Plasmodium spp. protozoa cause malaria and are known to infect humans and a variety of animal species including macaque monkeys. Here we report both our experience with malaria recrudescence in cynomolgus monkeys (Macaca fascicularis) in a toxicity study and the results of a survey on Plasmodium infection in cynomolgus monkeys imported to Japan for laboratory use. A cynomolgus monkey from the toxicity study presented with severe anemia and Plasmodium protozoa in erythrocytes on a thin blood smear and was subsequently diagnosed with symptomatic malaria. In this animal, congestion and accumulation of hemozoin (malaria pigment) in macrophages were noted in the enlarged and darkly discolored spleen. As a follow-up for the experience, spleen sections from 800 cynomolgus monkeys in toxicity studies conducted between 2003 and 2013 were retrospectively examined for hemozoin deposition as a marker of Plasmodium infection. The origin of the animals included Cambodia, China, Indonesia, and Vietnam. Hemozoin deposition was confirmed in 44% of all examined monkeys. Monkeys from Indonesia showed the highest incidence of hemozoin deposition (approx. 80%). A high prevalence of Plasmodium infection in laboratory monkeys was also confirmed with polymerase chain reaction (PCR) by using Plasmodium genus-specific primers. Although Japan is not a country with endemic malaria, it is important to be aware of the prevalence and potential impact of background infection with Plasmodium spp. and recrudescence of symptomatic malaria in imported laboratory monkeys on pharmaceutical toxicity studies. 相似文献

Salmonella (S.) Typhimurium and S. Enteritidis are the major causative agents of food-borne illnesses worldwide. Currently, a rapid detection system using multiplex real-time polymerase chain reaction (PCR) has been applied for other food-borne pathogens such as Escherichia coli, Staphylococcus aureus and Streptococcus spp. A multiplex real-time PCR was developed for the simultaneous detection of Salmonella spp., especially S. Typhimurium and S. Enteritidis, in beef and pork. For the specific and sensitive multiplex real-time PCR, three representative primers and probes were designed based on sequence data from Genbank. Among the three DNA extraction methods (boiling, alkaline lysis, and QIAamp DNA Mini Kit), the QIAamp DNA Mini Kit was the most sensitive in this study. The optimized multiplex real-time PCR was applied to artificially inoculated beef or pork. The detection sensitivity of the multiplex real-time PCR was increased. The specificity of the multiplex real-time PCR assay, using 128 pure-cultured bacteria including 110 Salmonella isolates and 18 non-Salmonella isolates, was 100%, 100% and 99.1% for Salmonella spp., S. Typhimurium and S. Enteritidis, respectively. The sensitivity was 100%, 100% and 91.7% for Salmonella spp., S. Typhimurium and S. Enteritidis, respectively. The multiplex real-time PCR assay developed in this study could detect up to 0.54 ± 0.09 and 0.65 ± 0.07 log10 CFU/ml for S. Typhimurium and S. Enteritidis for beef, 1.45 ± 0.21 and 1.65 ± 0.07 log10 CFU/ml for S. Typhimurium and S. Enteritidis for pork, respectively, with all conditions optimized. Our results indicated that the multiplex real-time PCR assay developed in this study could sensitively detect Salmonella spp. and specifically differentiate S. Typhimurium from S. Inhibins are members of the TGFβ superfamily and act as suppressors
of follicle stimulating hormone (FSH) secretion from pituitary glands
via a negative feedback mechanism to regulate folliculogenesis. In
this study, the INHBB gene was knocked down by three
RNAi-Ready pSIREN-RetroQ-ZsGreen vector- mediated recombinant plasmids
to explore the effects of INHBB silencing on
granulosa cell (GC) cell cycle, apoptosis and steroid production
in vitro. Quantitative real-time polymerase chain
reaction, Western blot, flow cytometry and ELISA were performed to
evaluate the role of INHBB in the mouse GC cell
cycle, apoptosis and steroid production in vitro. The
results showed that the relative mRNA and protein expression of
INHBB in mouse GCs can be significantly reduced by
RNAi with pshRNA-B1, pshRNA-B2 and pshRNA-B3 plasmids, with pshRNA-B3
having the best knockdown efficiency. Downregulation of the expression
of INHBB significantly arrests cells in the G1 phase
of the cell cycle and increases the apoptosis rate in GCs. This was
further confirmed by downregulation of the protein expressions of
Cyclin D1, Cyclin E and Bcl2, while the protein expression of Bax was
upregulated. In addition, specific downregulation of
INHBB markedly decreased the concentration of
estradiol and progesterone, which was further validated by the
decrease in the mRNA levels of CYP19A1and
CYP11A1. These findings suggest that inhibin βB is
important in the regulation of apoptosis and cell cycle progression in
granulosa cells. Furthermore, the inhibin βB subunit has a role in the
regulation of steroid hormone biosynthesis. Evidence is accumulating
to support the concept that inhibin βB is physiologically essential

Present study was undertaken to study the prevalence of β-haemolytic streptococci in
equine of northern temperate region of Jammu and Kashmir, India. One hundred and forty one
samples were collected in duplicate from nasopharyngeal tract of diseased (53) and
apparently healthy equine (88) for isolation and direct PCR. A total of 77 isolates of
streptococci were recovered from 141 samples with an overall prevalence rate of 54.60%.
Out of these 77 isolates, 52 were from diseased and 25 from apparently healthy animals. Of
the 77 isolates, 4 were identified as Streptococcus equi subsp.
equi, 56 as S. equi subsp.
zooepidemicus and 17 as S. dysgalactiae subsp.
equisimilis. Thus the overall prevalence of S. equi subsp.
equi, S. equi subsp. zooepidemicus
and S. dysgalactiae subsp. equisimilis was 2.83, 39.71
and 12.05% respectively. The sensitivity of the PCR for the detection of S.

Biotite and bentonite are phyllosilicate minerals that were originally used in
industrial applications. Several beneficial activities of them have recently been
reported, especially regulation of the immune system and antimicrobial effects. Therefore,
we investigated the immune-enhancing and bacterial clearance effects of a biotite and
bentonite mixture (BBM) on experimental infection of Salmonella enterica
serovar Typhimurium (S. Typhimurium) to determine whether the BBM could
be used as an alternative antibiotic. We administered 1% or 2% BBM as a feed supplement.
We then evaluated the bacterial clearance effects of the BBM against S.
Typhimurium. We also evaluated the immune-enhancing effect of the BBM through several
immunological experiments that included examination of the lysozyme activity,
CD4+/CD8+ T lymphocyte ratio and the T-helper type 1 (Th 1)
cytokine profile. The clinical signs of S. Typhimurium and the number of
viable bacteria in feces and tissues were significantly decreased in both BBM groups,
especially in the 2% BBM group. The BBM also markedly enhanced the lysozyme activity,
CD4+/CD8+ T lymphocyte ratio and expression levels of IFN-γ and
IL-12 in S. Typhimurium-challenged pigs. Therefore, the BBM could be a
good candidate as an alternative antibiotic that improves Th 1-specific immune responses

The prevalence of resistant
genes against β-lactams in 119 Aeromonas strains was determined. A large
number (99.2%) of the present fish strains were resistant to one or more β- lactams
including ceftiofur, amoxicillin-clavulanic acid, ampicillin, piperacillin and
cefpodoxime. Among antibiotic resistance phenotypes, the simultaneous resistance to all
β-lactams occurred in 25.2% (n=30) of all strains, which consisted of 18 strains of
A. dhakensis, 8 strains of A. caviae, 2 strains of
A. hydrophila and only one strain of A. veronii. For
exploring genetic background of the antibiotic resistances, multiple PCR assays were
subjected to detect β-lactamase-encoding genes, blaTEM,
blaOXA-B and blaCTX-M. In the
results, the blaTEM-1 gene was harbored in all strains,
whereas only 3 strains harbored blaOXA gene. In the case of
blaCTX-M gene, the gene was detected in 21.0% (25 out of
119) of all strains, which countered with 80% (20 out of 25) of A.
dhakensis, 8% (2 out of 25) of A. caviae and 12% (3 out of 25)
of A. hydrophila. In addition, most of the
blaCTX-M positive strains showed simultaneous resistance to
all β-lactams (18 out of 30 strains). In sequence analysis for
blaCTX-M genes detected, they were CTX-M group 1-encoding
genes including blaCTX-M-33 from 3 eel strains of A.
dhakensis. Therefore, A. dhakensis obtained from cultured fish
could represent a reservoir for spreading genes encoding CTX-M group 1 enzymes and hence
should be carefully monitored, especially for its potential risk to public health. 相似文献

Feline upper respiratory tract infection due to Aspergillus spp. is
considered an emerging disease, with the number of reported cases continuing to rise. In
this study, we report the first case of feline sinonasal aspergillosis caused by
Aspergillus fischeri in Japan. The patient presented after 2 months of
progressive facial deformity around the nose and nasal discharge. The isolate from this
case was susceptible to itraconazole (ITZ), voriconazole and micafungin, but was resistant
to amphotericine B. However, the infected cat died approximately 1 month after referral,
despite treatment for 12 days ITZ administered orally at 10 mg/kg. 相似文献

A total of 349 Salmonella enterica subspecies enterica
serovar Choleraesuis (S. Choleraesuis) strains, which were isolated
between 2008 and 2012 from 349 pigs at two slaughterhouses in Okinawa Prefecture, Japan,
were investigated for antimicrobial susceptibility and the presence of antimicrobial
resistance genes. All isolates were resistant to at least four antimicrobial agents. The
antimicrobial agents for which isolates showed a high incidence of resistance were as
follows: ampicillin (100%) and streptomycin (100%), followed by gentamicin (99.7%),
oxytetracycline (99.7%), sulfamethoxazole/trimethoprim (99.4%), nalidixic acid (40.1%) and
oxolinic acid (40.1%). All isolates were sensitive to cefuroxime, ceftiofur, colistin,
fosfomycin, enrofloxacin, orbifloxacin and danofloxacin. The predominant resistance
phenotypes and genotypes were: resistance to ampicillin, streptomycin, gentamicin,
oxytetracycline and sulfamethoxazole/trimethoprim (58.5%, 204/349) and
blaTEM-strA-strB-aadA1-aadA2-aacC2-tet
(B)-sul1-sul2-dhfrXII-dhfrXIII (36.1%, 126/349). The quinolone
resistance-determining regions (QRDRs) of gyrA, gyrB, parC and
parE of the quinolone-resistant isolates (n=12) showed amino acid
substitutions of Ser-83→Phe or Asp-87→Tyr in GyrA and Ser-107→Ala in ParC. To our
knowledge, this is the first report on the molecular characterization of antimicrobial

Forty‐seven elective surgery dogs allocated to 4 groups: Seronegative control (n = 12), A. platys (n = 10), E. canis (n = 14) and A. platys, and E. canis (n = 11) seroreactive.Methods
Preoperatively, hemostasis was assessed by platelet count, prothrombin time, activated partial thromboplastin time, and buccal mucosal bleeding time. Intra‐ and postoperative bleeding scores were subjectively assigned. Blood, spleen, bone marrow, and lymph node aspirates were tested by PCR.Results
Bleeding scores in dogs coseroreactive for A. platys and E. canis were higher (P = .015) than those of seronegative dogs. A. platys DNA was amplified from 7/21 (33%) A. platys seroreactive dogs and from 1 E. canis seroreactive dog; E. canis DNA was amplified from 21/25 (84%) E. canis seroreactive dogs. E. canis DNA was amplified most often from blood, whereas A. platys DNA was amplified most often from bone marrow.Conclusions and Clinical Importance
Apparently healthy, free‐roaming dogs coseropositive for A. platys and E. canis may have increased intraoperative bleeding tendencies despite normal hemostatic parameters. Future investigations should explore the potential for vascular injury as a cause for bleeding in these dogs. Improved tick control is needed for dogs in Grenada. 相似文献10.

The present report describes Syphacia
(Syphacia) maxomyos sp. n. (Nematoda:
Oxyuridae) from two species of spiny rats, Maxomys musschenbroekii from
Sulawesi and M. whiteheadi from Sumatra. It is characterized by a
cephalic plate extending laterally with dorsoventral constriction and stumpy eggs with an
operculum rim reaching pole. It is readily distinguishable by the former feature from all
of hitherto known representatives of this genus in Indonesia, but it resembles parasites
in Murini and Hydromyni rodents in continental Asia and Sahul. This is the first
Syphacia species distributed in both the Sunda Shelf and Sulawesi with
the exception of Syphacia muris, a cosmopolitan pinworm found in rodents
of the of genus Rattus. It is surmised that S. maxomyos
is specific to Maxomys and that it was introduced to Sulawesi by
dispersal of some Maxomys from the Sunda Shelf. 相似文献

Ticks carry and transmit a remarkable array of pathogens including bacteria, protozoa and
viruses, which may be of veterinary and/or of medical significance. With little to no
information regarding the presence of tick-borne zoonotic pathogens or their known vectors
in southern Africa, the aim of our study was to screen for Anaplasma
phagocytophilum, Borrelia burgdorferi, Coxiella
burnetii, Rickettsia species and Ehrlichia
ruminantium in ticks collected and identified from ruminants in the Eastern
Cape, Free State, KwaZulu-Natal and Mpumalanga Provinces of South Africa. The most
abundant tick species identified in this study were Rhipicephalus evertsi
evertsi (40%), Rhipicephalus species (35%), Amblyomma
hebraeum (10%) and Rhipicephalus decoloratus (14%). A total of
1634 ticks were collected. DNA was extracted, and samples were subjected to PCR
amplification and sequencing. The overall infection rates of ticks with the target
pathogens in the four Provinces were as follows: A. phagocytophilum, 7%;
C. burnetii, 7%; E. ruminantium, 28%; and
Rickettsia spp., 27%. The presence of B. burgdorferi
could not be confirmed. The findings of this study show that zoonotic pathogens are
present in ticks in the studied South African provinces. This information will aid in the
epidemiology of tick-borne zoonotic diseases in the country as well as in raising
awareness about such diseases in the veterinary, medical and tourism sectors, as they may

Bartonella rochalimae was first isolated from the blood of a human who traveled to Peru and was exposed to multiple insect bites. Foxes and dogs are likely natural reservoirs for this bacterium. We report the results of experimental inoculation of two dogs, five cats and six guinea pigs with the only human isolate of this new Bartonella species. Both dogs became bacteremic for 5–7 weeks, with a peak of 103–104 colony forming units (CFU)/mL blood. Three cats had low bacteremia levels (< 200 CFU/mL) of 6–8 weeks’ duration. One cat that remained seronegative had two bacterial colonies isolated at a single culture time point. A fifth cat never became bacteremic, but seroconverted. None of the guinea pigs became bacteremic, but five seroconverted. These results suggest that dogs could be a reservoir of this strain of B. rochalimae, in contrast to cats and guinea pigs. 相似文献

To search natural compounds having inhibitory effect on bacterial growth is important,
particularly in view of growing multidrug resistant (MDR) strains of bacterial pathogens.
Like other bacterial pathogens, MDR Vibrio cholerae, the causative agent
of diarrheal disease cholera, is becoming a great concern. As an approach of searching new
antimicrobial agents, here, we show that anethole, a well-studied natural component of
sweet fennel and star anise seeds, could potentially inhibit the growth of MDR O1 El Tor
biotype, the ongoing 7th cholera pandemic variant strains of toxigenic V.
cholerae. The minimum inhibitory concentration (MIC) of anethole against
diverse O1 El Tor biotype strains is evaluated as 200
µg/ml. Moreover, the effect of anethole is
bactericidal and exerts rapid-killing action on V.
cholerae cells. This study is the first report which demonstrates
that anethole, purified from natural compound, is a potent inhibitor of growth of
toxigenic V. cholerae. Our data suggest that anethole could be a
potential antimicrobial drug candidate, particularly against MDR V.

X-chromosome inactivation (XCI) is an epigenetic process that equalizes expression of X-borne genes between
male and female eutherians. This process is observed in early eutherian embryo development in a
species-specific manner. Until recently, various pluripotent factors have been suggested to regulate the
process of XCI by repressing XIST expression, which is the master inducer for XCI. Recent
insights into the process and its regulation have been restricted in mouse species despite the evolutionary
diversity of the process and molecular mechanism among the species. OCT4A is one of the
represented pluripotent factors, the gate-keeper for maintaining pluripotency, and an XIST
repressor. Therefore, in here, we examined the relation between OCT4A and X-linked genes in
porcine preimplantation embryos. Three X-linked genes, XIST,
LOC102165544, and RLIM, were selected in present study because their
orthologues have been known to regulate XCI in mice. Expression levels of OCT4A were
positively correlated with XIST and LOC102165544 in female blastocysts.
Furthermore, overexpression of exogenous human OCT4A in cleaved parthenotes generated
blastocysts with increased XIST expression levels. However, increased XIST
expression was not observed when exogenous OCT4A was obtained from early blastocysts. These
results suggest the possibility that OCT4A would be directly or indirectly involved in
XIST expression in earlier stage porcine embryos rather than blastocysts. 相似文献

Gongylonema pulchrum is an important parasite of captive
primates. Twelve rabbits were infected with 30 third-stage larvae of G.
pulchrum. At 4–7 months post-infection, animals were administered levamisole at
a single dose of 12 mg/kg, levamisole at 8 mg/kg three times at 2-day intervals,
levamisole at a single dose of 8 mg/kg after administration of mebendazole at 70 mg/kg for
3 days or 8 ml of distilled water for 3 days (control). Necropsy at 14
days after treatment revealed that single and multiple dosages of levamisole reduced
nematode burdens by 68.4% and 89.5%, respectively. The combined regimen of mebendazole and
levamisole exhibited high efficacy for treating G. pulchrum located
widely within the upper digestive tract, with a reduction of 98.2%. These results suggest
that this combined chemotherapy treatment may be effective against G.
pulchrum infection, including buccal and lingual gongylonemiasis in

The Asian seabass is euryhaline, therefore it is interesting to describe the infestation and survival of caligids at varying salinity on the host. In this study, two different brackish water culture systems with monoculture and polyculture practices were investigated for the occurrence of Caligus spp. on Lates calcarifer. Polyculture practices mainly consisted of snapper (Lutjanus spp.), grouper (Epinephelus spp.) and seabass (L. calcarifer), while the monoculture was stocked with only seabass. A total of 777 Caligus spp. specimens were isolated from the sampling in 2009, consisting of three species; Caligus chiastos, Caligus epidemicus and Caligus rotundigenitalis. In 2011, the total specimen was increased to 3110 and two additional species were found; Caligus punctatus and one unknown species (Caligus sp.). A 98.6% of the total examination was represented by C. epidemicus. Constant presence of C. epidemicus was observed throughout the study, regardless the differences in between culturing practices and systems. This species was able to survive within wide salinity range, from 5 to 28 ppt. The other isolated species (C. chiastos, C. punctatus, C. rotundigenitalis and Caligus sp.) were only found infesting in polyculture cages with the salinity ranging from 25 to 28 ppt. Despite accounts for less than 2% of the total specimens, these species may able to produce a challenge for L. calcarifer polyculture farming activity due to their capability for host switching. The present study revealed the potential risk for cross-species transmission in polyculture practices. 相似文献

Salmonella Typhimurium strain expressing the Actinobacillus
pleuropneumoniae antigens, ApxIA, ApxIIA, ApxIIIA and OmpA, was previously
constructed as a vaccine candidate for porcine pleuropneumonia. This strain was a live
attenuated (∆lon∆cpxR∆asd)Salmonella as a delivery host
and contained a vector containing asd. An immunological study of
lymphocyte proliferation, T-lymphocyte subsets and cytokines in the splenocytes of a mouse
model was carried out after stimulation with the candidate Salmonella
Typhimurium by intranasal inoculation. The splenic lymphocyte proliferation and the levels
of IL-4, IL-6 and IL-12 of the inoculated mice were significantly increased, and the T-
and B-cell populations were also elevated. Collectively, the candidate may efficiently

Between 2011 and 2012, a total of 896 pig fecal samples were collected from nine
provinces in Korea, and 50 salmonella enterica susp.
enterica serovar Typhimurium (S. Typhimurium) was
isolated. The characteristics of the 50 strains were analyzed, and 4 strains were
identified as Salmonella enterica subsp. enterica
serovar 4,[5],12:i:-. Salmonella 4,[5],12:i:- could not be distinguished
from S. Typhimurium through phage typing, antimicrobial resistance
testing or multiple-locus variable-number tandem repeat analysis (MLVA). However, among
the four Salmonella 4,[5],12:i:- strains, one (KVCC-BA1400078) was
identified as a Salmonella 4,[5],12:i:- clone isolated from humans in the
United States, and another (KVCC-BA1400080) was identified as DT193, which has been
primarily isolated from humans and animals in European countries. The presence of
Salmonella 4,[5],12:i:- in Korea poses a significant threat of

The nematode genus Baylisascaris (order Ascaridida, superfamily Ascaridoidea) contains nine relatively host-specific, parasite species of carnivores, omnivores, herbivores, carnivorous marsupials or rodents. They have a facultative heteroxenous life cycle, at least under experimental conditions. Eggs passed in faeces embryonate in the environment and the second-stage larva infective for both definitive and intermediate hosts develops. In intermediate hosts larvae migrate extensively through tissues, where they grow and moult to the third-stage, causing extensive damage. All Baylisascaris spp. are considered a potential cause of visceral, ocular and/or neural larval migrans in mammals including humans and in birds. This paper summarises our current knowledge on the prevalence, biology, pathogenicity and zoonotic significance of three Baylisascaris species: B. transfuga, B. schroederi and B. procyonis which have as definitive hosts bears, giant pandas and raccoons (occasionally dogs), respectively. 相似文献
