摘    要:通过对板栗种子超低温保存前后种子发芽率和脱水耐性,以及其离体胚的脯氨酸含量和丙二醛含量的测定分析,来研究板栗种子的超低温保存技术,结果表明:含水量水平是影响板栗种子发芽率以及超低温保存效果的重要因素,超低温保存时板栗种子和离体胚应进行适度脱水,超低温保存宜采用20%~25%含水量范围;采取适当脱水方式和含水量水平,能够较好地保持板栗种子的发芽率。适当的超低温保存因素的组合有利于板栗种子活力的维持、离体胚脯氨酸的诱导和减少丙二醛含量的积累。

Abstract:This study is mainly about the cryo-preservation technology of Castanea mollissima.The study was conducted by measuring and analyzing the relationship between seed germination rates,desiccation endurance,as well as the proline content and the MDA content of the excised embryos in seeds before and after the cryo-preservation.Results showed that moisture content(Mc) was the critical factor determining the seed germination rates and cryo-preservation effects;seeds and embryos should be desiccated down to and within 20%~25% MC levels during the cryo-preservation;proper desiccating methods and MC levels should be chosen for the sake of the fair maintenance of the seed germination rates;a proper combination of cryo-preservation factors was of benefit to the maintenance of the seed vigor,as well as the inducement of more proline content and the reduced accumulation of MDA content of excised embryos during the cryo-preservation.
