百岁老人肠道双歧杆菌分离鉴定研究

摘    要:为了建立百岁老人肠道双歧杆菌高效分离的技术体系,首先筛选了合理的大便样品采样液,以确保双歧杆菌的数量和活性;其次将杂菌抑制技术和显色技术相结合,快速得到显色菌株,并在初步鉴定的基础上采用16S rRNA对显色菌株进行了鉴定.结果 表明,使用双歧杆菌BS培养基作为采样液能够更有效地分离双歧杆菌,百岁老人的肠道菌相较于婴儿更难分离;使用X-Gal作为指示剂在粪便中分离双歧杆菌的过程中,不只有双歧杆菌菌株显色,得出了显色菌株的大致种类,为肠道益生菌的分离鉴定提供理论和技术支持.

Abstract:In order to establish a technical system for efficient separation of Bifidobacteria from the intestinal tract of centenarians,this study firstly screened reasonable stool sample samples to ensure the quantity and activity of Bifidobacteria.Secondly,the chromogenic strain was rapidly obtained by combining heterozygous inhibition technology with chromogenic technology,and 16S rRNA was used to identify the chromogenic strain on the basis of preliminary identification.The results showed that the Bifidobacteria BS medium was used as a sampling medium to isolate bifidobacterium more effectively.Intestinal bacteria of centenarians were more complex and difficult to isolate than those of infants.In the process of separating Bifidobacteria in feces with X-Gal as an indicator,not only Bifidobacteria strains developed color,but also the general species of color-developing strains was obtained.The results of this study would provide theoretical and technical support for the isolation and identification of intestinal probiotics.
