为了探究肌肉生长抑制素(myostatin,MSTN)基因在肌肉和脂肪中的DNA甲基化模式及mRNA表达水平,试验以5月龄巴什拜羊羔羊为研究对象,采用亚硫酸氢盐测序法(bisulfite sequencing PCR,BSP)检测MSTN基因启动子区和第1外显子甲基化模式,并通过实时荧光定量PCR检测MSTN基因在巴什拜羊股二头肌、股三头肌、半腱肌、半膜肌、背最长肌和尾脂中的mRNA表达水平。结果显示,肌肉组织甲基化概率高于脂肪组织,其中,股二头肌、股三头肌、半腱肌、半膜肌、背最长肌和尾脂的甲基化概率分别为74.2%、74.2%、83.2%、83.7%、82.1%和25.3%,MSTN基因在股二头肌、股三头肌、半腱肌、半膜肌、背最长肌中的表达水平显著低于尾脂(P < 0.05),股二头肌、股三头肌、半腱肌、半膜肌和背最长肌之间差异不显著(P > 0.05),巴什拜羊肌肉和脂肪MSTN基因DNA甲基化水平与MSTN基因表达量呈显著负相关(r=-0.886,P < 0.05)。  相似文献

本研究以2个骨骼肌表型差异明显的特克塞尔和乌珠穆沁绵羊为试验对象,研究DLK1和MSTN基因在不同妊娠阶段胎儿不同部位骨骼肌中表达的调控机制及早期肌肉发育规律。在绵羊妊娠第85、100、120和135天时,对胎儿的半腱肌、半膜肌、背最长肌、股四头肌和股二头肌肌肉质量进行方差分析以及对DLK1和MSTN基因在5种骨骼肌中的表达量进行研究。结果表明,妊娠85天时特克塞尔羊的半膜肌、背最长肌、股四头肌和股二头肌质量显著或极显著(P<0.05或P<0.01)高于乌珠穆沁羊;妊娠100天时,5种骨骼肌组织生长发育在品种间的差异达到最大(P<0.05或P<0.01)。实时荧光定量分析表明,随着胎儿日龄的增加,DLK1基因在半腱肌、半膜肌、背最长肌、股四头肌和股二头肌骨骼肌中的表达量有增加的趋势,同时在特克塞尔羊中的表达量高于乌珠穆沁绵羊。在妊娠120天的半腱肌和半膜肌、妊娠100和135天的背最长肌、妊娠85天股四头肌和妊娠135天的股二头肌中DLK1表达量在2个品种间差异显著(P<0.05)。MSTN的表达量在特克塞尔羊中高于乌珠穆沁羊,但整体较低。妊娠135天时背最长肌和股四头肌中MSTN基因的表达量在2个品种间差异显著(P<0.05)。结果显示,DLK1和MSTN基因在不同日龄不同肌肉组织中的差异表达可能与肌肉早期发育有关。  相似文献

本研究以育肥的小尾寒羊和寒泊羊的羔羊为试验动物,采集背最长肌、臂三头肌、股二头肌和臀中肌样品,测定了肌纤维横截面积、直径、粗脂肪含量和氨基酸种类及含量。结果表明,寒泊羊背最长肌肌纤维直径和横截面积均小于小尾寒羊(P<0.01),股二头肌肌纤维直径和横截面积大于小尾寒羊 (P<0.05)。寒泊羊背最长肌和股二头肌粗脂肪含量高于小尾寒羊(P<0.01,P<0.05),臂三头肌和臀中肌粗脂肪含量略高于小尾寒羊(P>0.05)。小尾寒羊肉中牛磺酸含量高于寒泊羊(P<0.01),其余17种氨基酸含量两品种羊肉间差异不显著(P>0.05)。与肉鲜味有关的5种氨基酸,寒泊羊肌肉的含量高于小尾寒羊。除赖氨酸和色氨酸外,其余6种人体必需氨基酸寒泊羊肌肉的含量高于小尾寒羊。  相似文献

The objectives of this experiment were to determine the extent of variation in proteolysis, sarcomere length, and collagen content among pork muscles and the association of those factors with tenderness variation among muscles at 1 d postmortem. Twenty-three white composite barrows were slaughtered and carcasses (66 kg) were chilled at 0 degrees C for 24 h. At 1 d postmortem, the longissimus lumborum, biceps femoris, semimembranosus, semitendinosus, and triceps brachii, long head were dissected from one side of each carcass and frozen. Trained sensory panelists evaluated tenderness, amount of connective tissue, juiciness, and pork flavor intensity of grilled (70 degrees C) chops on 8-point scales. Raw chops were used for total collagen content, sarcomere length, and the extent of desmin proteolysis. Tenderness ratings were highest (P < .05) for semitendinosus (7.2) and triceps brachii (7.1), followed by longissimus lumborum (6.4) and semimembranosus (5.7) and were lowest (P < .05) for biceps femorus (4.0). The simple correlations between longissimus lumborum tenderness and the tenderness of other muscles were .54 (semimembranosus), .34 (semitendinosus), .36 (triceps branchii), and .17 (biceps femorus). Total collagen was highest (P < .05) for biceps femorus (7.1 mg/g muscle), followed by triceps branchii (6.0 mg/g) and semitendinosus (5.3 mg/g), and lowest for semimembranosus (4.5 mg/g) and longissimus lumborum (4.1 mg/g). Sarcomere length was longest (P < .05) for semitendinosus (2.5 microm) and triceps branchii (2.4 microm), followed by semimembranosus (1.8 microm), longissimus lumborum (1.8 microm), and biceps femorus (1.7 microm). Proteolysis of desmin was greatest (P < .05) in longissimus lumborum (39.3%), followed by semimembranosus (21.0%) and biceps femoris (18.5%), then semitendinosus (.2%) and triceps brachii (.2%). Multiple linear regression using total collagen, sarcomere length, and proteolysis accounted for 57% of the variation in tenderness rating among all samples. Piecewise linear regression was used to account for the interaction of sarcomere length with proteolysis and collagen. This analysis accounted for 72% of the variation in tenderness rating. Variation in collagen, proteolysis, and sarcomere length and the degree of their interaction with one another determine the tenderness of individual muscles.  相似文献

In order to investigate skeletal muscle development mechanism,the expression of follistatin (FST) and fibromodulin (FMOD) gene were comparatively analyzed in fetus different skeletal muscles and different stages between Ujumqin sheep and Texel sheep during early stage.Two gene expression were analyzed including longissimusdorsi,semimembranosus,semitendinosus quadricepsfemoris and biceps femoris muscle were compared by variance analysis methods of four stages (85,100,120 and 135 d).The results of quantitative Real-time PCR showed that the expression of FST gene showed a tendency to increase with the addition of stage,and the expression of FST gene in Texel sheep was higher than that in Ujumqin sheep in semitendinosus (100,120 d),semimembranosus (120 d),and biceps femoris (120 d) (P<0.05).The expression of FST gene in Texel sheep was extremly higher than that of Ujumqin sheep in semitendinosus (100,135 d),longissimusdorsi (85 d),quadricepsfemoris (100 d) and biceps femoris (100 d) (P<0.01).As a whole,the expression of FMOD gene was the highest in quadriceps femoris,and was lower in other four muscles,it's expression in Ujumqin sheep was higher than that in Texel sheep.It was inferred that FST and FMOD genes might be related to muscle development during early stage.  相似文献

本研究以骨骼肌表型差异明显的两个绵羊品种特克塞尔羊和乌珠穆沁羊为试验对象,通过对两个与生长发育相关的基因卵泡抑素(follistatin,FST)与纤维蛋白聚糖基因(fibromodulin,FMOD)在不同日龄、不同骨骼肌部位表达的比较分析,探讨绵羊骨骼肌早期发育规律;同时对绵羊胎儿的85、100、120和135日龄4个阶段,在背最长肌、半腱肌、股四头肌、半膜肌和股二头肌5个部位肌肉FSTFMOD基因的表达量进行了比较研究。实时荧光定量PCR结果表明,随着胎儿日龄的增加,FST基因在半腱肌、股四头肌中表达量有增加的趋势,同时在特克塞尔绵羊中的表达量高于乌珠穆沁羊。在100和120日龄半腱肌、120日龄半膜肌、120日龄股二头肌中FST基因表达量在两个品种间差异显著(P<0.05),在100和135日龄半膜肌、85日龄背最长肌、100日龄股四头肌、100日龄股二头肌中FST基因表达量在两个品种间差异极显著(P<0.01)。FMOD基因在股四头肌中表达量较高,而在其他4种肌肉组织中相对表达量较低,且在乌珠穆沁羊中的表达量高于特克塞尔羊中的表达量。FSTFMOD基因在绵羊早期骨骼肌发育中起着重要作用。  相似文献

【目的】 对藏羊肌肉生长抑制素(myostatin,MSTN)基因进行克隆和生物信息学分析,检测其在藏羊不同组织中的表达,为探究MSTN基因在藏羊中的生物学功能提供参考。【方法】 以藏羊背最长肌组织cDNA为模板,克隆藏羊MSTN基因完整CDS区序列并测序,用SeqMan程序对测序结果进行拼接,并用BLAST在线程序对组装后的序列进行分析鉴定。用生物信息学软件进行相似性比对、系统进化树构建及生物信息学分析,用实时荧光定量PCR检测MSTN基因在藏羊不同组织中的表达量。【结果】 藏羊MSTN基因CDS全长为1 128 bp,编码375个氨基酸。藏羊MSTN基因氨基酸序列与绵羊、牦牛、牛、猪、恒河猴、人、黑猩猩、犬、鸡及斑马的相似性依次为100.0%、93.4%、93.4%、95.2%、94.4%、94.2%、94.4%、93.1%、87.8%和87.5%;系统进化树分析结果表明,藏羊与绵羊的亲缘关系最近,与斑马和鸡的亲缘关系最远。藏羊MSTN蛋白属于亲水性分泌蛋白,且具有不稳定性,不含跨膜结构,含1个信号肽,存在31个潜在的磷酸化位点、2个N-糖基化修饰位点,主要分布在线粒体和细胞质中;MSTN蛋白二级结构以无规卷曲为主,其次是α-螺旋、延伸链和β-转角;三级结构预测结果与二级结构一致。实时荧光定量PCR结果显示,MSTN基因在藏羊不同组织中均有表达,其中在臂三头肌和半腱肌的表达量显著高于肺脏、下丘脑、心脏、肝脏、十二指肠、瘤胃和肾脏(P<0.05)。【结论】 成功克隆了藏羊MSTN基因;该基因在藏羊肌肉中的表达量高于内脏组织。该结果为进一步研究MSTN基因对藏羊肌肉生长发育的调控机制奠定了基础。  相似文献

为了研究湖羊肌肉营养特点及肌纤维组织学特性,分别测定了不同年龄(初生、断奶、成年)湖羊肌肉中常规营养成分(水分、粗蛋白质、粗脂肪、粗灰分、钙、磷)、18种氨基酸的含量以及不同年龄湖羊不同部位肌肉(臂三头肌、股二头肌、背最长肌)的肌纤维组织学特性(肌纤维直径、肌纤维密度)。结果表明:随着年龄的增长,肌肉中水分含量显著降低(P0.05),粗脂肪、粗蛋白质、粗灰分、钙和磷的含量逐渐增加,且成年羊与初生羔羊和断奶羔羊之间差异显著(P0.05)。湖羊肌肉中苏氨酸、缬氨酸、蛋氨酸、异亮氨酸、亮氨酸、苯丙氨酸、赖氨酸、组氨酸和精氨酸的含量随着年龄的增长而显著增加(P0.05),非必需氨基酸含量随着年龄的增长大致呈增加趋势。在湖羊肌肉的所有氨基酸中含量最高的是谷氨酸,其次是天冬氨酸。不同年龄湖羊的肌纤维直径均表现为背最长肌股二头肌臂三头肌,肌纤维密度均表现为背最长肌股二头肌臂三头肌,且同龄湖羊3个部位之间的肌纤维密度均差异显著(P0.05)。相同部位的肌肉,其肌纤维直径随年龄的增长而增大,肌纤维密度随年龄的增长而减小,且不同年龄间差异显著(初生羔羊与断奶羔羊的臂三头肌除外)(P0.05)。综上可知,湖羊肌肉营养丰富,氨基酸种类齐全,肌纤维组织学特性在不同年龄和不同部位间存在差异,断奶羔羊肉营养更丰富,更具香、嫩等特点。  相似文献

试验旨在克隆山羊CMKLR1基因序列并进行生物信息学分析,研究其表达与肌内脂肪(IMF)沉积的相关性。采集山羊皮下脂肪组织,应用RT-PCR克隆山羊CMKLR1基因序列;以GAPDH(GenBank登录号:AJ431207.1)为内参基因,实时荧光定量PCR检测其在山羊不同组织中的表达量及在肌肉组织中的时序表达谱(以肝脏表达量为基准);测定肌肉组织中的IMF含量,分析其与CMKLR1 mRNA表达量的相关性。结果显示,克隆获得了山羊CMKLR1基因,1 089 bp的CDS区,GenBank登录号:KT165374。实时荧光定量PCR分析显示,CMKLR1基因在24月龄山羊心脏、肝脏、脾脏、肺脏、肾脏、脂肪、背最长肌、股二头肌和臂三头肌中均存在不同程度的表达,其中在肺脏中表达量显著高于其他组织(P< 0.05);CMKLR1基因在1~3和24月龄的山羊背最长肌、臂三头肌和股二头肌中表达趋势相同,均是在股二头肌中表达量最高,而8~10月龄则是在臂三头肌中表达量最高。山羊IMF含量以24月龄背最长肌含量最高。山羊背最长肌、股二头肌和臂三头肌中CMKLR1 mRNA表达与IMF含量相关性不显著(P> 0.05)。CMKLR1基因不参与山羊IMF沉积作用,试验结果为进一步研究CMKLR1基因奠定理论基础。  相似文献

In order to enrich basic date in goat (Capra hirus) CMKLR1 gene and investigate the correlation between CMKLR1 gene expression and intramuscular fat (IMF) content in muscles, the CMKLR1 gene was cloned from the goat of subcutaneous adipose tissue by RT-PCR, characterized by bioinformatics methods. The expression profiles of CMKLR1 gene of goat in various tissues were constructed liver was benchmark, the GAPDH (GenBank accession No.: AJ431207.1) as a reference gene.Then, the correlation between CMKLR1 mRNA expression and IMF content in muscles was analyzed. The results showed that the CMKLR1 gene CDS of goat (GenBank accession No.: KT165374) was 1 089 bp. Real-time PCR indicated that CMKLR1 was widely expressed in various tissues, including heart, liver, spleen, lung, kidney, adipose, longissimus dorsi, biceps femoris and triceps brachii, and was the highest in lung (P< 0.05). Similar expression variation trend of CMKLR1 was observed between the muscles from 1 to 3 and 24 months old goat, and was the highest in biceps femoris. Reversely, in 8 to 10 months old CMKLR1 was the highest in triceps brachii. The IMF of longissimus dorsi from 24 months old was the highest. Correlation analysis demonstrated that different correlations were observed between expression of CMKLR1 mRNA and IMF content in longissimus dorsi, biceps femoris, and triceps brachii in goat. The CMKLR1 gene did not participate in the deposition of IMF in goats. The research built the theoretical basis for further studies about the CMKLR1 gene.  相似文献

[目的]研究MS删基因在不同月龄滩羊不同肌肉组织中的表达差异。[方法]根据GenBank已发表的绵羊MSTN基因序列(NM001009428.1)设计l对引物,以滩羊肌肉组织cDNA为模板,采用实时荧光定量PCR技术(Real—timePCR)检测MS7Yv基因在不同月龄滩羊的背最长肌、腿肌和胸肌中的表达量。[结果]MS驯基因在背最长肌中的表达量最高,其次为胸肌,腿肌中的表达量最低;随着月龄的增加,滩羊3种肌肉组织中的表达量均呈先升高、后降低、再升高的趋势。[结论]不同月龄滩羊肌肉中的MSTNmRNA表达量存在差异.这些差异可能会影响其生长发育和产肉性能等指标。  相似文献

Left sides from 18 beef carcasses (9 steers and 9 heifers), selected to represent a wide range of marbling scores, were evaluated to determine the relationship between longissimus composition and the composition of other major muscles. The adductor (A), biceps femoris (BF), deep pectoral (DP) gluteal group (GL), infraspinatus (I), longissimus (L), psoas major (PM), rectus abdominis (RA), rectus femoris (RF), semimembranosus (SM), semitendinosus (ST), serratus ventralis (SV), spinalis (SP), supraspinatus (SU) and triceps brachii (TB) were removed, trimmed of external fat, weighed and ground for proximate analysis. Fat content of all muscles was related linearly (P less than .001) to L fat content (R2 values ranged from .67 to .84). The ST had the lowest mean fat content (4.4%) and SP had the highest mean percentage of fat (16.1%). The L ranged from 3.59% to 15.42% fat with a mean of 8.61%. Longissimus fat percentage can be used to predict the fat content of the other major muscles of the beef carcass.  相似文献

本研究旨在探讨胰岛素样生长因子结合蛋白1(Insulin-like growth factor-binding protein 1,IGFBP-1)基因在山羊出生后不同组织的发育性表达模式.在出生后0、15、30、60、90和120 d共屠宰36只南江黄羊羔羊(公、母羔羊各18只),取心脏、肝脏、脾脏、肺脏、背最长肌、半膜肌、臂三头肌和股二头肌样品以抽提RNA,进行克隆及荧光定量PCR分析.山羊IGFBP-1基因的ORF(Open reading frame)长792 bp,共编码263个氨基酸,其核苷酸和氨基酸序列相似性在反刍动物(牛、绵羊)之间远大于其它物种.羔羊肝脏IGFBP-1 mRNA表达量最高(P<0.01);肺脏、脾脏、心脏表达水平中等;肌肉中最低且在4种肌肉间表达差异不显著(P>0.05).出生后尤其是60 d内羔羊IGFBP-1mRNA丰度变化很大,且明显呈现持续下降(肝脏)、先升后降(心脏)以及下降-上升-下降(脾脏、肺脏和肌肉)3种模式的变化.结果提示,IGFBP-1基因CDS区在物种间保守性较强,肝脏是山羊合成IGFBP-1 mRNA的主要部位,IGFBP-1基因在出生后羔羊的早期生长中发挥着重要的作用且其mRNA发育性表达模式具有组织器官特异性.  相似文献

Forty USDA Select and 40 upper two-thirds USDA Choice beef carcasses were used to determine the effects of postmortem aging on tenderness of 17 individual beef muscles. Biceps femoris-long head, complexus, gluteus medius, infraspinatus, longissimus dorsi, psoas major, rectus femoris, semimembranosus, semitendinosus, serratus ventralis, spinalis dorsi, supraspinatus, tensor fasciae latae, teres major, triceps brachii-long head, vastus lateralis, and vastus medialis muscles were removed from each carcass. Seven steaks (2.54-cm thick) were cut from every muscle, and each steak was assigned to one of the following postmortem aging periods: 2, 4, 6, 10, 14, 21, or 28 d postmortem. After completion of the designated aging period, steaks were removed from storage (2 degrees C, never frozen), cooked to a peak internal temperature of 71 degrees C, and evaluated using Warner-Bratzler shear force (WBSF). Analysis of WBSF revealed a 3-way interaction (P = 0.004) among individual muscle, USDA quality grade, and postmortem aging period. With the exception of the Select teres major, WBSF of all muscles (both quality grades) decreased with increasing time of postmortem storage. Nonlinear regression was used to characterize the extent (aging response) and rate of decrease in WBSF from 2 through 28 d postmortem for each muscle within each quality grade. In general, WBSF of upper two-thirds Choice muscles decreased more rapidly from 2 to 10 d postmortem than did corresponding Select muscles. Muscles that had greater aging responses generally had greater 2-d WBSF values. The upper two-thirds Choice psoas major, serratus ventralis, and vastus lateralis muscles required similar aging times to complete a majority of the aging response (< or =0.1 kg of aging response remaining) compared with analogous Select muscles. The upper two-thirds Choice complexus, gluteus medius, semitendinosus, triceps brachii-long head, and vastus medialis muscles required 4 to 6 d less time to complete a majority of the aging response than did comparable Select muscles. Aging times for Select biceps femoris-long head, infraspinatus, longissimus dorsi, rectus femoris, semimembranosus, spinalis dorsi, supraspinatus, and tensor fasciae latae muscles were > or =7 d longer than those for corresponding upper two-thirds Choice muscles. Results from this study suggest that muscle-to-muscle tenderness differences depend on quality grade and aging time and that postmortem aging should be managed with respect to individual muscle and USDA quality grade.  相似文献

A split-plot design was used to study effects of male status (bull vs steer), zeranol implants, breed types (Angus vs Limousin) and electrical stimulation on palatability traits of five major muscles when all animals were fed to a similar fat-thickness endpoint. Warner-Bratzler shear (WBS) determinations and taste-panel evaluations were performed on the longissimus, adductor, semimembranosus, semitendinosus and biceps femoris muscles removed 24 h postmortem. Breed was the most consistent main effect difference, with Angus cattle having lower WBS values than Limousin for the longissimus and semitendinosus muscles. Angus cattle also were favored in many sensory traits. Few significant differences were noted between bulls and steers in taste-panel traits. Differences due to electrical stimulation were most evident in the longissimus muscle, with higher panel-evaluation scores, an enhancement of marbling and lean color and a lower incidence of heat ring. The implant tended to have negative effects on palatability in this trial. An overall ranking of the five muscles based on WBS from most tender to least tender was: adductor, semimembranosus, longissimus, semitendinosus and biceps femoris. Correlations of palatability traits of one muscle to other muscles within an experimental unit were found to be low.  相似文献

为探究饲养方式对滩羊羔羊肌肉和脂肪组织中脂肪酸组成的影响,试验选择24只健康且体重相近(14.16 kg±0.58 kg)的2月龄滩羊羔羊随机分成舍饲组和放牧组,每组12只,于4月龄屠宰取其背最长肌、股二头肌、膈肌、皮下脂肪、肾周脂肪、尾部脂肪,用气相色谱仪测定脂肪酸含量。结果表明:①在肌肉组织饱和脂肪酸中,与放牧组相比,舍饲组滩羊羔羊股二头肌丁酸、棕榈酸含量显著提高(P<0.05),膈肌辛酸、月桂酸、肉豆蔻酸含量显著降低(P<0.05)。在肌肉组织不饱和脂肪酸中,与放牧组比较,舍饲组滩羊羔羊背最长肌棕榈油酸、花生四烯酸含量显著降低(P<0.05),γ-亚麻酸、α-亚麻酸含量显著提高(P<0.05),二十碳五烯酸(EPA)、n-3多不饱和脂肪酸(n-3 PUFA)极显著提高(P<0.01);股二头肌花生四烯酸含量显著降低(P<0.05),十七碳一烯酸、α-亚麻酸、EPA、n-3 PUFA含量极显著提高(P<0.01);膈肌棕榈油酸含量显著降低(P<0.05)。②在脂肪组织饱和脂肪酸中,与放牧组比较,舍饲滩羊羔羊皮下脂肪十一碳酸、十五碳酸含量显著降低(P<0.05),十三碳酸含量极显著降低(P<0.01);肾周脂肪葵酸、肉豆蔻酸含量显著降低(P<0.05),丁酸、硬脂酸含量极显著提高(P<0.01);尾脂十一碳酸含量显著降低(P<0.05)。在脂肪组织不饱和脂肪酸中,与放牧组比较,舍饲滩羊羔羊皮下脂肪十八碳二烯酸含量极显著提高(P<0.01),肉豆蔻油酸、棕榈油酸、γ-亚麻酸含量极显著降低(P<0.01);肾周脂肪十八碳一烯酸、α-亚麻酸、n-3PUFA含量显著提高(P<0.05),肉豆蔻油酸含量显著降低(P<0.05),十七碳一烯酸、二十碳二烯酸、十八碳二烯酸含量极显著提高(P<0.01);尾脂肉豆蔻油酸、棕榈油酸含量显著降低(P<0.05),十八碳二烯酸含量极显著提高(P<0.01)。综上,不同饲养方式对滩羊羔羊肌肉和脂肪组织脂肪酸含量存在一定影响,舍饲滩羊羔羊股二头肌中饱和脂肪酸含量及肾周脂肪中不饱和脂肪酸含量显著高于放牧滩羊羔羊。  相似文献

为了明确云岭牛不同解剖部位肉的品质特性,本试验测定了云岭牛冈上肌(SU)、冈下肌(IF)、臀三头肌(TB)、菱形肌(RH)、颈腹侧锯肌(SVC)、夹肌(SP),中部胴体的腰大肌(PM)、背最长肌(LD)、背阔肌(LA),后部胴体的半膜肌(SM)、半腱肌(ST)、股二头肌(BF)、臀中肌(GM)、股直肌(RF)、股外侧肌(VL)和阔筋膜张肌(TFL)共16个解剖部位肉的压力失水率、蒸煮损失、剪切力、pH、亮度(L*)、红度(a*)、黄度(b*)、粗蛋白质、脂肪、水分10项品质指标,并通过方差分析、相关性分析和标准化分析研究其品质特性。结果显示,云岭牛不同解剖部位肉之间的10项品质指标均存在显著差异(P<0.05)。冈上肌、冈下肌、腰大肌、颈腹侧锯肌和阔筋膜张肌的嫩度较好,其剪切力均低于4 kg。夹肌、半腱肌和股外侧肌的保水性较差,半腱肌的L*值最高。相关性分析结果表明,剪切力和压力失水率、蒸煮损失呈显著正相关。标准化分析发现,背最长肌和半腱肌具有相似的品质特征;股二头肌、臀中肌和股外侧肌具有相似的品质特征。结果表明,部位因素对云岭牛肉品质具有显著影响,云岭牛胴体前部肉品质较好,可以作为开发高档产品的原料来源。  相似文献

Activities of mu- and m-calpain and of calpastatin were measured at four different times during postmortem storage (0, 1, 3, and 10 d) in three muscles from either callipyge or noncallipyge (normal) sheep. The weights of two muscles, the biceps femoris and the longissimus, are greater in the callipyge phenotype, whereas the weight of the infraspinatus is not affected. The activity of m-calpain was greater (P < 0.05) in the biceps femoris and longissimus from callipyge than in those from normal sheep, but it was the same in the infraspinatus in the two phenotypes. The extractable activity of m-calpain did not change (biceps femoris and infraspinatus) or decreased slightly (longissimus) during postmortem storage. Extractable activity of mu-calpain decreased to zero or nearly zero after 10 d postmortem in all muscles from both groups of sheep. The rate of decrease in mu-calpain activity was the same in muscles from the callipyge and normal sheep. At all time points during postmortem storage, calpastatin activity was greater (P < 0.05) in the biceps femoris and longissimus from the callipyge than from the normal sheep, but it was the same in the infraspinatus from callipyge and normal sheep. Calpastatin activity decreased (P < 0.05) in all three muscles from both phenotypes during postmortem storage; the rate of this decrease in the callipyge biceps femoris and longissimus and in the infraspinatus from both the callipyge and normal sheep was slow, especially after the first 24 h postmortem, whereas calpastatin activity in the biceps femoris and longissimus from the normal sheep decreased rapidly. During postmortem storage, the 125-kDa calpastatin polypeptide was degraded, but the 80-kDa subunit of mu-calpain was cleaved only to 76- and 78-kDa polypeptides even though extractable mu-calpain activity declined nearly to zero. Approximately 50 to 60% of total mu-calpain became associated with the nonextractable pellet after 1 d postmortem. The myofibril fragmentation index for the biceps femoris and longissimus from normal sheep increased significantly during postmortem storage. The fragmentation index for the infraspinatus from the callipyge and normal sheep increased to an intermediate extent, whereas the index for the biceps femoris and longissimus from the callipyge did not change during 10-d postmortem storage. The results suggest that postmortem tenderization is related to the rate of calpastatin degradation in postmortem muscle and that calpastatin inhibition of the calpains in postmortem muscle is modulated in some as yet unknown manner.  相似文献

为了研究滩羊和小尾寒羊不同部位脂肪酸组成的差异,试验对滩羊和小尾寒羊股二头肌、背最长肌和臂三头肌3个部位31种脂肪酸的停留时间和浓度进行了对比分析。结果表明,31种脂肪酸在滩羊和小尾寒羊不同部位羊肉中的停留时间均为差异不显著(P>0.05),但脂肪酸停留时间随着脂肪酸中碳含量的增加而增加;脂肪酸C16:0、C18:0、C18:2n6c在滩羊和小尾寒羊不同部位羊肉中的浓度均高于其他脂肪酸,脂肪酸C17:0在滩羊和小尾寒羊不同部位羊肉中的浓度呈现极显著差异(P<0.01)。  相似文献

DNA甲基化作为真核生物基因组重要的表观遗传学修饰,对生物体基因的表达有重要的调控作用。为了研究五指山猪肌肉组织基因组DNA的甲基化程度,试验利用MSAP技术,使用筛选的10对引物扩增,检测五指山猪背最长肌和半腱肌的甲基化程度。结果显示:1月龄、2月龄、4月龄五指山猪背最长肌的平均甲基化率分别是44.78%、41.58%、38.92%;1月龄、2月龄、4月龄五指山猪半腱肌的平均甲基化率分别是41.70%、39.39%、38.81%。研究结果为五指山猪生长发育规律、系统选育及矮小机制等方面的研究提供表观遗传学依据。  相似文献
