穿山薯蓣不定芽的诱导与植株再生

摘    要:以穿山薯蓣带腋芽茎段为外植体进行离体培养,通过正交试验筛选出最佳的诱导和增殖培养基,利用单因子试验筛选出最佳生根培养基。结果表明,培养基MS BA2.0mg/L NAA0.5mg/L为不定芽诱导的最佳培养基,诱导率为100%;培养基MS BA2.0mg/L NAA0.1mg/L最有利于植株再生,增殖倍数达4.9;培养基1/2MS NAA0.4mg/L对穿龙薯蓣试管苗生根培养较适合,生根率达86.7%;在塑料拱棚中利用蛭石:泥炭土:珍珠岩=3:3:1为移栽基质,移栽成活率可达85%以上。

Abstract:The adventitious bud of Discorea nipponica as explantswere cultured in vitro.The most suitable media for induction and multiplication were selected respectively through orthogonal experiments.The most suitable medium for root generation was also selected through unifactor experiment. The results showed that the MS+BA2.0mg/L+NAA0.5mg/L medium was most suitable for the induction of adventitious bud with the induction rate of 100%.The MS+BA2.0mg/L+NAA0.1mg/L medium was most suitable for plantlet regeneration with multiplication rate of 4.9.The 1/2MS+NAA0.4mg/L medium was most suitable for root generation with rooting rate of 86.7%. In the plastic arch shed, the seedlings were transplanted on substrate composing of vermiculite, turf soil and perlitein proportion of 3:3:1,and the survive rate of transplant seedlings was more than 85%.
