为给油桐ALDH基因的结构与功能研究提供理论依据,以葡萄桐的近成熟种子为材料,根据油桐转录组测序结果设计引物,采用RT-PCR技术克隆了油桐ALDH22A1基因和ALDH2C4基因的全长cDNA序列。ALDH22A1的cDNA序列全长1 782 bp,编码593个氨基酸,该基因编码蛋白质的相对分子质量为65.60 kDa,理论等电点为6.62,蛋白二级结构以α螺旋为主,具有1个明显的跨膜结构,是稳定的非分泌蛋白。ALDH2C4的cDNA序列全长1 506 bp,编码501个氨基酸,该基因编码蛋白质的相对分子质量为54.82 kDa,理论等电点为6.58,蛋白二级结构以α螺旋为主,是不具有跨膜结构的膜外蛋白。BLASTp分析结果表明,这2个基因所编码序列与麻疯树、蓖麻的乙醛脱氢酶一致性最高,高达80%以上,含有醛脱氢酶基因家族的保守结构域。  相似文献

根据植物肌动蛋白(Actin)基因编码区的保守序列设计引物,以山茶属植物荔波连蕊茶幼嫩茎段为材料,提取总RNA,进行RT-PCR。采用RACE技术扩增获得1 631 bp的Actin基因全长cDNA序列,命名为ClActin1(GenBank登录号KF366912)。序列分析表明,ClActin1开放阅读框(ORF)为1 134 bp,编码377个氨基酸,5’非编码区90 bp,3’非编码区407 bp。预测的ClActin1蛋白分子量为41.69 kD,理论等电点为5.31,具有Actin超基因家族的保守结构域和肌动蛋白家族特有的特征信号序列。ClActin1与GenBank中收录的其它植物肌动蛋白核苷酸序列的相似性在82%以上,氨基酸序列的相似性在97%以上。与其它植物肌动蛋白比较并构建系统进化树,结果显示山茶肌动蛋白与茶树和杨树的肌动蛋白的亲缘关系最为密切。实时荧光定量PCR结果显示,该基因在荔波边蕊茶不同组织器官及不同发育时期的表达量稳定,表明ClActin1基因可作为内参基因。  相似文献

为了解LEC1(Leafy Cotyledon 1)基因对油桐油脂合成过程的调控机理,以油桐种仁转录组数据库中基因的部分c DNA序列为基础,采用RT-PCR技术从油桐种子中克隆LEC1基因的全长c DNA,并对其进行序列分析。油桐LEC1基因全长c DNA为1 152 bp,编码区为729 bp(142~870),编码243个氨基酸,5′端非编码区和3′端非编码区分别为141 bp和263 bp。该基因编码蛋白质的相对分子质量为27 025.4 Da,理论等电点为6.97;蛋白二级结构以不规则卷曲和α螺旋为主,是不具有跨膜结构的膜外蛋白。经对比发现,油桐LEC1具有典型的HAP3结构特点,在不保守的N端和C端中间有1个十分保守的结构域,与拟南芥、玉米、麻风树、黄连木等的LEC1氨基酸序列高度同源。  相似文献

在转录组测序结果分析基础上,以山鸡椒cDNA 为模板,克隆得到山鸡椒1-脱氧木酮糖-5-磷酸还原异构酶DXR 基因cDNA 全长,以山鸡椒基因组DNA 为模板,设计引物、扩增拼接后获得山鸡椒DXR 基因全长,命名为LcDXR。序列分析表明,LcDXR cDNA 全长为1 501 bp,5'非编码区长34 bp,3'非编码区长53 bp,开放阅读框长1 413 bp,预测编码含有470个氨基酸残基的蛋白质,等电点为6.62,分子量为51.12 kD。LcDXR 基因全长为12 601 bp,其中外显子12 个,内含子11 个。对来自10个种源的LcDXR 基因编码区单核苷酸变异位点进行分析表明:在cDNA 区间内共发现10 个SNP(single nucleotide polymorphism)位点,其中有4 个单核苷酸变异导致了所编码的氨基酸的改变,为了分析氨基酸突变导致的蛋白质精细结构的变化,利用Swiss-PDB Viewer 模拟4 个突变位点氨基酸残基的替换。其中江西安远(AY)的突变Lys119Thr 引起了氢键的变化,推测可能对酶的活性产生影响。研究结果为深入研究山鸡椒脱氧木酮糖5-磷酸还原异构酶的活性和功能奠定了基础,同时为山鸡椒遗传育种提供理论依据。  相似文献

以油茶近成熟种子为材料,根据油茶EST文库中已知的脂氢过氧化物裂解酶EST序列,利用3’RACE与5’RACE技术,克隆到脂氢过氧化物裂解酶的全长cDNA序列。该基因全长1648bp,,包含一个1476bp开放阅读框长,编码491个氨基酸,5’与3’非编码分别为52bp、121bp。预测该蛋白相对分子量为54.7779KDa,等电点为6.77,是个叶绿体转运肽,N端包含有22个疏水氨基酸残基组成的序列,不仅具有转运肽富含的丝氨酸,还含有大量转运肽中很少存在的脯氨酸。多序列比对发现油茶脂氢过氧化物裂解酶核苷酸序列与茶的相似性达到96%,因此将该基因命名coHPL。  相似文献

杜仲幼果和成熟果转录组测序的基础上,分离鉴定了幼果和成熟果MVA途径ACOT、HMGS、HMGR基因全长cDNA序列,并对基因的核苷酸序列及其相应氨基酸序列的组成成分、跨膜结构域、疏水性/亲水性、蛋白质二、三级结构、分子系统进化等进行分析。EuACOT基因全长cDNA为1 248 bp,编码416个氨基酸,属于稳定疏水性蛋白,无明显的跨膜区,在进化上分属于不同的分类群,与烟草和番茄的ACOT蛋白间的亲缘关系最为接近;EuHMGS基因全长cDNA为1 356 bp,编码452个氨基酸,属于不稳定的疏水性蛋白,无明显的跨膜区,在进化上分属于不同的分类群,与茶树的HMGS蛋白间的亲缘关系最为接近;EuHMGR基因全长cDNA为1 770 bp,编码590个氨基酸,属于不稳定的疏水性蛋白,有两个较明显的跨膜区,在进化上分属于不同的分类群,与杜仲的HMGR蛋白间的亲缘关系最为接近;以上三个蛋白的二级结构均是以α-螺旋和螺环结构为主的混合型结构。  相似文献

为了探明胼胝质对玫瑰授粉亲和性的影响,本研究以‘唐红’玫瑰花柱为试材,采用RT-PCR和RACE方法获得了玫瑰β-1,3-葡聚糖合成酶基因的cDNA全长,命名为RrCalS。该基因全长5742 bp,开放阅读框5295 bp,编码1764个氨基酸。推导编码蛋白的分子量为204.7kD, PI值为9.00,在164-265位具有pfam14288结构域,在869-1642位具有pfam02364保守结构域,属于葡聚糖合成酶超家族;该蛋白属于疏水性、非分泌型蛋白,具有16个跨膜结构域,含有32个Ser磷酸化位点、21个Thr磷酸化位点、12个Tyr磷酸化位点;该蛋白的α-螺旋占49.49%,无规则卷曲占22.68%,β-转角占7.94%;该蛋白与Fragaria vesca等8种植物的Cals氨基酸序列同源性达73%以上,且它们的系统进化关系与传统分类结果一致。本研究为进一步深入研究玫瑰授粉不亲和机理,提高玫瑰育种理论和技术水平奠定了基础。  相似文献

α-桐酸合成酶是控制亚油酸向α-桐酸(18:3Δ9cis,11trans,13trans)转化的关键酶。本研究以发育中的油桐种子为材料,利用改良TRIzoL法提取总RNA,并根据GenBank中已经登录的α-桐酸合成酶基因FADX的序列,设计特异性引物,采用RT-PCR方法,克隆得到FADX基因全长cDNA,长度为1221bp。通过生物信息学分析结果表明,该序列5’端和3’端非编码区序列长度分别为13bp和47bp,含有一个开放阅读框(14~1174bp),编码386个氨基酸,含有典型的脂肪酸脱氢酶结构域。将所得基因经BamHI和SacI酶切后插入表达载体质粒pBI121,构建反义表达载体pBI121fadx。  相似文献

以油茶‘湘林1号’品种的近成熟种子为材料,采用简并PCR、RACE、交错PCR等技术,获得了油茶乙酰辅酶A羧化酶BC亚基基因的全长cDNA克隆。该基因cDNA序列全长1 901 bp,含有一个1 599 bp的ORF,编码533个氨基酸残基。BC蛋白的等电点pI为6.88,分子量为58 509.3 u,含两个跨膜结构域,是一个不稳定的非分泌蛋白。模体搜索结果表明在BC上具有N-糖基化位点、蛋白激酶C磷酸化位点、ATP结合位点等多个功能结构域。同源建模的模型中发现12个α-螺旋,整个BC蛋白为一个内凹的结构。该基因已登录到GenBank,并被命名为co-bc。  相似文献

为研究A功能基因在山茶花重瓣形成过程中所发挥的功能,利用同源克隆和RACE扩增方法,从重瓣山茶花品种‘金盘荔枝’发育早期的花芽中获得了山茶花AP基因全长cDNA,命名为CjAPL1,GenBank登录号JX657332。该基因全长1 149 bp,其中,开放阅读框(ORF)长度为741 bp,5’非编码区长度206 bp,3’非编码区长度202 bp。经氨基酸序列分析表明:CjAPL1基因编码一条含有246个氨基酸的蛋白质,与猕猴桃、八仙花等植物的Ap1蛋白同源性均在75%以上。Rea-time PCR结果显示,CjAPL1基因在‘金盘荔枝’不同发育时期花芽中的表达量为:花芽发育早III期>花芽发育早II期>花芽发育早I期>现蕾期,表达趋势表现为先逐渐升高而后急剧降低;花器官不同部位中的表达量为花柱最高,其次为子房和萼片,在雄蕊下部和外轮花上部中的表达量最低。这些差异表明,CjAPL1基因可能在‘金盘荔枝’重瓣花形成中发挥作用。  相似文献

APETALA2(AP2)基因在植物生长发育过程中发挥着重要作用.利用RT-PCR和RACE方法,从毛竹中克隆到1个AP2同源基因的全长cDNA序列,命名为PeAP2.序列分析表明:PeAP2基因全长1 750 bp,其中,5′端非编码区106bp,3′端非编码区174 bp,开放阅读框1 470 bp,编码1个489 aa的蛋白,该蛋白含有2个AP2结构域,属于AP2/EREBP家族的AP2亚家族.PeAP2蛋白与来自其它单子叶植物的AP2蛋白均有着较高同源性,其中,与二穗短柄草的AP2蛋白同源性最高,达74.85%.实时定量PCR分析显示:PeAP2基因在毛竹的根、茎、叶、鞘和节5种器官中均有表达,其中,叶片中的表达丰度最高,鞘中次之,而在根、茎、节中的表达丰度接近,均较低.利用hiTAIL-PCR方法克隆获得了PeAP2上游启动子区序列1 359 bp,分析显示其含有光、激素等多种信号应答相关的作用元件.  相似文献

根据平榛花芽转录本高通量测序的结果,采用RACE-PCR技术克隆到一个全长675 bp,编码179个氨基酸的平榛WRKY基因,命名为ChWRKY2。ChWRKY2蛋白序列中只含有一个WRKY结构域,锌指结构为C-X4-C-X23-H-X1-H,属于WRKY家族第Ⅱ类成员。对ChWRKY2基因进行实时荧光定量PCR表达分析,结果表明:平榛雌花芽中ChWRKY2在12月份的表达量最高,随后表达量逐渐下降。4℃低温胁迫处理根蘖苗4 h后,叶片中ChWRKY2的表达量快速升高,并在8 h达到最大表达量。此外,ChWRKY2在不同器官中的表达具有差异性,雄花序中表达量最高,其次是雌花芽,树皮(韧皮部)中表达量最低。  相似文献

Agrobacteriurn tumefaciens-mediated transformation (AtMT) method was used to study the genetic transformation in Rhizopus arrhizus of antisense expression vector pBI121-fad2 of VeFAD2 gene. We studied the key factors influencing transformation, such as A.tumefaciens strains,bacterial cell volume initially used, co-cultivation time, effective period and concentration of AS. The results shows that the ideal A.tumefaciens strain is AGL-1, the optimum bacterial cell volume initially used is 50-100 μL and co-culture 24 hours will get the most transformants. At co-culture period, AS induction is indispensable. It is beneficial to improve transformation ratio by adding to 200 μmol·L-1 AS at preincubate period and increasing AS concentration to 400-600 μmol·L-1 at co-culture period. It determined that antisense VeFAD2 gene has been integrated into the genome of R. arrhizus by PCR-Southern detection.  相似文献

Five aromatic plants, Carum carvi (caraway), Apium graveolens (celery), Foeniculum vulgare (fennel), Zanthoxylum limonella (mullilam) and Curcuma zedoaria (zedoary) were selected for investigating larvicidal potential against mosquito vectors. Two laboratory-reared mosquito species, Anopheles dirus, the major malaria vector in Thailand, and Aedes aegypti, the main vector of dengue and dengue hemorrhagic fever in urban areas, were used. All of the volatile oils exerted significant larvicidal activity against the two mosquito species after 24-h exposure. Essential oil from mullilam was the most effective against the larvae of A. aegypti, while A. dirus larvae showed the highest susceptibility to zedoary oil.  相似文献

Two new 2-aryl benzofuran derivatives, 3-[7-methoxy-2-(3,4-methylenedioxyphenyl) benzofuran-5-yl]propyl 3-[7-methoxy-2-(3,4-methylenedioxyphenyl)benzofuran-5-yl]propanoate (1) and demethoxy egonol gentiobioside (2), were isolated from the aqueous ethanolic extract of the seeds of Styrax macranthus, together with 7-methoxy-2-(3,4-methylenedioxyphenyl)benzofuran-5-carbaldehyde, egonol, demethoxy egonol, demethyl egonol, egonol glucoside, egonol gentiobioside, egonol gentiotrioside, stigmasterol, 2,3-dihydroxypropyl tetracosoate, and daucosterol. The Eastern Black Sea Region has an extensive flora because of ample rainfall lasting all year. In this study, antimicrobial effects of 74 crude extracts of 22 endemic plants were investigated. Among the 30 active crude extracts, water-insoluble crude extracts from Betula medwediewii, Heracleum platytaenium, Primula longipes, Anthemis cretica ssp. argaea and Centaurea helenioides were the prominent ones with their MIC values.  相似文献

从落叶松梢栖真菌筛选出对落叶松枯梢病病原菌有抑制作用的菌株,对落叶松枯梢病进行生物防治。将自落叶松梢部分离到的11种梢栖真菌与落叶松枯梢病病原菌进行对峙培养,根据被覆盖程度、抑菌率、拮抗系数和防治预试验的结果,综合评价后发现:粪生粪壳、深绿木霉和球毛壳3种梢栖真菌对落叶松枯梢病有良好的拮抗作用。利用这3个菌株在黑龙江省勃利县通天一林场国富沟进行林间防治试验,结果表明:防治效果为粪生粪壳>代森锰锌400倍液>深绿木霉>球毛壳;深绿木霉和粪生粪壳喷洒浓度50%和球毛壳喷洒浓度75%时,真菌多样性最多,梢栖真菌系统较其他浓度更为稳定,与化学防治相比,既达到了防治目的又具有不污染环境的优势。  相似文献

Efforts in Europe to convert Norway spruce (Picea abies) plantations to broadleaf or mixed broadleaf-conifer forests could be bolstered by an increased understanding of how artificial regeneration acclimates and functions under a range of Norway spruce stand conditions. We studied foliage characteristics and leaf-level photosynthesis on 7-year-old European beech (Fagus sylvatica) and pedunculate oak (Quercus robur) regeneration established in open patches and shelterwoods of a partially harvested Norway spruce plantation in southwestern Sweden. Both species exhibited morphological plasticity at the leaf level by developing leaf blades in patches with an average mass per unit area (LMA) 54% greater than of those in shelterwoods, and at the plant level by maintaining a leaf area ratio (LAR) in shelterwoods that was 78% greater than in patches. However, we observed interspecific differences in photosynthetic capacity relative to spruce canopy openness. Photosynthetic capacity (A1600, net photosynthesis at a photosynthetic photon flux density of 1600 μmol photons m−2 s−1) of beech in respect to the canopy gradient was best related to leaf mass, and declined substantially with increasing canopy openness primarily because leaf nitrogen (N) in this species decreased about 0.9 mg g−1 with each 10% rise in canopy openness. In contrast, A1600 of oak showed a weak response to mass-based N, and furthermore the percentage of N remained constant in oak leaf tissues across the canopy gradient. Therefore, oak photosynthetic capacity along the canopy gradient was best related to leaf area, and increased as the spruce canopy thinned primarily because LMA rose 8.6 g m−2 for each 10% increase in canopy openness. These findings support the premise that spruce stand structure regulates photosynthetic capacity of beech through processes that determine N status of this species; leaf N (mass basis) was greatest under relatively closed spruce canopies where leaves apparently acclimate by enhancing light harvesting mechanisms. Spruce stand structure regulates photosynthetic capacity of oak through processes that control LMA; LMA was greatest under open spruce canopies of high light availability where leaves apparently acclimate by enhancing CO2 fixation mechanisms.  相似文献

A screening was conducted with 26 plants collected in the Brazilian southeast region, to identify plant extracts with antibacterial properties against Aeromonas hydrophila, Bacillus subtilis, Pseudomonas aeruginosa and Staphylococcus aureus. Initially, the agar diffusion method was employed. Then, those extracts presenting activity were submitted to a broth microdilution assay to determine the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). It was observed that 13 of the tested extracts showed antibacterial activity. The best results were obtained with those from Lantana lilacina and Phyllanthus tenellus.  相似文献
