蚕蛹蛋白双酶法水解工艺优化

摘    要:研究了碱性蛋白酶和中性蛋白酶双酶法水解蚕蛹蛋白的工艺条件.在单因素试验的基础上,以水解度为考察指标,研究温度、脱脂蚕蛹粉浓度、水解时间、加酶量、酶质量比(碱性蛋白酶:中性蛋白酶)对蚕蛹蛋白水解效果的影响,通过正交试验优化水解工艺条件.结果表明,优化的工艺条件为脱脂蚕蛹粉浓度30 g/L,水解时间6h(其中碱性蛋白酶的水解时间为4.5 h,中性蛋白酶的为1.5 h),加酶量3%,温度55℃,酶质量比3:1,碱性蛋白酶处理时pH9.0、中性蛋白酶处理时pH7.5.在此工艺条件下蚕蛹蛋白水解度可达22.99%.

Abstract:The technology for silkworm pupa protein hydrolysis by double enzymatic(alkaline protease and neutral protease) was studied.The effects of temperature,concentration of skim silkworm powder,time,enzyme dosage and mass ratio of alkaline protease and neutral protease on the degree of hydrolysis were studied by single factor tests.And the hydrolytic conditions were optimized through orthogonal design.The results showed that the optimal process conditions were,concentration of skim silkworm powder,30 g/L;time,6 h(alkaline protease 4.5 h,neutral protease 1.5 h);amount of enzyme,3%;temperature,55℃;and malkaline protease ∶ mneutral protease,3∶1;pH for alkaline protease,9.0 and for neutral protease,pH 7.5.The hydrolysis degree of silkworm pupa protein under these conditions could be 22.99%.
