为明确上海地区鲜食玉米穗腐病致病镰孢菌的组成和分布,2020年在上海地区各鲜食玉米种植区采集66份穗粒腐果穗病样,通过组织分离、单孢纯化,获得272株单孢分离物。采用形态学和分子生物学等方法对单孢分离物进行鉴定,共鉴定到8个镰孢种203株镰孢菌,其中拟轮枝镰孢、变红镰孢、亚洲镰孢、产黄色镰孢、木贼镰孢、层出镰孢、新知镰孢和居群镰孢的分离频率依次为67.98%、9.36%、6.90%、5.91%、5.42%、2.96%、0.99%和0.49%。在不同采样区拟轮枝镰孢菌占比存在差异,崇明、奉贤和青浦地区采集的样品中拟轮枝镰孢菌分别占76.25%,71.62%和48.98%。结果表明,拟轮枝镰孢在上海地区鲜食玉米种植区广泛分布,为上海地区鲜食玉米穗腐病的主要优势致病菌。  相似文献

我国冬小麦主产省小麦茎基腐镰孢菌的组成及其致病力   总被引:2,自引:0,他引:2  
为了解我国小麦茎基腐病的致病菌镰孢菌(Fusariumspp.)的组成,2009-2012年从江苏、安徽、河南、河北、山东等5个冬小麦主产省采集茎基腐病样本,从中分离了135个镰孢菌菌株,用PCR-RFLP和ITS序列分析的方法进行了种群鉴定,并分别在温室和田间条件下对不同种群及化学型菌株进行致病力测定。结果表明,在分离的135株病菌中,103株为F.asiaticum,17株为F.graminearum,其余为F.acuminatum、F.pseudoraminearum和F.avenaceum,说明引起我国小麦茎基腐的镰孢菌主要由F.asiaticum和F.graminearum组成,且以F.asiaticum种群分布为主。在120个镰孢菌综合物种菌株(F.asiaticum和F.graminearum)中,79株为3-AcDON化学型,17株为15-AcDON化学型,24株为NIV化学型。在F.graminearum菌株中只发现15-AcDON化学型;在F.asiaticum菌株中存在2种化学型,以3-AcDON化学型菌株为主,少部分为NIV化学型菌株。F.asiaticum种群致病力略高于F.graminearum种群,DON化学型菌株致病力略高于NIV化学型菌株,但均无显著性差异。  相似文献

鲁西南地区小麦茎基腐病病原菌鉴定及其致病力分析   总被引:1,自引:0,他引:1  
为明确山东小麦茎基腐病害主要致病菌组成及其致病力,于2016年在山东省聊城、德州、临沂市共7个地点采集了具有典型茎基腐症状的病株并对其主要致病菌进行了分离、鉴定和致病力分析。结果共分离到45株病原真菌,其中有22株为镰孢菌,分别属于假禾谷镰孢菌(Fusarium pseudograminearum)、禾谷镰孢菌(F.graminearum)和亚洲镰孢菌(F.asiaticum),以假禾谷镰孢菌(F.pseudograminearum)分离频率最高,占所有镰孢菌株的68.18%;其余菌株属于链格孢菌(Alternaria spp.)。对22株镰孢菌进行小麦苗期致病力及重分离率测定发现,假禾谷镰孢菌(F.pseudograminearum)的致病力明显强于禾谷镰孢菌和亚洲镰孢菌,后两者的致病力相差不大;3种病原菌的重分离率均大于40%,病原菌测序鉴定结果与接种病原菌一致。  相似文献

Fusarium sambucinum is one of the most important causal agents that not only cause the dry rot disease of potato tubers in fields and stores worldwide but also capable of producing secondary metabolites toxic for people and animals. Here we present the first draft genome sequence of the strain (F-4) estimated to be around appx. 42.0 Mb. The genome has 12,845 protein coding genes with more than 35,900 exons and gene density of 3.13 per 10Kb. F. sambucinum is evolutionary more close to the F. graminearum among the Fusarium species complex. The genome sequence represents a valuable resource for understanding the pathogenecity and virulence factors, and their evolution within the complex and highly plastic genus Fusarium.  相似文献

Fusarium semitectum is one of the important causal agents of dry rot of potato tubers in the world. In order to determine genetic variability among 41 isolates of F. semitectum, morphological and molecular studies were carried out. All F. semitectum isolates were recovered from infected potato tubers with dry rot symptoms collected from four provinces in Iran. According to macroscopic and microscopic characteristics, 41 isolates of F. semitectum were classified in two morphotypes (morphotypes I and II). All 41 isolates were evaluated for their pathogenicity on healthy potato tubers. Tuber rot symptoms were observed on the 21st day after inoculation of Fusarium isolates on the tubers tested. The measurement was done by comparing the depth and width of lesion expansion among the isolates. Molecular characterization through PCR-IGS-RFLP analysis by six restriction enzymes (AluI, BsuRI, Eco88I, MspI, TaqI and PstI) divided the 41 isolates of F. semitectum into two separated clusters that were in accordance with the morphological characterization.  相似文献

通过形态学和分子生物学方法对2016年采自吉林省36个村镇的玉米根腐病样品进行镰孢菌分离和鉴定,并通过田间试验筛选有效药剂。结果发现,共分离获得84株镰孢菌,主要包括尖孢镰孢、禾谷镰孢、拟轮枝镰孢、层出镰孢、三线镰孢等11种镰孢菌,其中尖孢镰孢平均分离频率最高,为45.2%;其次是禾谷镰孢,平均分离频率为20.2%。从分布区域看,尖孢镰孢是白城、吉林、辽源和通化地区玉米根腐病的主要致病菌,分离频率分别为75%、54.5%、85.7%和45.5%;禾谷镰孢是松原和四平地区玉米根腐病的主要致病菌,分离频率分别为41.7%和41.7%。田间试验表明,5种药剂的防治效果为16.35%~81.26%,药种比为10 g/kg的10%咯菌腈悬浮种衣剂对玉米根腐病防治效果最好,防效达81.26%。吉林省玉米根腐病的主要病原菌为尖孢镰孢,药种比为10 g/kg的10%咯菌腈悬浮种衣剂可用于玉米根腐病的田间防治。  相似文献

为明确甘肃省玉米根腐病致病镰孢菌种类,2017~2019年在甘肃省不同地区采集玉米植株进行组织分离,采用形态学和分子生物学鉴定方法对分离物进行鉴定,按照柯赫氏法则对其致病性进行测定。结果表明,引起甘肃省玉米根腐病的致病镰孢菌为厚垣镰孢Fusarium chlamydosporum、禾谷镰孢F.graminearum、尖镰孢F.oxys-porum、层出镰孢F.proliferatum、茄镰孢F.solani、三线镰孢F.tricinctum、拟轮枝镰孢F.verticillioides和产黄色镰孢F.thapsinum,其中尖镰孢和拟轮枝镰孢为优势镰孢菌,分离频率分别为22.5%和20%。通过致病性测定发现,所有分离的镰孢菌都会引起玉米根系不同程度变褐,导致根系生长缓慢、茎基部溢缩和腐烂、近地部叶片黄化、叶边缘干枯等症状。  相似文献

Progeny tubers from seed potatoes originating from either a traditional clonal selection method or the more modernin vitro tissue culture method of propagation were tested for storage rot response following inoculation of three tuber sites with twoFusarium species. Significant differences were found among disease responses for the twoFusarium species and for the four cultivars tested. Disease symptoms were less severe forF. solani var.coeruleum than forF. sambucinum. Fusarium sambucinum caused less disease in Superior than Kennebec, Russet Burbank and Sebago. Seed propagation method did not significantly affect disease response except for one case;in vitro culture of Kennebec had less disease due toF. sambucinum at the tuber side inoculation site than clonal selection. Kennebec and Superior inoculatedF. sambucinum had significant differences with respect to the number of years of field propagation for the tuber side inoculation site and for the tuber mean rot index. Significant differences in number of years of field propagation were also found for eye-ends of Superior tubers and stem ends of Russet Burbank tubers inoculated withF. solani var.coeruleum.  相似文献

Several species ofFusarium cause both dry rot in storage and seedpiece decay in the field, which results in severe losses in all major potato-growing regions. In temperate climates twoFusarium species are the principle causes of tuber dry rot-F.sambucinum andF. coeruleum. Resistance toF. sambucinum was identified in cultivated and wildSolanum species. Relatively high levels of resistance exist in the following 2EBN species:S. boliviense (PI#’s 265860, 310975, 310974, 545853, 545889, 545964),S. gourlayi ssp.gourlayi (PI#’s 473344, 545865),S. microdontum (PI#’s 310979, 473171),S. sancta-rosae (PI# 498392),S. kurtzianum (PI# 472930),S. fendleri ssp.fendleri (PI# 262895),S. gandarillasii (PI#’s 265866, 283076),S. oplocense (PI# 498281) andS. vidaurrei (PI# 498332). Summary The majority of isolates ofFusarium avenaceum caused dry rot on potato tubers; many were as pathogenic asF. coeruleum. Pathogenicity ofF. avenaceum was not related to the plant species from which the isolates originated. Tubers of potato cv. Cara were more susceptible than those of cvs Romano or Maris Piper. Temperature (5–15° C) had no effect on lesion depth but lesions tended to be slightly wider at the lowest temperature. Two isolates (of 61) from wheat and white lupin had a moderate level of resistance to thiabendazole. The results are discussed in relation to the control of dry rot in stored potato tubers.  相似文献

Fungi Causing Dry Tuber Rots of Seed Potatoes in Storage in Scotland   总被引:2,自引:0,他引:2  
The occurrence, pathogenicity (1997 isolates only) and fungicide sensitivity of pathogens causing dry tuber rots in Scottish seed potatoes was investigated over three storage seasons between 1997 and 2000 in relation to region of production, cultivar, class and generation of seed potatoes, type of temperature control in store and earthiness of tubers. A total of 156 samples, each comprising up to ten rotted tubers, was received over the three seasons. The relative importance of each pathogen in causing rots was summarised by calculating a mean rot index that combined the prevalence of a pathogen in the samples with the incidence of tubers affected by the pathogen within those samples. Phoma foveata (gangrene) had the highest rot index, which was five times greater than for P. exigua and ten times greater than for P. eupyrena. This relationship was mirrored by the relative pathogenicities of P. foveata and P. eupyrena, as measured by size of rots developing at inoculated wounds in test tubers. Fusarium avenaceum appeared to be the greatest cause of Fusarium dry rots, having a rot index at least twice as great as that for F. solani var. coeruleum. Infection by F. sulphureum was relatively uncommon. In the pathogenicity test, F. avenaceum, F. solani var. coeruleum and F. sulphureum produced rots of similar depths and widths and larger than those of F. culmorum. The mean rot index for Cylindrocarpon spp. was slightly more than that for F. avenaceum, whereas C. destructans produced smaller rots in the tuber pathogenicity test. Region of production affected the prevalence of P. foveata and F. avenaceum, but only the occurrence of P. foveata was affected by class and generation of seed potatoes. Isolate sensitivity to thiabendazole and imazalil was examined in vitro over 2 years. Six out of seven isolates of F. sulphureum were resistant to thiabendazole, with the remaining isolate being partially resistant. The growth of two out of 34 isolates of F. avenaceum was inhibited by more than 50% only at 100 mg thiabendazole l−1. All isolates, except those of F. avenaceum,, were inhibited in their growth by more than 50% at either 1 or 10 mg imazalil l−1. However, the growth of 40% of isolates of F. avenaceum was inhibited only at 100 mg l−1. The various changes in pathogen prevalence and isolate sensitivity to fungicides recorded in this study highlight the need for regular monitoring programmes to be conducted in order that disease-control strategies can remain effective.  相似文献

Summary Virulence of 12Fusarium oxysporum isolates was determined on three potato cultivars Late Harvest, BP1 and Kimberley Choice. The production of fusaric acid by the 12F. oxysporum isolates was also determined. The aim of the study was to determine whether a correlation exists between fusaric acid production and virulence ofF. oxysporum isolates. Late Harvest was the most tolerant and BP1 the most susceptible cultivar. Virulence ranking, order of theF. oxysporum isolates with the different cultivars corresponded with the mean of value of the ranking order for all three cultivars. A correlation was found between virulence of isolates and fusaric acid production except for Late Harvest. Fusaric acid production may play a major role in the development of dry rot in potato tubers.  相似文献

禾谷镰孢菌(Fusarium graminearum)是小麦上的重要病原真菌。通过致病性测定从江苏小麦茎基腐病病害样本分离菌中筛选到了对小麦赤霉病和茎基腐病都有强致病力的F.graminearum菌株GF1117。为了对F.graminearum进行生物防治,利用稀释涂布平板法和平板对峙法从小麦不同生境中分离筛选到35株对GF1117具有明显拮抗效果的细菌菌株,分别在田间和温室进行了小麦赤霉病和茎基腐病的生物防治试验。结果表明,35株拮抗细菌对小麦赤霉病均有一定的防治效果,且在小麦感病品种和中抗品种上对茎基腐病的防效不同;菌株1-8对两种病害的防效都在45%以上。  相似文献

Fusarium spp. is among the largest and most important pathogen groups that attack major grain crops including sorghum. The objectives of this study were to compare the virulence of Fusarium spp. associated with sorghum and examine the mode of resistance of genotypes to the disease. Eight Fusarium species – F. verticillioides, F. thapsinum, F. andiyazi, F. proliferatum, F. nyagamai, F. pseudoanthophilum, F. brevicatenulatum, and F. pseudonygamai – were studied using three sorghum genotypes under greenhouse condition. Three of these pathogens (F. verticillioides, F. thapsinum, and F. andiyazi) were selected for genetic analysis of resistance under field conditions. Sorghum genotypes with contrasting stalk rot reactions were selected for use in both the greenhouse and field experiments. Two weeks after flowering, plants were inoculated with liquid inoculum culture (5 × 104 conidia ml−1) of the different pathogens. Plants were harvested 4 weeks after inoculation and rated for disease severity on the basis of lesion length and number of nodes crossed by the lesion. Among the pathogens, F. thapsinum resulted in consistently higher disease scores in all genotypes under all environments. Likewise, genotype SC599 showed the greatest and most stable resistance as inbred as well as in hybrid combinations as shown by consistently high GCA for resistance to all pathogen species. We recommend that future screening exercises for Fusarium stalk rot resistance utilize F. thapsinum as the causal organism and include the resistant genotype SC599 as a control.  相似文献

Soil suppressiveness to Fusarium root rot of soybean had been observed in a black soil field after a long-term fertilization with nitrogen (N) and phosphorus (P) fertilizer combined with pig manure as organic amendment (NPM), rather than that with only nitrogen and phosphorus fertilizer (NP) or no fertilizer (NF). To determine the microbial role on this suppressiveness, fungal and bacterial community characteristics in NPM, NP and NF treatments were investigated by qPCR and DGGE. Compared with the similar bacterial community characteristics among 3 treatments, fungal community, especially Fusarium population size and community composition in NPM treatment were different with those of NP and NF groups. Based on the isolation and pathogenicity test, pathogenic F. oxysporum, F. graminearum, F. verticillioide and F. lateritium absolutely dominated Fusarium community in NF and NP groups. Nonpathogenic F. avenaceum, F. equiseti, F. culmorum, F. redolens, F. solani and F. tricinctum dominated Fusarium community in NPM group. Isolation rate of pathogenic Fusarium in NPM reduced from 100% to 38% in NF. These results suggested that the dominance of soil non-pathogenic Fusarium population induced by organic amendment might play an important role on suppressing Fusarium root rot in the tested field.  相似文献

The level ofFusarium dry-rot resistance found in important potato varieties has been improved with agronomically acceptableSolanum tuberosum genotypes. Three breeding selections, A74114-4, A76260-16, and BR6316-7 were highly resistant to the two commonFusarium dry-rot species (F. roseum Lk ex Fr. f. sp.sambucinum [Fs], andF. solani var.coeruleum (Sacc), Booth [Fc]). Resistance was determined separately for each of these species. Resistance to Fs was genetically distinct from resistance to Fc since there was no correlation between Fs and Fc reaction for 180 progeny clones from six crosses representing all combinations of resistance. Resistance to each of theseFusarium species was readily transmitted to progeny. Seventy-five percent of the progeny from resistant X resistant parents were also resistant to Fs, while 95% of the progeny from susceptible X susceptible parents were susceptible. Mixed infections withErwinia atroseptica and eitherFusarium species resulted in a qualitative change from a resistant reaction to a susceptible reaction. Bacterial interaction withFusarium spp. is, therefore, an important consideration in determining storage-rot resistance.  相似文献

Résumé La capacité infectieuse de 5 espèces ou variétés deFusarium responsables de la pourriture des tubercules est étudiée.F. roseum var.sambucinum possède la capacité infectieuse la plus importante.F.r. var.arthrosporioides etF.r. var.culmorum sont, à l'inverse, caractérisés par une capacité infectieuse faible, ils ne produisent des pourritures qu'à des doses élevées de spores et sont rapidement limités par la cicatrisation. Enfin,F. solani var.coeruleum etF.r. var.graminearum possèdent un comportement intermédiaire. Le niveau de ‘virulence’ des 5Fusarium apprécié sur 9 cultivars, est identique mais leur niveau ‘d'agressivité’ varie; ils sont doués d'une agressivité non-spécifique (l'ensemble desFusarium provoque sur un cultivar déterminé, le même niveau de maladie) ou spécifique (une espèce ou variété deFusarium possède un comportement déterminé sur un cultivar qui ne correspond pas à celui des autresFusarium). Il est traité dans la discussion-conclusion de l'apport de ces résultats dans un programme de sélection vis-à-vis des Fusarioses.
Summary The objective of this study is to confirm the infective potential of 5 species or varieties ofFusarium causing dry rot in potato tubers (F. roseum var.sambucinum (F.r. samb.), F. solani var.coeruleum (F.s. coer.), F. roseum var.arthrosporioides (F.r. arth.), F. roseum var.culmorum (F.r. culm.) andF. roseum var.graminearum (F.r. gram.)) in relation to different factors: inoculum level, wound type, time taken for wound healing, physiological age of tubers and cultivar. At low inoculum levels, certainFusarium can be highly aggressive, for exampleF.r. samb. and to a lesser extentF.s. coer. andF.r. gram., whereas others (F.r. arth. andF.r. culm.) can initiate a rot only when the inoculum level is high (Fig. 1 and Table 1). With respect to wounding (Tables 2 and 3),F.r. samb. causes extensive lesions regardless of wound type in contrast to the otherFusarium which do so only when the wounds are deeper than 2 mm. The reaction of the differentFusarium to wound healing is variable (Fig. 2). Some(F.r. culm. andF.r. arth.) are usually not affected whereas with others infection is arrested after a longer healing time: 8 days in the case ofF.s. coer. andF.r. samb. and longer withF.r. gram. Wound healing reaction can modify the relative ranking order of cultivars; a cultivar classified as resistant to aFusarium in the absence of healing can find itself grouped with the less resistant ones after wound healing probably because of a rate of healing lower than that of the other cultivars (Table 4). Behaviour of the different species and varieties ofFusarium is not affected differently by the physiological age of tubers; disease level is lower in young than in older tubers (Table 5). Moreover, cultivar reaction is more pronounced on young than on older tubers.

Die Vernarbungsreaktion kann das ungef?hre Sortenklassement ver?ndern; eine für ein Fusarium wenig empfindliche Sorte kann zu den Empfindlichsten geh?ren durch ihres Verhalten bei der Wundheilung, wenn ihre Vernarbungszeit l?nger dauert als die anderer Sorten (Tab. 4). Die verschiedenen Arten oder Sorten desFusarium reagieren auf gleiche Weise was das physiologische Alter der Knollen betrifft. Der Krankheitsbestand ist auf jüngeren Knollen schw?cher als auf ?lteren (Tab. 5). Weiter ist das Verhalten der verschiedenen Sorten deutlicher bei den jungen Knollen als bei alten.

Fusarium wilt is a disease that restricts carnation (Dianthus caryophyllus L.) yield worldwide. Efficacies in reducing the Fusarium wilt of carnation (FWC), of various types of organic amendments (fresh or pelletized poultry manure, pelletized Brassica carinata and olive residue compost) combined with soil solarization, were compared in two biennial field trials conducted in a greenhouse with a history of carnation monoculture over 8 years. Soil treatments combining organic amendments and soil solarization significantly reduced disease incidence (86–99%) and increased the number of commercial carnation stems by 5–9 times compared to non-treated plots. Twenty-one Fusarium spp. isolates, with different colony morphologies were recovered from soil samples taken in the greenhouse, before the application of treatments in June 2013. Nineteen of them were morphological and molecularly characterized. Additionally, two pathogenicity tests with 17 isolates recovered from greenhouse soils and two isolates recovered from organic amendments were performed. Fusarium species associated with carnation cultivation were identified as Fusarium oxysporum (43%), Fusarium proliferatum (24%), and Fusarium solani (33%). The phylogenetic analysis of the translation elongation factor 1 alpha (EF-1α) region distinguished highly aggressive isolates of F. oxysporum f. sp. dianthi, from low aggressive isolates. The pathogenicity tests showed that FWC has a complex etiology, with several Fusarium spp. identified as causal agents. F. proliferatum and F. solani are associated with carnation wilt for the first time in Spain.  相似文献

Fungi of the genusFusarium Link isolated in Perú from potatoes (Solanum tuberosum L. subsp.andigena Juz. & Buk.) affected by: i) wilt, ii) dry rot of stored tubers, iii) secondary rot in tubers affected by “cork” disease (potato mop-top virus?), caused dry rot when inoculated to tubers of Renaacimiento variety. The identification of some of theseFusaria suggests thatFusarium solani Snyder & Hansen is associated with wilt and dry rot, andFusarium oxysporum Snyder & Hansen with rot secondary to cork disease. Tubers of eight potato varieties inoculated with fourFusaria representative of those isolated showed a great range in levels of susceptibility to colonization by theseFusaria, Sapa being the most susceptible variety, and Mantaro appearing to be the most resistant. These results suggest that useful levels of resistance toFusarium tuber rot are available in the Peruvian germ-plasm.  相似文献

A total of 137 actinomycetes cultures, isolated from 25 different herbal vermicomposts, were characterized for their antagonistic potential against Fusarium oxysporum f. sp. ciceri (FOC) by dual-culture assay. Of the isolates, five most promising FOC antagonistic isolates (CAI-24, CAI-121, CAI-127, KAI-32 and KAI-90) were characterized for the production of siderophore, cellulase, protease, hydrocyanic acid (HCN), indole acetic acid (IAA) and antagonistic potential against Rhizoctonia bataticola, which causes dry root rot in chickpea (three strains viz. RB-6, RB-24 and RB-115) and sorghum (one strain). All of the five FOC antagonistic isolates produced siderophore and HCN, four of them (except KAI-90) produced IAA, KAI-32 and KAI-90 produced cellulase and CAI-24 and CAI-127 produced protease. In the dual-culture assay, three of the isolates, CAI-24, KAI-32 and KAI-90, also inhibited all three strains of R. bataticola in chickpea, while two of them (KAI-32 and KAI-90) inhibited the tested strain in sorghum. When the FOC antagonistic isolates were evaluated further for their antagonistic potential in the greenhouse and wilt-sick field conditions on chickpea, 45-76% and 4-19% reduction of disease incidence were observed, respectively compared to the control. The sequences of 16S rDNA gene of the isolates CAI-24, CAI-121, CAI-127, KAI-32 and KAI-90 were matched with Streptomyces tsusimaensis, Streptomyces caviscabies, Streptomyces setonii, Streptomyces africanus and an identified species of Streptomyces, respectively using the BLAST searching. This study indicated that the selected actinomycete isolates have the potential for biological control of Fusarium wilt disease in chickpea.  相似文献
