基于28S rDNA D2区序列的陕南凹缘菱纹叶蝉分子系统学与遗传多样性研究

摘    要:为探讨陕南桑园凹缘菱纹叶蝉(Hishimonus sellatus Uhler)系统关系与遗传多样性,对陕南7个主要蚕桑产区共132头凹缘菱纹叶蝉的 28S rDNA基因D2区序列进行核苷酸多样性和遗传差异分析,同时研究单倍型之间的分子系统关系和网络进化图。结果表明,总群体共存在多态性位点63个,单倍型8个,优势单倍型H_1占单倍型总数的74.2%,遗传分化程度较低。分子方差分析显示:种群内变异占总组分的98.94%,差异不显著,且其基因流系数Nst和种群地理距离之间不存在相关性(R~2=0.019 7),说明各个地理种群之间存在渐渗杂交。

Abstract:The nucleotide diversity and genetic difference were analyzed based on28S rDNA D2 sequence of 132 Hishimones sellatus Uhler individuals from 7 major sericulture-producing regions in South Shaanxi.And then, the molecular phylogenetic relationship among haplotypes of the population were studied.The results showed that a total of 63 polymorphic loci existed in the total population, and formed 8 haplotypes, which the dominant haplotype H_1 accounted for 74.2% of the total haplotype, with a lower genetic differentiation.The molecular variance analysis showed that the variation within population accounted for 98.94% of the total component, and the difference was not significant, and there was no correlation between the gene flow coefficient Nst and the population geographic distance(R=0.019 7).All of these indicated that there was an introgression hybridization between the various geographical populations, it would provid a reference for further study of the population evolution.
