DDT: interaction with nerve membrane conductance changes   总被引:5,自引:0,他引:5  
The falling phase of action potentials of lobster giant axons is prolonged by DDT; finally a plateau phase is produced like cardiac action potentials. In axons poisoned with DDT, peak transient (sodium) currents associated with step depolarizations are turned off very slowly, and steady-state (potassium) currents are markedly suppressed. These two changes would cause the prolongation of action potentials and are considered the major ionic mechanisms of DDT action.  相似文献

Acetylcholine is transported along insect chemoreceptor axons at a rate of 12 to 13 centimeters per day after peripheral uptake of choline. Colchicine, vinblastine sulfate, and cytochalasin B all block transport, but transport continues in axons separated surgically from the cell body. These data from an insect are in accord with many studies on vertebrates which have implicated intracellular microtubules in the transport mechanism. The peripheral uptake of choline and its acetylation and transport to nerve terminals in the brain are consistent withthe suggestion that acetylcholine is an antennal sensory transmitter in insects.  相似文献

Saxitoxin and tetrodotoxin: comparison of nerve blocking mechanism   总被引:4,自引:0,他引:4  
Saxitoxin at concentrations of 3 x 10(-8) to 3 x 10(-7) mole per liter blocks the conduction of lobster giant axon with no change in resting potential. Recovery of washed axons is faster in those that had been treated with saxitoxin than it is in those that were treated with tetrodotoxin. Peak transient increase in nerve membrane conductance is selectively blocked by saxitoxin with no change in late steady-state increase in conductance. The major mechanism of saxitoxin blockage is the same that of tetrodotoxin blockage.  相似文献

The nerve-impuilse-blocking actions of derivatives of tetrodotoxin have been tested on lobster and squid axons. The block produced by deoxytetrodotoxin was similar to that produced by tetrodotoxin and was probably caused by tetrodotoxin contamination. Tetrodaminotoxin and anhydrotetrodotoxin also produced a similar block but at such high concentrations that tetrodotoxin contamination cannot be ruled out. The hydroxyl group of C(4) and the hemilactal oxygen links play an important role for the nerve-blocking action.  相似文献

Tritiated norepinephrine was injected into the lateral ventricles of rats, and its localization in the hypothalamus was determined by light and electron-microscopic autoradiography. Eighty percent of the autoradiographic grains were located over nerve endings and unmyelinated axons. Large, dense synaptic vesicles were present in most of the endings and axons with activity. Grains were rarely seen over myelinated axons, glia, or blood vessels.  相似文献

Formation of myelin, the electrical insulation on axons produced by oligodendrocytes, is controlled by complex cell-cell signaling that regulates oligodendrocyte development and myelin formation on appropriate axons. If electrical activity could stimulate myelin induction, then neurodevelopment and the speed of information transmission through circuits could be modified by neural activity. We find that release of glutamate from synaptic vesicles along axons of mouse dorsal root ganglion neurons in culture promotes myelin induction by stimulating formation of cholesterol-rich signaling domains between oligodendrocytes and axons, and increasing local synthesis of the major protein in the myelin sheath, myelin basic protein, through Fyn kinase-dependent signaling. This axon-oligodendrocyte signaling would promote myelination of electrically active axons to regulate neural development and function according to environmental experience.  相似文献

Electron microscopical studies revealed the presence of dendritic nerve processes in sensory organs of the third- and fourth-stage juveniles of Haemonchus contortus, which contained structures resembling modified cilia. With few exceptions, the outer circle of fibers consisted of ten doublets, and in place of typical cilia-like central fibers were small microtubules or vesicles varying in number from zero to five.  相似文献

We present video-rate (28 frames per second) far-field optical imaging with a focal spot size of 62 nanometers in living cells. Fluorescently labeled synaptic vesicles inside the axons of cultured neurons were recorded with stimulated emission depletion (STED) microscopy in a 2.5-micrometer by 1.8-micrometer field of view. By reducing the cross-sectional area of the focal spot by about a factor of 18 below the diffraction limit (260 nanometers), STED allowed us to map and describe the vesicle mobility within the highly confined space of synaptic boutons. Although restricted within boutons, the vesicle movement was substantially faster in nonbouton areas, consistent with the observation that a sizable vesicle pool continuously transits through the axons. Our study demonstrates the emerging ability of optical microscopy to investigate intracellular physiological processes on the nanoscale in real time.  相似文献

Experiments with vesicles containing N-methyl-D-aspartate (NMDA) receptor 2B (NR2B subunit) show that they are transported along microtubules by KIF17, a neuron-specific molecular motor in neuronal dendrites. Selective transport is accomplished by direct interaction of the KIF17 tail with a PDZ domain of mLin-10 (Mint1/X11), which is a constituent of a large protein complex including mLin-2 (CASK), mLin-7 (MALS/Velis), and the NR2B subunit. This interaction, specific for a neurotransmitter receptor critically important for plasticity in the postsynaptic terminal, may be a regulatory point for synaptic plasticity and neuronal morphogenesis.  相似文献

In vitro pharmacological studies demonstrated that exogenously applied vasoactive intestinal polypeptide (VIP) relaxes the smooth muscle cells of cat cerebral arteries, whereas substance P constricts them. Ultrastructural-immunocytochemical techniques show that a VIP-like substance is present in the large granular vesicles of nonsympathetic nerve axons and terminals in the cerebral arterial walls. These results provide strong evidence in favor of the hypothesis that a VIP-like substance is the transmitter for vasodilation in cerebral blood vessels.  相似文献

本文研究了贻贝、牡蛎、杂虾、杂蟹、杂鱼和配合饲料6种饵料对中国龙虾的饲养效果。结果表明:饵料对中国龙虾的蜕壳率、蜕壳后体重增长率、生长率和饵料转换效率有显著的影响,而对中国龙虾蜕壳后体长增长率和存活率影响并不明显。各种试验饵料中翡翠贻贝是饲养中国龙虾的最好饵料,其次是牡蛎、杂虾、杂蟹、杂鱼只能作为辅助性饵料,配合饲料的某些成分及其适品性、诱食性尚需改善和提高。  相似文献

[目的]通过腹腔注射的致敏方式建立中国龙虾食物变态反应Balb/c小鼠模型,探讨中国龙虾食物变态反应体外鉴定与评价方法。[方法]将40只雄性Balb/c小鼠分为卵清蛋白(OVA)阳性对照组、Coca’s液阴性对照组、空白对照组和模型组,以OVA、中国龙虾粗提蛋白,加氢氧化铝佐剂腹腔注射免疫Balb/c小鼠,建立食物变态反应小鼠模型。ELISA法测定第二次致敏激发后血清中IgE与组胺水平并进行被动皮肤过敏试验(PCA)确定特异性IgE抗体滴度,同时观察脾指数、小肠组织学变化及激发后的食物变态反应症状。[结果]末次激发后1 h采血,中国龙虾粗提蛋白组血清IgE含量为236.75(±73.39)μg/L,与阳性对照OVA组无区别,与阴性对照Coca’s液组和正常对照组相比显著升高(P〈0.01);中国龙虾粗提蛋白组的组胺含量406.55(±232.79)μg/L与正常对照组相比显著升高(P〈0.01);PCA反应中国龙虾粗提蛋白组血清特异性IgE抗体滴度达到1/16;中国龙虾粗提蛋白组和OVA致敏组小鼠脾指数明显大于Coca’s液或正常对照组(P〈0.01)且前两组小肠粘膜固有层皆出现淋巴细胞、嗜酸性粒细胞等炎性细胞浸润。[结论]建立了一种中国龙虾食物变态反应小鼠模型,通过ELISA测定血清IgE和组胺以及PCA确定特异性IgE抗体滴度可作为一种鉴定与评价中国龙虾食物变态反应的方法。  相似文献

Hemocyanins are large multi-subunit copper proteins that transport oxygen in many arthropods and molluscs. Comparison of the amino acid sequence data for seven different subunits of arthropod hemocyanins from crustaceans and chelicerates shows many highly conserved residues and extensive regions of near identity. This correspondence can be matched closely with the three domain structure established by x-ray crystallography for spiny lobster hemocyanin. The degree of identity is particularly striking in the second domain of the subunit that contains the six histidines which ligate the two oxygen-binding copper atoms. The polypeptide architecture of spiny lobster hemocyanin appears to be the same in all arthropods. This structure must therefore be at least as old as the estimated time of divergence of crustaceans and chelicerates, about 540 to 600 million years ago.  相似文献

The plant cytoskeleton is a highly dynamic and versatile intracellular scaffold composed of microtubules and microfilaments, serving a multiplicity of functions in plant cells. To reveal the relationship between the cytoskeleton in wheat (Triticum aestivum L.) cv. Suwon 11 attacked by the non-host pathogen Sphaerotheca fuliginea and the initiation of the hypersensitive response, the microtubule inhibitor oryzalin was injected into the wheat leaves immediately prior to inoculation. The incidence of hypersensitive cell death was significantly lower than that in water-treated control. In addition, the occurrence of hypersensitive cell death was also delayed and S. fuliginea was able to penetrate and form haustoria in epidermal tissues of wheat. All the results above indicated that hypersensitive cell death was associated with depolymerisation of microtubules, suggesting that microtubules might play an important role in the expression of non-host resistance of wheat.  相似文献

Like the chelipeds, the claw closer muscles of the adult lobster are asymmetric (dipmorphic). In the crusher claw the closer muscle is composed entirely of slow fibers, and in the cutter claw it has 65 to 75 percent fast fibers and 25 to 35 percent slow fibers. While claw placement in the adult is essentially random, it can be demonstrated in two ways that the muscle fiber properties are not genetically fixed: (i) if one claw is removed in the fourth and early fifth stages, the remaining closer muscle develops all slow muscle fibers, and (ii) if the animals are raised in smooth-bottomed containers, both claws can become cutter types, having closer muscles with more than 50 percent fast fibers. Thus, as in vertebrate skeletal muscle, the properties of lobster closer muscle fibers can be transformed by various experimental manipulations.  相似文献

The asymmetric localization of messenger RNA (mRNA) and protein determinants plays an important role in the establishment of complex body plans. In Drosophila oocytes, the anterior localization of bicoid mRNA and the posterior localization of oskar mRNA are key events in establishing the anterior-posterior axis. Although the mechanisms that drive bicoid and oskar localization have been elusive, oocyte microtubules are known to be essential. Here we report that the plus end-directed microtubule motor kinesin I is required for the posterior localization of oskar mRNA and an associated protein, Staufen, but not for the anterior-posterior localization of other asymmetric factors. Thus, a complex containing oskar mRNA and Staufen may be transported along microtubules to the posterior pole by kinesin I.  相似文献
