酸处理的沙门氏菌作为异种细菌抗原的免疫载体

用酸对鼠伤寒沙门氏菌进行处理,制成裸细菌,作为空肠弯曲菌共同抗原(CA)的载体.用这种裸细菌和CA的复合物免疫小鼠,以间接ELISA和琼脂糖凝胶扩散沉淀反应检测免疫反应的变化及血清效价.结果表明,用这种方法免疫的小鼠,其血清抗体的ELISA滴度与对照组(福氏完全佐剂+CA组,CA组)相差不显著,但能与可溶性CA形成沉淀线,并含有IgM和IgG两类抗体;而对照组则不与CA出现沉淀反应,且只含IgM一类抗体.这表明,酸处理的沙门氏菌作为异种细菌抗原的载体,在改变机体对某种抗原的免疫类型上具有一定的作用.

Acid-treated Salmonella typhimurium as Immune Carrier for Het- erologous Bacterial Antigens

Immune carrier for heterologous bacterial antigen was prepared from S. ty-phimurium with an acid treatment procedure. The common antigens (CA) of Campylobacter jejuni were conjugated to the acid-treated bacteria (naked bacteria) and injection of the complex into mice was done for determining its antigenicity. The antibody responses were assayed with indirect ELISA and agarose gel diffusion test. The experimental results indicated that there were no significant differences in anti-CA antibody titer among the sera from the animals immunized with naked bacteria-CA complex, CA plus adjuvant and CA alone; the first one antiserum reveared precipitation line with CA in agarose gel diffusion, whereas the other two immune sera did not; besides, IgG and IgM classes of antibody were found in the serum against carrier-CA complex, only IgG, however, in those of the other two control groups. Accordingly, it suggested that acid-treated bacteria as immune carrier could be of potential for altering the pattern of immune response to some antigens.
