sambucinum is evolutionary more close to the F. graminearum among the Fusarium species complex. The genome sequence represents a valuable resource for understanding the pathogenecity and virulence factors, and their evolution within the complex and highly plastic genus Fusarium.  相似文献

The Quaggy Joe potato variety is high-yielding and medium to medium-late maturing with attractive, round-oval, white-skinned, white-fleshed tubers with medium-shallow pink eyes. Its major use is expected to be as a maincrop tablestock variety. Quaggy Joe is resistant to the potato leafroll virus, and is also moderately resistant to early blight (Alternaria solani), dry rot (Fusarium sambucinum), and soft rot (Erwinia carotovora subsp.carotovora). Quaggy Joe tubers have shown very little shatter or blackspot bruising, but sprout earlier than Atlantic. Young plants have shown sensitivity to the herbicide metribuzin.  相似文献

The Mainestay potato variety is high-yielding and late-maturing producing attractive, round, white-skinned, white-fleshed tubers with medium-shallow eyes. Its major use is expected to be as a maincrop tablestock variety. Mainestay appearance ratings are similar to those of Katahdin, and greening in Mainestay tubers is less than in Kennebec. Most boil and bake qualities were rated equal to Katahdin in at least half of the tests, and Mainestay usually has a low incidence of internal defects. However, purple streak, heat necrosis, vascular discoloration, and heat sprouts have been seen on occasion. Mainestay tubers do not show the net necrosis caused by potato leafroll virus, and have shown very little hollow heart or blackspot bruising. Mainestay is susceptible to common scab and acid scab, but moderately resistant to verticillium wilt (Verticillium, albo-atrum andV. dahliae), early blight (Alternaria solani), rhizoctonia (Rhizoctonia solani), dry rot (Fusarium sambucinum), and pink rot (Phytophthora erythroseptica). It is moderately susceptible to the common race of late blight (Phytophthora infestans). Mainestay tubers are slow to mature after vine kill, and additional time is required prior to harvest in order to allow for proper periderm development and to reduce storage losses.  相似文献

Arbuscular mycorrhizae (AM) have been shown to increase plant resistance to root-rotting pathogens. This study was conducted to determine whether a commercial peat-based medium containing the AM fungusGlomus intraradix (Schenck & Smith) could influence postharvest progression of tuber dry rot in prenuclear minitubers of potato (Solanum tuberosum). Minitubers grown in this medium had significantly less tuber dry rot (20–90% reduction) when later inoculated with the dry rot fungusFusarium sambucinum relative to minitubers grown in an identical peat-based medium without the AM fungus. This disease suppression was also demonstrated in a high-input commercial greenhouse, and occurred despite only trace levels of AM colonization of the parent plants, and with no evidence of enhanced plant phosphorus nutrition or differences in minituber mineral content. These results suggest that the AM fungal inoculum has potential for use in suppression of tuber dry rot of potato.  相似文献

Fusarium semitectum is one of the important causal agents of dry rot of potato tubers in the world. In order to determine genetic variability among 41 isolates of F. semitectum, morphological and molecular studies were carried out. All F. semitectum isolates were recovered from infected potato tubers with dry rot symptoms collected from four provinces in Iran. According to macroscopic and microscopic characteristics, 41 isolates of F. semitectum were classified in two morphotypes (morphotypes I and II). All 41 isolates were evaluated for their pathogenicity on healthy potato tubers. Tuber rot symptoms were observed on the 21st day after inoculation of Fusarium isolates on the tubers tested. The measurement was done by comparing the depth and width of lesion expansion among the isolates. Molecular characterization through PCR-IGS-RFLP analysis by six restriction enzymes (AluI, BsuRI, Eco88I, MspI, TaqI and PstI) divided the 41 isolates of F. semitectum into two separated clusters that were in accordance with the morphological characterization.  相似文献

Lack of effective chemicals impedes control of Fusarium dry rot of stored potato tubers destined for processed and table stock use. Biological control of dry rot incited byGibberella pulicaris (anamorph=Fusarium sambucinum) has been demonstrated in laboratory studies but not in commercial storage environments. Several Gram-negative bacterial strains that were efficacious and amenable to production in liquid culture in laboratory studies were selected for pilot studies in Idaho and for bin trials at commercial storage houses in Idaho and North Dakota. In the first year of pilot studies,Pseudomonas fluorescens S22:T:04 (≈1 x 108 cfu/ml) decreased dry rot caused byG. pulicaris by 19% when coinoculated with the pathogen compared to controls and to the fungicide thiabendazole (TBZ) applied at label rates. In second year pilot studies,P. fluorescens P22:Y:05 andEnterobacter cloacae S11:T:07 (≈4 x 108 cfu/ml) reduced the severity of disease incited byG. pulicaris (25% and 17% average disease decrease, respectively) when antagonists were applied after pathogen inoculum. Treatment effects on dry rot that developed from inoculation withNectria haematococca (anamorph=Fusariumsolani var.coeruleum) were variable and influenced by interactions between antagonists, a wetting agent, and TBZ. In commercial storage bin trials,E. cloacae S11:T:07 reduced naturally occurring levels of dry rot by an average of 21% compared to 14% for TBZ, demonstrating that this antagonist was effective when produced using a liquid culture medium and methodology that approximates commercial practices for producing biomass.  相似文献

Laboratory and field studies with the Russet Burbank (RB) potato provide evidence for synergism betweenErwinia carotovora var. atroseptica (Ea) andFusarium roseunt ‘Sambucinum’ (Fs). When these pathogens were inoculated together, the severity of tuber rot was significantly greater than when either pathogen was inoculated separately. Similarly, these pathogens interacted to reduce yield. When both organisms were uniformly applied to puncture wounds on potato seed (inoculum suspension consisted of 108 cells/ml Ea and 105 cells/ml Fs), the total yield was reduced by 46% and U.S. #1 yield by 53%. These reductions occurred even though blackleg symptoms (caused by Ea) were negligible (< 1%). In contrast, inoculations withErwinia carotovora var.carotovora (Ec) and Fs did not interact to reduce potato yield. Potato yields were also not influenced when these pathogens (Ea, Ec, Fs) were separately inoculated.Verticillium wilt (Verticillium dahliae) was significantly less when tubers were inoculated with either Ea, Ec, Fs, Ea + Fs, or Ec + Fs than with uninoculated tubers.  相似文献

Frontier Russet, a new, medium-early potato variety, was jointly released to growers by the U.S. Department of Agriculture and the Agricultural Experiment Stations of Idaho, Oregon, Washington, and Colorado on April 9, 1990. It was tested in irrigated yield trials since 1979. Frontier Russet produces a moderately high yield of high quality, oblong to long, cylindrical, russet-skinned tubers. The tubers are well suited for baking and for processing into french fries at harvest. Frontier Russet is resistant to Fusarium dry rot and more resistant to Verticillium wilt than other early russets.  相似文献

Progeny tubers from seed potatoes originating from either a traditional clonal selection method or the more modernin vitro tissue culture method of propagation were tested for storage rot response following inoculation of three tuber sites with twoFusarium species. Significant differences were found among disease responses for the twoFusarium species and for the four cultivars tested. Disease symptoms were less severe forF. solani var.coeruleum than forF. sambucinum. Fusarium sambucinum caused less disease in Superior than Kennebec, Russet Burbank and Sebago. Seed propagation method did not significantly affect disease response except for one case;in vitro culture of Kennebec had less disease due toF. sambucinum at the tuber side inoculation site than clonal selection. Kennebec and Superior inoculatedF. sambucinum had significant differences with respect to the number of years of field propagation for the tuber side inoculation site and for the tuber mean rot index. Significant differences in number of years of field propagation were also found for eye-ends of Superior tubers and stem ends of Russet Burbank tubers inoculated withF. solani var.coeruleum.  相似文献

Summary The production of extensive soft rot in potato tubers which were wounded, inoculated withErwinia carotovora var.atroseptica and stored inc. 100% RH at 20° was greatly increased by replacing air in containers with N2. Accumulation of CO2 due to tuber respiration did not significantly affect the production of rots in these conditions. When tubers were inoculated with sterile water in place of theErwinia and held under anaerobic conditions, spreading soft rots were also produced andClostridia, but not soft rotErwinia, were isolated from the rotted tissue. The potato cultivar Defender is high-yielding, white-skinned, and notable for having foliar and tuber resistance to late blight infection caused byPhytophthora infestans (Mont.) de Bary. It was released in 2004 by the USDA-ARS and the agricultural experiment stations of Idaho, Oregon, and Washington. Defender is suitable for processing into french fries and other frozen potato products directly from the field or from storage. Defender also may be used for fresh markets in regions such as California, where cultivars with long tubers and white skin are traditionally grown. Resistances to late blight and other potato diseases make Defender an ideal candidate for organic potato production. Defender consistently produced greater total and U.S. No. 1 yields than ‘Russet Burbank’ in Idaho trials. In early harvest trials conducted in the western U.S., average total yields of Defender were 17% and 23% greater than yields for ‘Ranger Russet’ and ‘Shepody’, respectively. In full-season trials conducted in the western U.S., Defender averaged 10% and 15% higher yields than Ranger Russet and Russet Burbank, respectively. Specific gravity of Defender is consistently high, with values comparable to those of Ranger Russet; tuber ascorbic acid (Vitamin C) levels are also high. In addition to late blight, Defender also is resistant to tuber early blight (Alternaria solani, (Ellis & G. Martin), L.R. Jones & Grout), potato virus X, and net necrosis; it has moderate levels of resistance to Verticillium wilt (Verticillium dahliae, Kleb), pink rot, foliar early blight, corky ringspot, and Erwinia soft rot. Defender is susceptible to scab (common and powdery) and potato leafroll virus; it has moderate susceptibility to dry rot (Fusarium spp.) and potato virus Y. Susceptibilities to internal necrosis, tuber greening, and blackspot bruise also have been noted, but may be minimized through cultural and harvest practices.  相似文献

Summary In artificial inoculations it was found thatFusarium oxysporum f.sp.tuberosi is able to penetrate through potato leaves. The fungus was isolated from the stem of infected plants from all inoculated cultivars. Tubers were completely destroyed if they were inoculated before sprouting. Not any destruction was observed if inoculation was made before young sprouts are longer than 1–3 cm; plants from these sprouts were infected. Lesioned tubers were more sensitive than unlesioned ones, and tuber rot and sprout damage were increased significantly.  相似文献

Summary The majority of isolates ofFusarium avenaceum caused dry rot on potato tubers; many were as pathogenic asF. coeruleum. Pathogenicity ofF. avenaceum was not related to the plant species from which the isolates originated. Tubers of potato cv. Cara were more susceptible than those of cvs Romano or Maris Piper. Temperature (5–15° C) had no effect on lesion depth but lesions tended to be slightly wider at the lowest temperature. Two isolates (of 61) from wheat and white lupin had a moderate level of resistance to thiabendazole. The results are discussed in relation to the control of dry rot in stored potato tubers.  相似文献

Mechanically harvested tubers of 14 potato cultivars grown on both loamy sand and silt loam soils were evaluated for resistance to bacterial soft rot caused byErwinia carotovora subsp.atroseptica. Cultivars were also assayed for calcium and dry matter content to determine possible correlations with soft rot resistance. Resistance of potato tubers to bacterial soft rot was assayed after harvest by inoculating tubers and determining percent surface area decayed after four days in a mist chamber. Tubers of the cultivars differed widely in resistance to bacterial soft rot. The ranking of the resistance of tubers from different cultivars to bacterial soft rot was similar for both soils, although disease severity in tubers from a particular soil type varied with the season. Of the cultivars tested, tubers of Butte, Russet Burbank, and Nor-gold Russet were ranked as the most resistant, whereas tubers of Redsen, Norchip, Norland and Belchip were rated as the most susceptible. Tubers of Rhinered, Red La Soda, Superior, Pontiac, Atlantic, and Nooksack were intermediate in resistance. Neither calcium concentration in medullar tissues nor dry matter of potato tubers alone could be correlated with ranking of the cultivars with respect to severity of bacterial soft rot. However, a multiple linear regression model incorporating both variables indicated that tubers with a high content of both calcium and dry matter were likely to be less susceptible to bacterial soft rot than were tubers with lower percentages of these variables.  相似文献
