杜仲叶中绿原酸和黄酮同时提取分离方法的研究

摘    要:目的]对同时提取分离杜仲叶中的绿原酸和黄酮的方法进行了研究。方法]研究了以超声波辅助乙醇同时提取杜仲叶中绿原酸和黄酮的工艺条件,以绿原酸得率为考察指标,用紫外分光光度法测定绿原酸和黄酮的含量。结果]最佳提取工艺条件为:提取溶剂为50%乙醇,原料与溶剂比例为1∶10(g∶ml),在60℃下超声提取50 min。将提取液浓缩后用乙酸乙酯萃取,酯层纯化后得黄酮提取物,黄酮含量为37.6%;水层酸化后用乙酸乙酯萃取,该酯层纯化后得绿原酸提取物,绿原酸含量为23.4%。结论]该工艺操作简单,成本低,提取分离效率高。

Study on Extracting Chlorogenic Acid and Flavonoids from Eucommia Leaves

Abstract: Objective] Extraction of chlorogenic acid and flavonoids from Eucommia leaves is studied. Method ] The process condition of extracting chlorogenic acid and flavonoids from Eucommia leaves by ethanol with ultrasonic is studied. Evaluation index is the yield of chloro-genic acid. Chlorogenic acid and flavonoids contents are measured by UV-spectrophotometry. Result] The optimum process condition is ap-ply ultrasonic extraction at 60 ℃ for 50 rain, using 50% ethanol as solvent with 1:10 (g:ml) material to solvent ratio. Extract is concentrated and extracted by ethyl acetate. The content of flavonoids extracts is 37.61% , obtained from the upper ester layer after purification. The lower water layer is acidified and then extracted by ethyl acetate, the content of chlorogenic acid extracts is 23.4% , obtained from ester layer after purification. Conclusion] The operation is simple, low cost, high extraction and separation effect.
