摘 要: | 由水稻花粉株系9211215及品种早雪的细胞悬浮物游离原生质体,采用改良的RY—2培养基以琼脂糖珠包埋法进行原生质体培养,发生了持续分裂,早雪植板率达5%以上。ABA对恢复和提高原生质体再生愈伤组织的分化能力作用显著。两个材料均成功地再生出植株,再生频率9211215为8.9%,而早雪高达33.3%。60余株再生植株移栽田间后能正常开花结实。这是我省第一次水稻原生质体再生植株的报道。

Studies on Protoplast Culture of Rice (Oryza Sativa L. ) and Plant Regeneration From Protoplast-derived Calli |

Abstract: | Protoplasts isolated from suspension cells of two rice varieties (9211215 and Zaoxue) were cultured in modified RY- 2 medium with agarose- ball embedding culture method. The plating efficiency of Zaoxue was as high as 5. 0%. Plants were regenerated from two tested cultivars with frequencies of 8. 9% (9211215) and 33. 3% (Zaoxue) through preculture using ABA -added media in culturing protoplast-derived calli. Over 60 protoplast-derived plants were transplanted to field and grew up to mature normally. This is the first report of the plant regeneration from rice protoplast culture in Jiangxi. |
