木聚糖酶降解玉米秸秆的工艺研究

摘    要:[目的]利用木聚糖酶降解玉米秸秆中的木聚糖,为玉米秸秆的生物降解提供参考。[方法]将玉米秸秆烘干、磨碎、过筛。采用单因素和正交试验研究酶添加量、pH值、酶解时间及酶解温度对玉米秸秆木聚糖降解率的影响。[结果]各因素对木聚糖降解率的影响由大到小为:pH值>酶解温度>酶添加量>酶解时间。酶解温度为50℃时,木聚糖降解率最大。木聚糖酶添加量为65×104U/g,酶解时间为20 h及pH值为4.0时,秸秆木聚糖的降解率最大,分别为27.80%、27.00%、26.37%。[结论]木聚糖酶降解玉米秸秆中木聚糖的最适条件为:酶添加量65×104U/g、pH值4.0、酶解时间20 h、酶解温度50℃,该条件下玉米秸秆木聚糖的降解率为26.61%。

Process Research on Degradation of Corn Stalk by Xylanase
Abstract:[Objective] The objective was to use xylanase to degrade the xylan in corn stalk and provide the reference for the biodegradation of corn stalk.[Method] Corn stalk was dried,grinded and sieved.A single factor test and orthogonal test were taken to study the effect of enzyme addition,pH value,enzymolysis time and temperature on the degradation rate of xylan in corn stalk.[Result] The effect of factors on degradation rate in order was pH value>enzymolysis temperature>enzyme addition>enzymolysis time.When the enzymolysis temperature was 50 ℃,the degradation rate of xylan was the biggest.When the enzyme addition was 65×104 U/g,the enzymolysis time was 20 h and pH value was 4.0,the degradation rate of xylan was the biggest,being 27.80%,27.00%,26.37% resp.[Conclusion] The optimum conditions for the degradation of xylan in corn stalk by xylanase was enzyme addition of 65×104 U/g,pH value of 4.0,enzymolysis time of 20 h and enzymolysis temperature of 50 ℃,under which the degradation rate of xylan in corn stalk was 26.61%.
