大孔树脂法纯化红花芸豆色素及初步鉴定

摘    要:该文通过静态吸附和解吸实验,选择了合适的大孔树脂纯化红花芸豆色素,优化了吸附和解吸条件,并应用纸色谱法和紫外—可见光谱法对纯化后的色素进行了初步的分离鉴定。结果表明,HPD400型大孔树脂对pH值为1的红花芸豆色素酸提液吸附率为88.38%,pH值2的75%乙醇可较好的洗脱色素。纯化后色素有2个主要成分,初步鉴定为天竺葵定-3-葡萄糖苷和芍药定-3-葡萄糖苷。

Abstract:In order to determine the optimum macroporous resin and condition for purifying pigments extracted from red speckled kidney bean(Phaseolus vulgaris L.), the adsorption and desorption properties of five different resins were studied. Also, paper chromatography and ultraviolet-visible spectrum methods were applied to identify the pigments purified by macroporous resins. Results show that macroporous resin HPD400 is favorable in purification of the kidney bean pigments: adsorption rate of pigments(solvent pH 1) to HPD400 resin is 88.38%. 75% ethanol(pH 2) is suitable for elution. And, there are two main components of the purified pigments, which are primarily identified as pelargonidin -3-glucoside and peonidin-3-glucoside.
