GM-CSF与SS融合表达质粒对小鼠淋巴细胞增殖及GH和IGF-1分泌的影响

摘    要:以小白鼠为试验对象,分别肌肉注射生理盐水及pcS/2SS、pGM-CSF/SS、pGM-CSF pcS/2SS质粒,每只50 μg,免疫后第3周以相同剂量加强免疫1次,测定不同质粒对小鼠淋巴细胞增殖反应及血浆生长激素(GH)、胰岛素样生长因子-1 (IGF-1)分泌水平,探讨GM-CSF对生长抑素DNA疫苗的免疫增强作用.结果表明: GM-CSF可增强小鼠脾脏淋巴细胞的增殖反应,以pGM-CSF pcS/2SS组的增殖能力最强,显著高于pcS/2SS组和生理盐水组;同时注射2种表达质粒(pGM-CSF pcS/2SS)的GH分泌水平和融合表达质粒(pGM-CSF/SS)的IGF-1分泌水平明显高于生理盐水组和pcS/2SS免疫组.这证明GM-CSF对生长抑素DNA疫苗有一定的免疫增强作用,可促进生长抑素DNA疫苗的免疫反应,提高血浆GH和IGF-1的分泌水平.

Abstract:A total of 40 mice at the age of 22 days was divided into 4 groups to study the immuno-enhancement of genetic adjuvant GM-CSF on somatostatin(SS)DNA vaccine.Ten mice in each group were given two intramuscular injections at interval of 2 weeks with saline solution as control(group 1),pcS/2SS(group 2),pGM-CSF/SS(group 3),and pGM-CSF+pcS/2SS(group 4),respectively.The results showed that lymphocyte proliferation response of pcS/2SS along with pGM-CSF was higher than that of the others,indicating that GM-CSF pos...
