应用参考家系信息构建猪12号染色体部分微位星位点遗传图的研究

为了提高猪１２号染色体遗传图的精度，选择德系大约克与二花脸猪通过回交方法建立了参考家系，采取了两胎共１５４头个体的血样，全血裂解法提取ＤＮＡ后，对每头猪的ＤＮＡ样品进行ＰＣＲ扩增后应用银染法检测了每头个体在猪１２号染色体上３个微卫星位点（ＳＷ１５５３，ＳＷ８７４，ＳＷ１３５０）的基因型，应用ＣＲＩＭＡＰ程序进行了统计分析，得到的３位点间的最佳排序为：ＳＷ１５５３－ＳＷ１３５０－ＳＷ８７４。其中在两个美国肉畜研究中心图中重组率为０的位点（ＳＷ１５５３，ＳＷ１３５０）上发现了重组，并与其他位点做了排序，提高了这一区域的定位精度。

CONSTRACTION OF GENETIC LINKAGE MAP OF MICROSATELLITES ON PORCINE CHROMOSOME 12 BY USING REFERENCE FAMILY INFORMATION

In order to improve marker order on porcine chromosome 12, a backcross reference family was established by using Large White boar and Erhualian dams. Blood samples of approximately 154 individuals of three generations were collected and whole blood lysis method was used to isolate genomic total DNA. Each sample was amplified by using primers of 3 microsatellite loci (SW1553, SW874 and SW1350) on chromosome12 and genotypes were identified through silver staining method after polyarcrylamide gel electrophoresis. Data were scored and linkage computations were performed by using CRIMAP 2.4. The most likely order is: SW1553 SW1350 SW874. Recombinant events were found between SW1553 and SW1350 and marker order was improved in this region in contrast to the result that no recombination was found between the two loci in USDA MARC reference family.
