摘    要:目的]研究反相高效液相色谱法同时测定金银花中的绿原酸和木犀草苷的优化色谱条件.方法]采用反相高效液相色谱法(RP-HPLC)同时测定金银花中绿原酸和木犀草苷含量,并对色谱条件进行优化.结果]最佳色谱条件为色谱柱Waters SunFireTMC18(4.6mm×150 mm,3.5 μm)、柱温40℃,流动相为乙腈-0.5%冰醋酸,梯度洗脱为0~15 min(10:90~20:80)、16 ~ 17 min(20:80 ~ 10:90),流速1.0 ml/min,检测波长0~15 min 328 nm、16 ~ 17 min 350 nm.结论]优化后缩短了测试时间,为金银花质量的快速检测提供参考依据.

Optimization of the Determination Method of the Chlorogenic Acid and Luteolin-7-O-glucoside in Flos Lonicerae by RP-HPLC

Abstract:The research aimed to study the optimal chromatographic conditions of the deter mination method of the ehlorogenie acid and luteolin-7-O-glueoside in flos lonieerae by RP-HPLC. Method] A RP-HPLC method with UV detector was established and optimized to de- ter mine the ehlorogenie acid and luteolin-7-O-glueoside in Flos Lonieerae. Result ] The chromatographic conditions were: Waters SunFireT- MCI8 (4.6 mmx 250 mm, 3.5 p,m) used for column( at 40 ~C ) with 0.5% acetic - acetonitrile. Linear gradient elution: 0 - 15 rain( 10:90 - 20 : 80), 16 - 17 min(20:80 ~ 10: 90), flow rate :1 ml/min, deterction wavelength:0 - 15 rain 328 nm, 16 - 17 rain 350 nm. Conclusion] After optimization, the test time was shortened. The study provides the reference for deter ruination method of rapid detection of flos lonicerae quality.
