五味子叶枯病菌拮抗放线菌A-25-8的鉴定及发酵条件优化

摘    要:经五味子根际土中分离获得1株高效拮抗五味子叶枯病菌的放线菌A-25-8,对其进行鉴定并研究其最佳发酵条件。根据菌株的形态与培养特征、生理生化特性、16SrDNA序列分析对其进行鉴定,应用牛津杯法研究A-25-8菌株抑菌活性及最优发酵条件。A-25-8菌株与Streptomyces anulatus(环圈链霉菌)的亲缘关系接近;最佳发酵条件:发酵培养基为马铃薯葡萄糖液体培养基,碳源为葡萄糖,氮源为KNO_3,发酵培养基初始pH为7.5,发酵温度为28℃,发酵时间为3 d,接种量为10%,摇床转速为160 r·min~(-1)。拮抗放线菌A-25-8鉴定为环圈链霉菌(Streptomyces anulatus),优化后A-25-8无菌发酵滤液对五味子叶枯病菌显示出更强的抑菌活性。

Abstract:An actinomycetes strain A-25-8 with strong antifungal activity against leaf blight pathogens of Schisandra chinensis was isolated from the rhizosphere soil of Schisandra chinensis.Its fermentation conditions were optimized.A-25-8 was identified based on its 16S rRNA sequence,together with the morphological,physiological and biochemical characteristics.The antagonistic activity of strain A-25-8 and its optimal fermentation conditions were also investigated by Oxford plate method.As a result,A-25-8 appeared to have a close relation to Streptomyces anulatus.The optimum fermentation conditions for strain A-25-8 were as follows:PD culture medium;glucose for carbon source;KNO3 for nitrogen source;pH at 7.5;temperature at 28℃;culture time at 3 d;inoculation amount of 10% (V/V) and rotational speed of 160 rpm.In conclusion,the antagonistic actinomycetes A-25-8 was identified as Streptomyces anulatus.A-25-8 strain shows stronger antagonistic activities against leaf blight pathogens of Schisandra chinensis under optimized fermentation conditions.
