奶山羊周期黄体中催产素的免疫组化定位

运用免疫组化 ABC法对奶山羊周期黄体中催产素 (OT)的定位进行了研究。结果表明 ,奶山羊周期黄体中存在数量较多的 OT免疫反应阳性细胞。OT阳性细胞根据胞质内反应颗粒着色的深浅可分为强阳性、中等阳性和弱阳性细胞 3种。阳性细胞在形态上以圆形、卵圆形为主 ,另有相当数量的细胞具有突起 ,小梁两侧及黄体中央近腔区域的阳性细胞则多呈现为长梭形。从分布上看 ,强阳性细胞主要分布于黄体周边 ,少数散布于中央区域 ,中等阳性及弱阳性细胞则均匀分布于整个黄体组织中。连续切片 HE染色的对照观察显示 ,奶山羊周期黄体中的 OT主要是由大黄体细胞分泌 ,小黄体细胞也可产生少量的 OT。

Immunohistochemical localization of oxytocin in dairy goat corpus luteum during estrous cycle

The localization of oxytocin(OT) in dairy goat corpus luteum(CL) during estrous cycle was studied by using avidin- biotin- peroxidase complex(ABC) method of immunohistochemistry.The results showed that there were many oxytocin- immunoreactive(OT- IR) cells in CL.According to the staining color of reactive granules,the positive cells could be divided into intensely,medium and lightly stained cells.The OT- IR cells were mainly spherical and ovoid,in addition,some of them had projects.The positive cells which surrounded the trabecula and located near the lumen were spindle- shaped.Most of intensely stained cells occured in the peripheral area of CL and few of them were located atcentral area,the medium and lightly stained cells were equally distributed in whole CL tissue.According to observation of serial sections staining with haematoxylin and eosin,OT in CL during estrous cycle were mainly produced by the large luteal cells,but the small luteal cells could not be excluded.
