Processing tomato lines suppressed in the accumulation of ripening-related polygalacturonase or expansin were generated by introduction of transgenes to silence expression of the LePG and LeExp1 genes, respectively. The rheological properties of juice and juice reconstituted from paste produced from lines suppressed in one of these genes, or in both, were compared with azygous controls. When assayed by measuring Bostwick consistency, paste produced from either suppressed LePG or suppressed LeExp1 lines and diluted to 5 degrees Brix was approximately 18% more viscous than that produced from controls. Simultaneous suppression of LePG and LeExp1 produced a small additional increase in viscosity of 4%. Rheometric flow analysis at 5 or 10 degrees Brix also showed substantial increases in the consistency index due to suppression of either LePG or LeExp1 alone, and a small additional increase when both genes were suppressed in the same transgenic line. Measurements by laser diffraction and [1H]NMR showed that suppression of LePG or LeExp1 accumulation altered the size distribution of insoluble particles and modified their surface properties. The data are consistent with suppression of LePG increasing serum viscosity, and suppression of either LePG or LeExp1 altering the properties of the insoluble particles and improving some aspect of particle-particle or particle-serum interaction, or both. However, relative to that caused by suppression of either gene alone, the additional increase in viscosity caused by simultaneous suppression of LePG and LeExp1 together was slight.  相似文献

Suppression of the expression of a ripening-related expansin gene, LeExp1, in tomato enhanced fruit firmness and overexpression of LeExp1 resulted in increased fruit softening. Because of the incompletely understood relationship between fresh fruit texture and the consistency of processed products, we examined the effects of LeExp1 overexpression on the processing characteristics of tomato fruit. As determined by Bostwick consistency and by controlled strain rheometry, juices and pastes prepared from transgenic tomatoes with suppressed LeExp1 expression had a higher viscosity than preparations from control fruits. However, the viscosity of juice and paste prepared from fruit overexpressing LeExp1 was significantly greater than products from controls or lines with reduced LeExp1. Bostwick consistency increased by 9% (juice) and 6% (paste) in lines with suppressed LeExp1 expression but increased by 27.5% (juice) and 19.5% (paste) in lines overexpressing LeExp1, relative to controls. Determined by laser diffraction, the particles in juice and paste prepared from transgenic fruits with reduced LeExp1 expression were smaller, and preparations from fruits overexpressing LeExp1 had a size distribution indicating more large particles. Analysis of cell wall polysaccharides size indicated that LeExp1 overexpression enhanced depolymerization of water soluble pectins as well as tightly bound matrix glycans. LeExp1 overexpression may allow increased cell wall hydration, resulting in expanded particle size and increased viscosity of products. Because either LeExp1 suppression or overexpression leads to improved consistency, the interactions that contribute to optimal product rheological properties are complex.  相似文献

Processing effects on lycopene content and antioxidant activity of tomatoes   总被引:8,自引:0,他引:8  
Consumption of tomato products has been associated with decreased risk of some cancer types, and the tomato antioxidant, lycopene, is thought to play an important role in the observed health effects. In this study, four carotenoids, trans-lycopene, phytofluene, phytoene, and zeta-carotene, were quantified in tomato products. Samples of raw tomatoes, tomato juice after hot break scalder, and final paste were obtained from two different processing plants over two years. Comparison of carotenoid levels throughout processing indicated that lycopene losses during processing of tomatoes into final paste (25-30 degrees Brix) ranged from 9 to 28%. The initial Brix level of the raw tomatoes appeared to influence the amount of lycopene loss that occurred, possibly due to the differences in processing time required to achieve the final desired Brix level of the paste. In general, no consistent changes in the other carotenoids were observed as a function of processing. The antioxidant activity of fresh tomatoes, tomato paste, and three fractions obtained from these products (i.e., aqueous, methanol, and hexane fractions) was also determined. In both a free radical quenching assay and a singlet oxygen quenching assay, significant antioxidant activity was found in both the hexane fraction (containing lycopene) and the methanol fraction, which contained the phenolic antioxidants caffeic and chlorogenic acid. The results suggest that in addition to lycopene, polyphenols in tomatoes may also be important in conferring protective antioxidative effects.  相似文献

Commercial scale pulsed electric field processing of tomato juice   总被引:8,自引:0,他引:8  
Effects of commercial scale pulsed electric field (PEF) processing on the quality of tomato juice were studied and compared with those of thermal processing. Tomato juice was prepared by hot break at 88 degrees C for 2 min or by cold break at 68 degrees C for 2 min and then thermally processed at 92 degrees C for 90 s or PEF processed at 40 kV/cm for 57 micros. Thermally processed, PEF processed, and unprocessed control juices were packed into 50 mL sterilized polypropylene tubes in a sanitary glovebox and stored at 4 degrees C for 112 days. Both thermally and PEF processed juices showed microbial shelf life at 4 degrees C for 112 days. The lipoxygenase activities of thermally and PEF processed juices were 0 and 47%, respectively. PEF processed juice retained more ascorbic acid than thermally processed juice at 4 degrees C for 42 days (p < 0.05). No significant differences were observed in the concentration of lycopene, degrees Brix, pH, or viscosity between thermally and PEF processed juices during the storage (p > 0.05). Sensory evaluations indicated that flavor and overall acceptability of PEF processed juice were preferred to those of thermally processed juice (p < 0.05).  相似文献

Effects of pulsed electric fields (PEF) at 35 kV/cm for 59 micros on the quality of orange juice were investigated and compared with those of heat pasteurization at 94.6 degrees C for 30 s. The PEF treatment prevented the growth of microorganisms at 4, 22, and 37 degrees C for 112 days and inactivated 88% of pectin methyl esterase (PME) activity. The PEF-treated orange juice retained greater amounts of vitamin C and the five representative flavor compounds than the heat-pasteurized orange juice during storage at 4 degrees C (p < 0.05). The PEF-treated orange juice had lower browning index, higher whiteness (L), and higher hue angle (theta) values than the heat-pasteurized orange juice during storage at 4 degrees C (p < 0. 05). The PEF-treated orange juice had a smaller particle size than the heat-pasteurized orange juice (p < 0.05). degrees Brix and pH values were not significantly affected by processing methods (p > 0. 05).  相似文献

Sea buckthorn juice is one product that can be derived from the sea buckthorn berry, a new alternative crop for the Canadian western provinces. Fresh pressed juice separates into three phases when allowed to stand overnight in the refrigerator: an upper cream phase, juice in the middle portion, and a sediment at the bottom. Enzymatic hydrolysis with commercial, broad spectrum carbohydrate hydrolyzing enzyme preparations reduced the juice viscosity, assisted juice separation, and provided an opalescent juice. Soluble solids averaged 10.2 degrees Brix, pH averaged 3.13, ascorbic acid averaged 174.2 mg/100 mL, and titratable acidity averaged 1.97% as malic acid all determined on centrifuged (10 000 rpm, 15 min) juice. Soluble sugars included glucose, fructose, and an unidentified component that was not sucrose or other common soluble monomeric or dimeric sugar. Quinic acid was quantitatively most important, while malic was next, and oxalic, citric, and tartaric acids were minor components. Washing berries by dipping reduced soluble solids (degrees Brix) in juice suggesting uptake of wash water.  相似文献

An HPLC-PDA method for the determination of ethylenethiourea (ETU), the main degradation product of the organic fungicides ethylene bis(dithiocarbamate)s (EBDCs), in tomatoes and tomato products is reported. Solid-matrix liquid-liquid (l-l) partitioning and separatory funnel l-l partitioning for the cleanup were examined. The effect of salt addition, pH, and phase ratio on analyte recovery at the cleanup step was studied. It was found that solid-matrix l-l partitioning afforded higher precision and more selective separation of the analyte. According to the method proposed, the samples were extracted with methanol/water (3:1, v/v) and cleaned up on an Extrelut 20 column. ETU was eluted with dichloromethane and separated on a reversed phase HPLC column. For tomato products with degrees Brix > 20 further purification through silica cartridge was adopted. The method was validated over the following ranges of concentrations: 0.01-0.5 mg/kg for tomatoes, 0.01-0.1 mg/kg for tomato juice, and 0.05-0.25 mg/kg for tomato paste. The accuracy (recoveries > 70%) and the precision obtained (%RSD < 10%) were satisfactory.  相似文献

The effect of the addition of commercial pectin to orange juice that was de-esterified by alkaline treatment or pectinmethylesterase was evaluated. Pectin at various degrees of esterification (DE) was added to reconstituted frozen orange juice concentrate, and the extent of cloud loss was determined after centrifugation. Of the five pectin treatments, percent transmittance (% T) of juice remained <5%, except for the pectin treatment with a residual methoxyl content (% DE) of <5%. Addition of 100--600 ppm of 5% DE pectin to juice resulted in % T between 36 and 73%. Despite the absence of clarification, increases in cloud particle size were observed and depended on the % DE, the amount of pectin added, and the method used to modify the pectins.  相似文献

The rheological properties of tomato juice containing 1.5% soy germ were compared to plain tomato juice with and without soy protein isolate. This novel product was developed to provide a delivery system of carotenoids, soy protein, and significant isoflavone content without compromising the perceived juice characteristics of tomato product. Rheological tests depicted physical gel characteristics for all three products. Dynamic tests as a function of temperature showed that the stability and the compatibility between tomato juice and soy germ were higher as compared to soy protein isolate. The hydrophobic and electrostatic interactions between pectin and protein in the tomato soy protein isolate system were weakened as the temperature was increased. In the case of tomato juice with soy germ, the viscosity did not change during heating. The addition of soy germ increased the viscosity of tomato juice reinforcing the entire system without major qualitative effects on the rheological properties of plain tomato juice.  相似文献

Tomato products and especially concentrated tomato paste are important sources of antioxidants in the Mediterranean diet. Tomato fruit contain well-known antioxidants such as vitamin C, carotenoids, flavonoids, and hydroxycinnamic acids. The industrial processing of this fruit into tomato paste involves several treatments that potentially affect the final profile of antioxidants and other metabolites in the commercial product. Here we have used both biochemical and metabolomic techniques to assess the effect of each separate step in the industrial production chain starting from fresh fruit to the final tomato paste. Material was collected from five independent tomato paste production events spread over two successive years. Samples comprised the intact ripe fruits and semifinished products after fruit-breaking, separation of the pulp from skin and seeds, evaporation, and finally after canning and pasteurization. The effect of each processing step was determined by different types of analysis. First, the total antioxidant capacity and total phenolic content were determined by commonly used spectrophotometric methods. Second, individual antioxidants in the extracts were identified and compared using an HPLC with online antioxidant detection. Third, in each sample the levels of the major individual antioxidants present, i.e., vitamin C, phenolic compounds (such as rutin and chlorogenic acid), tocopherols, and carotenoids, were quantified. Fourth, an untargeted metabolomic approach using LC-QTOF-MS was used to identify those production steps that have the largest impact on the overall metabolic profile in the final paste as compared to the original fruits. This multifaceted approach has revealed that each processing step induces specific alterations in the metabolic profile, as determined by the different analysis procedures, and that in particular the fruit-breaking step and the removal of seed and skin significantly affect the levels of antioxidants and many other metabolites present in commercial tomato paste.  相似文献

The effect of vacuum infusion on eggplant quality of a commercial fungal (Aspergillus niger) and citrus pectinmethylesterase (PME) with calcium chloride (4000 ppm) was investigated after processing and during storage. Firmness of infused eggplants using fungal or citrus PME was significantly increased compared to controls (fresh noninfused and water-infused control) after processing and during storage for 7 days at 4 degrees C. Activity of fungal PME-infused eggplant increased almost 32 times, whereas activity of eggplant infused with Marsh grapefruit PME increased 2-fold. Degree of esterification of pectin of eggplants infused with fungal or citrus PME decreased slightly. Cryo-SEM showed that samples treated with fungal PME/ CaCl2 displayed more integrity among cells as compared with water-infused control. The change of pectin in the cell wall was visualized using monoclonal antibodies JIM5 (low-esterified pectin) and JIM7 (high-esterified pectin). JIM5 showed more binding than JIM7 with the cell walls of eggplant tissues from fungal PME/ CaCl2 treatment.  相似文献

为弄清橙汁加工过程中农药炔螨特残留的动态,通过田间喷施浓度为5倍于最高推荐剂量的农药溶液以强化炔螨特在甜橙上的残留,然后按照橙汁商业化加工过程进行加工,采用QuEChERS前处理技术结合气相色谱-串联质谱法检测炔螨特的含量,来考察橙汁商业化加工过程对炔螨特残留的影响。结果表明:炔螨特残留主要分布于甜橙果皮中,果肉中炔螨特的残留量不足全果的5%。清洗能除去全果中32.5%的炔螨特残留。初榨果汁、精滤果汁、非浓缩橙汁(NFC橙汁)和浓缩橙汁中的残留量分别为原料果的1.98%、1.95%、1.73%、1.37%,其中NFC橙汁和浓缩橙汁的加工因子分别为0.0173和0.0137,但炔螨特在果渣和精油中发生富集,加工因子分别为1.2822和18.4947。研究结果为橙汁加工工艺的优化和炔螨特残留的膳食暴露评估提供参考。  相似文献

The aim of this research was to evaluate the suitability of uracil as an hygienic quality index of tomato products. Whereas uridine was naturally present throughout tomato fruits' ripening, uracil appeared only after microbial contamination. In tomato pulp inoculated with nine different microbial strains, all five lactic acid bacteria (LAB) studied released relevant quantities of uracil (150-1040 mg/kg of dm), with a correlated partial or total decrease of uridine. Uracil production by yeasts and molds was very low or nonexistent; the starting uridine concentration (approximately 960 mg/kg of dm) remained constant or increased. Uracil thermostability was also verified. Twenty-six samples of tomato paste (30 degrees Brix) were collected from bag-in-drums produced in an industrial processing plant, some with evident swelling symptoms. All of the samples with high microbial count presented uracil. Uracil was also present in samples with microbial contamination under the detection limit and Howard mold count below legislation limits, implying the reprocessing, at least partial, of altered tomato product. The results indicate that uracil presence in tomato products is an index of LAB contamination that has occurred before heat treatment.  相似文献

Hand-squeezed navel orange juice contains 839 mg/L phenolics, including flavanones, flavones, and hydroxycinnamic acid derivatives. The flavanones are the main phenolics in the soluble fraction (648.6 mg/L) and are also present in the cloud fraction (104.8 mg/L). During refrigerated storage of fresh juice (4 degrees C), 50% of the soluble flavanones precipitate and integrate into the cloud fraction. Commercial orange juices contain only 81-200 mg/L soluble flavanones (15-33%) and the content in the cloud is higher (206-644 mg/L) (62-85%), showing that during industrial processing and storage the soluble flavanones precipitate and are included in the cloud. An in vitro simulation of orange juice digestion shows that a serving of fresh orange juice (240 mL) provides 9.7 mg of soluble hesperidin (4'-methoxy-3',5,7-trihydroxyflavanone-7-rutinoside) and 4.7 mg of the C-glycosylflavone vicenin 2 (apigenin, 6,8-di-C-glucoside) for freshly squeezed orange juice, whereas pasteurized commercial juices provide 3.7 mg of soluble hesperidin and a higher amount of vicenin 2 (6.3 mg). This means that although orange juice is a very rich source of flavanones, only a limited quantity is soluble, and this might affect availability for absorption (11-36% of the soluble flavanones, depending on the juice). The flavanones precipitated in the cloud are not available for absorption and are partly transformed to the corresponding chalcones during the pancreatin-bile digestion.  相似文献

The changes in volatile flavor components of guava juice during pressure processing (25 degrees C, 600 MPa, 15 min), heat processing (95 degrees C, 5 min), and storage at 4 and 25 degrees C were evaluated by purge and trap/gas chromatography/mass spectrometry. Esters were the major volatile fraction in guava juice, and alcohols were the second. Pressure processing could maintain the original flavor distribution of the juice. Heat processing (95 degrees C, 5 min) caused decreases in the majority of flavor components in the juice when compared with freshly extracted juice. High-pressure treatment at 600 MPa for 15 min can effectively sterilize microbes but partially inactivate enzymes of guava juice; therefore, volatile components in pressure-treated juice gradually changed during storage periods. Pressure-treated guava juice showed increases in methanol, ethanol, and 2-ethylfuran with decreases in the other components during storage period. Nevertheless, the volatile distribution of 600 MPa treated guava juice was similar to that of freshly extracted juice when stored at 4 degrees C for 30 days.  相似文献

The present work aimed to identify the key odorants of tomato sauces responsible for the flavor change during storage. Products made from paste or canned tomatoes were stored at 25 and 40 degrees C. Sensory properties and quantification of the key odorants were measured and correlated. Significant sensory changes appeared after 1 and 3 months at 25 degrees C in the respective dice and paste sauces (p < 0.01). The dice sauce was characterized by a steep loss of the sensory quality during the early storage and then by identical changes within the same time span at 25 and 40 degrees C. In the paste sauce the sensory deterioration was slower than for the dice sauce and occurred more extensively at 40 degrees C than at 25 degrees C. Correlation between sensory and instrumental data revealed that the source of sensory changes should be (E,E)-deca-2,4-dienal in the dice sauce. The sensory change in the paste sauce could be due to acetaldehyde, methylpropanal, 3-methylbutanal, oct-1-en-3-one, 3-methylbutanoic acid, deca-2,4-dienal, 2-methoxyphenol, and beta-damascenone.  相似文献

Even though several types of phosphorus fertilizers are used in crop production, the influence of phosphorus on produce quality is not well understood. Several quality attributes of tomato juice were analyzed in relation to phosphorus supplementation during a three-year field study (2000-2002). In addition to the recommended phosphorus fertilization, phosphorus supplementations, either through soil (low and high) or through foliar spray (hydrophos, seniphos), were tested. In general, soil and foliar phosphorus supplementation did not provide a statistically significant increase in yield. Tomato juice was evaluated for various quality characteristics including pH, titratable acidity, precipitate weight ratio, total solids, serum viscosity, Brookfield viscosity, color, lycopene levels, vitamin C, and flavor volatiles. Changes observed in several quality parameters were marginal, statistically insignificant and influenced by the season. Therefore, it appears that phosphorus supplementation may not significantly affect the processing quality parameters in tomato fruits.  相似文献

A methodology for flavor and composition assessment of processed tomato juice samples was developed using a wide range of commercial processing tomato varieties (Lycopersicon esculentum) grown in Spain and the United States. Fruitiness intensity was found by a trained panel to best describe overall tomato flavor. For two consecutive years, fruitiness intensity was significantly dependent on growing location and variety, and it was consistently linked to increased levels of glucose and reducing sugars and decreased glutamic acid content. Using the same procedure on a population of 176 breeding lines derived from the wild species of Lycopersicon pimpinellifolium, it was shown that tomato fruitiness intensity was significantly correlated to reducing sugars/glutamic acid ratio and glucose and glutamic acid contents. The definition of markers for tomato flavor of processed juice can provide the tomato breeder and processor with reliable analytical tools that can be applied in a straightforward way for the identification of raw materials that can be processed into juice with predictably high or low fruitiness.  相似文献

This study was conducted to investigate the antioxidant vitamin C, the polyphenol content, and the hydrophilic antioxidant capacity of tomato juice, baked tomatoes, tomato sauce, and tomato soup. During the production of tomato juice and during the preparation of the other tomato products, samples were taken after different times, respectively, after each particular production step. High-performance liquid chromatography was used to determine the content of vitamin C. The total phenolics content was analyzed spectrophotometrically by using the Folin-Ciocalteu method. The hydrophilic antioxidant capacity was measured by using three different methods: the Trolox equivalent antioxidant capacity assay, the ferric reducing antioxidant power test, and the photochemiluminescence assay. The vitamin C contents of the tomato products decreased during the thermal processing of tomatoes. In contrast, the total phenolics concentration and the water soluble antioxidant capacity increased.  相似文献
