荷叶中生物碱的分离纯化及脂肪酶活性抑制作用研究

摘    要:荷叶中含有生物碱等多种有效成份,同时具有药食两用的功效.本文研究了荷叶提取物的分离和纯化条件与荷叶乙醇提取物对脂肪酶活性的抑制作用,发现荷叶中各生物碱的含量为0.1~0.6%,经大孔树脂预纯化后,生物碱的含量提高近100倍.当流动相中三乙胺含量增加至0.2%时,峰形明显改善,各色谱峰重叠部分小,高纯度收集部分加宽;1 mg/ml荷叶提取物对脂肪酶的抑制作用为11.25%,荷叶碱的抑制活性为25.77%,N-降荷叶碱的抑制活性为21.37%,O-降荷叶碱的抑制活性为24.63%.

Abstract:Lotus leaves contained multiple useful components such as alkaloid, have edible and drug double effects in the mean time. The conditions for separation and purification of lotus leaf extracts and the inhibition effects of the ethanol extracts of lotus leaves on lipase activity were studied in the paper. The results showed that the content of all kinds of alkaloid in lotus leaves is within 0.1-0.6% and it can be enhanced to approximately 100 times after pre-purification with the macroporous absorption resin. When the content of triethylamine in mobile phase increased to 0.2 %, the ehromatogram speak shape was distinctly improved with little peak overlap and the sample collection part was stretched. The inhibition effects of lmg/ml lotus leaf extracts on lipase activity was 11.25%, and the inhibition effects of nuciferine, N-nornuciferine and O-nornuciferin were 25.77%, 21.37% and 24.63% respectively.
