丁内酯-I对绵羊卵母细胞体外成熟及发育能力的影响

摘    要:研究从丁内酯-I(BL-I)的浓度筛选、作用时间以及作用解除后的成熟培养时间等方面分别进行了试验,以观察其对绵羊卵母细胞核成熟的抑制程度以及对卵母细胞体外成熟及发育能力的影响。结果表明:150μmol/L的BL-I能将62.9%的卵母细胞抑制在GV期。BL-I抑制8、16 h和24 h后绵羊卵母细胞停滞在GV期的比率差异不显著(P>0.05);抑制24 h时卵裂率和囊胚率低于对照组,16 h的囊胚率也低于对照组,抑制8 h的卵裂率和囊胚率与对照组差异不显著(P>0.05)。去除抑制后卵母细胞的成熟能力不受影响。由此可得出,丁内酯-I能可逆性抑制绵羊卵母细胞的核成熟,但未能提高其发育潜力。

Abstract:Butyrolactone(BL) I,a selective inhibitor of cyclin dependent kinases which can competitive bind p34cdc2,inhibits meiotic resumption.In this experiment,the effect of butyrolactone I with different concentration,inhibition durations and removal of inhibition for different durations on maturation in vitro and developmental competence of ovine oocytes was assessed.Results showed that GVBD was prevented in 62.9% of the oocytes after 24 h of culture with 150 μmol/L BL-I.There were no significant differences in the GV rates among inhibited 8,16 and 24 h groups;the cleavage and blastocyst rates of inhibited 24 h group was lower than that of the control,but there were no significant differences between the inhibited 8 h group and the control.Mature competence was not affected,but the proceeding of nuclear maturation was accelerated following the withdrawal of BL-I.We can draw a conclusion that BL-I can reversibly inhibit IVM of ovine oocytes;ovine oocytes cultured with BL-I were not enhanced developmental competence.
