| 摘 要： | 利用根癌农杆菌介导法对仙客来叶片进行遗传转化,研究影响仙客来转化的若干因素,建立一套高效的遗传转化体系.结果表明:2 d的预培养,OD600值约为0.8的工程菌液浓度,侵染时间15 min,4 d的共培养,延迟筛选10 d有利于获得较高的转化效率.PCR检测表明,APX基因已整合到仙客来基因组中,共获得4株转基因植株,转化率为9.3%.

| Abstract: | In order to establish an efficent genetic transformation system with Cyclamen perslcum leaves,several factors affected genetic transformation mediated by Agrobacterlum tumefacien were studied.The results showed that highest transformation frequency was obtained when the leaves was precultivated 2 days,the ODeoo of Agrobacterlum was about 0.8,the Agrobacterlum infected 15 min,4 days coculture,and 10 days of detention screening.Results of PCR proved that the APX gene had integrated into Cyclamen perslcum genome,and four transgenic lines were obtained,the transformation rate is 9.3%. |
