Abstract:The aim of this study was to investigate the effect of HOXC8 DNA methylation during the differentiation of ovine preadipocytes.In this study,ovine subcutaneous preadipocytes were used as research material.The CpG islands and methylation levels in the promoter and the first exon sequence of HOXC8 were analyzed by bioinformatics software and bisulfite sequencing PCR(BSP).The expression of HOXC8 mRNA was detected by qPCR.The results showed that there was a CpG island in the promoter region and the first exon of HOXC8 respectively.The expression level of HOXC8 before differentiation was extremely significantly higher than after differentiation in ovine preadipocyte(P<0.01).Between day 0 and day 2 of ovine preadipocyte differentiation,no significant difference of methylation level was revealed in the promoter,but there was significant difference in the methylation level of the first exon(P<0.05).The methylation level of HOXC8 first exon was extremely significantly higher than that of the promoter region(P<0.001).According to the linear regression analysis of the methylation level of HOXC8 first exon and its mRNA expression,there was a high positive correlation between them(r=0.994,P<0.0001).HOXC8 may play an important role in ovine preadipocyte before differentiation.The methylation of the first exon of HOXC8 positively regulates its expression at transcription level during the differentiation of ovine preadipocytes.
