| 摘 要： | 根据GenBank中轮状病毒VP6序列设计1对引物，从pGEM—T—VP6质粒中扩增VP6PCR产物。将VP6克隆到PBI121质粒中，转化根癌农杆菌LBA4404，挑取阳性菌落的质粒进行酶切鉴定。用叶盘法转化烟草，获得再生苗，用RT—PCR、PCR和PCR—southern杂交方法进行鉴定，结果表明，VP6基因已成功地转入烟草中。 |

| Abstract: | According to the sequence of VP6 in GenBank,a pair of primer for amplifying the VP6 gene bas been designed,with which the predicted fragment bad been amplified by PCR from Pgem-T-VP6 plasmid.The VP6 gene was cloned into PBI121 plasmid and transformed into agrobacterium tumefaciens strain LBA4404,positive clones were selected and identified by restriet enzyme.Then transformed into tobacco by the leaf disc infection mehtod and the regenerated plants were obtained.The result was analyzed by RT-PCR,PCR and PCR-southern.The result showed that the VP6 gene was successfully integrated into the genomes of tobacco and it will provide the basic data for the development of a new style vaccine. |
