青藏高原海东鸡CAPN3基因第1外显子多态性研究

[目的]对青藏高原海东鸡CAPN3基因第1外显子的多态性进行研究。[方法]采用PCR-SSCP技术检测海东鸡CAPN3基因第1外显子的多态性。[结果]海东鸡CAPN3基因第1外显子存在c.223GA的错义突变,对应2种等位基因G和A,等位基因G为优势等位基因;该群体CAPN3基因检测区域0.25PIC≤0.5为中度多态,且偏离Hardy-Weinberg平衡状态(P0.01)。[结论]该研究为海东鸡功能基因的分子遗传研究提供了基础资料。

Study on Polymorphism of First Exon of CAPN3 Gene in Haidong Chicken on Tibetan Plateau

[ Objective ] The aim was to study the polymorphism of first exon of CAPN3 gene in Haidong chicken on Tibetan Plateau. [ Meth-od] Polymorphism of first exon of CAPN3 gene in Haidong chicken was detected by PCR-SSCP. [ Result] The first exon of CAPN3 gene in Haidong chicken had the missense mutation of c. 223G 〉 A, corresponding to the 2 alleles of G and A, and the allele G was the dominant al-lele; the detection region of CAPN3 gene with 0.25 〈 P1C≤0. 5 was moderate polymorphism, and deviated fi＇om the equilibrium state of Hardy - Weinberg （P 〈0.01 ）. [ Conclusion] The study provided basic data for the molecular genetic research of Haidong chicken＇ s function genes.
