微波辅助提取桔子皮中多糖的研究

摘    要:[目的]为桔子皮的综合利用提供技术支撑。[方法]将桔子皮干燥粉碎,以乙醇为溶剂加热超声辅助提取其中的多糖,采用苯酚-硫酸法测定提取液中多糖的含量,并分析不同提取条件对多糖提取率的影响。[结果]单因素试验结果表明,多糖提取率最高的条件分别为:加热浸提时间4h,浸提温度80℃,料液比1∶50,微波辐射功率500W,辐射时间5min,浸提次数6次,乙醇浓度100%。最佳提取条件为:加热浸提时间4h,浸提温度80℃,料液比1∶50,浸提次数4次,乙醇浓度80%,微波辐射功率500W,辐射时间5min。此条件下多糖提取率达15.23%。[结论]该研究确定了桔子皮中多糖的最佳提取条件。

Abstract:[Objective]The study was to provide the technical support for the comprehensive utilization of orange peel.[Method]The orange peel was crushed and dried.The polysaccharide in it was extracted with ethanol as solvent assisted by heating and ultrasonic.The content of polysaccharide in the extract was measured by phenol-sulfuric acid method.The effects of different extraction conditions on extraction rate of polysaccharides were analyzed.[Results] The results of single factor experiment showed that when the heating extraction time was 4 h,the extraction temperature was 80 ℃,the solid-liquid ratio was 1∶50,the microwave power was 500 W,the irradiation time was 5 min,the extracting times was 6 times,the ethanol concns.was 100% resp.the extraction rate of polysaccharide was the highest.The optimum extraction conditions was heating extraction time of 4 h,extraction temperature of 80 ℃,solid-liquid ratio of 1∶50,extraction times of 4 times,ethanol concns.of 80%,microwave power of 500 W,irradiation time of 5 min.Under this condition,the extraction rate of polysaccharide reached 15.23%.[Conclusion] This study identified the optimum extraction conditions of polysaccharide in orange peel.
